    Microbial Community Composition during a Bloom of Purple Bacteria in Intertidal Sediments in Vigo (Northwest Spain)

    In summer 2019, a large, bright pink microbial mat was visible on top of macroalgal deposits in muddy sediments of an urban beach (Playa do Adro, Vigo). In order to characterize the dominant organisms in these colored mats, results from microscopic observations, photosynthetic pigments, and molecular analysis were gathered. Light microscopy examination revealed pinkish microbial aggregates with minor contributions of larger protists and cyanobacteria. High-performance liquid chromatography (HPLC) pigment analysis documented the dominance of bacteriochlorophyll a and carotenoids whose spectra were compatible with those described in photosynthetic purple bacteria. 16S rRNA gene amplicon sequencing confirmed that the vast majority of reads belonged to Proteobacteria (73.5%), and among them, nearly 88% of those reads belonged to purple sulfur bacteria (Gammaproteobacteria). A single family, Chromatiaceae, constituted the bulk of this assemblage, including the genera Thiohalocapsa (32%), Marichromatium (12.5%), Phaeochromatium (5%), and Halocromatium (2%) as main contributors. Nonetheless, a considerable number of sequences could not be assigned to a particular genus, stressing the large biological diversity in these microbial mats and the potential presence of novel taxa of purple sulfur bacteria.En prens

    Neck circumference is associated with nutritional status in elderly nursing home residents

    Objectives: Anthropometry is an easy and noninvasive method to evaluate nutritional status in institutionalized elderly people who are often bedridden. The aim of this study was to investigate the relationship between the neck circumference (NC) and nutritional status of elderly nursing home residents and to find cutoff points for NC size to identify individuals at risk of malnutrition. Methods: A cross-sectional study was developed with data collected from 352 elderly people living in five public nursing homes. Different anthropometric measures and the Mini Nutritional Assessment (MNA) were used to determine nutritional status. Receiver operating characteristic (ROC) curves were built for each anthropometric variable to determine their sensitivity and specificity for predicting the risk of malnutrition according to the MNA. Results: The mean age of the participants (59% females) was 83 years old. In total, 48.3% of women and 45.5% of men were at risk of malnutrition according to their MNA scores. All anthropometric measurements were highly intercorrelated in both men and women, indicating a high degree of collinearity. Bootstrapped linear regression was used to assess the strength of the association between an individuals’ nutritional status and their anthropometric parameters. Calf circumference and NC presented the best predictive value with the highest sensitivity for diagnosing the risk of malnutrition in both institutionalized elderly men and women. The best cutoff points of NC to identify elderly nursing home residents at risk of malnutrition were 35.2 cm for females and 37.8 cm for males. Conclusions: NC is associated with other classical anthropometric parameters and malnutrition status in elderly people living in nursing homes

    PTRF acts as an adipokine contributing to adipocyte dysfunctionality and ectopic lipid deposition

    Adipose tissue (AT) expands under obesogenic conditions. Yet, when the growth exceeds a certain limit, AT becomes dysfunctional and surplus lipids start depositing ectopically. Polymerase I and transcription release factor (PTRF) has been proposed as a mechanism leading to a dysfunctional AT by decreasing the adipogenic potential of human adipocyte precursors. However, whether or not PTRF can be secreted by the adipocytes into the bloodstream is not yet known. For this work, PTRF presence was investigated in plasma. We also produced a recombinant PTRF (rPTRF) and examined its impact on the functional interactions between the adipocyte and the hepatocyte in vitro. We demonstrated that PTRF can be found in human plasma, and is at least in part, carried by exosomes. In vitro treatment with rPTRF increased the hypertrophy and senescence of 3T3-L1 adipocytes. In turn, those rPTRF-treated adipocytes increased lipid accumulation in hepatocytes. Lastly, we found a positive correlation between circulating PTRF and the concentration of PTRF in the visceral fat depot. All these findings point toward the presence of an enlarged and dysfunctional visceral adipose tissue which secretes PTRF. This circulating PTRF behaves as an adipokine and may partially contribute to the well-known detrimental effects of visceral fat accumulation

    Apolipoprotein E (APOE) is a multifunctional protein synthesized and secreted by multiple mammalian tissues. Although hepatocytes contribute about 75% of the peripheral pool, APOE can also be expressed in adipose tissue, the kidney, and the adrenal glands, among other tissues. High levels of APOE production also occur in the brain, where it is primarily synthesized by glia, and peripheral and brain APOE pools are thought to be distinct. In humans, APOE is polymorphic, with three major alleles (e2, e3, and e4). These allelic forms dramatically alter APOE structure and function. Historically, the vast majority of research on APOE has centered on the important role it plays in modulating risk for cardiovascular disease and Alzheimer's disease. However, the established effects of this pleiotropic protein extend well beyond these two critical health challenges, with a demonstrated roles for APOE across a wide spectrum of biological conditions, including adipose tissue function and obesity, metabolic syndrome and diabetes, fertility and longevity, and immune function. While the spectrum of biological systems in which APOE plays a role seems implausibly wide at first glance, there are some potential unifying mechanisms that could tie these seemingly disparate disorders together. In the current review, we aim to concisely summarize a wide breadth of APOE-associated pathologies and to analyze the influence of APOE in the development of several distinct disorders in order to provide insight into potential shared mechanisms implied in these various pathophysiological processes

