    Mitotic entry in the presence of DNA damage is a widespread property of aneuploidy in yeast

    Genetic instability is a hallmark of aneuploidy in budding and fission yeast. All aneuploid yeast strains analyzed to date harbor elevated levels of Rad52-GFP foci, a sign of DNA damage. Here we investigate how continuously elevated levels of DNA damage affect aneuploid cells. We show that Rad52-GFP foci form during S phase, consistent with the observation that DNA replication initiation and elongation are impaired in some aneuploid yeast strains. We furthermore find that although DNA damage is low in aneuploid cells, it nevertheless has dramatic consequences. Many aneuploid yeast strains adapt to DNA damage and undergo mitosis despite the presence of unrepaired DNA leading to cell death. Wild-type cells exposed to low levels of DNA damage exhibit a similar phenotype, indicating that adaptation to low levels of unrepaired DNA is a general property of the cell's response to DNA damage. Our results indicate that by causing low levels of DNA damage, whole-chromosome aneuploidies lead to DNA breaks that persist into mitosis. Such breaks provide the substrate for translocations and deletions that are a hallmark of cancer

    Uncertainty in the measurement of indoor temperature and humidity in naturally ventilated dairy buildings as influenced by measurement technique and data variability

    [EN] The microclimatic conditions in dairy buildings affect animal welfare and gaseous emissions. Measurements are highly variable due to the inhomogeneous distribution of heat and humidity sources (related to farm management) and the turbulent inflow (associated with meteorologic boundary conditions). The selection of the measurement strategy (number and position of the sensors) and the analysis methodology adds to the uncertainty of the applied measurement technique. To assess the suitability of different sensor positions, in situations where monitoring in the direct vicinity of the animals is not possible, we collected long-term data in two naturally ventilated dairy barns in Germany between March 2015 and April 2016 (horizontal and vertical profiles with 10 to 5 min temporal resolution). Uncertainties related to the measurement setup were assessed by comparing the device outputs under lab conditions after the on-farm experiments. We found out that the uncertainty in measurements of relative humidity is of particular importance when assessing heat stress risk and resulting economic losses in terms of temperature-humidity index. Measurements at a height of approximately 3 m-3.5 m turned out to be a good approximation for the microclimatic conditions in the animal occupied zone (including the air volume close to the emission active zone). However, further investigation along this cross-section is required to reduce uncertainties related to the inhomogeneous distribution of humidity. In addition, a regular sound cleaning (and if possible recalibration after few months) of the measurement devices is crucial to reduce the instrumentation uncertainty in long-term monitoring of relative humidity in dairy barns (C) 2017 The Authors.     Are transnational tobacco companies' market access strategies linked to economic development models? A case study of South Korea.

    Transnational tobacco companies (TTCs) have used varied strategies to access previously closed markets. Using TTCs' efforts to enter the South Korean market from the late 1980s as a case study, this article asks whether there are common patterns in these strategies that relate to the broader economic development models adopted by targeted countries. An analytical review of the existing literature on TTCs' efforts to access emerging markets was conducted to develop hypotheses relating TTCs' strategies to countries' economic development models. A case study of Korea was then undertaken based on analysis of internal tobacco industry documents. Findings were consistent with the hypothesis that TTCs' strategies in Korea were linked to Korea's export-oriented economic development model and its hostile attitude towards foreign investment. A fuller understanding of TTCs' strategies for expansion globally can be derived by locating them within the economic development models of specific countries or regions. Of foremost importance is the need for governments to carefully balance economic and public health policies when considering liberalisation

    Anaerobic digestion of whole-crop winter wheat silage for renewable energy production

    With biogas production expanding across Europe in response to renewable energy incentives, a wider variety of crops need to be considered as feedstock. Maize, the most commonly used crop at present, is not ideal in cooler, wetter regions, where higher energy yields per hectare might be achieved with other cereals. Winter wheat is a possible candidate because, under these conditions, it has a good biomass yield, can be ensiled, and can be used as a whole crop material. The results showed that, when harvested at the medium milk stage, the specific methane yield was 0.32 m3 CH4 kg–1 volatile solids added, equal to 73% of the measured calorific value. Using crop yield values for the north of England, a net energy yield of 146–155 GJ ha–1 year–1 could be achieved after taking into account both direct and indirect energy consumption in cultivation, processing through anaerobic digestion, and spreading digestate back to the land. The process showed some limitations, however: the relatively low density of the substrate made it difficult to mix the digester, and there was a buildup of soluble chemical oxygen demand, which represented a loss in methane potential and may also have led to biofoaming. The high nitrogen content of the wheat initially caused problems, but these could be overcome by acclimatization. A combination of these factors is likely to limit the loading that can be applied to the digester when using winter wheat as a substrat

    Cruise Report: EX-17-11 Gulf of Mexico 2017 (ROV and Mapping)

