    The Effect of Hive Volume on Efficiency and Strength Conservation and Restoration of Food Supplies During the Wintering in Langstroth Hives

    The influence of volume of the hive on wintering, colony strength and food supply was observed during the three-year period. Standard LR hives were used for the experiment. Colonies were tested based on the number of hive bodies and the number of frames in the brood hive bodies. The colonies were divided into two groups based on the number of frames: group I had eight frames in the brood bodies, the second group included 10 frames in the brood bodies. Based on the number of hive bodies, colonies were also divided into two groups: colonies with a single hive body and colonies with two hive bodies. The inspections were carried out in the autumn in late August and early September, and in the spring in late March and early April. Colony strength was determined by the amount of brood and food supplies through the amount of honey and pollen. It was found that the number of frames in the brood bodies had almost no impact on the colony strength and the amount of food in the autumn and spring inspections. Percentage ratio in spring and autumn inspections was more favourable in ten-frame hives considering the amount of bees and pollen. The ratio for the amount of brood between the two inspections was higher in eight-frame hives. The colonies replenished bees more rapidly and foraged pollen more efficiently in ten-frame hives while the colonies in eight-frame hives replenished brood quicker. It was found that, during the three-year period, the hive volume had impact on strength and food supply of colonies. Colonies that have wintered in two hive bodies had favourable ratio of spring and autumn inspections for the amount of bees, brood and pollen, while the ratio for the amount of honey was more favourable in the colonies with a single hive body.     Ligand-Bound GeneSwitch Causes Developmental Aberrations in Drosophila that Are Alleviated by the Alternative Oxidase

    Culture of Drosophila expressing the steroid-dependent GeneSwitch transcriptional activator under the control of the ubiquitous -tubulin promoter was found to produce extensive pupal lethality, as well as a range of dysmorphic adult phenotypes, in the presence of high concentrations of the inducing drug RU486. Prominent among these was cleft thorax, seen previously in flies bearing mutant alleles of the nuclear receptor Ultraspiracle and many other mutants, as well as notched wings, leg malformations, and bristle abnormalities. Neither the -tubulin-GeneSwitch driver nor the inducing drug on their own produced any of these effects. A second GeneSwitch driver, under the control of the daughterless promoter, which gave much lower and more tissue-restricted transgene expression, exhibited only mild bristle abnormalities in the presence of high levels of RU486. Coexpression of the alternative oxidase (AOX) from Ciona intestinalis produced a substantial shift in the developmental outcome toward a wild-type phenotype, which was dependent on the AOX expression level. Neither an enzymatically inactivated variant of AOX, nor GFP, or the alternative NADH dehydrogenase Ndi1 from yeast gave any such rescue. Users of the GeneSwitch system should be aware of the potential confounding effects of its application in developmental studies.Peer reviewe

    Influence of Higenamine on Exercise Performance of Recreational Female Athletes: A Randomized Double-Blinded Placebo-Controlled Trial

    The aim of this study was to determine the ergogenic effects and the safety profile of a one-component higenamine supplement in female recreational athletes. Twelve recreational female basketball players (age 29–41 years, oxygen consumption (VO2max) > 30 ml⋅kg–1⋅min–1, with training > 5 h wk–1) were randomized either to the higenamine group, or to the placebo group for 3 weeks. In order to determine ergogenic effects and safety profile of higenamine administration, we assessed the following variables before and after 3 weeks of supplementation: anthropometric parameters, resting metabolic rate (RMR), exercise testing variables, serum free fatty acids (FFAs), blood pressure, enzyme activity, urea, lipid profile, and complete blood count. There were no differences between groups in anthropometric parameters, including basal metabolic rate (BMR), RMR and body fat [p = 0.706 (Cohen’s d 0.223), p = 0.169 (Cohen’s d 0.857), and p = 0.223 (Cohen’s d 0.750), respectively], FFAs [0.43 ± 0.03 vs. 0.54 ± 0.23, p = 0.206 (Cohen’s d 0.540)], neither significant differences in cardiopulmonary parameters after the intervention period. Furthermore, all measured outcome variables in the safety assessment were not significant, with values remaining stable during the intervention period for participants in both groups. This is the first study to document the effects and the safety profile of higenamine-based dietary supplements at a specified dose in female recreational athletes. Our data indicate that 21-day of supplementation with 75 mg higenamine would not result in improving cardiopulmonary exercise fitness and weight loss in female recreational athletes. Moreover, supplementation with 75 mg higenamine is safe and well-tolerated in younger recreational female athletes

