    Near future climate change projections with implications for the agricultural sector of three major Mediterranean islands

    AbstractThe paper presents the analysis of a sub-set of high-resolution bias-adjusted simulations from the EURO-CORDEX initiative, in order to examine the changes in the mean climate and the extremes in three Mediterranean islands, namely, Sicily, Crete and Cyprus, in the near future (2031–2060) compared to the present climate (1971–2000), under two future scenarios, i.e. RCP4.5 and RCP8.5. The analysis entails commonly used climatic indices of interest related to the islands' agricultural sector. The results indicate robust increases for both the mean maximum and minimum temperatures on a seasonal basis, as well as for the temperature related extremes under both climate scenarios. On the contrary, the changes in precipitation are less pronounced as the changes in the seasonal precipitation are not found statistically significant for the three islands under both scenarios. The projected warming combined with the projected unchanged precipitation pattern in the future, especially in spring and summer, might expose the crops to conditions with a negative impact on the plants' phenology, leading to implications on crop production and quality. The results presented here might be the basis for the development of an adaptation strategy specifically targeted on the three islands but also replicable to other Mediterranean islands

    Assessing climate change impacts on crops by adopting a set of crop performance indicators

    AbstractThe impact of climate change on the agricultural systems of three major islands in the Mediterranean basin, namely Sicily, Crete and Cyprus, was evaluated using a suite of specifically calibrated crop models and the outputs of a regional circulation model for Representative Concentration Pathway (RCP) 4.5 and 8.5 downscaled to 12 km of resolution and tested for its effectiveness in reproducing the local meteorological data. The most important annual (wheat, barley, tomato and potato) and perennial (grapevine and olive tree) crops were selected to represent the agricultural systems of the islands. The same modelling framework was used to test the effectiveness of autonomous adaptation options, such as shifting sowing date and the use of varieties with different growing season length. The results highlighted that, on average, warmer temperatures advanced both anthesis and maturity of the selected crops, but at different magnitudes depending on the crop and the island. Winter crops (barley, wheat and potato) experienced the lowest impact in terms of yield loss with respect to the baseline, with even some positive effects, especially in Sicily where both wheat and barley showed a general increase of 9% as compared to the baseline, while potato increased up to + 17%. Amongst perennial crops, olive tree showed low variation under RCP 4.5, but on average increased by 7% under RCP 8.5 on the three islands. Climate change had a detrimental effect specifically on tomato (− 2% on average in RCP 8.5 and 4.5 on the three islands) and grapevine (− 7%). The use of different sowing dates, or different varieties, revealed that for winter crops early autumn sowing is still the best option for producing wheat and barley in future periods on the three islands under both future scenarios. For tomato and potato, advancing sowing date to early winter is a winning strategy that may even increase final yield (+ 9% for tomato and + 17% for potato, on average). For grapevine, the use of late varieties, while suffering the most from increasing temperatures and reduced rainfall (− 15%, on average), is still a valuable option to keep high yield levels with respect to earlier varieties, which even if showing some increases with respect to the baseline have a generally much lower production level. The same may be applied to olive tree although the production differences between late and early varieties are less evident and climate change exerts a favourable influence (+ 4 and + 3% for early and late varieties, respectively)

    MicroRNA-Restricted Transgene Expression in the Retina

    Background: Gene transfer using adeno-associated viral (AAV) vectors has been successfully applied in the retina for the treatment of inherited retinal dystrophies. Recently, microRNAs have been exploited to fine-tune transgene expression improving therapeutic outcomes. Here we evaluated the ability of retinal-expressed microRNAs to restrict AAV-mediated transgene expression to specific retinal cell types that represent the main targets of common inherited blinding conditions. Methodology/Principal Findings: To this end, we generated AAV2/5 vectors expressing EGFP and containing four tandem copies of miR-124 or miR-204 complementary sequences in the 39UTR of the transgene expression cassette. These vectors were administered subretinally to adult C57BL/6 mice and Large White pigs. Our results demonstrate that miR-124 and miR-204 target sequences can efficiently restrict AAV2/5-mediated transgene expression to retinal pigment epithelium and photoreceptors, respectively, in mice and pigs. Interestingly, transgene restriction was observed at low vector doses relevant to therapy.     Genetic prediction of ICU hospitalization and mortality in COVID-19 patients using artificial neural networks

