    Aplicações sucessivas de dejetos suínos alteram a dinâmica de carbono, nitrogênio e frações da matéria orgânica dos agregados do solo

    Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Ciências Agrárias, Programa de Pós-Graduação em Agroecossistemas, Florianópolis, 2017.Aplicações continuadas de dejetos suínos no solo podem promover alterações nos atributos químicos, físicos e biológicos, os quais podem ser utilizados como indicadores de qualidade do solo. A matéria orgânica do solo (MOS), assim como as suas frações, e também a agregação do solo, com destaque para os agregados biogênicos, são considerados excelentes indicadores de qualidade do solo, sendo utilizados para evidenciar diferenças em sistemas de uso do solo sob condições diversificadas. Tais condições podem acarretar em modificações nos atributos edáficos, assim como nas vias de formação dos agregados, que podem ser classificados como biogênicos e fisiogênicos, sendo estes padrões estabelecidos conforme a sua morfologia. Para aumentar a agregação do solo em agroecossistemas, o manejo deve ter como um dos objetivos o aumento das entradas de carbono no solo, a diminuição de perturbações e das taxas de perdas desse elemento por processos como a decomposição e a erosão. Este trabalho visa avaliar os efeitos de diferentes fontes e quantidades de dejetos suínos sobre os teores de carbono orgânico total (COT) e nitrogênio total (NT) e das frações da MO dos agregados biogênicos e fisiogênicos em um Argissolo com histórico de aplicações continuadas de dejetos suínos por 10 anos. Foram coletadas amostras indeformadas de solo nas profundidades de 0-5 e 5-10 cm, nos tratamentos sem aplicação de dejetos (TESTEMUNHA), com aplicação de dejeto líquido de suíno (DLS) e cama sobreposta de suíno (CSS) em dose equivalente a uma e duas vezes a recomendação de nitrogênio para o milho e aveia (DLS1X, DLS2X, CSS1X e CSS2X, respectivamente). Os agregados foram separados conforme a via de formação em biogênicos e fisiogênicos. Posteriormente, em cada via de formação foram quantificados o COT, NT, assim como o C e N das frações químicas da MO, humina (C-HUM/N-HUM), ácidos fúlvicos (C-FAF/N-FAF) e ácidos húmicos (C-FAH/N-FAH), e das frações granulométricas, MO particulada (C-MOP/N-MOP) e associada aos minerais (C-MOM/N-MOM). A aplicação de CSS, independentemente da dose, aumentou os teores totais de C e N dos agregados e das frações húmicas da MO, bem como C e N da MOP e MOM. A aplicação de diferentes formas e quantidades de dejetos suínos aumentaram o C e N em agregados biogênicos, em comparação com agregados fisiogênicos.Abstract : Continuous applications of pig manure in soil lead to changes in chemical, physical, and biological attributes, which can be used as indicators of soil quality. Soil organic matter (SOM) and its fractions, as well as soil aggregation, especially biogenic aggregates, are indicators of soil quality. They may be used to show differences in soil under diverse management systems, which lead to changes in soil attributes, as well as in aggregate formation pathways. Aggregates are classified as biogenic or physicogenic, according to their morphology. In order to increase soil aggregation in agroecosystems, management should aim to increase soil carbon inputs, reduce disturbances and reduce soil loss by processes such as decomposition and erosion. This work aimed to evaluate total organic carbon (TOC), total nitrogen (TN), and organic matter (OM) fractions in biogenic and physicogenic aggregates in soil from an area with a history of continuous applications of swine manure for 10 years. Undisturbed soil samples were collected (0-5 cm and 5-10 cm depths) in plots without waste application (CONTROL), with application of pig slurry (PS) and deep litter (DL) at doses equivalent to one and two times the recommended nitrogen dose for corn and oats (PL1X, PL2X, DL1X and DL2X, respectively). Aggregates were separated according to the biogenic and physiogenic formation pathways. Soil TOC, TN, were measured, as well as carbon (C), and nitrogen (N) in the OM fractions humin (C-HUM/N-HUM), fulvic acids (C-FAF/N-FAF), and humic acids (C-HAF/N-HAF). C and N were also determined in particulate OM (C-POM/N-POM) or mineral-associated OM (C-MOM/N-MOM). Deep litter application, regardless of dose, increased soil TOC and TN, and C and N in all OM fractions, as well as C and N in POM and MOM. Pig manure application, in all forms and doses, increased C and N in biogenic aggregates, as compared to physicogenic aggregates