    Alteraciones del metabolismo lipídico producidas por el virus de la hepatitis C en pacientes con infección crónica

    Fundamentos: Es conocido que la infección crónica por el virus de la hepatitis C (VHC) genera un perfil lipídico aparentemente favorable. Paradójicamente, estos pacientes presentan un aumento de eventos cardiovasculares concomitantes. Los objetivos de la presente revisión fueron analizar y sintetizar los estudios que indagasen en las modificaciones que produce el VHC sobre el metabolismo lipídico de los pacientes con infección crónica, así como estudiar si esas modificaciones pueden asociarse a episodios posteriores de enfermedad cardiovascular. Métodos: Se realizó una búsqueda bibliográfica en las bases de datos de Medline y Scopus de los artículos publicados desde enero de 2008 hasta febrero de 2019. Se identificaron un total de 901 publicaciones, de las cuales se revisaron 10 estudios que cumplieron con los criterios de inclusión y exclusión propuestos. Resultados: Se encontró que en los pacientes con infección crónica por el VHC estaban disminuidos los niveles de colesterol total y sus fracciones lipídicas. No existió una clara asociación con los niveles de triglicéridos. Además, parecía haber una asociación entre la infección crónica por VHC y un aumento del riesgo de padecer aterosclerosis y de desarrollar enfermedades cardiovasculares. Conclusiones: La infección crónica por el VHC tiene un efecto hipolipemiante y aumenta el riesgo cardiovascular. Se precisan estudios prospectivos que analicen el efecto de las nuevas terapias con antivirales de acción directa sobre el metabolismo lipídico y el riesgo cardiovascular. Background: Chronic infection with the hepatitis C virus (HCV) is known to generate an apparently favorable lipid profile. Paradoxically, these patients present an increase in concomitant cardiovascular events. The objectives of the present review were to analyze and synthesize studies that inquired into the changes produced by hepatitis C virus (HCV) in the lipid metabolism of patients with chronic infection, and about whether these modifications can be associated with subsequent episodes of cardiovascular diseases. Methods: A bibliographic search was carried out in the Medline and Scopus databases of the articles published from January 2008 to February 2019. A total of 901 publications were identified, of which 10 studies that fulfilled the inclusion and exclusion criteria were reviewed. Results: It was found that the levels of total cholesterol and its lipid fractions were decreased in patients with chronic HCV infection. There was no clear association with triglyceride levels. In addition. there seemed to he an association between chronic HCV infection and an increased risk of developing atherosclerosis and cardiovascular diseases. Conclusions: Chronic HCV infection has a lipid-lowering effect and increases cardiovascular risk. Prospective studies am needed to analyze the effect of new therapies with direct-acting antivirals on lipid metabolism and cardiovascular risk

    Amino acid profile in malnourished patients with liver cirrhosis and its modification with oral nutritional supplements: Implications on minimal hepatic encephalopathy

    Low plasma levels of branched chain amino acids (BCAA) in liver cirrhosis are associated with hepatic encephalopathy (HE). We aimed to identify a metabolic signature of minimal hepatic encephalopathy (MHE) in malnourished cirrhotic patients and evaluate its modification with oral nutritional supplements (ONS) enriched with ß-Hydroxy-ß-methylbutyrate (HMB), a derivative of the BCAA leucine. Post hoc analysis was conducted on a double-blind placebo-controlled trial of 43 individuals with cirrhosis and malnutrition, who were randomized to receive, for 12 weeks, oral supplementation twice a day with either 220 mL of Ensure® Plus Advance (HMB group, n = 22) or with 220 mL of Ensure® Plus High Protein (HP group, n = 21). MHE evaluation was by psychometric hepatic encephalopathy score (PHES). Compared to the HP group, an HMB-specific treatment effect led to a larger increase in Val, Leu, Phe, Trp and BCAA fasting plasma levels. Both treatments increased Fischer’s ratio and urea without an increase in Gln or ammonia fasting plasma levels. MHE was associated with a reduced total plasma amino acid concentration, a reduced BCAA and Fischer´s ratio, and an increased Gln/Glu ratio. HMB-enriched ONS increased Fischer´s ratio without varying Gln or ammonia plasma levels in liver cirrhosis and malnutrition, a protective amino acid profile that can help prevent MHE. © 2021 by the authors.     Spatial and temporal variability in coccolithophore abundance and distribution in the NW Iberian coastal upwelling system