    From November 29, 2017 to December 21, 2017, the NOAA Office of Ocean Exploration and Research (OER) and partners conducted a telepresence-enabled ocean exploration expedition on NOAA Ship Okeanos Explorer to collect critical baseline data and information and to improve knowledge about unexplored and poorly understood deepwater areas of the Gulf of Mexico. The Gulf of Mexico 2017 (EX-17-11) expedition was part of a series of expeditions between 2017 and 2018 that explored deepwater areas in the Gulf of Mexico. During 23 days at sea, 17 remotely operated vehicle (ROV) dives were completed off the Western Florida Escarpment and in the central and western Gulf of Mexico. Over 93 hours of ROV bottom time were logged at depths between 300 and 2,321 meters. Over 20,000 square kilometers of seafloor were mapped. A total of 138 biological and 11 geological samples were collected. The expedition gathered over 280,000 live video views worldwide and the OER website received over 35,600 views. A core onshore science team of over 80 participants from around the world collaborated and supported real-time ocean exploration science. The data associated with this expedition have been archived and are publicly available through the NOAA Archives

    Topological signature of deterministic chaos in short nonstationary signals from an optical parametric oscillator

    Although deterministic chaos has been predicted to occur in the triply resonant optical parametric oscillator (TROPO) fifteen years ago, experimental evidence of chaotic behavior in this system has been lacking so far, in marked contrast with most nonlinear systems, where chaos has been actively tracked and found. This situation is probably linked to the high sensitivity of the TROPO to perturbations, which adversely affects stationary operation at high power. We report the experimental observation in this system of a burst of irregular behavior of duration 80 microseconds. Although the system is highly nonstationary over this time interval, a topological analysis allows us to extract a clearcut signature of deterministic chaos from a time series segment of only 9 base cycles (3 microseconds). This result suggests that nonstationarity is not necessarily an obstacle to the characterization of chaos

    KiDS-i-800: Comparing weak gravitational lensing measurements in same-sky surveys

    We present a weak gravitational lensing analysis of 815 square degree of ii-band imaging from the Kilo-Degree Survey (KiDS-ii-800). In contrast to the deep rr-band observations, which take priority during excellent seeing conditions and form the primary KiDS dataset (KiDS-rr-450), the complementary yet shallower KiDS-ii-800 spans a wide range of observing conditions. The overlapping KiDS-ii-800 and KiDS-rr-450 imaging therefore provides a unique opportunity to assess the robustness of weak lensing measurements. In our analysis, we introduce two new `null' tests. The `nulled' two-point shear correlation function uses a matched catalogue to show that the calibrated KiDS-ii-800 and KiDS-rr-450 shear measurements agree at the level of 1±41 \pm 4\%. We use five galaxy lens samples to determine a `nulled' galaxy-galaxy lensing signal from the full KiDS-ii-800 and KiDS-rr-450 surveys and find that the measurements agree to 7±57 \pm 5\% when the KiDS-ii-800 source redshift distribution is calibrated using either spectroscopic redshifts, or the 30-band photometric redshifts from the COSMOS survey.Comment: 24 pages, 20 figures.     Photometric redshifts for the Kilo-Degree Survey. Machine-learning analysis with artificial neural networks

    We present a machine-learning photometric redshift analysis of the Kilo-Degree Survey Data Release 3, using two neural-network based techniques: ANNz2 and MLPQNA. Despite limited coverage of spectroscopic training sets, these ML codes provide photo-zs of quality comparable to, if not better than, those from the BPZ code, at least up to zphot<0.9 and r<23.5. At the bright end of r<20, where very complete spectroscopic data overlapping with KiDS are available, the performance of the ML photo-zs clearly surpasses that of BPZ, currently the primary photo-z method for KiDS. Using the Galaxy And Mass Assembly (GAMA) spectroscopic survey as calibration, we furthermore study how photo-zs improve for bright sources when photometric parameters additional to magnitudes are included in the photo-z derivation, as well as when VIKING and WISE infrared bands are added. While the fiducial four-band ugri setup gives a photo-z bias ÎŽz=−2e−4\delta z=-2e-4 and scatter σz<0.022\sigma_z<0.022 at mean z = 0.23, combining magnitudes, colours, and galaxy sizes reduces the scatter by ~7% and the bias by an order of magnitude. Once the ugri and IR magnitudes are joined into 12-band photometry spanning up to 12 ÎŒ\mu, the scatter decreases by more than 10% over the fiducial case. Finally, using the 12 bands together with optical colours and linear sizes gives ÎŽz<4e−5\delta z<4e-5 and σz<0.019\sigma_z<0.019. This paper also serves as a reference for two public photo-z catalogues accompanying KiDS DR3, both obtained using the ANNz2 code. The first one, of general purpose, includes all the 39 million KiDS sources with four-band ugri measurements in DR3. The second dataset, optimized for low-redshift studies such as galaxy-galaxy lensing, is limited to r<20, and provides photo-zs of much better quality than in the full-depth case thanks to incorporating optical magnitudes, colours, and sizes in the GAMA-calibrated photo-z derivation.Comment: A&A, in press.