    A presente dissertação parte de dois temas-chave, essências para a sua elaboração: a arquitectura e o vinho. Pretende-se com isto criar uma reflexão crítica sobre a arquitectura como experiência para os sentidos, tendo como enquadramento o Douro e a produção vinícola, dados base para o desenvolvimento projectual de uma adega. Para além do interesse disciplinar, a escolha deste tema permitirá a oportunidade de estudar essa indústria, em constante evolução, assim como diferentes atitudes arquitectónicas com que tem sido abordada, com intensa carga conceptual e sensorial. A análise e definição dos diferentes elementos da paisagem vinhateira duriense, a compreensão do lugar, e a interpretação do enquadramento actual da cultura do vinho na arquitectura, permitirá também perceber, ainda que implicitamente, o fenómeno do enoturimo enquanto veículo de aproximação entre as pessoas e as adegas.Todos estes elementos contribuirão para uma melhor definição de uma estratégia projectual, e a construção de um programa que possa relacionar os vários âmbitos. O âmbito do turismo rural, que visa requalificar um espaço devoluto da propriedade (promovendo a integração de espaços pré-existentes num programa actual) e o âmbito do turismo de produção que visa a apresentação de um projecto para uma adega contemporânea. Estes procurarão ser veículos para conjugar as vertentes funcionais e perceptivas da arquitectura, aspectos técnicos e poéticos, como proposta para a materialização de ambientes singulares.The present dissertation starts from two key themes, essences for its elaboration: the architecture and the wine. This is intended to create a critical reflection on architecture as an experience for the senses, having as a framework Douro and wine production, basic data for the development of a winery. In addition to the disciplinary interest, the choice of this theme will allow the opportunity to study this industry, in constant evolution, as well as different architectonic attitudes with which it has been approached, with an intense conceptual and sensorial load. The analysis and definition of the different elements of Douro wine landscape, the understanding of the place, and the interpretation of the current framework of wine culture in architecture, will also allow us to perceive, albeit implicitly, the phenomenon of wine tourism as a vehicle for rapprochement between people and the cellars.All these elements will contribute to a better definition of a project strategy, and the construction of a program that can relate to the various spheres. The scope of rural tourism, which aims to requalify a vacant property space (promoting the integration of pre-existing spaces in a current program) and the scope of production tourism aimed at presenting a project for a contemporary winery. These will seek to be vehicles to combine the functional and perceptive aspects of architecture, technical and poetic aspects, as a proposal for the materialization of singular environments

    DSD and CAD/CAM integration in the planning and execution of an oral rehabilitation procedure: a case report

    This article presents a clinical case involving the integrated application of CAD/CAM and DSD. DSD – Digital Smile Design – assists in obtaining a treatment plan adjusted to the patient’s facial features, achieving the best aesthetic potential. The integration between DSD and CAD/CAM allows for greater fidelity in respect to the original plan. A patient received a rehabilitation with veneers milled out of leucite-reinforced feldspathic ceramics, using the CAD/CAM system and a DSD-obtained digital wax-up. This workf low simplifies design and fabrication, providing greater accuracy and predictability to the rehabilitation process. The digital planning and development of the mockup also makes clinical conditions more predictable. Making ceramic veneers using the CAD/CAM technique requires judicious adhesive cementation, and also an accurate practical and theoretical knowledge on the part of the professional.Este artigo apresenta um caso clínico envolvendo a aplicação integrada de CAD/CAM e DSD. O DSD – Digital Smile Design – auxilia na obtenção de um plano de tratamento ajustado às características faciais do paciente, alcançando o melhor potencial estético. A integração entre DSD e CAD/CAM permite maior fidelidade em relação ao plano original. Um paciente recebeu uma reabilitação com facetas fresadas de cerâmica feldspática, reforçada com leucita usando o sistema CAD/CAM, e um encerado digital obtido por DSD. Esse fluxo de trabalho simplifica o projeto e a fabricação, proporcionando maior precisão e previsibilidade ao processo de reabilitação oral. O planejamento digital e o desenvolvimento da maquete também tornam as condições clínicas mais previsíveis.     Emprendimiento sociolaboral de cerámica: una experiencia de capacitación solidaria en el hospital A. Korn

    En el siguiente trabajo presentamos la experiencia de un emprendimiento sociolaboral iniciado en el Hospital A. Korn en contexto de pandemia y su desarrollo al momento actual. Se describe el surgimiento de esta experiencia formativa desarrollada en el edificio histórico La Barraca, de la cual participan personas usuarias del servicio de salud mental. Así mismo su articulación con políticas públicas que contribuyen al fortalecimiento del proceso de salud y continuidad de cuidados. Se retomarán los distintos aspectos de la Economía Social y Solidaria (ESS), en la co-construcción de un espacio de aprendizaje colectivo, valorando los vínculos interpersonales en un universo social de personas con padecimiento subjetivo. Se analiza el factor social del emprendimiento productivo como aporte a la inclusión y participación social desde una perspectiva de derechos, a fin de fortalecer el proceso salud/enfermedad/atención/cuidado. Compartimos algunas reflexiones, tensiones y desafíos utilizando como metodología cualitativa la investigación acción participativa invitándonos a identificar estrategias que protejan y estimulen los vínculos sociales, laborales y afectivos acorde con los principios de la ESS.Facultad de Trabajo Socia

