Vertaisarvioitu. English summary. SyövÀn lÀÀkehoitojen kehitys on tuonut vaihtoehtoja aivoetÀpesÀkkeiden hoitoon. Erityisesti melanooman aivoetÀpesÀkkeiden hoidossa immuno-onkologinen yhdistelmÀhoito (nivolumabi-ipilimumabi) nÀyttÀisi tulokselliselta

Objective. Resistance to standard chemotherapy poses a major clinical problem in the treatment of ovarian cancer patients. Adult-type granulosa cell tumor (AGCT) is a unique ovarian cancer subtype for which efficient treatment options are lacking in advanced disease. To this end, systematic drug response and transcriptomics profiling were performed to uncover new therapy options for AGCTs. Methods. The responses of three primary and four recurrent AGCTs to 230 anticancer compounds were screened in vitro using a systematic drug sensitivity and resistance testing (DSRT) platform, coupled with mRNA sequencing. The responses of the AGCTs were compared with those of human granulosa luteal cells and bone marrow mononuclear cells. Results. Patient-derived AGCT cells showed selective sensitivity to the Src family tyrosine kinase inhibitor dasatinib. A combination of either dasatinib or an mTOR-inhibitor everolimus with paclitaxel resulted in synergistic inhibition of AGCT cell viability. The key kinase targets of dasatinib and members of the mTOR pathway were constantly expressed at mRNA and protein levels, indicating multikinase signal addictions in the AGCT cells. Transcriptomic characterization of the tumors revealed no known oncogenic mutations, suggesting that the drug sensitivity of AGCTs was rather conveyed by selective target expression. Conclusions. We used a systematic functional approach to reveal novel treatment options for a unique gynecological cancer. The selective synergy found between taxanes and dasatinib or mTOR inhibitors warrants further clinical investigations of these combinations in relapsed or aggressive AGCTs and demonstrate that high throughput drug screening and molecular profiling can provide an effective approach to uncover new therapy options. Publisher Copyright: © 2021 The Authors. Molecular Ecology Resources published by John Wiley & Sons Ltd.To associate specimens identified by molecular characters to other biological knowledge, we need reference sequences annotated by Linnaean taxonomy. In this study, we (1) report the creation of a comprehensive reference library of DNA barcodes for the arthropods of an entire country (Finland), (2) publish this library, and (3) deliver a new identification tool for insects and spiders, as based on this resource. The reference library contains mtDNA COI barcodes for 11,275 (43%) of 26,437 arthropod species known from Finland, including 10,811 (45%) of 23,956 insect species. To quantify the improvement in identification accuracy enabled by the current reference library, we ran 1000 Finnish insect and spider species through the Barcode of Life Data system (BOLD) identification engine. Of these, 91% were correctly assigned to a unique species when compared to the new reference library alone, 85% were correctly identified when compared to BOLD with the new material included, and 75% with the new material excluded. To capitalize on this resource, we used the new reference material to train a probabilistic taxonomic assignment tool, FinPROTAX, scoring high success. For the full-length barcode region, the accuracy of taxonomic assignments at the level of classes, orders, families, subfamilies, tribes, genera, and species reached 99.9%, 99.9%, 99.8%, 99.7%, 99.4%, 96.8%, and 88.5%, respectively. The FinBOL arthropod reference library and FinPROTAX are available through the Finnish Biodiversity Information Facility (www.laji.fi) at https://laji.fi/en/theme/protax. Overall, the FinBOL investment represents a massive capacity-transfer from the taxonomic community of Finland to all sectors of society.Peer reviewe

