    A review on polycyclic aromatic hydrocarbons distribution in freshwater ecosystems and their toxicity to benthic fauna

    Polycyclic aromatic hydrocarbons (PAHs) are a group of organic compounds, found ubiquitously in all environmental compartments. PAHs are considered hazardous pollutants, being of concern to both the environmental and human health. In the aquatic environment, PAHs tend to accumulate in the sediment due to their high hydrophobicity, and thus sediments can be considered their ultimate sink. Concurrently, sediments comprise important habitats for benthic species. This raises concern over the toxic effects of PAHs to benthic communities. Despite PAHs have been the subject of several reviews, their toxicity to freshwater benthic species has not been comprehensively discussed. This review aimed to provide an overview on PAHs distribution in freshwater environments and on their toxicity to benthic fauna species. The distribution of PAHs between sediments and the overlying water column, given by the sediment-water partition coefficient, revealed that PAHs concentrations were 2 to 4 orders of magnitude higher in sediments than in water. The sediment-water partition coefficient was positively correlated to PAHs hydrophobicity. Toxicity of PAHs to benthic fauna was addressed through Species Sensitivity Distributions. The derived hazardous concentration for 5% of the species (HC5) decreased as follows: NAP (376 μg L-1) > PHE > PYR > FLT > ANT (0.854 μg L-1), varying by 3 orders of magnitude. The hazardous concentrations (HC5) to benthic species were inversely correlated to the hydrophobicity of the individual PAHs. These findings are pertinent for environmental risk assessment of these compounds. This review also identified future challenges regarding the environmental toxicity of PAHs to freshwater benthic communities, namely the need for updating the PAHs priority list and the importance of comprehensively and more realistically assess the toxicity of PAHs in combination with other stressors, both chemical and climate-related.publishe

    Exploring the natural history of intrinsic capacity impairments: Longitudinal patterns in the 10/66 study

    BACKGROUND: intrinsic capacity (IC) is a construct encompassing people\u27s physical and mental abilities. There is an implicit link amongst IC domains: cognition, locomotion, nutrition, sensory and psychological. However, little is known about the integration of the domains. OBJECTIVES: to investigate patterns in the presentation and evolution of IC domain impairments in low-and-middle-income countries and if such patterns were associated with adverse outcomes. METHODS: secondary analyses of the first two waves of the 10/66 study (population-based surveys conducted in eight urban and four rural catchment areas in Cuba, Dominican Republic, Puerto Rico, Venezuela, Peru, Mexico and China). We applied latent transition analysis on IC to find latent statuses (latent clusters) of IC domain impairments. We evaluated the longitudinal association of the latent statuses with the risk of frailty, disability and mortality, and tested concurrent and predictive validity. RESULTS: amongst 14,923 participants included, the four latent statuses were: high IC (43%), low deterioration with impaired locomotion (17%), high deterioration without cognitive impairment (22%), and high deterioration with cognitive impairment (18%). A total of 61% of the participants worsened over time, 35% were stable, and 3% improved to a healthier status.Participants with deteriorated IC had a significantly higher risk of frailty, disability and dementia than people with high IC. There was strong concurrent and predictive validity. (Mortality Hazard Ratio = 4.60, 95%CI 4.16; 5.09; Harrel\u27s C = 0.73 (95%CI 0.72;0.74)). CONCLUSIONS: half of the study population had high IC at baseline, and most participants followed a worsening trend. Four qualitatively different IC statuses or statuses were characterised by low and high levels of deterioration associated with their risk of disability and frailty. Locomotion and cognition impairments showed other trends than psychological and nutrition domains across the latent statuses

    Laser-Assisted Production of Carbon-Encapsulated Pt-Co Alloy Nanoparticles for Preferential Oxidation of Carbon Monoxide

    C-encapsulated highly pure PtxCoy alloy nanoparticles have been synthesized by an innovative one-step in-situ laser pyrolysis. The obtained X-ray diffraction pattern and transmission electron microscopy images correspond to PtxCoy alloy nanoparticles with average diameters of 2.4 nm and well-established crystalline structure. The synthesized PtxCoy/C catalyst containing 1.5 wt% of PtxCoy nanoparticles can achieve complete CO conversion in the temperature range 125–175°C working at weight hourly space velocities (WHSV) of 30 L h−1g−1. This study shows the first example of bimetallic nanoalloys synthesized by laser pyrolysis and paves the way for a wide variety of potential applications and metal combinations

    Unraveling the ecotoxicity of deep eutectic solvents using the mixture toxicity theory

