The sense of sounds: brain responses to phonotactic frequency, phonological grammar and lexical meaning
Two outstanding questions in spoken-language comprehension concern (1) the interplay of phonological grammar (legal vs. illegal sound sequences), phonotactic frequency (high- vs. low-frequency sound sequences) and lexicality (words vs. other sound sequences) in a meaningful context, and (2) how the properties of phonological sequences determine their inclusion or exclusion from lexical-semantic processing. In the present study, we used a picture-sound priming paradigm to examine the ERP responses of adult listeners to grammatically illegal sound sequences, to grammatically legal sound sequences (pseudowords) with low- vs. high-frequency, and to real words that were either congruent or incongruent to the picture context. Results showed less negative N1-P2 responses for illegal sequences and low-frequency pseudowords (with differences in topography), but not high-frequency ones. Low-frequency pseudowords also showed an increased P3 component. However, just like illegal sequences, neither low- nor high-frequency pseudowords differed from congruent words in the N400. Thus, phonotactic frequency had an impact before, but not during lexical-semantic processing. Our results also suggest that phonological grammar, phonotactic frequency and lexicality may follow each other in this order during word processing
Catching the news: Processing strategies in listening to dialogs as measured by ERPs
<p>Abstract</p> <p>Background</p> <p>The online segmentation of spoken single sentences has repeatedly been associated with a particular event-related brain potential. The brain response could be attributed to the perception of major prosodic boundaries, and was termed Closure Positive Shift (CPS). However, verbal exchange between humans is mostly realized in the form of cooperative dialogs instead of loose strings of single sentences. The present study investigated whether listeners use prosodic cues for structuring larger contextually embedded utterances (i.e. dialogs) like in single sentence processing.</p> <p>Methods</p> <p>ERPs were recorded from listeners (n = 22) when presented with question-answer dialogs in German. The prosody of the answer (target sentence) either matched the context provided by a question or did not match the context question.</p> <p>Results</p> <p>CPS responses to the processing of the target sentences are elicited, first, when listeners encounter information comprising 'novelties', i.e. information not mentioned in the preceding question but facts corrected between context and target. Thereby it is irrelevant whether the actual prosody of the target sentence is in congruence with the informative status or not. Second, when listeners encounter target sentences which do not convey any novelties but only previously 'given' already known information, the structuring of the speech input is driven by prosody again. The CPS is then elicited when listeners perceive major prosodic boundaries similar as for the processing of context-free single sentences.</p> <p>Conclusion</p> <p>The study establishes a link between the on-line structuring of context-free (single sentences) and context-embedded utterances (dialogs) as measured by ERPs. Moreover, the impact of prosodic phrasing and accentuation on the perception of spoken utterances on and beyond sentence level is discussed.</p
EEG potentials associated with artificial grammar learning in the primate brain
AbstractElectroencephalography (EEG) has identified human brain potentials elicited by Artificial Grammar (AG) learning paradigms, which present participants with rule-based sequences of stimuli. Nonhuman animals are sensitive to certain AGs; therefore, evaluating which EEG Event Related Potentials (ERPs) are associated with AG learning in nonhuman animals could identify evolutionarily conserved processes. We recorded EEG potentials during an auditory AG learning experiment in two Rhesus macaques. The animals were first exposed to sequences of nonsense words generated by the AG. Then surface-based ERPs were recorded in response to sequences that were ‘consistent’ with the AG and ‘violation’ sequences containing illegal transitions. The AG violations strongly modulated an early component, potentially homologous to the Mismatch Negativity (mMMN), a P200 and a late frontal positivity (P500). The macaque P500 is similar in polarity and time of occurrence to a late EEG positivity reported in human AG learning studies but might differ in functional role
Sensory quality of turnip greens and turnip tops grown in northwestern Spain
