    Profiles of human capital and strategic technological decisions on turbulence environment

    [EN] The objective of this article is to examine the absorptive capacity in the technology industry and aspires to recognize how firms can manage their strategic decisions in the turbulence contexts. In particular, we examine how organizations can strengthen their organizational contexts in order to absorb knowledge. From the knowledge management literature, this investigation extends our perception of the relationship between the human capital profiles (organization, research and development unit, and recent incorporations) and technological decision-making. Through the SEPI Foundation, a balanced panel of 1,220 Spanish industrial companies has used that answer to the Survey of Business Strategies (SBS) for a threeyear period, which signifies a total of 3,660 cases. The principal finding is the presence of high levels of human resources to understand a decision efficiency process. It also highlights its relationship to the firm’s technological committee. These contributions are notable for both researchers and practitioners.     Evaluation of polyherbal methionine and choline in feedlot rations for lambs

    Requirements of lambs for choline and methionine have not been clearly established, but because of their metabolic relationship, the availability of these two nutrients may affect protein synthesis and energy balance. Therefore, a trial was carried out to evaluate the effect on productive performance and blood metabolites of including methionine and choline from polyherbal mixtures in finishing lamb diets. Forty Hampshire x Suffolk lambs weighing 26.9 ± 2.8 kg were used for a 45-day experiment. The treatments were arranged as a 2x2 factorial, in which the factors were dietary supplementation with herbal methionine and choline at two levels (0% and 0.4% of dry matter (DM)). The treatments had no effects on the lambs’ productive variables (daily feed intake, average daily gain, and feed to gain ratio), carcass characteristics and lipid metabolites (P >0.05). The results indicated that the inclusion of polyherbal mixtures containing methionine, choline and their combination at 0.4% of DM in finishing lamb diets showed no benefits in productive response or in blood metabolites related to lipid metabolism.     Correlative light and electron microscopy is an increasingly popular technique to study complex biological systems at various levels of resolution. Fluorescence microscopy can be employed to scan large areas to localize regions of interest which are then analyzed by electron microscopy to obtain morphological and structural information from a selected field of view at nm-scale resolution. Previously, an integrated approach to room temperature correlative microscopy was described. Combined use of light and electron microscopy within one instrument greatly simplifies sample handling, avoids cumbersome experimental overheads, simplifies navigation between the two modalities, and improves the success rate of image correlation. Here, an integrated approach for correlative microscopy under cryogenic conditions is presented. Its advantages over the room temperature approach include safeguarding the native hydrated state of the biological specimen, preservation of the fluorescence signal without risk of quenching due to heavy atom stains, and reduced photo bleaching. The potential of cryo integrated light and electron microscopy is demonstrated for the detection of viable bacteria, the study of in vitro polymerized microtubules, the localization of mitochondria in mouse embryonic fibroblasts, and for a search into virus-induced intracellular membrane modifications within mammalian cells

    Noble gas signals in corals predict submarine volcanic eruptions

    Deep-water corals growing in close proximity to the 2011 submarine eruption at Tagoro (El Hierro Island; Canary Archipielago, Spain) have revealed their ability to record the magmatic helium (3He) signal during a period of months prior to the eruption with magmatic 3He/4He ratios of 3.6-5.0 RA This is similar to the range of He isotope values (3-10 Ra) obtained from olivine samples from basinites erupted during the 2011 Tagoro eruption. Whip-like black corals (Anthozoa: Antipatharia: Antipathidae: Stichopathes) growing on basaltic rocks trapped 3He within their skeletons. We used a theoretical growth rate to estimate the time of trapping of 3He between release from the magmatic source at depth to trapping by the aquatic organisms. Our findings suggest that magmatic 3He emission occurred a few months before the beginning of the seismic activity in the region, the latter occurring four months in advance of the beginning of the eruption. This discovery implies that corals living in submarine volcanic areas may act as archives of noble gases released before the beginning of an eruption, and that the continuous monitoring of 3He may help to constrain the arrival time of a subaqueous eruption with potential applications in volcano eruption forecasting.En prens

    EPIdemiology of Surgery-Associated Acute Kidney Injury (EPIS-AKI) : Study protocol for a multicentre, observational trial

