    Předmětem diplomové práce je urbanistické řešení území, vymezeného ulicemi Sportovní, Drobného, tř. Gen. Píky, Porgesova. Důraz se klade především na obnovu a zatraktivnění ústředního bodu jímž je fotbalový stadion, zefektivnění napojení na okolní dopravní uzly a revitalizace zeleně, nově vybudovanými parky. Předpokládaný výsledek je navrácení sportovní funkce oblasti, jako je celkové zvýšení zájmu veřejnosti o toto území.The subject of the masters thesis is an urban solution of the area circumscribed by the streets Sportovní, Drobného, tř. Gen. Píky, Porgesova. The emphasis is put primarily on restoring and enhancing the central point, which is the football stadium, building more effective conection to nearby traffic hubs and revitalising the surrounding parks. Estimated result is to bring back the sport function to the area as well as overall increase of the interest from public.

    The topic of this bachelor's thesis is the function of the reccomendation algorithm on the YouTube platform and its preferential treatment of misinformation and alternative content over reliable sources. It focuses on Czech-language YouTube. This thesis is also concerned with the theory of the filter bubble, to which this topic is also connected. The main question posed in this bachelor's thesis is whether the algoritm purposely encloses the user in the filter bubble or not. Executed research follows the method of gathering recommended videos propagated by Guillame Chaslot, and focuses on the analysis of the data acquired. The results, presented in charts and accompanied by sufficient commentary, are then compared and critically evaluated. I believe that the results will prove valuable for future research into this topic.Tato bakalářská práce se zabývá fungováním doporučovacího algoritmu na platformě YouTube a jeho upřednostňováním dezinformačního a alternativního obsahu před ověřenými a spolehlivými zdroji. Soustředí se na české prostředí YouTube. S danou problematikou souvisí informační bublina, které se práce v teoretické rovině také věnuje. Hlavní otázkou bakalářské práce je, zda algoritmus funguje tak, že uživatele do informační bubliny aktivně uzavírá. Provedený výzkum napodobuje metodu sběru doporučených videí tak, jak ji navrhuje Guillame Chaslot, a věnuje se analýze získaných dat. Výsledky jsou prezentovány v grafech a patřičně okomentovány, porovnávány mezi sebou a kriticky hodnoceny. Prostor uvažované stavby se nachází na neudržovaném rovinatém pozemku. Objekt má pravidelný obdélníkový půdorysný tvar. Skládá se ze dvou bloků a má tří podlažní. Navazuje na uliční řadovou zástavbu a svou výškou nenarušuje okolí. Fasáda bude prosklená. Nosnou konstrukcí je železobetonový monolitický skelet v modulu 6x6m. Výplňové zdivo je z tvárnic YTONG. Jako zastřešení je zvolena plochá střecha. Založení objektu je provedeno pomocí železobetonových patek a pásů.Home Business is situated in a part of Brno-South.     Praca recenzowana / Peer-reviewed paperThis publication includes opinion and research papers written by authors with academic backgrounds from the Czech Republic, Slovakia and Poland. The monograph consists of 14 chapters and the order of them refers to the relationship between the family and the health of its members. The first seven texts concern children’s health problems. The next two concern widely defined issues of nutrition and adult nursing. The next four discuss health issues of the elderly, while the last describes the dilemma of bereaved family members who have to decide about organ donation. The individual chapters show different aspects of family participation in the treatment and care of children, the elderly, the chronically ill, and the disabled. The discussed topics are a part of the wider issue of social medicine. A very important issue is the interdisciplinary problem of interaction between health and illness and their influence on the family, which is on one hand embedded in medical science, while on the other hand it refers to the social sciences, especially family sociology, psychology, pedagogy, and also social policy to some extent. The authors of the monograph also discuss the issue of life quality of the elderly living in domestic environments and the health and social problems they face. The authors also analyse the role of nurses in promoting health, such as immunisation, rational nutrition, and ethical aspects such as the screening of newborns. This monograph is addressed to all who work or want to work with families and support them in the difficult challenges posed by disease.     Secondary structure is required for 3′ splice site recognition in yeast

    Higher order RNA structures can mask splicing signals, loop out exons, or constitute riboswitches all of which contributes to the complexity of splicing regulation. We identified a G to A substitution between branch point (BP) and 3′ splice site (3′ss) of Saccharomyces cerevisiae COF1 intron, which dramatically impaired its splicing. RNA structure prediction and in-line probing showed that this mutation disrupted a stem in the BP-3′ss region. Analyses of various COF1 intron modifications revealed that the secondary structure brought about the reduction of BP to 3′ss distance and masked potential 3′ss. We demonstrated the same structural requisite for the splicing of UBC13 intron. Moreover, RNAfold predicted stable structures for almost all distant BP introns in S. cerevisiae and for selected examples in several other Saccharomycotina species. The employment of intramolecular structure to localize 3′ss for the second splicing step suggests the existence of pre-mRNA structure-based mechanism of 3′ss recognition

    Interlaboratory Coverage Test on Plant Food Bioactive Compounds and their Metabolites by Mass Spectrometry-Based Untargeted Metabolomics.