    The interest on deep eutectic solvents (DES) has been increasing. However, the ecotoxicological profile of DES is scarcely known. Also, despite previous studies showed that DES components dissociate in water, none assessed DES toxicity using the classical and adequate models for mixture toxicity prediction - concentration addition (CA) and independent action (IA). This study evaluates the ecotoxicological profile of DES based on [N1111]Cl, [N2222]Cl and [N3333]Cl as hydrogen bond acceptors (HBA) combined with hydrogen-bond donors (HBD) vis. ethylene glycol and 1-propanol, through the Microtox® Acute Toxicity Test. CA and IA with deviations describing synergism/antagonism, dose-ratio and dose-level effects were fitted to the toxicity data. Neither the starting materials nor DES were found hazardous to Aliivibrio fischeri, in this specific case agreeing with the claimed "green character" of DES. Among the starting materials, ethylene glycol was the least toxic, whereas [N3333]Cl was the most toxic (30 min-EC50 = 96.49 g L-1 and 0.5456 g L-1, respectively). DES toxicity followed the same trend as observed for the salts: [N1111]Cl-based DES < [N2222]Cl-based DES < [N3333]Cl-based DES. The IA model, with specific deviations, adjusted better in 5 out of 6 DES. Antagonism was observed for [N1111]Cl-based DES, and synergism for [N3333]Cl-based DES and for 1-propanol:[N2222]Cl. The application of the mixture toxicity models represents a breakthrough in the problematic of assessing the toxicity of the countless number of DES that can be created with the same starting materials, since they provide the expected toxicity of any virtual combination between HBA and HBD.publishe

    New genes of Xanthomonas citri subsp. citri involved in pathogenesis and adaptation revealed by a transposon-based mutant library

    <p>Abstract</p> <p>Background</p> <p>Citrus canker is a disease caused by the phytopathogens <it>Xanthomonas citri </it>subsp. <it>citri</it>, <it>Xanthomonas fuscans </it>subsp. <it>aurantifolli </it>and <it>Xanthomonas alfalfae </it>subsp. <it>citrumelonis</it>. The first of the three species, which causes citrus bacterial canker type A, is the most widely spread and severe, attacking all citrus species. In Brazil, this species is the most important, being found in practically all areas where citrus canker has been detected. Like most phytobacterioses, there is no efficient way to control citrus canker. Considering the importance of the disease worldwide, investigation is needed to accurately detect which genes are related to the pathogen-host adaptation process and which are associated with pathogenesis.</p> <p>Results</p> <p>Through transposon insertion mutagenesis, 10,000 mutants of <it>Xanthomonas citri </it>subsp. <it>citri </it>strain 306 (Xcc) were obtained, and 3,300 were inoculated in Rangpur lime (<it>Citrus limonia</it>) leaves. Their ability to cause citrus canker was analyzed every 3 days until 21 days after inoculation; a set of 44 mutants showed altered virulence, with 8 presenting a complete loss of causing citrus canker symptoms. Sequencing of the insertion site in all 44 mutants revealed that 35 different ORFs were hit, since some ORFs were hit in more than one mutant, with mutants for the same ORF presenting the same phenotype. An analysis of these ORFs showed that some encoded genes were previously known as related to pathogenicity in phytobacteria and, more interestingly, revealed new genes never implicated with <it>Xanthomonas </it>pathogenicity before, including hypothetical ORFs. Among the 8 mutants with no canker symptoms are the <it>hrpB4 </it>and <it>hrpX </it>genes, two genes that belong to type III secretion system (TTSS), two hypothetical ORFS and, surprisingly, the <it>htrA </it>gene, a gene reported as involved with the virulence process in animal-pathogenic bacteria but not described as involved in phytobacteria virulence. Nucleic acid hybridization using labeled cDNA probes showed that some of the mutated genes are differentially expressed when the bacterium is grown in citrus leaves. Finally, comparative genomic analysis revealed that 5 mutated ORFs are in new putative pathogenicity islands.</p> <p>Conclusion</p> <p>The identification of these new genes related with Xcc infection and virulence is a great step towards the understanding of plant-pathogen interactions and could allow the development of strategies to control citrus canker.</p

    Treatment outcomes of pulp revascularization in traumatized immature teeth using calcium hydroxide and 2% chlorhexidine gel as intracanal medication

    Objective: Pulp revascularization is an effective treatment for immature necrotic teeth. Calcium hydroxide has been used in pulp revascularization as an intracanal medication due to its antimicrobial action and the non-exhibition of crown discoloration and cytotoxicity for stem cells from the apical papilla. Our study aimed to investigate the clinical success and quantitative radiographic changes of root development in immature traumatized teeth using calcium hydroxide plus 2% chlorhexidine gel as intracanal medication. Methodology: In this retrospective study, 16 patients were treated with a standardized pulp revascularization protocol. Calcium hydroxide and 2% chlorhexidine gel were manipulated in a 1:1 (v/v) ratio and inserted into root canals with Lentulo spirals (Dentsply Maillefer, Baillaigues, Switzerland). Patients were followed up for a period from 9 to 36 months for the evaluation of clinical and radiological data. Radiographic measurements of root length, root width, apical diameter, and MTA placement from the apex were quantified using software ImageJ. Wilcoxon test and t-test were used, according to nonparametric or parametric data, respectively, for changes over time in root length, root width, and apical diameter. Results: Fifteen teeth survived during the follow-up period (93.75%) and met the criteria for clinical success. Although the changes seem to be very small in many cases, significant increases in the average root length (14.28%, p&lt;0.0001), root width (8.12%, p=0.0196), and decrease in apical diameter (48.37%, p=0.0007) were observed. MTA placement from the apex and age at the time of treatment was not significantly associated with the quantitative radiographic outcomes. Conclusions: Pulp revascularization in traumatized immature teeth treated with calcium hydroxide plus 2% chlorhexidine gel as intracanal medication had high success and survival rates, showing periodontal healing and resolution of signs and symptoms. However, concerning the continued root development, the outcomes can still be considered unpredictable

