Towards Amyotrophic Lateral Sclerosis Interpretable Diagnosis Using Surface Electromyography
Amyotrophic Lateral Sclerosis (ALS) is a fast-progressing disease with no cure. It is
diagnosed through the assessment of clinical exams, such as needle electromyography,
which measures themuscles’ electrical activity by inserting a needle into themuscle tissue.
Nevertheless, surface electromyography (SEMG) is emerging as a more practical and less
painful alternative. Even though these exams provide relevant information regarding the
electric structures conducted in the muscles, ALS symptoms are similar to those of other
neurological disorders, preventing a faster detection of the disease.
This dissertation focuses on implementing and analyzing innovative SEMG features
related to the morphology of the functional structures present in the signal. To assess the
efficiency of these features, a framework is proposed, aiming to distinguish healthy from
pathological signals through the use of a classification algorithm. The classification task
was performed using SEMG signals acquired from the upper limb muscles of healthy and
ALS subjects.
The results show the utility of employing the proposed set of features for ALS diagnosis,
with an F1 Score higher than 80% in most experimental conditions. The novel features
improved the model’s overall performance when combined with other state-of-art SEMG
features and also demonstrated efficiency when used individually. These outcomes are
of significant importance in supporting the use of SEMG as a complementary diagnosis

seu diagnóstico é feito através da avaliação de exames clínicos como a eletromiografia de
profundidade, que mede a atividade elétrica muscular com agulhas inseridas no músculo.
No entanto, a eletromiografia de superfície (SEMG) surge como uma alternativa mais prática
e menos dolorosa. Embora ambos os exames forneçam informações relevantes sobre
as estruturas elétricas conduzidas nos músculos, os sintomas da ELA são semelhantes aos
de outras doenças neurológicas, impedindo uma identificação mais precoce da doença.
Esta dissertação foca-se na implementação e análise de atributos inovadores de SEMG
relacionados com a morfologia das estruturas funcionais presentes no sinal. Para avaliar
a eficiência destes atributos, é proposto um framework, com o objetivo de distinguir sinais
saudáveis e sinais patológicos através de um algoritmo de classificação. A tarefa de classificação
foi realizada utilizando sinais de SEMG adquiridos dos músculos dos membros
superiores de indivíduos saudáveis e com ELA.
Os resultados demonstram a utilidade do conjunto de atributos proposto para o diagnóstico
de ELA, com uma métrica de classificação F1 superior a 80% na maioria das
condições experimentais. Os novos atributos melhoraram o desempenho geral do modelo
quando combinados com outros atributos de SEMG do estado da arte, e também se comprovaram
eficientes quando aplicados individualmente. Estes resultados são de grande
importância na justificação da aplicabilidade da SEMG como um exame complementar
de diagnóstico da ELA. Os atributos apresentados demonstram ser promissores para um
melhor e mais rápido diagnóstico, e facilitam a explicação dos resultados da classificação
devido à sua interpretabilidade
Biosynthesis optimization of STEAP1 protein in Pichia pastoris X33 MUT+
Prostate cancer (PCa) is the most common type of cancer in men over the age of 50 in Western countries. The current therapies available are, in many cases, limited and ineffective. Therefore, it is necessary to develop alternative therapies, that can fight specifically and effectively cancer cells. The Six Transmembrane Epithelial Antigen of Prostate 1 (STEAP1) is a protein with six transmembrane domains located in the cell-cell junctions of the secretory epithelium of the prostate. Some studies have reported it as overexpressed in PCa but in normal tissues, its expression is minimal. The structure and cell location of STEAP1 suggest that it may function as a channel for ions. Moreover, it was demonstrated that STEAP1 overexpression induces proliferation of cancer cells. The differential expression of STEAP1 in tissues, associated with its localization, suggests its potential use as a target for cancer therapy. In order to delineate novel strategies for targeting STEAP1 protein, it is necessary to obtain high levels of purified protein. Therefore, the aim of this study is to produce STEAP1 protein in Pichia pastoris X33 Mut+. The use of a eukaryotic system will allow the correct folding of the protein since Pichia pastoris is able to perform