    Bioactive compounds present in plant-based foods, and their metabolites derived from gut microbiota and endogenous metabolism, represent thousands of chemical structures of potential interest for human nutrition and health. State-of-the-art analytical methodologies, including untargeted metabolomics based on high-resolution mass spectrometry, are required for the profiling of these compounds in complex matrices, including plant food materials and biofluids. The aim of this project was to compare the analytical coverage of untargeted metabolomics methods independently developed and employed in various European platforms. In total, 56 chemical standards representing the most common classes of bioactive compounds spread over a wide chemical space were selected and analyzed by the participating platforms (n = 13) using their preferred untargeted method. The results were used to define analytical criteria for a successful analysis of plant food bioactives. Furthermore, they will serve as a basis for an optimized consensus method

    Human SKIP/NcoA-62/SNW1 ortholog proteins are nuclear factors which have been found in connection with the regulation of gene expression and subsequent modifications of primary transcripts. Human representative SKIP acts primarily as a transcription factor during the processes involving differentiation, carcinogenesis and other cellular signaling. It operates in the signaling pathways, eg TGFß/Smad, E2F/pRb, Notch/CBF1 and nuclear hormone receptors. SKIP protein is also essential for pre-mRNA splicing and recently its contribution during the elongation phase of RNA polymerase II transcription and contribution to some HIV genes alternative transcripts formation has been identified. The ortholog of S. cerevisiae FUN2O/Prp45 has been shown to participate in pre-mRNA splicing, others processes mentioned have not yet been associated with the function of this factor. Pre-mRNA splicing is a complicated process leading to the formation of native mRNA lacking non-coding regions, introns. In terms of composition of the spliceosome subunits and biochemical principle of its function is this process very well described but despite this knowledge the specific roles of most splicing factors are not known.     Cílem této bakalářské práce je především ověřit míru informovanosti spotřebitelů o negativních dopadech padělaného zboží a na základě toho navrhnout řešení, jak informovanost o tomto problému zlepšit a přispět tak ke snížení poptávky po padělcích. Práce rovněž tyto negativní dopady shrnuje a vymezuje, jak se na prodeji padělků podílejí spotřebitelé svojí neklesající poptávkou. V první polovině práce je popsána aktuální situace na trhu padělků, nové trendy v oblasti padělání i současná opatření, kterými se snaží různé instituce rostoucí množství padělků řešit. Ve druhé polovině práce je za pomoci dotazníkového šetření a rozhovoru s odborníkem potvrzena domněnka, že spotřebitelé by padělky kupovali méně, kdyby byli při svém nákupním rozhodnutí o uváděných negativních dopadech dostatečně informovaní. Na závěr jsou sepsána doporučení, jak lze tohoto zjištění v době rozvoje internetu využít. Obzvláště v současném období koronavirové krize, kdy se mnozí občané cítí vládou nadměrně omezováni, by nebylo vhodné zavádět další radikální omezující opatření. Proto je pracováno s doporučeními, která navrhují řešení problému nenásilnou cestou.The aim of this bachelor thesis is to test the consumers knowledge of the main issues of counterfeiting and suggest a solution to improve the awareness in order to limit the demand for counterfeit goods. The thesis also summarizes the negative effects of counterfeit trade and determines the role played by the consumer in mitigating these. In the first half of this paper, the current situation on the counterfeit market is reviewed and new trends as well as measures taken by the institutions to stop the trade are evaluated. In the second half, results of a survey and an interview confirm the tested hypothesis that consumers would reduce their spending on counterfeit goods if they were more informed about the negative consequences. In conclusion, recommendations to achive higher awareness and limit the impact conservatively are presented. Especially in the current period of the coronavirus crisis, when many citizens feel overly constrained by the government, introducing more radical restrictive measures would not be appropriate. Therefore solutions to the problem in non-violent way are suggested