In Galicia (northwestern Spain), Brassica rapa var. rapa L. includes turnip greens and turnip tops as vegetable products. They are characterized by a particular sulfurous aroma, pungent flavor, and a bitter taste. In this work twelve local varieties grown as turnip greens and turnip tops were evaluated to define the sensory attributes, to relate them with secondary metabolites, and to select those sensorial traits that better describe these crops. Results showed differences in the sensory profiles of B. rapa varieties. Turnip greens were significantly differed for aroma intensity, leaf color, and salty taste, while turnip tops were for color and firmness of leaves, moistness and fibrosity in mouth, sharpness, and bitter taste. Secondary metabolites as glucosinolates in turnip greens and phenolic compounds in turnip tops were highly correlated with texture and flavor. Glucosinolates especially progoitrin (in turnip greens) and gluconapin (in turnip tops) showed correlation with bitter taste and aftertaste persistence. Correlation between sensory traits showed highest values between leaf firmness and stalk firmness (0.94**), leaf firmness and fibrosity (R=0.92**), aftertaste persistence and bitterness (R=0.91**) and between bitterness and moistness (R=-0.89**).Research supported by the Xunta de Galicia (PGIDIT06RAG40302PR) and Excma. Dicer and microRNAs protect adult dopamine neurons
MicroRNAs (miRs) are important post-transcriptional regulators of gene expression implicated in neuronal development, differentiation, aging and neurodegenerative diseases, including Parkinson's disease (PD). Several miRs have been linked to PD-associated genes, apoptosis and stress response pathways, suggesting that deregulation of miRs may contribute to the development of the neurodegenerative phenotype. Here, we investigate the cell-autonomous role of miR processing RNAse Dicer in the functional maintenance of adult dopamine (DA) neurons. We demonstrate a reduction of Dicer in the ventral midbrain and altered miR expression profiles in laser-microdissected DA neurons of aged mice. Using a mouse line expressing tamoxifen-inducible CreERT2 recombinase under control of the DA transporter promoter, we show that a tissue-specific conditional ablation of Dicer in DA neurons of adult mice led to decreased levels of striatal DA and its metabolites without a reduction in neuronal body numbers in hemizygous mice (Dicer(HET)) and to progressive loss of DA neurons with severe locomotor deficits in nullizygous mice (Dicer(CKO)). Moreover, we show that pharmacological stimulation of miR biosynthesis promoted survival of cultured DA neurons and reduced their vulnerability to thapsigargin-induced endoplasmic reticulum stress. Our data demonstrate that Dicer is crucial for maintenance of adult DA neurons, whereas a stimulation of miR production can promote neuronal survival, which may have direct implications for PD treatment.Peer reviewe
The Marburgvirus-Neutralizing Human Monoclonal Antibody MR191 Targets a Conserved Site to Block Virus Receptor Binding
Since their first identification 50 years ago, marburgviruses have emerged several times, with 83%–90% lethality in the largest outbreaks. Although no vaccines or therapeutics are available for human use, the human antibody MR191 provides complete protection in non-human primates when delivered several days after inoculation of a lethal marburgvirus dose. The detailed neutralization mechanism of MR191 remains outstanding. Here we present a 3.2 Å crystal structure of MR191 complexed with a trimeric marburgvirus surface glycoprotein (GP). MR191 neutralizes by occupying the conserved receptor-binding site and competing with the host receptor Niemann-Pick C1. The structure illuminates previously disordered regions of GP including the stalk, fusion loop, CX_6CC switch, and an N-terminal region of GP2 that wraps about the outside of GP1 to anchor a marburgvirus-specific “wing” antibody epitope. Virus escape mutations mapped far outside the MR191 receptor-binding site footprint suggest a role for these other regions in the GP quaternary structure
German S3 guideline "actinic keratosis and cutaneous squamous cell carcinoma" – long version of the update 2023
Actinic keratosis (AK) are common lesions in light-skinned individuals that can potentially progress to cutaneous squamous cell carcinoma (cSCC). Both conditions may be associated with significant morbidity and constitute a major disease burden, especially among the elderly. To establish an evidence-based framework for clinical decision making, the guideline “actinic keratosis and cutaneous squamous cell carcinoma” was updated and expanded by the topics cutanepus squamous cell carcinoma in situ (Bowen’s disease) and actinic cheilitis. This guideline was developed at the highest evidence level (S3) and is aimed at dermatologists, general practitioners, ear nose and throat specialists, surgeons, oncologists, radiologists and radiation oncologists in hospitals and office-based settings, as well as other medical specialties, policy makers and insurance funds involved in the diagnosis and treatment of patients with AK and cSCC

Hayes/Lahiri 1991). Gut untersucht ist die Beziehung zwischen der Unterteilung von Sätzen in prosodische Phrasen und der Fokusstruktur (vgl. Nespor/Vogel 1986). Dies trifft ebenso auf die Analyse der Akzentpositionen in Verbindung mit der Informationsstruktur zu (Féry 1993). Bezüglich der Annahmen zur prosodischen Phrasierung und ihrem Zusammenhang mit der Akzentplazierung läßt sich ein solcher minimaler Konsens nur schwer feststellen. Übereinstimmung besteht lediglich darin, daß durch Grenzsignale (Pausen, Glottisverschlußinsertion, Grenztone) die prosodische Phrasierung manifestiert wird. Ich möchte hier zeigen, daß zwischen der Akzentplazierung und der prosodischen Phrasierung ebenfalls ein Zusammenhang besteht, und Möglichkeiten erörtern, diese Verbindung sprachübergreifend und experimentell nachzuweisen.
