    O presente artigo tem como objetivo delinear a trajetória do Programa de Pós-Graduação da PUC de São Paulo, que celebra seus 50 anos em 2022. Para isso, foi elaborado um panorama geral tanto da história da educação superior no Brasil como das reformas universitárias que pautaram a criação e desenvolvimento deste Programa. A narração dos fatos e eventos que marcaram o Programa de Pós-Graduação da PUC-SP baseou-se, em grande medida, na memória não só das autoras, mas também naquela de diversos colegas. O artigo ganha especial relevo em um contexto de profunda crise pela qual passa a produção de conhecimento científico no Brasil

    Epigenome-wide DNA methylation profiling of periprostatic adipose tissue in prostate cancer patients with excess adiposit : a pilot study

Here, we sought to investigate whether excess adiposity modulates the methylome of PPAT in PCa patients. DNA methylation profiling was performed in PPAT from obese/overweight (OB/OW, BMI > 25 kg m−2) and normal weight (NW, BMI < 25 kg m−2) PCa patients. Significant differences in methylated CpGs between OB/OW and NW groups were inferred by statistical modeling. Results: Five thousand five hundred twenty-six differentially methylated CpGs were identified between OB/OW and NW PCa patients with 90.2% hypermethylated. Four hundred eighty-three of these CpGs were found to be located at both promoters and CpG islands, whereas the representing 412 genes were found to be involved in pluripotency of stem cells, fatty acid metabolism, and many other biological processes; 14 of these genes, particularly FADS1, MOGAT1, and PCYT2, with promoter hypermethylation presented with significantly decreased gene expression in matched samples. Additionally, 38 genes were correlated with antigen processing and presentation of endogenous antigen via MHC class I, which might result in fatty acid accumulation in PPAT and tumor immune evasion. Conclusions: Results showed that the whole epigenome methylation profiles of PPAT were significantly different in OB/OW compared to normal weight PCa patients. The epigenetic variation associated with excess adiposity likely resulted in altered lipid metabolism and immune dysregulation, contributing towards unfavorable PCa microenvironment, thus warranting further validation studies in larger samples.This work was supported in part by the Natural Sciences and Engineering Research Council of Canada, Manitoba Research Health Council, University of Manitoba, and China Scholarship Council.info:eu-repo/semantics/publishedVersio

    mHealth in urology

    Introduction: Smartphones are increasingly playing a role in healthcare and previous studies assessing medical applications (apps) have raised concerns about lack of expert involvement and low content accuracy. However, there are no such studies in Urology. We reviewed Urology apps with the aim of assessing the level of participation of healthcare professionals (HCP) and scientific Urology associations in their development. Material and Methods: A systematic search was performed on PubMed, Apple's App Store and Google's Play Store, for Urology apps, available in English. Apps were reviewed by three graders to determine the app's platform, target customer, developer, app type, app category, price and the participation of a HCP or a scientific Urology association in the development. Results: The search yielded 372 apps, of which 150 were specific for Urology. A fifth of all apps had no HCP involvement (20.7%) and only a third had been developed with a scientific Urology association (34.7%). The lowest percentage of HCP (13.4%) and urological association (1.9%) involvement was in apps designed for the general population. Furthermore, there was no contribution from an Urology society in "Electronic Medical Record" nor in "Patient Information" apps. A limitation of the study is that only Android and iOS apps were reviewed. Conclusions: Despite the increasing Mobile Health (mHealth) market, this is the first study that demonstrates the lack of expert participation in the design of Urology apps, particularly in apps designed for the general public. Until clear regulation is enforced, the urological community should help regulate app development. Maintaining a register of certified apps or issuing an official scientific seal of approval could improve overall app quality. We propose that urologists become stakeholders in mHealth, shaping future app design and promoting peer-review app validation

    Semen parameters in testicular tumor patients before orchiectomy: What is the impact of testicular tumor stage and histology?