    A systematic investigation of the spatial and temporal variability in coccolithophore abundance and distribution through the water column of the NW Iberian coastal up-welling system was performed. From July 2011 to June 2012, monthly sampling at various water depths was conducted at two parallel stations located at 42 degrees N. Total coccosphere abundance was higher at the outer-shelf station, where warmer, nutrient-depleted waters favoured coccolithophore rather than phytoplanktonic diatom blooms, which are known to dominate the inner-shelf location. In seasonal terms, higher coccosphere and coccolith abundances were registered at both stations during upwelling seasons, coinciding with high irradiance levels. This was typically in conjunction with stratified, nutrient-poor conditions (i.e. relaxing upwelling conditions). However, it also occurred during some upwelling events of colder, nutrient-rich subsurface waters onto the continental shelf. Minimum abundances were generally found during downwelling periods, with unexpectedly high coccolith abundance registered in subsurface waters at the inner-shelf station. This finding can only be explained if strong storms during these downwelling periods favoured resuspension processes, thus remobilizing deposited coccoliths from surface sediments, and hence hampering the identification of autochthonous coccolithophore community structure. At both locations, the major coccolithophore assemblages were dominated by Emiliania huxleyi, small Gephyrocapsa group, Gephyrocapsa oceanica, Florisphaera profunda, Syracosphaera spp., Coronosphaera mediterranea, and Calcidiscus leptoporus. Ecological preferences of the different taxa were assessed by exploring the relationships between environmental conditions and temporal and vertical variability in coccosphere abundance. These findings provide relevant information for the use of fossil coccolith assemblages in marine sediment records, in order to infer past environmental conditions, of particular importance for Paleoceanography. Both E. huxleyi and the small Gephyrocapsa group are proposed as proxies for the upwelling regime with a distinct affinity for different stages of the upwelling event: E. huxleyi was associated with warmer, nutrient-poor and more stable water column (i.e. upwelling relaxation stage) while the small Gephyrocapsa group was linked to colder waters and higher nutrient availability (i.e. early stages of the upwelling event), similarly to G. oceanica. Conversely, F. profunda is suggested as a proxy for the downwelling regime and low-productivity conditions. The assemblage composed by Syracosphaera pulchra, Coronosphaera mediterranea, and Rhabdosphaera clavigera may be a useful indicator of the presence of subtropical waters conveyed northward by the Iberian Poleward Current.     Response of phytoplankton to enhanced atmospheric and riverine nutrient inputs in a coastal upwelling embayment

    Original research paperOver the past decades, as a consequence of human activity, there was an increase in nutrient inputs to the ocean and they are expected to enhance even more in the future. Coastal areas, accounting for a significant proportion of marine primary productivity, are the most vulnerable zones to anthropogenic impacts. The response of phytoplankton communities to an increase in organic and inorganic nutrients levels from natural allochthonous sources was assessed in microcosm experiments conducted in a coastal system affected by intermittent upwelling events (Ría de Vigo, NW Iberia). Three nutrient addition experiments were performed in spring, summer and autumn, when surface water was supplemented with 5 and 10% of atmospheric and riverine matter. Pico-, nano- and microphytoplankton abundances, chlorophyll a concentration (Chl a) and primary production rates (PP) were measured and compared with those in the control seawater sample (without additions) after 48 h of incubation. Simultaneous experiments with controlled additions of inorganic and organic nutrients were also performed in order to describe the limiting nutrient for phytoplankton growth at each experiment. The composition of the matter inputs and the structure of the phytoplankton communities determined the type of response observed. Phytoplankton responses varied among seasons, being positively correlated with dissolved inorganic nitrogen (DIN) concentrations. As expected, the phytoplankton responses to external nutrient inputs were stronger under low nutrient levels (summer) than when phytoplankton was already growing in nutrient replete conditions (spring). Null and negative responses to the natural inputs were observed in autumn, which suggests that the oceanic phytoplankton advected to this coastal system during downwelling events could be occasionally inhibited by these nutrient inputs.     Combined assessment of DYRK1A, BDNF and homocysteine levels as diagnostic marker for Alzheimer’s disease

    Early identification of Alzheimer’s disease (AD) risk factors would aid development of interventions to delay the onset of dementia, but current biomarkers are invasive and/or costly to assess. Validated plasma biomarkers would circumvent these challenges. We previously identified the kinase DYRK1A in plasma. To validate DYRK1A as a biomarker for AD diagnosis, we assessed the levels of DYRK1A and the related markers brain-derived neurotrophic factor (BDNF) and homocysteine in two unrelated AD patient cohorts with age-matched controls. Receiver-operating characteristic curves and logistic regression analyses showed that combined assessment of DYRK1A, BDNF and homocysteine has a sensitivity of 0.952, a specificity of 0.889 and an accuracy of 0.933 in testing for AD. The blood levels of these markers provide a diagnosis assessment profile. Combined assessment of these three markers outperforms most of the previous markers and could become a useful substitute to the current panel of AD biomarkers. These results associate a decreased level of DYRK1A with AD and challenge the use of DYRK1A inhibitors in peripheral tissues as treatment.