    Identification of emerging hazards in mussels by the Galician Emerging Food Safety Risks Network (RISEGAL). A first approach

    Emerging risk identification is a priority for the European Food Safety Authority (EFSA). The goal of the Galician Emerging Food Safety Risks Network (RISEGAL) is the identification of emerging risks in foods produced and commercialized in Galicia (northwest Spain) in order to propose prevention plans and mitigation strategies. In this work, RISEGAL applied a systematic approach for the identification of emerging food safety risks potentially affecting bivalve shellfish. First, a comprehensive review of scientific databases was carried out to identify hazards most quoted as emerging in bivalves in the period 2016–2018. Then, identified hazards were semiquantitatively assessed by a panel of food safety experts, who scored them accordingly with the five evaluation criteria proposed by EFSA: novelty, soundness, imminence, scale, and severity. Scores determined that perfluorinated compounds, antimicrobial resistance, Vibrio parahaemolyticus, hepatitis E virus (HEV), and antimicrobial residues are the emerging hazards that are considered most imminent and severe and that could cause safety problems of the highest scale in the bivalve value chain by the majority of the experts consulted (75%).     Questions in Science textbooks: development and validation of a checklist

    Questões são enunciadas que requerem uma resposta refletida e que desempenham diversas funções educativas, incluindo o despoletar novas aprendizagens. Neste caso, o seu uso enquadra-se na Aprendizagem Baseada na Resolução de Problemas (ABRP, do inglês, Problem-Based Learning, PBL), que coloca o aluno no centro dos processos de ensino e aprendizagem, atribuindo-lhe um papel ativo na aprendizagem de novo conhecimento. Atendendo à influência que os manuais exercem nas práticas de ensino, a questão que se coloca é a de saber em que medida as questões incluídas nos manuais escolares de Ciências podem fomentar um ensino orientado para a ABRP. Neste texto, descrever-se-á o processo de construção e validação de uma grelha de análise de questões incluídas em manuais escolares de Ciências com essa finalidade, apresentando-se não só essa grelha, mas também, a título ilustrativo, uma aplicação da mesma a um tópico de Ciências, 8º ano, abordado nas duas disciplinas desta área.Questions require a thoughtful answer and can play diverse educational roles, such as stimulating the learning of new knowledge, as is the case in Problem-Based Learning (PBL) contexts. In this context, the student is placed at the center of the teaching and learning processes and plays an active role in the learning of new knowledge. Bearing in mind the influence that textbooks exert on teaching practices, it is worthwhile questioning to which extent questions included in school science textbooks promote a PBL oriented teaching approach. This paper describes the development and validation of a checklist for analyzing the questions included in science textbooks.     Avaliação dos efeitos dos incêndios florestais na espécie bentónica Chironomus riparius

    Os incêndios florestais constituem uma fonte de contaminação difusa, sendo reconhecidos como uma ameaça à biodiversidade dos ecossistemas aquáticos. No entanto, os seus efeitos nos organismos aquáticos permanecem pouco estudados. Neste trabalho avaliou-se o efeito dos contaminantes associados aos incêndios florestais em organismos bentónicos da espécie Chironomus riparius. Para tal, foram recolhidos sedimentos em vários locais de um curso de água afetado por um incêndio: um local de referência a montante da área ardida (M), dois locais no interior da área ardida (um local afetado, A+, e outro não afetado por outras fontes evidentes de poluição, A) e um local a jusante da área ardida (J). Em laboratório, espécimes de C. riparius foram introduzidos nos sedimentos recolhidos nos locais de amostragem e acompanhados ao longo do seu ciclo de vida (28 dias). Comparativamente ao local de referência, verificou-se um atraso significativo (26%) no tempo até à primeira emergência dos adultos no local A+. Neste local observou-se também uma redução de 78% na taxa de emergência de adultos, enquanto que nos outros locais a taxa de emergência não foi significativamente diferente. Ainda assim, os locais no interior da área ardida foram os que apresentaram as taxas de emergência mais baixas. Em termos de reprodução, o número de ovos por massa de ovos foi 59% inferior no local A+ comparativamente ao local de referência. Estes resultados sugerem que os incêndios florestais podem constituir uma fonte adicional de poluição quando em combinação com outros contaminantes de origem antropogénica presentes nos ecossistemas aquáticos.publishe
