    Retención del conocimiento de los enfermeros sobre derivación ventricular externa

    Introduction: Hydrocephalus is a common complication among neurological patients whose treatment in acute cases is done through an external ventricular drain catheter which allows monitoring and controlling intracranial hypertension. This study aimed at describing the knowledge retention by nurses after an educational intervention on nursing care for an external ventricular drain. Materials and Methods: A quasi-experimental study with nurses at an adult intensive care unit was conducted in which knowledge retention on the topic was evaluated at different three times: before, one week and three months after training. Results: After one week, significant retention was observed but not after three months, given that after this period the question success rate was significantly low with respect to the previous phases. Participants’ performance was better in those questions related to system management. Discussion: Time is a factor that interferes with learning since studies show that knowledge at three months significantly decreases and is almost completely gone after six months and a year. Practical behaviors based on assistance guidelines have better retention among professionals, underlining that it is easier to learn technical skills that are associated with professional experience. Conclusions: A significant knowledge retention was observed among professionals in the first week after training but not after three months. O objetivo desse estudo foi descrever a retenção do conhecimento dos enfermeiros após intervenção educativa sobre cuidados com derivação ventricular externa. Materiais e Métodos: Estudo quase experimental com enfermeiros de uma unidade de terapia intensiva adulto, em que foi avaliado a retenção de conhecimento sobre o tema em três momentos: antes, uma semana e três meses após o treinamento. Resultados: Houve retenção significativa após uma semana, mas não aos três meses, sendo que aos três meses o índice de acertos nas questões foi expressivamente baixo em relação às fases anteriores. O desempenho dos participantes foi melhor nas questões relacionadas ao manuseio do sistema. Discussão: O tempo é um fator que interfere no aprendizado e os estudos mostram que o conhecimento aos 3 meses diminui significativamente, sendo que aos 6 meses e um ano quase que completamente. Condutas práticas baseadas em diretrizes de assistência tem melhor retenção entre os profissionais, ressaltando que há mais facilidade no aprendizado de habilidades técnicas que estão associadas a vivência profissional. Conclusões: Houve retenção de conhecimento significativa entre os profissionais na primeira semana após o treinamento, mas não aos três meses após o treinamento. Discusión: El tiempo es un factor que interfiere en el aprendizaje y los estudios muestran que el conocimiento a los tres meses disminuye significativamente, y después de seis meses y un año casi por completo. Las conductas prácticas basadas en directrices de asistencia tienen mejor retención entre los profesionales, resaltando que resulta más fácil aprender habilidades técnicas que están asociadas con la vivencia profesional. Conclusiones: Se observó una retención de conocimiento significativa entre los profesionales en la primera semana posterior al entrenamiento, pero no a los tres meses después del entrenamiento.     GCN2 kinase plays an important role triggering the remission phase of experimental autoimmune encephalomyelitis (EAE) in mice

    Experimental autoimmune encephalomyelitis (EAE) has been widely employed as a model to study multiple sclerosis (MS) and indeed has allowed some important advances in our comprehension of MS pathogenesis. Several pieces of evidence suggest that infiltrating Th1 and Th17 lymphocytes are important players leading to CNS demyelination and lesion during the peak of murine EAE. Subsequently, effector T cell responses rapidly decline and the recovery phase of the disease strongly correlates with the expression of anti-inflammatory cytokines and the enrichment of Foxp3+ regulatory T (Treg) cells within the target organ. However, the mechanisms leading to the increased presence of Treg cells and to the remission phase of the disease are still poorly understood. Recent researches demonstrated that chemically induced amino-acid starvation response might suppress CNS immune activity. Here we verified an important participation of the general control nonrepressible 2 (GCN2), a key regulator kinase of the amino-acid starvation response, in the development of the remission phase of EAE in C57BL/6 mice. By immunizing wild type C57BL/6 (WT) and GCN2 knock-out mice (GCN2 KO) with myelin oligodendrocyte glycoprotein peptide (MOG(35-55)), it was noticed that GCN2 KO mice did not develop the remission phase of the disease and this was associated with higher levels of CNS inflammation and increased presence of effector T cells (Th1/Th17). These animals also showed lower frequency of Treg cells within the CNS as compared to WT animals. Higher expression of indoleamine 2,3-dioxygenase (IDO) and higher frequency of plasmacytoid dendritic cells (pDCs) were found at the peak of the disease in the CNS of WT animals. Our results suggest that the GCN2 kinase-dependent sensing of IDO activity represents an important trigger to the EAE remission phase. the IDO-mediated immunoregulatory events may include the arresting of effector T cell responses and the differentiation/expansion of Treg cells within the target organ.     The gut microbiome and metabolome of two riparian communities in the Amazon

