    Effects of simvastatin/ezetimibe on microparticles, endothelial progenitor cells and platelet aggregation in subjects with coronary heart disease under antiplatelet therapy

    It is not known whether the addition of ezetimibe to statins adds cardiovascular protection beyond the expected changes in lipid levels. Subjects with coronary heart disease were treated with four consecutive 1-week courses of therapy (T) and evaluations. The courses were: T1, 100 mg aspirin alone; T2, 100 mg aspirin and 40 mg simvastatin/10 mg ezetimibe; T3, 40 mg simvastatin/10 mg ezetimibe, and 75 mg clopidogrel (300 mg initial loading dose); T4, 75 mg clopidogrel alone. Platelet aggregation was examined in whole blood. Endothelial microparticles (CD51), platelet microparticles (CD42/CD31), and endothelial progenitor cells (CD34/CD133; CDKDR/CD133, or CD34/KDR) were quantified by flow cytometry. Endothelial function was examined by flow-mediated dilation. Comparisons between therapies revealed differences in lipids (T2 and T3T1 and T4, P=0.001). Decreased platelet aggregation was observed after aspirin (arachidonic acid, T1<T3 and T4, P=0.034) and clopidogrel (adenosine, T3 and T4<T1 and T2, P<0.0001) therapy. Simvastatin/ezetimibe diphosphate did not change platelet aggregation, the amount of circulating endothelial and platelet microparticles, or endothelial progenitor cells.     Oilfield carbonated produced water recycling coupled to exopolysaccharide transformation by lelliottia amnigena

    It was tested the production of a viscous exopolysaccharide (EPS) by Lelliottia amnigena using different concentrations (10, 15, 25, 50 and 100%) of produced water (PW) and Dialyzed PW (DPW) at 100% concentration, with a nutrient medium based on glycerin and sucrose as carbon sources in 44 h batches. There was an increase in EPS recovery when used PW up to a concentration of 25%. The viscosity of the EPS solutions was maintained with DPW and up to 15% of PW, such a condition resulted in 9.34 gL− 1 of EPS, representing an increase of 84.9% compared to the control (P < 0.0001). The PW 15% increased the viscosity per produced liter by 114.3% (P < 0.0001). Scanning electron microscopy and Energy-dispersive X-ray spectroscopy analysis showed that EPS adsorbed mineral constituents of the substrate and its characterization by Fourier Transform Infrared Spectroscopy was similar to an external control of a commercial EPS produced by an Enterobacter A47 strain.     Photobiomodulation Therapy in the Proliferation and Differentiation of Human Umbilical Cord Mesenchymal Stem Cells: An In Vitro Study

    Introduction: Since photobiomodulation therapy (PBMT) favors in vitro mesenchymal stem cell (MSC) preconditioning before MSC transplantation, increasing the proliferation of these cells without molecular injuries by conserving their characteristics, in the present in vitro study we analyzed the effect of PBMT on the proliferation and osteogenic differentiation of human umbilical cord mesenchymal stem cells (hUCMSCs).Methods: Irradiation with an InGaAIP Laser (660 nm, 10 mW, 2.5 J/cm2, 0.08 cm2 spot size, and 10 s) was carried out. The cells were divided into four groups: CONTROL [cells grown in Dulbecco’s Modified Eagle Medium (DMEM)], OSTEO (cells grown in an osteogenic medium); PBMT (cells grown in DMEM+PBMT), and OSTEO+PBMT (cells grown in an osteogenic medium-plus PBMT). The cell proliferation curve was obtained over periods of 24, 48 and 72 hours using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Osteogenic differentiation was analyzed by the formation of calcium nodules over periods of 7, 14 and 21 days. Morphometric analysis was performed to quantify the total area of nodular calcification.Results: The highest cell proliferation and cell differentiation occurred in the OSTEO+PBMT group, followed by the PBMT, OSTEO, and CONTROL groups respectively, at the observed times (P &lt; 0.05).Conclusion: PBMT enhanced the osteogenic proliferation and the differentiation of hUCMSCs during the periods tested, without causing damage to the cells and preserving their specific characteristics, a fact that may represent an innovative pretreatment in the application of stem cells

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Professores universitários com deficiência: trajetória escolar e conquista profissional

    Este estudo tem como foco a educação escolar de pessoas com deficiência. Investigou-se a trajetória escolar de professores universitários, com o objetivo de compreender influências sobre desempenho escolar e identificar razões da distinção que os tornou minoria dentre os milhares de outras pessoas com deficiência que não atingem tal patamar de educação formal e profissionalização. O estudo tem como base teórica os trabalhos de Bourdieu. Optou-se por trabalhar com procedimentos metodológicos de entrevista, questionário e análise de documentos biográficos. Participaram desta pesquisa três professores. O estudo indica que a valorização familiar pelo estudo, a condescendência da instituição escolar e a distribuição do capital cultural objetivado são indicativos do desempenho escolar e profissional destes sujeitos