post-translation modifications. Our results showed that the target protein is being produced by Pichia pastoris strain and it was identified in the pellet fraction centrifuged at 16000g. Western blot analysis revealed the protein in the correct molecular weight (~ 40 kDa) The best conditions for protein production is a 6 hours fermentation process in BMMH medium with 1.25% methanol and 6% DMSO at 30ºC, 250rpm. The solubilization process should be done using SDS in a concentration of 1-3%.O cancro da próstata é o tipo de cancro mais comum entre os homens com mais de 50 anos nos países ocidentais. As terapias atuais disponíveis são, em alguns casos, limitadas e ineficazes. Por isso, é necessário desenvolver terapias alternativas que possam combater de forma mais específica e eficaz as células tumorais. A proteína Six Transmembrane Epithelial Antigen of the Prostate 1 (STEAP1) é formada por seis domínios transmembranares, localizada nas junções célula-célula do epitélio prostático. Diversos estudos indicam que a STEAP1 se encontra sobre-expressa no cancro da próstata mas em tecidos normais, a sua expressão é mínima. A sua localização e estrutura celular sugere que esta proteína pode funcionar como um canal iónico. Logo, a expressão diferencial da STEAP1 entre tecidos, associada com a sua localização, sugerem o seu uso como possível alvo terapêutico do cancro. Para delinear novas formas de terapia usando como alvo a STEAP1, é necessário obter-se elevados níveis de proteína purificada. Portanto, o objetivo deste estudo é produzir a proteína STEAP1 em Pichia pastoris X33. A utilização de um sistema eucariótico irá permitir o enovelamento correto da proteína, visto a Pichia pastoris ser capaz de realizar reações pós-tradução. Os nossos resultados demonstraram que a proteína alvo é eficazmente produzida em Pichia pastoris e foi identificada na fração centrifugada a 16000g. Análise por Western blot revela a proteína com o peso molecular correto (~40 kDa). As melhores condições para a produção da proteína são um processo de fermentação de 6 horas em meio BMMH com 1,25% de metanol e 6% de DMSO a 30ºC, 250 rpm. O passo de solubilização deve ser efetuado utilizando SDS numa concentração de 1-3%
Export Market Effectiveness: The Role of Export Commitment, Innovativeness and Marketing Capabilities
Drawing on the resource-based view, this study investigates the way that export commitment
and innovativeness contribute to marketing capabilities development, and the effect of all
these factors on export market effectiveness. We use a survey data of 202 exporting
manufacturing firms based in Portugal to test the relationships between the constructs
analyzed in this study. The findings demonstrate that a high export commitment tends to
cultivate a higher degree of innovativeness, which in turn allows firms to develop superior
marketing capabilities (i.e., pricing, new product development, marketing communication and
distribution capabilities). Export commitment and new product development capability have a

O papel do Empreendedorismo Social na sustentabilidade do Associativismo: O caso da Associação EcoGerminar
Nos dias de hoje, estamos perante uma sociedade cada vez mais globalizada, marcada
pelo desenvolvimento, que salienta a importância de uma reflexão sobre o
Empreendedorismo Social, que tem vindo a assumir relevo perante o número crescente
de iniciativas. Nos últimos anos, o Empreendedorismo Social é um fenómeno que tem
vindo a suscitar uma crescente atenção por parte de académicos, profissionais e políticos
dos diversos quadrantes.
Portugal enfrenta a situação económica e social mais crítica da sua história recente,
perante a pandemia atual do Covid-19 antecipada da crise financeira vivida nos últimos
anos. Esta crise económica agudiza a situação de cada vez mais pessoas, o que leva a que
as entidades que trabalham na área social a dedicar uma especial atenção aos que se
encontram em desvantagem económico-social.
As entidades, mais conhecidas por ONG´s (Organizações Não Governamentais), como
por exemplo as associações, não conseguem fazer milagres, pois também se deparam
com problemas diariamente, despertando para o risco da sustentabilidade social e
financeiro, e consequentemente, atingindo os seus projetos/iniciativas. Uma das formas
usadas para ultrapassar estas dificuldades é concorrendo a concursos e candidaturas
para financiamento de projetos, sejam eles locais, nacionais, europeus e mesmo
mundiais, pois não apresentam capacidade financeira autossuficiente para garantir a sua
sustentabilidade do projeto/iniciativa por meios próprios.