    During the last decades it has become increasingly clear that the microbes that live on and in humans are critical for health. The communities they form, termed microbiomes, are involved in fundamental processes such as the maturation and constant regulation of the immune system. Additionally, they constitute a strong defense barrier to invading pathogens, and are also intricately linked to nutrition. The parameters that affect the establishment and maintenance of these microbial communities are diverse, and include the genetic background, mode of birth, nutrition, hygiene, and host lifestyle in general. Here, we describe the characterization of the gut microbiome of individuals living in the Amazon, and the comparison of these microbial communities to those found in individuals from an urban, industrialized setting. Our results showed striking differences in microbial communities from these two types of populations. Additionally, we used high-throughput metabolomics to study the chemical ecology of the gut environment and found significant metabolic changes between the two populations. Although we cannot point out a single cause for the microbial and metabolic changes observed between Amazonian and urban individuals, they are likely to include dietary differences as well as diverse patterns of environmental exposure. To our knowledge, this is the first description of gut microbial and metabolic profiles in Amazonian populations, and it provides a starting point for thorough characterizations of the impact of individual environmental conditions on the human microbiome and metabolome

    Annexin A1 protein attenuates cyclosporine-induced renal hemodynamics changes and macrophage infiltration in rats

    Cyclosporine (CsA) remains an important immunosuppressant for transplantation and for treatment of autoimmune diseases. The most troublesome side effect of CsA is renal injury. Acute CsA-induced nephrotoxicity is characterized by reduced renal blood flow (RBF) and glomerular filtration rate (GFR) due to afferent arteriole vasoconstriction. Annexin A1 (ANXA1) is a potent anti-inflammatory protein with protective effect in renal ischemia/reperfusion injury. Here we study the effects of ANXA1 treatment in an experimental model of acute CsA nephrotoxicity. Salt-depleted rats were randomized to treatment with VH (vehicles 1 mL/kg body weight/day), ANXA1 (Ac2-26 peptide 1 mg/kg body weight/day intraperitoneally), CsA (20 mg/kg body weight/day subcutaneously) and CsA + ANXA1 (combination) for seven days. We compared renal function and hemodynamics, renal histopathology, renal tissue macrophage infiltration and renal ANXA1 expression between the four groups. CsA significantly impaired GFR and RBF, caused tubular dilation and macrophage infiltration and increased ANXA1 renal tissue expression. Treatment with ANXA1 attenuated CSA-induced hemodynamic changes, tubular injury and macrophage infiltration.     LEMON GRASS ESSENTIAL OIL (CYMBOPOGUM FLEXUOSUS) IN HIGH-CONCENTRATE DIETS FOR LAMBS

    The objective of this study was to evaluate the effect of the inclusion of two doses of monensin and lemon grass essential oil (Cymbopogum flexuosus) on nutrient intake and digestibility, ruminal fermentation and ingestive behavior of lambs fed a high-concentrate diet. Four ruminally cannulated ram lambs (44.7 ± 3.72 kg of BW and approximately 5 mo. old) in a 4 x 4 Latin square experimental design were used. The control diet (CONT) contained 15% of oat hay, 85% of concentrate and no additives. In the remaining diets it was included 12.5 (12.5MON) or 25.0 (25MON) mg/kg of monensin (as-fed basis) or 1 mL/kg of DM of lemon grass essential oil (EO). The treatments had no effect on the nutrient intake and digestibility. The lambs that received the EO treatment showed a higher ruminal pH in comparison to those that received the control diet. However, they did not differ from treatments with monensin. A quadratic response was observed for ruminal pH, with the highest value observed in the animals fed the diet 12.5MON. The lambs fed EO had a higher ruminal concentration of acetate than the diet with monensin. There was a quadratic response of monensin on ruminal concentration of acetate, with the lowest value observed for 12.5MON. The acetate:propionate ratio was lower in lambs fed 12.5MON compared to the control. EO decreased the ruminal concentration of butyrate; however, its concentration was higher in lambs fed monensin than EO diets. A lower concentration of total short chain fatty acids (SCFA) was observed in lambs fed 12.5MON. The alanine aminotransferase (ALT) blood concentration was higher in the lambs fed EO compared to the control. EO increased rumen concentration of blood glucose compared to the treatments containing monensin and decreased the number of eggs per gram of feces (EGF). In addition, there was a quadratic response of monensin on the EGF, with a higher value in the lambs fed 12.5MON. In conclusion, half of the daily dose of monensin was more effective at increasing ruminal pH than the full dose, with no negative reflection on nutrient intake and digestibility, as well as on the ingestive behavior and parasitic load of the animals. Thus, 12.5MON may be indicated for lambs feed high concentrate diet. EO proved to be a good natural source to replace ionophores in diets for lambs

    Leishmania spp.: isolamento de parasitos pela inoculação de macerados de biopsias de pacientes em camundongos deficientes em interferon gama