    Enhanced insulin sensitivity associated with provision of mono and polyunsaturated fatty acids in skeletal muscle cells involves counter modulation of PP2A

    International audienceAims/Hypothesis: Reduced skeletal muscle insulin sensitivity is a feature associated with sustained exposure to excess saturated fatty acids (SFA), whereas mono and polyunsaturated fatty acids (MUFA and PUFA) not only improve insulin sensitivity but blunt SFA-induced insulin resistance. The mechanisms by which MUFAs and PUFAs institute these favourable changes remain unclear, but may involve stimulating insulin signalling by counter-modulation/repression of protein phosphatase 2A (PP2A). This study investigated the effects of oleic acid (OA; a MUFA), linoleic acid (LOA; a PUFA) and palmitate (PA; a SFA) in cultured myotubes and determined whether changes in insulin signalling can be attributed to PP2A regulation. Principal Findings: We treated cultured skeletal myotubes with unsaturated and saturated fatty acids and evaluated insulin signalling, phosphorylation and methylation status of the catalytic subunit of PP2A. Unlike PA, sustained incubation of rat or human myotubes with OA or LOA significantly enhanced Akt-and ERK1/2-directed insulin signalling. This was not due to heightened upstream IRS1 or PI3K signalling nor to changes in expression of proteins involved in proximal insulin signalling, but was associated with reduced dephosphorylation/inactivation of Akt and ERK1/2. Consistent with this, PA reduced PP2Ac demethylation and tyrosine 307 phosphorylation-events associated with PP2A activation. In contrast, OA and LOA strongly opposed these PA-induced changes in PP2Ac thus exerting a repressive effect on PP2A.Conclusions/Interpretation: Beneficial gains in insulin sensitivity and the ability of unsaturated fatty acids to oppose palmitate-induced insulin resistance in muscle cells may partly be accounted for by counter-modulation of PP2A

    Nuclear Charge Radii of Be-7,9,10 and the one-neutron halo nucleus Be-11

    Nuclear charge radii of 7,9,10,11^{7,9,10,11}Be have been determined by high-precision laser spectroscopy. On-line measurements were performed with collinear laser spectroscopy in the 2s1/22p1/22s_{1/2} \to 2p_{1/2} transition on a beam of Be+^{+} ions. Collinear and anticollinear laser beams were used simultaneously and the absolute frequency determination using a frequency comb yielded an accuracy in the isotope-shift measurements of about 1 MHz. Combination with accurate calculations of the mass-dependent isotope shifts yield nuclear charge radii. The charge radius decreases from 7^7Be to 10^{10}Be and then increases for the halo nucleus 11^{11}Be. When comparing our results with predictions of {\it ab initio} nuclear structure calculations we find good agreement. Additionally, the nuclear magnetic moment of 7^7Be was determined to be 1.3995(5)μN-1.3995(5)\mu_{\rm N} and that of 11^{11}Be from a previous β\beta-NMR measurement was confirmed.Comment: 4 pages, 2 figures calculated mass shift values have been re-evaluated with the latest mass values for the beryllium isotopes and the nuclear polarization contribution for Be-11, published by K.     Diiron centre mutations in Ciona intestinalis alternative oxidase abolish enzymatic activity and prevent rescue of cytochrome oxidase deficiency in flies