Este é o propósito que dá o mote a esta investigação, pretende-se identificar, analisar e
perceber o papel do Empreendedorismo Social na sustentabilidade da Associação
EcoGerminar, identificando a sua influência sobre a sustentabilidade da associação, pois
considero que existe a necessidade de promover a visibilidade do Empreendedorismo
Social na realidade de uma associação, como a EcoGerminar.Nowadays, we are facing an increasingly globalized society, marked by development,
which highlights the importance of a reflection on Social Entrepreneurship, which has
come to assume importance in the face of the growing number of initiatives. In recent
years, Social Entrepreneurship is a phenomenon that has been attracting growing
attention from academics, professionals and politicians from different areas.
Portugal is facing the most critical economic and social situation in its recent history,
given the current Covid-19 pandemic, anticipated in the financial crisis experienced in
recent years. This economic crisis is worsening the situation of more and more people,
which leads entities that work in the social area to pay special attention to those who are
economically and socially disadvantaged.
Entities, better known as NGOs (Non-Governmental Organizations), such as
associations, cannot work miracles, as they also face daily problems, awakening to the
risk of social and financial sustainability, and consequently, reaching their
projects/initiatives. One of the ways used to overcome these difficulties is competing for
tenders and applications for financing projects, whether local, national, European or
even global, as they do not have self-sufficient financial capacity to guarantee the
sustainability of the project/initiative by their own means.
This is the purpose that sets the tone for this investigation, it is intended to identify,
analyze and understand the role of Social Entrepreneurship in the sustainability of the
EcoGerminar Association, identifying its influence on the sustainability of the
association, as I believe there is a need to promote visibility of Social Entrepreneurship
in the reality of an association such as EcoGerminar
Alunos do 4º ano de escolaridade e o cálculo mental : Preferências, atitudes, desempenho e estratégias
Dissertação de Mestrado apresentada ao ISPA - Instituto UniversitárioO principal objectivo deste estudo consiste em caracterizar o perfil dos alunos do
4ºano do ensino básico relativamente ao cálculo mental. Mais especificamente pretendese
caracterizar as suas preferências e atitudes, o seu desempenho e as estratégias que
utilizam para resolver tarefas de cálculo mental.
Para tal, participaram no estudo 46 alunos do 4º ano de escolaridade de duas
escolas da região de Lisboa e foram recolhidos dados através do questionário de
preferências, do questionário de atitudes, do teste de cálculo mental e, por último,
através da realização de entrevistas.
Os principais resultados encontrados foram: 1) Os alunos portugueses preferem
utilizar o cálculo mental em detrimento de outro tipo de cálculo, e esta preferência é
mais evidente em alunos com um desempenho superior em tarefas de cálculo mental; 2)
Os alunos acham que o cálculo mental é mais interessante e mais desafiante do que o
cálculo escrito; 3) A maioria dos alunos afirma que faz mais cálculo escrito do que
mental na escola; 4) Os alunos não se percepcionam como sendo eficazes na realização
de tarefas de cálculo mental; 5) O desempenho dos alunos em tarefas de cálculo mental
é razoável; 6) Os alunos recorreram a um leque de estratégias pouco variado, e usam
frequentemente a “Imagem mental do algoritmo” para resolver os cálculos que lhes são
propostos; 7) São poucos os alunos que conseguem referir estratégias alternativas para a
realização dos cálculos.ABSTRACT: The main purpose of this study consists in characterizing the profile of the 4th
grade students in what concerns mental computation. More specifically it is intended to
characterize their preferences and attitudes, their performance and the strategies they
used to solve mental computation tasks.
Forty-six 4th grade students of the Lisbon area participated in this study. Data
was gathered through the use of the preferences questionnaire, the attitudes
questionnaire, the mental computation test and lastly through the realization of
interviews.
The main found results were: 1) The Portuguese students prefer to use mental
computation in detriment of other types of computation and this preference is clearer in
students whose performance is superior in mental computation tasks; 2) The students
find mental computation more interesting and more daring than other types of
computation; 3) Most students state that they do more written computation and less
mental computation at school; 4) The students don’t percept themselves as being
effective in the fulfilment of mental computation tasks; 5) Students performance in
mental computation tasks was reasonable; 6) The students turned to a little varied range
of strategies and frequently use the “Mental image of the algorithm” to solve the
proposed computations; 7) Few students manage to mention alternative strategies for
the realization of the computations

Orientação: Manuel TavaresUltimamente, as expectativas sociais sobre a docência têm aumentado, pedindo-se aos professores que desempenhem mais e diversificadas funções.