    Isolation of Leishmania parasite and species identification are important for confirmation and to help define the epidemiology of the leishmaniasis. Mice are often used to isolate pathogens, but the most common mouse strains are resistant to infection with parasites from the Leishmania (Viannia) subgenus. In this study we tested the inoculation of interferon gamma knockout (IFNγ KO) mice with biopsy macerates from Leishmania-infected patients to increase the possibility of isolating parasites. Biopsies from twenty five patients with clinical signs of leishmaniasis were taken and tested for the presence of parasites. Immunohistochemical assay (IHC) and conventional histopathology detected the parasite in 88% and 83% of the patients, respectively. Leishmania sp. were isolated in biopsy macerates from 52% of the patients by culture in Grace's insect medium, but 13% of isolates were lost due to contamination. Inoculation of macerates in IFNγ KO mice provides isolation of parasites in 31.8% of the biopsies. Most isolates belong to L. (Viannia) subgenus, as confirmed by PCR, except one that belongs to L. (Leishmania) subgenus. Our preliminary results support the use of IFNγ KO mice to improve the possibility to isolate New World Leishmania species.O isolamento e a identificação da espécie de parasito do gênero Leishmania são importantes para a confirmação e auxiliam na epidemiologia da leishmaniose. Os camundongos são freqüentemente utilizados para isolar patógenos, porém, as linhagens mais comuns de camundongos são resistentes à infecção por parasitos do subgênero Leishmania (Viannia). Neste estudo, avaliamos a inoculação de macerados de biópsias de pacientes infectados em camundongos deficientes do gene do interferon gama (IFNγ KO) como um método para aumentar a possibilidade de isolar Leishmania spp. Biópsias de 25 pacientes infectados com Leishmania sp. foram avaliadas para a presença de parasitos pelos métodos de imunohistoquímica (IHC) e histopatologia convencional. Os parasitos foram observados, respectivamente, em 88% e 83% das biópsias. Leishmania sp. foi isolada de macerados de biópsia de 52% dos pacientes infectados, quando cultivados em meio Grace, porém, 13% destes isolados foram perdidos devido a contaminações. Inoculação dos macerados em camundongos IFNγ KO proporcionou o isolamento de parasitos oriundos de 31,8% dos pacientes. A maioria dos isolados pertence ao subgênero L. (Viannia), exceto um que pertence ao subgênero L. (Leishmania), como confirmado pela reação da polimerase em cadeia. Nossos resultados preliminares sugerem que o uso de camundongos IFNγ KO pode ser útil para aumentar a possibilidade de isolamento de leishmânias encontradas nas Américas

    Data standardization of plant-pollinator interactions

    Background: Animal pollination is an important ecosystem function and service, ensuring both the integrity of natural systems and human well-being. Although many knowledge shortfalls remain, some high-quality data sets on biological interactions are now available. The development and adoption of standards for biodiversity data and metadata has promoted great advances in biological data sharing and aggregation, supporting large-scale studies and science-based public policies. However, these standards are currently not suitable to fully support interaction data sharing. Results: Here we present a vocabulary of terms and a data model for sharing plant–pollinator interactions data based on the Darwin Core standard. The vocabulary introduces 48 new terms targeting several aspects of plant–pollinator interactions and can be used to capture information from different approaches and scales. Additionally, we provide solutions for data serialization using RDF, XML, and DwC-Archives and recommendations of existing controlled vocabularies for some of the terms. Our contribution supports open access to standardized data on plant–pollinator interactions. Conclusions: The adoption of the vocabulary would facilitate data sharing to support studies ranging from the spatial and temporal distribution of interactions to the taxonomic, phenological, functional, and phylogenetic aspects of plant–pollinator interactions. We expect to fill data and knowledge gaps, thus further enabling scientific research on the ecology and evolution of plant–pollinator communities, biodiversity conservation, ecosystem services, and the development of public policies.     A list of land plants of Parque Nacional do Caparaó, Brazil, highlights the presence of sampling gaps within this protected area

    Brazilian protected areas are essential for plant conservation in the Atlantic Forest domain, one of the 36 global biodiversity hotspots. A major challenge for improving conservation actions is to know the plant richness, protected by these areas. Online databases offer an accessible way to build plant species lists and to provide relevant information about biodiversity. A list of land plants of “Parque Nacional do Caparaó” (PNC) was previously built using online databases and published on the website "Catálogo de Plantas das Unidades de Conservação do Brasil." Here, we provide and discuss additional information about plant species richness, endemism and conservation in the PNC that could not be included in the List. We documented 1,791 species of land plants as occurring in PNC, of which 63 are cited as threatened (CR, EN or VU) by the Brazilian National Red List, seven as data deficient (DD) and five as priorities for conservation.