    Purpose: To evaluate the influence of testicular cancer histology and stage on sperm parameters in cryopreserved samples collected prior to orchiectomy. Materials and methods: We conducted a retrospective analysis of tumor histology, stage and sperm parameters of patients who underwent pre-orchiectomy sperm cryopreservation for testicular cancer between March 2010 and March 2023. The World Health Organization (WHO) 2010 sperm reference values were used to identify patients with subnormal semen parameters and to further categorize patients by sperm alteration. Localized disease was classified as Stage I, while metastatic disease encompassed Stages II and III. Continuous variables were compared using t-test or Mann Whitney U test, and categorical variables using Chi-square and Fisher´s exact test. Results: A total of 64 patients was identified, 48 (75%) classified as stage I and 16 (25%) classified as stage II/III. No difference was found in semen parameters between patients with seminoma and patients with non-seminoma germ cell tumor (NSGCT). Patients with stage II/III disease had significantly lower percentages of progressive motility (36% vs 53%, p = 0.021) and total motility (60% vs 69%, p = 0.015) than stage I patients. When categorizing by sperm alterations according to WHO 2010 reference values, patients with stage II/III disease had significantly higher proportions of asthenozoospermia (38% vs 15%, p = 0.048) and teratozoospermia (63% vs 31%, p = 0.027) than stage I patients. Elevated tumor markers were not associated with sperm abnormalities. Conclusions: Patients with metastatic testicular cancer present with worse sperm quality than patients with localized disease. Sperm cryopreservation should be offered to all patients with testicular cancer, and especially emphasized in patients with metastatic disease

    Human periprostatic adipose tissue promotes prostate cancer aggressiveness in vitro

    Background - Obesity is associated with prostate cancer aggressiveness and mortality. The contribution of periprostatic adipose tissue, which is often infiltrated by malignant cells, to cancer progression is largely unknown. Thus, this study aimed to determine if periprostatic adipose tissue is linked with aggressive tumor biology in prostate cancer. Methods - Supernatants of whole adipose tissue (explants) or stromal vascular fraction (SVF) from paired fat samples of periprostatic (PP) and pre-peritoneal visceral (VIS) anatomic origin from different donors were prepared and analyzed for matrix metalloproteinases (MMPs) 2 and 9 activity. The effects of those conditioned media (CM) on growth and migration of hormone-refractory (PC-3) and hormone-sensitive (LNCaP) prostate cancer cells were measured. Results - We show here that PP adipose tissue of overweight men has higher MMP9 activity in comparison with normal subjects. The observed increased activities of both MMP2 and MMP9 in PP whole adipose tissue explants, likely reveal the contribution of adipocytes plus stromal-vascular fraction (SVF) as opposed to SVF alone. MMP2 activity was higher for PP when compared to VIS adipose tissue. When PC-3 cells were stimulated with CM from PP adipose tissue explants, increased proliferative and migratory capacities were observed, but not in the presence of SVF. Conversely, when LNCaP cells were stimulated with PP explants CM, we found enhanced motility despite the inhibition of proliferation, whereas CM derived from SVF increased both cell proliferation and motility. Explants culture and using adipose tissue of PP origin are most effective in promoting proliferation and migration of PC-3 cells, as respectively compared with SVF culture and using adipose tissue of VIS origin. In LNCaP cells, while explants CM cause increased migration compared to SVF, the use of PP adipose tissue to generate CM result in the increase of both cellular proliferation and migration. Conclusions - Our findings suggest that the PP depot has the potential to modulate extra-prostatic tumor cells' microenvironment through increased MMPs activity and to promote prostate cancer cell survival and migration.     The AlfaCrux CubeSat mission description and early results

    On 1 April 2022, the AlfaCrux CubeSat was launched by the Falcon 9 Transporter-4 mission, the fourth SpaceX dedicated smallsat rideshare program mission, from Space Launch Complex 40 at Cape Canaveral Space Force Station in Florida into a Sun-synchronous orbit at 500 km. AlfaCrux is an amateur radio and educational mission to provide learning and scientific benefits in the context of small satellite missions. It is an opportunity for theoretical and practical learning about the technical management, systems design, communication, orbital mechanics, development, integration, and operation of small satellites. The AlfaCrux payload, a software-defined radio hardware, is responsible for two main services, which are a digital packet repeater and a store-and-forward system. In the ground segment, a cloud-computing-based command and control station has been developed, together with an open access online platform to access and visualize the main information of the AlfaCrux telemetry and user data and experiments. It also becomes an in-orbit database reference to be used for different studies concerned with, for instance, radio propagation, attitude reconstruction, data-driven calibration algorithms for satellite sensors, among others. In this context, this paper describes the AlfaCrux mission, its main subsystems, and the achievements obtained in the early orbit phase. Scientific and engineering assessments conducted with the spacecraft operations to tackle unexpected behaviors in the ground station and also to better understand the space environment are also presented and discussed.Fundação de Apoio à Pesquisa del Distrito Federal (FAPDF), Brasil | Ref.     Impacto da pandemia de COVID-19 nas doenças respiratórias: Um estudo sobre bronquite e bronquiolite aguda em crianças na região norte do Brasil