A insatisfação profissional tornou-se um dos principais temas de investigação e um problema social relevante, dado que as exigências actuais e as mudanças socioeducativas influenciam como se vive a docência. A literatura considera, com certa relevância, algumas manifestações de insatisfação, tais como: a fadiga-exaustão ou desgaste docente, o mal-estar, o „stress‟, o absentismo, o desejo de abandono – abandono.
As questões da nossa pesquisa incidiram sobre a identificação dos factores de insatisfação e suas manifestações, dos paradoxos na docência e as causas actuais de abandono. O objectivo geral foi avaliar os factores de insatisfação docente nas escolas de um concelho do distrito de Santarém.
O estudo foi quantitativo e recorremos à aplicação de um questionário. Os dados foram tratados através do programa estatístico SPSS® versão 16.0.
Concluímos que a maioria dos professores sente-se realizada profissionalmente, mas a burocracia e o tempo de serviço para a aposentação são factores de insatisfação. 30,3% dos professores desejam abandonar a profissão, devido à desvalorização da profissão, à burocracia e à instabilidade profissional. Três razões principais condicionam essa decisão: dificuldade de emprego, razões económicas e idade.Social expectations on teaching have been increasing lately and teachers are asked to
play more diversified roles.
Professional dissatisfaction has become one of the top research themes and a relevant
social problem since the current demands and social and educational changes have an effect
on how teaching is faced. Research relevantly considers some signs of dissatisfaction, such as:
teaching fatigue-exhaustion, malaise, stress, absenteeism, the wish of resigning.
Our research questions were focused on the causes of dissatisfaction and its signs, teaching
paradoxes and the current causes of resignation. The main goal was to assess the causes of
teachers‟ dissatisfaction in a municipality in Santarem.
It was a quantitative study and we applied a questionnaire. The data was processed
with the statistical software SPSS, version 16.0.
We concluded that most teachers feel professionally satisfied but bureaucracy and
the working period to retirement are causes of dissatisfaction. 30,3% of the teachers want to
abandon their career because of its depreciation, bureaucracy and professional instability.

Conversão catalítica de sacarídeos em 2-furaldeído e 5-hidroximetil-2-furaldeído
Doutoramento em QuímicaThe conversion of plant biomass-derived carbohydrates (preferably non-edible) into added-value products is envisaged to be at the core of the future biorefineries. Carbohydrates are the most abundant natural organic polymers on Earth. This work deals with the chemical valorisation of plant biomass, focusing on the acid-catalysed conversion of carbohydrates (mono and polysaccharides) to furanic aldehydes, namely 2-furaldehyde (Fur) and 5-hydroxymethyl-2-furaldehyde (Hmf), which are valuable platform chemicals that
have the potential to replace a variety of oil derived chemicals and materials.
The investigated reaction systems can be divided into two types depending on the solvent used to dissolve the carbohydrates in the reaction medium: water or ionic liquid-based systems. The reaction temperatures were greater than 150 ºC when the solvent was water, and lower than 150 º C in the cases of the ionic liquid-based catalytic systems. As alternatives to liquid acids (typically used in the industrial production of Fur), solid acid catalysts were investigated in these reaction systems. Aiming at the identification of (soluble and insoluble) reaction products, complementary characterisation techniques were used namely, FT-IR spectroscopy, liquid and solid state NMR spectroscopy, TGA, DSC and GC´GC-ToFMS analyses. Complex mixtures of soluble reaction products were obtained and different types of side reactions may occur.
The requirements to be put on the catalysts for these reaction systems partly depend on the type of carbohydrates to be converted and the reaction conditions used. The thermal stability is important due to the fact that formation of humins and catalyst coking phenomena are characteristically inherent to
these types of reactions systems leading to the need to regenerate the catalyst which can be effectively accomplished by calcination. Special attention was given to fully inorganic nanoporous solid acids, amorphous or crystalline, and consisting of nano to micro-size particles. The investigated catalysts were
silicoaluminophosphates, aluminosilicates and zirconium-tungsten mixed oxides which are versatile catalysts in that their physicochemical properties can be fine-tuned to improve the catalytic performances in the conversion of different substrates (e.g. introduction of mesoporosity and modification of the
acid properties). The catalytic systems consisting of aluminosilicates as solid acids and water as solvent seem to be more effective in converting pentoses and related polysaccharides into Fur, than hexoses and related polysaccharides into Hmf. The investigated solid acids exhibited fairly good hydrothermal stabilities. On the other hand, ionic liquid-based catalytic systems can allow reaching simultaneously high Fur and Hmf yields, particularly when Hmf is obtained from D-fructose and related polysaccharides; however, catalyst
deactivation occurs and the catalytic reactions take place in homogeneous phase. As pointed out in a review of the state of the art on this topic, the development of truly heterogeneous ionic liquid-based catalytic systems for producing Fur and Hmf in high yields remains a challenge.Os carboidratos constituem os polímeros naturais mais abundantes na Terra, e a sua valorização química é de grande interesse no contexto das biorefinarias.