Internetin toiminnan mahdollistavista datakeskuksista on viime vuosien aikana tullut yksi IT-alan merkittävimmistä kasvihuonekaasupäästöjen aiheuttajista ja niiden energiankulutus on koko maailmankin mittakaavassa huomattava. Tutkimuksessa selviää, että vihreän IT:n hyödyntäminen voidaan jakaa kolmeen osa-alueeseen: IT-laitteisiin, niiden ohjelmistoihin ja IT:n epäsuoraan hyödyntämiseen. Esimerkkeinä IT-laitteista on serverien energiatehokkuuden parantaminen, ohjelmistoista virtualisointi, ja IT:n epäsuorasta hyödyntämisestä datakeskuksen toiminnan mittaaminen ja analysointi. Vihreän IT:n käyttämisen hyötyjä voidaan myös väljästi jaotella näihin osa-alueisiin. Hyötyjä ovat muun muassa kustannussäästöt, serverien laskentatehon kasvu, uusien laitteiden vähentynyt tarve ja kasvihuonekaasupäästöjen väheneminen.Climate change, contamination and other environmental harms caused by mankind have led environmental friendliness and a green way of thinking to be a very popular topic of conversation. These environmental problems have also been recognized inside the IT sector and green IT aims to play a part in solving them. The purpose of green IT is to use information and communication technology to reduce the harmful environmental impacts, either directly or indirectly. Data centers, an important part of the modern Internet, have in recent years become one of the biggest sources of greenhouse gases in the IT sector and the amount of energy they use is significant even on a world scale. The topic of this Bachelor’s Thesis is the use of green IT in data centers and the advantages of using it. The study was conducted as a literature review. The study reveals that the use of green IT can be grouped into three categories: IT devices, their software and the indirect utilization of IT. Corresponding examples of the three categories are improving the energy efficiency of servers, virtualization, and using metrics to monitor and analyze the data center’s performance. Benefits of using green IT can also be roughly grouped into these three categories. Some of the benefits include cost savings, increased computing power of the servers, reduced need of new equipment and reduced emission of greenhouse gases

Tutkielma tarjoaa tiettĂ€vĂ€sti ensimmĂ€isen kirjallisuuskatsauksen, jossa tekoĂ€lyn kĂ€yttöÀ olisi yleisesti tarkasteltu sisĂ€logistiikan automaatiojĂ€rjestelmissĂ€.Intralogistics is a part of organization’s logistics and supply chain management, which are central functions of modern organizations. Effective management of intralogistics helps an organization to gain competitive advantage over its competitors. Automation systems which replace human manual labor with machines are one way to make intralogistics functions more efficient. The rapid advancements of different artificial intelligence technologies and their successful applications in various domains has piqued the interest, whether artificial intelligence could also be used to make automation systems more efficient in the field of intralogistics. The purpose of this thesis is to research the aforementioned topic, and the research question is to figure out how artificial intelligence could be used in controlling intralogistics automation systems. The thesis is done as a commission for a Finnish company which makes intralogistics automation systems. Research method applied was Design Science Research Methodology process. The resulting artifact assesses the feasibility of genetic algorithms, one of the technologies included in the field of artificial intelligence, to solve the warehouse product assignment problem in a simulated portal robot warehouse. The created artifact proves the potential of genetic algorithms in a real world automation systems but using them in a production environment would still require making the simulation more accurate. In addition, the results of the thesis show that the use of artificial intelligence has mainly be researched in automation systems using high bay warehouses or self-driving AGVs. These studies use multi-agent systems, genetic algorithms and reinforcement learning among other technologies. All in all the topic has not received much research interest. As far as is known, the thesis offers the first literature review where the application of artificial intelligence is examined broadly in the domain of intralogistics automation systems

Digital flow control unit for controlling amount of water used for binding dust

    Get PDF
Rock drilling requires that the rock dust formed during drilling is removed with flushing. Nowadays, liquid mist is often employed to bind dust from the flushing air in overground drilling. The amount of the liquid, for example water, is precise, because too small or too large amount of liquid leads to unwanted result. Traditionally, the liquid amount is controlled by a manual valve in the control cabin of the drilling machine. This method has disadvantages and therefore, some alternative control systems have been considered. This paper presents a 7-bit Digital Flow Control Unit (DFCU) which was designed for controlling the amount of water in the dust binding system.Peer reviewe