    Prenatal diagnosis of mosaic tetrasomy 18p

    To report the case of a prenatally detected de novo mosaic tetrasomy 18p where a combination of different methodologies was used,including conventional cytogenetics and molecular genetics techniques. METHOD: Molecular rapid aneuploidy diagnosis, by quantitative fluorescent polymerase chain reaction (QF-PCR), conventional cytogenetics and fluorescence in situ hybridization (FISH). RESULTS:Amniocentesis was performed at 22 + 5 gestational weeks, on a 37-year-old woman, due to ultrasound detection of fetal interventricular communication, overlapped fingers and unilateral club foot. Molecular rapid aneuploidy testing, by QF-PCR, showed highly increased ratios for markers on the short arm of chromosome 18. Cytogenetic analysis revealed a male karyotype with mosaicism involving two cell lines: one with a supernumerary isochromosome 18p and another with two extra derivative chromosomes 18. Parental karyotypes were normal and QF-PCR analysis indicated that the extra chromosome 18p material was of maternal origin. After counseling the couple opted for pregnancy termination. Anatomopathological studies, as well as further characterization of the derivative chromosomes, are underway in order to provide more accurate genotype-correlation. CONCLUSIONS: The majority of tetrasomy 18p reported cases were ascertained postnatally, had a de novo occurrence and an isochromosome 18p in all cells examined. Prenatal diagnoses of mosaic tetrasomy 18p are, on the other hand, rarely described in the literature. The case herein reported, where a mosaic is present and none of the cell lines is chromosomally normal, may provide further insight on this rare syndrome and help in the knowledge of the associated phenotype. The application of a combined strategy, using QF-PCR, conventional cytogenetics and FISH analysis allowed not only for the identification of the extra chromosome 18p material, in the form of a mosaicism involving isochromosome 18p, but also for determining its parental origin. This information is of particular importance in recurrence risk assessment and therefore crucial for genetic counseling

    Prenatal diagnosis of terminal 11q deletion

    The majority of 11q deletion cases described may be included in the “distal 11q deletion syndrome”, or Jacobsen syndrome. This is a rare but clinically recognizable condition with an incidence of 1/ 100,000 births. The most common clinical features are psychomotor delay, characteristic facial dysmorphism and malformations of the heart, kidney, genitalia, central nervous system and skeleton. Patients usually have visible deletions of chromosomal bands 11q23, 11q24, and/or 11q25. Approximately 85% of the cases are de novo deletions, and only a few prenatal cases have been reported. We report the clinical case of a 30-year-old pregnant woman who was referred to our laboratory with a positive first trimester prenatal screening for Down syndrome. The cytogenetic analysis revealed a terminal deletion on the distal 11q chromosome. Fluorescence in situ hybridization using a whole-chromosome painting probe and subtelomeric probes confirmed the terminal deletion and excluded other material involvement. Parental karyotypes were normal. Second trimester ultrasound revealed clinodactyly and a cardiac defect described as a subvalvular and intraventricular communication. The couple opted for medical termination of pregnancy. The postmortem examination of the 22-week fetus showed facial dysmorphism, cardiac defects and uterus bicornis. There are few reports of prenatally diagnosed 11q−,and there seems to be a phenotypic variability. Some cases had a positive prenatal screening for Down syndrome and/or abnormal prenatal ultrasound with olygohydramnios, nuchal thickening, heart malformations and kidney anomalies. Other reports mention no structural fetal abnormalities. The present case, similar to others, had a positive first trimester screening. The fetus presented abnormal fingers, cardiac defects and malformations of genital tract, identified at the second trimester, which is consistent with the del11q− phenotype. This case reinforces the phenotypic variability associated with partial monosomy of distal 11q in the fetus and the difficulty of establishing genotype–phenotype correlations

    Gastrointestinal delivery of codfish skin-derived collagen hydrolysates: deep eutectic solvent extraction and bioactivity analysis

    The fishing industry produces substantial by-products, such as heads, skins, bones, and scales, rich in collagen—a prevalent protein in these materials. However, further application of deep eutectic solvent-based extraction remains unexplored. In this study, we extracted collagen with urea: propanoic acid mixture (U:PA; 1:2) with a 2.2% yield, followed by enzymatic hydrolysis with alcalase for 120 min. The resulting bioactive peptides demonstrated notable antioxidant activity (961 µmol TE) and antihypertensive properties (39.3% ACE inhibition). Subsequently, we encapsulated 39.3% of these hydrolysates in chitosan-TPP capsules, which released about 58% of their content, primarily in the intestine, as mimicked in the in vitro model of the gastrointestinal tract. Although the digestion process did not significantly alter the size of the non-encapsulated collagen peptides, it did influence their health benefits. The promising results suggest that further research could optimize the use of collagen from fish by-products, potentially offering a sustainable source for health products.N/