O objetivo deste trabalho centrou-se na conversão de carboidratos (monossacarídeos e polissacarídeos) em 2-furaldeído (Fur) e 5-hidroximetil-2-furaldeído (Hmf) na presença de catalisadores ácidos, em reatores
descontínuos. Fur e Hmf são considerados compostos “plataforma” porque podem ser convertidos numa grande variedade de produtos químicos e materiais (alternativos aos derivados do petróleo). Testaram-se catalisadores ácidos heterogéneos como alternativa aos ácidos minerais que são comumente usados como catalisadores homogéneos para a produção industrial do Fur. Por outro lado utilizou-se água ou um líquido iónico como solvente para a dissolução dos carboidratos no meio reacional. As temperaturas reacionais foram superiores a 150 ºC quando o solvente era a água, e inferiores a 150 ºC no caso
de líquidos iónicos. Com o intuito de identificar os produtos reacionais (solúveis e insolúveis), utilizaram-se diferentes técnicas nomeadamente espetroscopia de infravermelho, espetroscopia de RMN de estado líquido e sólido, TGA, DSC e GCxGC-ToFMS. Obtiveram-se misturas complexas de produtos reacionais e
discutiram-se aspetos mecanísticos.
A estabilidade térmica do catalisador é importante uma vez que a formação de matéria carbonácea insolúvel é característica destes sistemas reacionais tornando-se necessário proceder à regeneração do catalisador por calcinação.
Os catalisadores testados foram ácidos inorgânicos nanoporosos, cristalinos ou amorfos, com tamanho de partícula nano ou micrométrico, especificamente silicoaluminofosfatos, aluminossilicatos e óxidos mistos de zircónio e tungsténio.
Estes tipos de materiais são versáteis uma vez que as suas propriedades físicoquímicas podem ser modificadas no sentido de melhorar os seus desempenhos catalíticos na conversão de diferentes tipos de substratos (ex. através da criação de mesoporos nos materiais e/ou modificação das propriedades ácidas).
Os materiais testados exibiram melhores desempenhos catalíticos para a conversão de pentoses em Fur do que para a de hexoses em Hmf, quando o solvente era a água. Em suma, os catalisadores apresentaram boa estabilidade hidrotérmica. No caso dos sistemas reacionais à base de líquidos iónicos foram verificados elevados rendimentos em Fur e Hmf. especialmente quando os substratos eram a D-frutose ou polissacarídeos relacionados. Contudo, os catalisadores sofreram desativação tal que as reações catalíticas ocorreram em fase homogénea. Conforme explicado numa revisão bibliográfica sobre o estado da arte da conversão catalítica de carboidratos em Fur e Hmf usando líquidos iónicos, o desenvolvimento de sistemas catalíticos heterogéneos à base de líquidos iónicos representa um grande desafio
Diagnóstico molecular e citológico de herpesvirus felino tipo 1, chlamydophila felis e mycoplasma felis, em animais com conjutivite e/ou doença do trato respiratório superior
Orientação: Margarida Alves ; co-orientação: Odete AlmeidaA doença do trato respiratório superior (DTRS), apesar de muito comum em gatos, é, frequentemente, subvalorizada. Os sinais clínicos apresentados com mais frequência são espirros, tosse, secreção ocular e/ou nasal e conjuntivite. A conjuntivite felina apresenta, maioritariamente, uma natureza infeciosa, tendo como principais responsáveis o Herpesvírus felino-1, Chlamydophila felis e, possivelmente, Mycoplasma felis. Por vezes, o diagnóstico pode tornar-se desafiante, quer pela natureza dos agentes, quer pelos sinais clínicos inespecíficos.