    Respiratory diseases constitute a diverse set of conditions that affect the respiratory system, playing a significant role in healthcare systems. Bronchitis and acute bronchiolitis are two respiratory conditions that significantly impact lung health, especially in young children and some vulnerable groups. The association between bronchitis, acute bronchiolitis and COVID-19 is significant due to the similarities in the respiratory symptoms of these conditions. Both involve inflammation of the airways, resulting in coughing and difficulty breathing. Therefore, it is important to analyse epidemiological profiles before and after the pandemic and understand the impact of the pandemic on hospitalization and deaths associated with bronchitis and bronchiolitis. The main objective of this article was to analyse the prevalence and epidemiological profile of hospitalizations and deaths caused by bronchitis and acute bronchiolitis in children living in the Northern region of Brazil. This is a quantitative, retrospective study that analysed the number of hospitalizations and deaths due to bronchitis and acute bronchiolitis in the North region based on public data from the Hospital Information System (SIH) of the Unified Health System (SUS). During the analysis period, the number of hospitalizations decreased by 6%, but the number of deaths increased significantly. The state of Pará has the highest number of hospitalizations, and the state of Amazonas has the highest mortality rate. Both predominate in children under 1 year of age. Additionally, hospital spending increased by 32% during the COVID-19 pandemic. The significant incidence of bronchitis and acute bronchiolitis in children under one year of age stands out as a primary concern for child health. The inherent vulnerability of this age group, combined with frequent exposure to infectious agents, highlights the need for more effective preventive approaches and management strategies.As doenças respiratórias constituem um conjunto diversificado de condições que afetam o sistema respiratório, desempenhando um papel significativo nos sistemas de saúde. A bronquite e bronquiolite aguda são duas condições respiratórias que impactam significativamente a saúde pulmonar, especialmente em crianças pequenas e em alguns grupos vulneráveis. A associação entre bronquite, bronquiolite aguda e COVID-19 é significativa devido às semelhanças nos sintomas respiratórios dessas condições. Ambos envolvem inflamação das vias respiratórias, resultando em tosse e dificuldade respiratória. Dessa forma, é importante analisar perfis epidemiológicos antes e depois da pandemia e compreender o impacto da pandemia na hospitalização e óbitos associados à bronquite e bronquiolite. O objetivo principal deste artigo foi analisar a prevalência e o perfil epidemiológico das internações e óbitos causados por bronquite e bronquiolite aguda em crianças residentes na região Norte do Brasil. Trata-se de um estudo quantitativo, retrospectivo, que analisou o número de internações e óbitos por bronquites e bronquiolite aguda na região Norte com base em dados públicos do Sistema de Informações Hospitalares (SIH) do Sistema Único de Saúde (SUS). Durante o período de análise, o número de hospitalizações diminuiu 6%, mas o número de mortes aumentou significativamente. O estado do Pará tem o maior número de internações, e o estado do Amazonas tem a maior taxa de mortalidade. Ambos predominam em crianças menores de 1 ano. Além disso, os gastos hospitalares aumentaram 32% durante a pandemia de COVID-19. A significativa incidência de bronquites e bronquiolite aguda em crianças com menos de um ano destaca-se como uma preocupação primordial para a saúde infantil. A vulnerabilidade inerente a esse grupo etário, combinada com a exposição frequente a agentes infecciosos, evidencia a necessidade de abordagens preventivas e estratégias de manejo mais eficazes
