Application of artificial intelligence for Euler solutions clustering
International audienceResults of Euler deconvolution strongly depend on the selection of viable solutions. Synthetic calculations using multiple causative sources show that Euler solutions cluster in the vicinity of causative bodies even when they do not group densely about the perimeter of the bodies. We have developed a clustering technique to serve as a tool for selecting appropriate solutions. The clustering technique uses a methodology based on artificial intelligence, and it was originally designed to classify large data sets. It is based on a geometrical approach to study object concentration in a finite metric space of any dimension. The method uses a formal definition of cluster and includes free parameters that search for clusters of given properties. Tests on synthetic and real data showed that the clustering technique successfully outlines causative bodies more accurately than other methods used to discriminate Euler solutions. In complex field cases, such as the magnetic field in the Gulf of Saint Malo region (Brittany, France), the method provides dense clusters, which more clearly outline possible causative sources. In particular, it allows one to trace offshore the main inland tectonic structures and to study their interrelationships in the Gulf of Saint Malo. The clusters provide solutions associated with particular bodies, or parts of bodies, allowing the analysis of different clusters of Euler solutions separately. This may allow computation of average parameters for individual causative bodies. Those measurements of the anomalous field that yield clusters also form dense clusters themselves. Application of this clustering technique thus outlines areas where the influence of different causative sources is more prominent. This allows one to focus on these areas for more detailed study, using different window sizes, structural indices, etc
Experimental study of the role of physicochemical surface processing on the IN ability of mineral dust particles
During the measurement campaign FROST 2 (FReezing Of duST 2), the Leipzig Aerosol Cloud Interaction Simulator (LACIS) was used to investigate the influence of various surface modifications on the ice nucleating ability of Arizona Test Dust (ATD) particles in the immersion freezing mode. The dust particles were exposed to sulfuric acid vapor, to water vapor with and without the addition of ammonia gas, and heat using a thermodenuder operating at 250 °C. Size selected, quasi monodisperse particles with a mobility diameter of 300 nm were fed into LACIS and droplets grew on these particles such that each droplet contained a single particle. Temperature dependent frozen fractions of these droplets were determined in a temperature range between −40 °C ≤T≤−28 °C. The pure ATD particles nucleated ice over a broad temperature range with their freezing behavior being separated into two freezing branches characterized through different slopes in the frozen fraction vs. temperature curves. Coating the ATD particles with sulfuric acid resulted in the particles' IN potential significantly decreasing in the first freezing branch (T>−35 °C) and a slight increase in the second branch (T≤−35 °C). The addition of water vapor after the sulfuric acid coating caused the disappearance of the first freezing branch and a strong reduction of the IN ability in the second freezing branch. The presence of ammonia gas during water vapor exposure had a negligible effect on the particles' IN ability compared to the effect of water vapor. Heating in the thermodenuder led to a decreased IN ability of the sulfuric acid coated particles for both branches but the additional heat did not or only slightly change the IN ability of the pure ATD and the water vapor exposed sulfuric acid coated particles. In other words, the combination of both sulfuric acid and water vapor being present is a main cause for the ice active surface features of the ATD particles being destroyed. A possible explanation could be the chemical transformation of ice active metal silicates to metal sulfates. The strongly enhanced reaction between sulfuric acid and dust in the presence of water vapor and the resulting significant reductions in IN potential are of importance for atmospheric ice cloud formation. Our findings suggest that the IN concentration can decrease by up to one order of magnitude for the conditions investigated

Observations of radio sources near the Sun
Geodetic Very Long Baseline Interferometry (VLBI) data are capable of
measuring the light deflection caused by the gravitational field of the Sun and
large planets with high accuracy. The parameter of the parametrized
Post-Newtonian (PPN) formalism estimated using observations of reference radio
sources near the Sun should be equal to unity in the general relativity. We
have run several VLBI experiments tracking reference radio sources from 1 to 3
degrees from the Sun. The best formal accuracy of the parameter
achieved in the single-session mode is less than 0.01 percent, or better than
the formal accuracy obtained with a global solution included all available
observations at arbitrary elongation from the Sun. We are planning more
experiments starting from 2020 using better observing conditions near the

Transcellular migration of leukocytes is mediated by the endothelial lateral border recycling compartment
Leukocyte migration across endothelial cell borders (paracellular) and through endothelial cells (transcellular) appear to be distinct processes. During paracellular migration, membrane from a parajunctional reticulum of interconnected vesicles, the endothelial lateral border recycling compartment (LBRC), moves to surround the leukocyte in a kinesin-mediated, microtubule-dependent manner. We show that transcellular migration likewise requires targeted trafficking of LBRC membrane. We show that in addition to platelet/endothelial cell adhesion molecule (PECAM; CD31), CD99 and junctional adhesion molecule A (JAM-A), but apparently not vascular endothelial cell–specific cadherin (cadherin 5, CD144), are components of the LBRC. During transcellular migration, LBRC membrane invests the transmigrating leukocyte. Intracellular adhesion molecule 1 (ICAM-1) on the apical endothelial surface is enriched around adherent leukocytes. Depolymerization of microtubules has no effect on ICAM-1 enrichment but blocks targeted trafficking of LBRC membrane and transcellular migration by >90%. Similar to their effects on paracellular transmigration, antibodies against PECAM or CD99, but not JAM-A, block transcellular migration. We conclude that similar molecular mechanisms promote both para- and transcellular migration
Extracellular signal-regulated kinase 1/2 activity is not required in mammalian cells during late G2 for timely entry into or exit from mitosis

However, it is unclear whether this involvement reflects a direct requirement for ERK1/2 activity during these transitions or for activating gene transcription programs at earlier stages of the cell cycle. To examine these possibilities, we followed live cells in which ERK1/2 activity was inhibited through late G2 and mitosis. We find that acute inhibition of ERK1/2 during late G2 and through mitosis does not affect the timing of the G2/M or M/A transitions in normal or transformed human cells, nor does it impede spindle assembly, inactivate the p38 stress-activated checkpoint during late G2 or the spindle assembly checkpoint during mitosis. Using CENP-F as a marker for progress through G2, we also show that sustained inhibition of ERK1/2 transiently delays the cell cycle in early/mid-G2 via a p53-dependent mechanism. Together, our data reveal that ERK1/2 activity is required in early G2 for a timely entry into mitosis but that it does not directly regulate cell cycle progression from late G2 through mitosis in normal or transformed mammalian cells.This research was supported by National Institutes of Health