    The mitochondrial alternative oxidase, AOX, carries out the non proton-motive re-oxidation of ubiquinol by oxygen in lower eukaryotes, plants and some animals. Here we created a modified version of AOX from Ciona instestinalis, carrying mutations at conserved residues predicted to be required for chelation of the diiron prosthetic group. The modified protein was stably expressed in mammalian cells or flies, but lacked enzymatic activity and was unable to rescue the phenotypes of flies knocked down for a subunit of cytochrome oxidase. The mutated AOX transgene is thus a potentially useful tool in studies of the physiological effects of AOX expression.Peer reviewe

    IKKβ regulates essential functions of the vascular endothelium through kinase-dependent and -independent pathways

    Vascular endothelium provides a selective barrier between the blood and tissues, participates in wound healing and angiogenesis, and regulates tissue recruitment of inflammatory cells. Nuclear factor (NF)-κB transcription factors are pivotal regulators of survival and inflammation, and have been suggested as potential therapeutic targets in cancer and inflammatory diseases. Here we show that mice lacking IKKβ, the primary kinase mediating NF-κB activation, are smaller than littermates and born at less than the expected Mendelian frequency in association with hypotrophic and hypovascular placentae. IKKβ-deleted endothelium manifests increased vascular permeability and reduced migration. Surprisingly, we find that these defects result from loss of kinase-independent effects of IKKβ on activation of the serine-threonine kinase, Akt. Together, these data demonstrate essential roles for IKKβ in regulating endothelial permeability and migration, as well as an unanticipated connection between IKKβ and Akt signalling

    An acoustic emission landslide early warning system for communities in low-income and middle-income countries

Full details of this licence are available at: http://creativecommons.org/licenses/by/4.0/Early warning systems for slope instability are needed to alert users of accelerating slope deformation behaviour, enable evacuation of vulnerable people, and conduct timely repair and maintenance of critical infrastructure. Communities exposed to landslide risk in low- and middle-income countries seldom currently instrument and monitor slopes to provide a warning of instability because existing techniques are complex and prohibitively expensive. Research and field trials have demonstrated conclusively that acoustic emission (AE) monitoring can be an effective approach to detect accelerating slope movements and to subsequently communicate warnings to users. The objective of this study was to develop and assess a simple, robust, low-cost AE monitoring system to warn of incipient landslides, which can be widely deployed and operated by communities globally to help protect vulnerable people. This paper describes a novel AE measurement sensor that has been designed and developed with the cost constrained to a few hundred dollars (US). Results are presented from physical model experiments that demonstrate performance of the AE system in measuring accelerating deformation behaviour, with quantifiable relationships between AE and displacement rates. Exceedance of a pre-determined trigger level of AE can be used to communicate an alarm to users in order to alert them of a slope failure. Use of this EWS approach by communities worldwide would reduce the number of fatalities caused by landslides

    Highly Charged Ions in Rare Earth Permanent Magnet Penning Traps

    A newly constructed apparatus at the National Institute of Standards and Technology (NIST) is designed for the isolation, manipulation, and study of highly charged ions. Highly charged ions are produced in the NIST electron-beam ion trap (EBIT), extracted through a beamline that selects a single mass/charge species, then captured in a compact Penning trap. The magnetic field of the trap is generated by cylindrical NdFeB permanent magnets integrated into its electrodes. In a room-temperature prototype trap with a single NdFeB magnet, species including Ne10+ and N7+ were confined with storage times of order 1 second, showing the potential of this setup for manipulation and spectroscopy of highly charged ions in a controlled environment. Ion capture has since been demonstrated with similar storage times in a more-elaborate Penning trap that integrates two coaxial NdFeB magnets for improved B-field homogeneity. Ongoing experiments utilize a second-generation apparatus that incorporates this two-magnet Penning trap along with a fast time-of-flight MCP detector capable of resolving the charge-state evolution of trapped ions. Holes in the two-magnet Penning trap ring electrode allow for optical and atomic beam access. Possible applications include spectroscopic studies of one-electron ions in Rydberg states, as well as highly charged ions of interest in atomic physics, metrology, astrophysics, and plasma diagnostics.Comment: Proceedings of CDAMOP-2011, 13-16 Dec 2011, Delhi, India.