O objetivo primário do presente trabalho foi realizar o diagnóstico da infeção por estes agentes, através da citologia conjuntival e da Reação em Cadeia da Polimerase (PCR), em gatos com sinais clinicos de DTRS e/ou de conjuntivite. Foram, ainda, objetivos do presente estudo avaliar a concordância entre os resultados do diagnóstico molecular e citológico e caracterizar a população em função dos agentes causadores de infeção.
Para tal, numa população de 29 gatos com sinais clínicos de DTRS e/ou conjuntivite, foram realizadas duas recolhas de amostras conjuntivais, uma com zaragatoas e outra com escova de citologia, para o diagnóstico molecular e citológico, respetivamente. As citologias foram coradas pelo método de May Grunwald-Giemsa. Para cada amostra, procedeu-se a duas PCR convencionais, para amplificação do DNA de FHV-1 e M.felis e uma PCR em tempo real, para deteção de DNA de C.felis. Foram, também, recolhidos dados clínicos e epidemiológicos dos animais em estudo, aquando das recolhas das amostras.
Ao diagnóstico molecular, foi obtida uma frequência de infeção de 58,6% para FHV-1 e de 31,0% para C.felis. Não foi identificado nenhum gato com infeção por M.felis. No diagnóstico citológico conjuntival, apenas seis animais foram diagnosticados com C.felis, não tendo sido feita nenhuma observação suspeita de infeção para os restantes agentes.
Os resultados evidenciaram uma elevada frequência de infeção por FHV-1 e C.felis. A PCR mostrou ser um meio de diagnóstico mais eficaz para a deteção dos agentes em estudo, quando comparada com a citologia conjuntival. O presente estudo deixa em aberto a dúvida sobre o papel de M.felis enquanto agente etiológico primário, quer em conjuntivites, quer na DTRS.The upper respiratory tract disease (URTD) is very usual in cats, but quite often underestimated. Main clinical signs are sneezes, cough, ocular and/or nasal secretions and conjunctivitis. Feline conjunctivitis has, mostly, an infectious nature with feline-1 herpesvirus, Chlamydophila felis, and, possibly, Mycoplasma felis as the main causative agents. Diagnosis can, sometimes, become challenging, both by the nature of the agents and the nonspecific clinical signs.
The primary objective of this study was to diagnose these agents through conjunctival cytology and Polymerase Chain Reaction (PCR), in cats with clinical signs of URTD and/or conjunctivitis. The secondary objective of this study was to evaluate the concordance of molecular and cytological results and to characterize the population according to the causative agents of infection.
For this purpose, in a population of 29 cats with clinical signs of URTD and/or conjunctivitis, two conjunctival samples were taken, one with a swab, for PCR and another with a cytology brush, for cytological evaluation. Cytologies were stained with May Grunwald-Giemsa. Two conventional PCRs, for FHV-1 and M.felis, and a real-time PCR for C. felis, were performed for each sample. Clinical and epidemiological data were also collected from the animals, when samples were taken.
By PCR, an infection frequency of 58.6% was obtained for FHV-1 and 31.0% for C. felis. M.felis DNA was not detected in all animals. In the conjunctival cytologic diagnosis, only six animals were positive to C. felis, and no signs suggestive of infection were observed for the remaining agents.
The results evidenced a high frequency of infection by FHV-1 and C.felis. PCR has shown to be a more effective as a diagnostic tool for detecting the studied agents, compared to conjunctival cytology. This study also reinforces the doubt about the possibility of M.felis playing a primary role in both conjunctivitis and URTD.

