    Efficiency of Antimicrobial Electrospun Thymol-Loaded Polycaprolactone Mats in Vivo

    Due to the prevalence of antimicrobial resistant pathogens, natural products with long-term antimicrobial activities are considered as potential alternatives. In this work, polycaprolactone (PCL) electrospun fibers with mean diameters around 299 nm and loaded with 14.92 ± 1.31% w/w thymol (THY) were synthesized. The mats had appropriate elongation at break (74.4 ± 9.5%) and tensile strength (3.0 ± 0.5 MPa) to be potentially used as wound dressing materials. In vivo studies were performed using eight to ten week-old male SKH1 hairless mice. The infection progression was evaluated through a semiquantitative method and quantitative polymerase chain reaction. The analyses of post-mortem samples indicated that THY-loaded PCL fibers acted as inhibitors of Staphylococcus aureus ATCC 25923 strain growth being as efficient as chlorhexidine (CLXD). Histopathological and immunohistochemical studies showed that the PCL-THY-treated wounds were almost free of an inflammatory reaction.     Aphanius iberus (= Lebias ibera) is one of the most endangered Iberian vertebrates. It is an endemic fish and its drastic regression for the last two decades has involved the cataloguing of the species into the all national and international Red Listes. Because the area of this study is probably the area where the habitat´s species is most threatened, so the increment in conservation’s efforts on this species is absolutely necessary to secure its perpetuation. As a first step to develop a Recovery Plan for the species in the Murcian region, we have studied its distribution and assessed its conservation status of each locality in which it occurs. For that purpose, we reviewed historical references of the species and sampled all potential sites identified on 1:25000 scale maps using different sampling gears: trawls, fyke nets, minnow traps, hand nets and small bag seines. A.iberus occurred in 40 out of 266 localities visited, 39 localities (9 new records) were included in the Mar Menor coastal lagoon. We also found a new population for the species in the Chicamo Stream (30SXH749368), an isolated population outside of the distribution range known for the species in the Murcian Region. In the study area the regression of A. iberus is clear and coincides with the observed in other areas. Finally, we establish 6 Operational Population Groups (OPGs) as the management units to preserve the species in the Murcian region.Aphanius iberus (= Lebias ibera) es una de las especies de vertebrados ibéricos en mayor peligro de extinción. Su carácter endémico y la drástica regresión que ha sufrido en las dos últimas décadas ha conducido a la catalogación de la especie en todas las listas de especies amenazadas, nacionales e internacionales. El estado de conservación de la especie en el sureste de la Península Ibérica se ha visto deteriorado notablemente. En este sentido, como primer paso para la elaboración del Plan de Recuperación del Fartet en Murcia, se ha establecido la distribución de la especie y el estado de conservación preliminar de las localidades con presencia de la misma. Con esta finalidad han sido estudiadas 266 localidades, para lo que fueron visitadas la totalidad de zonas húmedas, litorales e interiores, con interés para esta especie en la Región de Murcia. Los muestreos fueron realizados con diferentes artes de pesca según las características del cuerpo de agua. A. iberus se presentó en 40 localidades, 39 dentro del área geográfica del Mar Menor y su entorno (9 nuevas citas para la especie). Resalta el hallazgo de una nueva población interior en el río Chícamo (30SXH749368). La regresión de la especie en la Región de Murcia es clara, y coincide con lo observado en otras zonas de la Península. Finalmente, han sido establecidos 6 Grupos Poblacionales Operativos (GPOs) como las unidades de manejo para la adecuada gestión de la especie en la Región de Murcia

    Synthesis and biological properties of β-turned Aβ31-35 constrained analogues

    A series of constrained pentapeptide analogues of the fragment Aβ31–35 has been prepared using solid phase synthesis protocols. The results of conformational studies and surface plasmon resonance (SPR) experiments seem to indicate that the affinity of these constrained analogues for immobilized Aβ25–35 peptide could be related to their ability to adopt a Leu34N-Ile31O β-turn-like folded conformation.This work has been supported by the Spanish Ministry of Education and Science (SAF 2006-01205) and the Comunidad de Madrid (GR/SAL/0846/2004).     Disease severity in familial cases of IBD