    There is an unmet need of models for early prediction of morbidity and mortality of Coronavirus disease-19 (COVID-19). We aimed to a) identify complement-related genetic variants associated with the clinical outcomes of ICU hospitalization and death, b) develop an artificial neural network (ANN) predicting these outcomes and c) validate whether complement-related variants are associated with an impaired complement phenotype. We prospectively recruited consecutive adult patients of Caucasian origin, hospitalized due to COVID-19. Through targeted next-generation sequencing, we identified variants in complement factor H/CFH, CFB, CFH-related, CFD, CD55, C3, C5, CFI, CD46, thrombomodulin/THBD, and A Disintegrin and Metalloproteinase with Thrombospondin motifs (ADAMTS13). Among 381 variants in 133 patients, we identified 5 critical variants associated with severe COVID-19: rs2547438 (C3), rs2250656 (C3), rs1042580 (THBD), rs800292 (CFH) and rs414628 (CFHR1). Using age, gender and presence or absence of each variant, we developed an ANN predicting morbidity and mortality in 89.47% of the examined population. Furthermore, THBD and C3a levels were significantly increased in severe COVID-19 patients and those harbouring relevant variants. Thus, we reveal for the first time an ANN accurately predicting ICU hospitalization and death in COVID-19 patients, based on genetic variants in complement genes, age and gender.     Cost-effective sequence analysis of 113 genes in 1,192 probands with retinitis pigmentosa and Leber congenital amaurosis

    Introduction: Retinitis pigmentosa (RP) and Leber congenital amaurosis (LCA) are two groups of inherited retinal diseases (IRDs) where the rod photoreceptors degenerate followed by the cone photoreceptors of the retina. A genetic diagnosis for IRDs is challenging since &gt;280 genes are associated with these conditions. While whole exome sequencing (WES) is commonly used by diagnostic facilities, the costs and required infrastructure prevent its global applicability. Previous studies have shown the cost-effectiveness of sequence analysis using single molecule Molecular Inversion Probes (smMIPs) in a cohort of patients diagnosed with Stargardt disease and other maculopathies. Methods: Here, we introduce a smMIPs panel that targets the exons and splice sites of all currently known genes associated with RP and LCA, the entire RPE65 gene, known causative deep-intronic variants leading to pseudo-exons, and part of the RP17 region associated with autosomal dominant RP, by using a total of 16,812 smMIPs. The RP-LCA smMIPs panel was used to screen 1,192 probands from an international cohort of predominantly RP and LCA cases. Results and discussion: After genetic analysis, a diagnostic yield of 56% was obtained which is on par with results from WES analysis. The effectiveness and the reduced costs compared to WES renders the RP-LCA smMIPs panel a competitive approach to provide IRD patients with a genetic diagnosis, especially in countries with restricted access to genetic testing.</p

    A blood atlas of COVID-19 defines hallmarks of disease severity and specificity.

    Treatment of severe COVID-19 is currently limited by clinical heterogeneity and incomplete description of specific immune biomarkers. We present here a comprehensive multi-omic blood atlas for patients with varying COVID-19 severity in an integrated comparison with influenza and sepsis patients versus healthy volunteers. We identify immune signatures and correlates of host response. Hallmarks of disease severity involved cells, their inflammatory mediators and networks, including progenitor cells and specific myeloid and lymphocyte subsets, features of the immune repertoire, acute phase response, metabolism, and coagulation. Persisting immune activation involving AP-1/p38MAPK was a specific feature of COVID-19. The plasma proteome enabled sub-phenotyping into patient clusters, predictive of severity and outcome. Systems-based integrative analyses including tensor and matrix decomposition of all modalities revealed feature groupings linked with severity and specificity compared to influenza and sepsis. Our approach and blood atlas will support future drug development, clinical trial design, and personalized medicine approaches for COVID-19