Síntese de novos compostos do tipo 1,5-benzodiazepina
Mestrado em Química de Produtos NaturaisA procura de novos compostos biologicamente activos e, consequentemente,
com possível utilização no tratamento de várias doenças tem aumentado nas
últimas décadas. Entre os compostos que têm sido alvo de intensas pesquisas
encontram-se as benzodiazepinas. Esta dissertação tem como objectivo a
síntese de novas 1,5-benzodiazepinas, que têm sido reportadas como uma
importante classe de benzodiazepinas. Na primeira parte deste trabalho
apresenta-se uma breve revisão bibliográfica sobre a estrutura química e
sistema de numeração, ocorrência natural, algumas metodologias de síntese e
aplicações de benzodiazepinas. A segunda parte descreve a síntese de 4-aril-
2-(2-hidroxifeniletilidenona)-3,4-di-hidro-1H,5H-benzo[b][1,5]-diazepinas,
4-estiril-2-(2-hidroxifenil)-3H-benzo[b][1,5]-diazepinas, e 1-acetil-4-estiril-
2-(2-hidroxifenil)-1H-benzo[b][1,5]-diazepinas obtidas por reacção de 5-aril-3-
hidroxi-1-(2-hidroxifenil)-2,4-pentadieno-1-onas com orto-fenilenodiamina. As
primeiras resultam de adição conjugada de orto-fenilenodiamina à unidade
carbonílica α,β-insaturada, e as segundas da condensação da ortofenilenodiamina
com a unidade β-dicetónica. As 5-aril-3-hidroxi-
1-(2-hidroxifenil)-2,4-pentadieno-1-onas foram previamente preparadas por um
rearranjo de Baker-Venkataraman de 2’-cinamoiloxiacetofenonas. Para além
das benzodiazepinas desejadas foram também obtidos produtos secundários,
dos quais se destacam as 2-estirilcromonas. Na terceira parte descreve-se
detalhadamente a caracterização estrutural das novas 1,5-benzodiazepinas e
dos produtos secundários que são compostos novos. Para tal, recorreu-se a
estudos de espectroscopia de ressonância magnética nuclear (RMN) os quais
incluíram a obtenção de espectros de 1H, 13C e estudos bidimensionais de
correlação espectroscópica homo- e heteronuclear (HSQC, HMBC).
ABSTRACT: The search of new biologically active compounds and consequently possible
use in the treatment of several diseases has been improving over the last
decades. Among the target compounds submitted to an intensive research,
benzodiazepines play an important role. This dissertation has as objective the
synthesis of new 1,5-benzodiazepines, which have been reported as an
important class of benzodiazepines. In the first part of this work, a review on
the chemical structure and numbering system, natural occurrence, some
methodologies of synthesis and their applications are presented. The second
part describes the synthesis of 4-aryl-2-(2-hydroxyphenylethylidenone)-
3,4-dihydro-1H,5H-benzo[b][1,5]-diazepines, 2-(2-hydroxyphenyl)-4-styryl-3Hbenzo[
b][1,5]-diazepines, and 1-acetyl-2-(2-hydroxyphenyl)-4-styryl-1H-benzo
[b][1,5]-diazepines, obtained by the reaction of 5-aryl-3-hydroxy-
1-(2-hydroxyphenyl)-2,4-pentadiene-1-one with ortho-phenylenediamine. The
former have been obtained from the conjugated addition of orthophenylenediamine
to the α,β-insaturated carbonyl moiety while the latter were
obtained by the condensation of ortho-phenylenediamine with β-diketone
moiety. The 5-aryl-3-hydroxy-1-(2-hydroxyphenyl)-2,4-pentadiene-1-ones were
previously prepared from the Baker Venkataraman rearrangement of
2-cinnamoyloxyacetophenones. Besides the desired benzodiazepines, some
by products have been obtained, being the 2-styrylchromones the most
important ones. In the third part the structural characterization of the new
1,5-benzodiazepines and other new compounds are described in detail. For
that, we used exhaustive nuclear magnetic resonance (NMR) spectroscopy
studies, including 1H, 13C, 2D-NMR homo- and heteronuclear correlated
spectroscopy (HSQC and HMBC)
INTERNATIONAL DIVERSIFICATION STRATEGIES AND THEIR ORGANIZATIONAL AND EXOGENOUS ANTECEDENTS
International operations, with its multiplicity of objectives, can be seen
as a diversification strategy, sometimes even as a strategic option taken
as an effective alternative to product diversification. Focusing on a sample of SMEs of Portuguese manufacturing industries, this study analyzes the international diversification strategies and their organizational and exogenous antecedents. Specifically, entrepreneurial orientation and dynamic capabilities of the firm are two factors of organizational nature, while the environmental uncertainty arising from the diversity of the different markets is studied as an exogenous antecedent. Data collected in 390 companies through an online questionnaire, its processing and analysis through the use of a structural equations model suggest the existence of different types of influence of the antecedents, according to companies’ options in various dimensions of the international diversification strategies. These results provide guidelines for managers and generate directions for future research