    Background: Phenotypic traits of familial IBD relative to sporadic cases are controversial, probably related to limited statistical power of published evidence. Aim: To know if there are phenotype differences between familial and sporadic IBD, evaluating the prospective Spanish registry (ENEIDA) with 11,983 cases. Methods: 5783 patients (48.3%) had ulcerative colitis (UC) and 6200 (51.7%) Crohn's disease (CD). Cases with one or more 1st, 2nd or 3rd degree relatives affected by UC/CD were defined as familial case. Results: In UC and CD, familial cases compared with sporadic cases had an earlier disease onset (UC: 33 years [IQR 25–44] vs 37 years [IQR 27–49]; p b 0.0001); (CD: 27 years [IQR 21–35] vs 29 years [IQR 22–40]; p b 0.0001), higher prevalence of extraintestinal immune-related manifestations (EIMs) (UC: 17.2% vs 14%; p = 0.04); (CD: 30.1% vs 23.6%; p b 0.0001). Familial CD had higher percentage of ileocolic location (42.7% vs 51.8%; p = 0.0001), penetrating behavior (21% vs 17.6%; p = 0.01) and perianal disease (32% vs 27.1%; p = 0.003). Differences are not influenced by degree of consanguinity. Conclusion: When a sufficiently powered cohort is evaluated, familial aggregation in IBD is associated to an earlier disease onset, more EIMs and more severe phenotype in CD.     Adipose-derived mesenchymal stromal cells for the treatment of patients with severe SARS-CoV-2 pneumonia requiring mechanical ventilation. A proof of concept study

    Background: Identification of effective treatments in severe cases of COVID-19 requiring mechanical ventilation represents an unmet medical need. Our aim was to determine whether the administration of adipose-tissue derived mesenchymal stromal cells (AT-MSC) is safe and potentially useful in these patients. Methods: Thirteen COVID-19 adult patients under invasive mechanical ventilation who had received previous antiviral and/or anti-inflammatory treatments (including steroids, lopinavir/ritonavir, hydroxychloroquine and/or tocilizumab, among others) were treated with allogeneic AT-MSC. Ten patients received two doses, with the second dose administered a median of 3 days (interquartile range-IQR- 1 day) after the first one. Two patients received a single dose and another patient received 3 doses. Median number of cells per dose was 0.98 × 106 (IQR 0.50 × 106) AT-MSC/kg of recipient's body weight. Potential adverse effects related to cell infusion and clinical outcome were assessed. Additional parameters analyzed included changes in imaging, analytical and inflammatory parameters. Findings: First dose of AT-MSC was administered at a median of 7 days (IQR 12 days) after mechanical ventilation. No adverse events were related to cell therapy. With a median follow-up of 16 days (IQR 9 days) after the first dose, clinical improvement was observed in nine patients (70%). Seven patients were extubated and discharged from ICU while four patients remained intubated (two with an improvement in their ventilatory and radiological parameters and two in stable condition). Two patients died (one due to massive gastrointestinal bleeding unrelated to MSC therapy). Treatment with AT-MSC was followed by a decrease in inflammatory parameters (reduction in C-reactive protein, IL-6, ferritin, LDH and d-dimer) as well as an increase in lymphocytes, particularly in those patients with clinical improvement. Interpretation: Treatment with intravenous administration of AT-MSC in 13 severe COVID-19 pneumonia under mechanical ventilation in a small case series did not induce significant adverse events and was followed by clinical and biological improvement in most subjects.     Water-saving techniques for restoring desertified lands : Some lessons from the field

Currently, one innovative solution is The Cocoon™, which has been designed as a new ecotechnology for improving seedling establishment. The Cocoon consists of a doughnut-shaped container made of recycled cardboard that provides water and shelter at least during the first year of a seedling, which is the most critical for plant establishment. To determine the effectiveness of this ecotechnology under different conditions, the Cocoon was tested on a variety of soils, climates, vegetation, and land uses. Six planting trials were performed in Spain and Greece, which covered a range from humid to arid climates. With the objective of studying its functionality, the survival of the seedlings, their vigor, and growth were monitored for 2 years. Compared with conventional planting systems, the Cocoon has effectively increased seedling survival, especially under dry growing conditions (low rainfall, soils with low water holding capacity). The Cocoon also allowed for higher growth of some species (olive trees, holm oaks, and Aleppo pines). Moreover, a positive correlation between the rainfall on the site and the biodegradation degree of the Cocoon device was observed. Overall, the Cocoon becomes more efficient in arid climates or adverse growing conditions

    El presente trabajo aborda el estudio detallado y preliminar del yacimiento arqueológico de Campo Real/Fillera (Sos del Rey Católico/Sangüesa, en el límite entre las actuales provincias de Zaragoza y Navarra) con especial atención a su etapa romana. Se procede a la revisión del material arqueológico y epigráfico procedente del lugar, se defiende la condición de enclave urbano del yacimiento y se plantea una hipótesis respecto de su identificación con las ciudades que las fuentes antiguas atribuyen a los Vascones. The following paper deals with the detailed and preliminary study of the archaeological site of Campo Real/Fillera (Sos del Rey Católico/Sangüesa, between today Zaragoza and Navarra provinces) specially focusing in its Roman period. The paper exposes a review of the archeological and epigraphical material from the site, proposes an urban condition for it and raises some hypothesis on its identification with one of the cities that ancient sources tribued to Vascones