    Η εργασία αυτή αφορά τη μελέτη της συνεργασίας μιας ομάδας εκπαιδευτικών. Συγκεκριμένα πρόκειται για μια συνεργασία που ξεκινά το Δεκέμβρη του 2018 όπου τα μέλη της συναντήθηκαν μέσω του προγράμματος educate. Οι εκπαιδευτικοί που έλαβαν μέρος μελέτησαν τις μαθηματικές και κοινωνικομαθηματικές νόρμες εστιάζοντας στη διαφοροποίηση και τη μαθηματική πρόκληση και συμμετέχοντας στο συνέδριο της ΕΝΕΔΙΜ. Δύο χρόνια αργότερα, το Φεβρουάριο του 2021, όπου λόγω των νέων συνθηκών που έφερε ο COVID 19 η εκπαίδευση γίνεται εξ αποστάσεως , η ομάδα συνεχίζει τη συνεργασία της με στόχο τη μελέτη των νορμών στην τηλεκπαίδευση και το ρόλο τους στην εμπλοκή των μαθητών. Η εργασία αυτή θα στοχεύσει στις κατανοήσεις που αναπτύσσουν οι εκπαιδευτικοί μέσα από τη συνεργασία τους ως προς το περιεχόμενο της διδασκαλίας και τη σημασία των νορμών στην εμπλοκή των μαθητών (Cao, Zhang, Chan &amp; Kang 2021, Kalogeropoulos, Roche 2021). Ακόμα, θα μελετήσουμε τον τρόπο που οι νόρμες διαφοροποιούνται στην εξ αποστάσεως εκπαίδευση. Οι εκπαιδευτικοί έχουν κεντρικό ρόλο στην καθιέρωση των νορμών (Bishop, 1985, Cobb , 2001), και η ικανότητα των εκπαιδευτικών να κατανοούν τη σημασία και τις επιπτώσεις των νορμών στη διδασκαλία και τη μάθηση είναι το πρώτο βήμα για τον καθορισμό τους στην τάξη. Η μελέτη θα πραγματοποιηθεί στηριζόμενη στην ανάλυση των συναντήσεων των εκπαιδευτικών, οι οποίες έχουν καταγραφεί και έχουν απομαγνητοφωνηθεί.The topic of this study is the cooperation among a group of educators. In particular, the study is based on the analysis of their collaboration that started in December of 2018 through the program educate. The educators studied the mathematical and sociomathematical norms focusing on the differentiating factors and challenges, while they also participated in ΕΝΕΔΙΜ conference. Two years later, in February 2021, the group continued the collaboration. This time, due to the new circumstances brought by COVID-19, they focused on norms related to distance learning. The present study aims to present and analyse the way in which the educators perceive the new norms and how they influence the degree of students’ involvement. Moreover, we will study the transformation of the old norms under these unprecedented conditions. Teachers have a central role in the establishment of these norms and their ability to understand their importance and consequences are the first step towards forming them.     Investigating the Climate-Related Risk of Forest Fires for Mediterranean Islands’ Blue Economy

    The Mediterranean islands’ blue economy and, more specifically, the tourism sector, largely regulate Europe’s gross product. Climate change threatens the ecological, societal, and economic sustainability of the islands in many ways, with increasing wildfires making up one of the most critical components of the climate change impacts on tourism. Here, we aim to identify and assess forest fire vulnerability and risk due to climate change for seven Mediterranean islands through the application of the “impact chain” conceptual framework. The backbone of this approach requires the integration of quantitative and qualitative data according to the three main risk components sensu the Intergovernmental Panel on Climate Change (IPCC), i.e., hazard, exposure, and vulnerability, with a structured participatory approach involving stakeholders and experts. Our results illustrate the islands with high potential for improvement in terms of adapting capacity and, by indicating the contribution of the different risk components, highlight the main environmental and socio-economic elements that affect the islands’ vulnerability and risk under climate change. The approach’s potentials and constraints are discussed, suggesting that the method can be handily used to point out the priorities that must be addressed by mitigation and adaptation policies and measures at the island level