    More than 300 million surgical procedures are performed each year. Acute kidney injury (AKI) is a common complication after major surgery and is associated with adverse short-term and long-term outcomes. However, there is a large variation in the incidence of reported AKI rates. The establishment of an accurate epidemiology of surgery-associated AKI is important for healthcare policy, quality initiatives, clinical trials, as well as for improving guidelines. The objective of the Epidemiology of Surgery-associated Acute Kidney Injury (EPIS-AKI) trial is to prospectively evaluate the epidemiology of AKI after major surgery using the latest Kidney Disease: Improving Global Outcomes (KDIGO) consensus definition of AKI. EPIS-AKI is an international prospective, observational, multicentre cohort study including 10 000 patients undergoing major surgery who are subsequently admitted to the ICU or a similar high dependency unit. The primary endpoint is the incidence of AKI within 72 hours after surgery according to the KDIGO criteria. Secondary endpoints include use of renal replacement therapy (RRT), mortality during ICU and hospital stay, length of ICU and hospital stay and major adverse kidney events (combined endpoint consisting of persistent renal dysfunction, RRT and mortality) at day 90. Further, we will evaluate preoperative and intraoperative risk factors affecting the incidence of postoperative AKI. In an add-on analysis, we will assess urinary biomarkers for early detection of AKI. EPIS-AKI has been approved by the leading Ethics Committee of the Medical Council North Rhine-Westphalia, of the Westphalian Wilhelms-University Münster and the corresponding Ethics Committee at each participating site. Results will be disseminated widely and published in peer-reviewed journals, presented at conferences and used to design further AKI-related trials. Trial registration number NCT04165369

    Dual targeting of histone methyltransferase G9a and DNMT1 for the treatment of experimental hepatocellular carcinoma

    Las modificaciones epigenéticas, como la metilación del ADN e histonas, cooperan funcionalmente para fomentar el crecimiento tumoral, incluido el carcinoma hepatocelular (HCC). La inhibición farmacológica de estos mecanismos puede abrir nuevas vías terapéuticas. El objetivo del trabajo desarrollado en la presente tesis doctoral fue determinar la eficacia terapéutica y el potencial mecanismo de acción de nuevos inhibidores duales de la histona-metiltransferasa G9a y la ADN-metiltransferasa 1 (DNMT1) en células HCC humanas y su interferencia con células fibrogénicas. Estos inhibidores han sido desarrollados en la propia Institución y sujetos a patente (EPOPatent application #14-382230.2-1462). La expresión de G9a y DNMT1, junto con la de su adaptador molecular UHRF1, se midió en muestras de HCC humanas (n = 268), tejidos peritumorales (n = 154) y líneas células de HCC (n = 32). Se evaluó el efecto de la inhibición individual y combinada de G9a y DNMT1 en el crecimiento de células HCC mediante enfoques farmacológicos y genéticos. La actividad de nuestro compuesto principal, CM-272, se examinó en células HCC bajo normoxia e hipoxia, células estelares hepáticas humanas y células LX2, y tumores de xenoinjerto formados por células HCC o células HCC + LX2 combinadas. Encontramos una sobreexpresión significativa y correlativa de G9a, DNMT1 y UHRF1 en HCC en asociación con un mal pronóstico. La inhibición farmacológica independiente de G9a y DNMT1 redujo sinérgicamente el crecimiento de las células HCC. CM-272 inhibió potentemente la proliferación de células HCC y LX2, y detuvo el crecimiento del tumor en xenoinjertos de HCC y HCC combinadas con LX2. CM-272 impidió la adaptación metabólica de las células HCC a la hipoxia, e indujo un fenotipo diferenciado en las células HCC y fibrogénicas. La expresión del gen supresor de tumores metabólicos fructosa-1,6-bifosfatasa (FBP1), reprimido epigenéticamente en el HCC, fue restaurada por CM-272. Como conclusión del trabajo, la inhibición combinada de G9a / DNMT1 con compuestos como CM-272 es una estrategia prometedora para el tratamiento del HCC.     Joining of ceramic matrix composites to high temperature ceramics for thermal protection systems

    The current work reports a novel approach for the integration of external protective SIC multilayers with ceramic matrix composite (C-f/SiC) with the view of application in aerospace heat protection systems. The integration method is based on diffusion brazing bonding. As a joining agent the MAX-Phase Ti3SiC2, produced by self-propagating high temperature synthesis, has been employed. The pressure applied during the joining process and its effect on the microstructure of the integrated structure is discussed. Microstructural analysis of the resulting joints is conducted using scanning electron microscopy coupled with energy dispersive spectroscopy and X-ray diffraction measurements. Analysis of the joints showed that the bonds are uniform, dense, with few crack vertical to the interface which are not detrimental for the performance of the joints. Ground re-entry tests showed that the joints survive 5 re-entry cycles at 1391 and 1794 degrees C without any detectable damage. (C) 2015 Elsevier Ltd.
