The p75 neurotrophin receptor is important in multiple physiological actions including neuronal survival and neurite outgrowth during development, and after central nervous system injury. We have discovered a novel piperazine-derived compound, EVT901, which interferes with p75 neurotrophin receptor oligomerization through direct interaction with the first cysteine-rich domain of the extracellular region. Using ligand binding assays with cysteine-rich domains-fused p75 neurotrophin receptor, we confirmed that EVT901 interferes with oligomerization of full-length p75 neurotrophin receptor in a dose-dependent manner. Here we report that EVT901 reduces binding of pro-nerve growth factor to p75 neurotrophin receptor, blocks pro-nerve growth factor induced apoptosis in cells expressing p75 neurotrophin receptor, and enhances neurite outgrowth in vitro Furthermore, we demonstrate that EVT901 abrogates p75 neurotrophin receptor signalling by other ligands, such as prion peptide and amyloid-β. To test the efficacy of EVT901 in vivo, we evaluated the outcome in two models of traumatic brain injury. We generated controlled cortical impacts in adult rats. Using unbiased stereological analysis, we found that EVT901 delivered intravenously daily for 1 week after injury, reduced lesion size, protected cortical neurons and oligodendrocytes, and had a positive effect on neurological function. After lateral fluid percussion injury in adult rats, oral treatment with EVT901 reduced neuronal death in the hippocampus and thalamus, reduced long-term cognitive deficits, and reduced the occurrence of post-traumatic seizure activity. Together, these studies provide a new reagent for altering p75 neurotrophin receptor actions after injury and suggest that EVT901 may be useful in treatment of central nervous system trauma and other neurological disorders where p75 neurotrophin receptor signalling is affected

TRAFIR: caractĂŠrisation des feux baladeurs dans les grands compartiments

Inspection of recent fire events in large compartments reveals them to have a great deal of non-uniformity, they generally burn locally and move across floor plates over a period of time. This phenomenon which generates transient heating of the structure is idealized as “travelling fire”. A first series of tests was launched to define a fire load representative of an office building according to Eurocodes. Additional tests where the fire dynamics were controlled were launched to develop an understanding of the fire exposure to steel structures. Then, a second series of large scale tests were performed in real building dimensions. These tests had no artificial control over the dynamics, which allowed a realistic characterization of the fire. The fire load was identical for all tests, only the openings were modified. CFD numerical models were developed to reproduce the experimental campaign and to launch parametric analyses. This allowed to provide information concerning the conditions which may lead (or not) to a travelling fire scenario. An analytical model for the characterization of a travelling fire was developed and implemented in a simple calculation tool. It allows to evaluate the fire location, the gas temperatures in the flames, the heat fluxes in the different parts of the compartment and the temperature in a steel member. In addition, the methodology is introduced in the FEM software "SAFIR" and "OpenSees". Ultimately, a design guide was prepared including worked examples which are detailed step by step and for which the influence of the inputs on the results is analysed

The DESI One-Percent Survey: Exploring the Halo Occupation Distribution of Luminous Red Galaxies and Quasi-Stellar Objects with AbacusSummit

We present the first comprehensive Halo Occupation Distribution (HOD) analysis of the DESI One-Percent survey Luminous Red Galaxy (LRG) and Quasi-Stellar Object (QSO) samples. We constrain the HOD of each sample and test possible HOD extensions by fitting the redshift-space galaxy 2-point correlation functions in 0.15 < r < 32 Mpc/h in a set of fiducial redshift bins. We use AbacusSummit cubic boxes at Planck 2018 cosmology as model templates and forward model galaxy clustering with the AbacusHOD package. We achieve good fits with a standard HOD model with velocity bias, and we find no evidence for galaxy assembly bias or satellite profile modulation at the current level of statistical uncertainty. For LRGs in 0.4 < z < 0.6, we infer a satellite fraction of fsat = 11+-1%, a mean halo mass of log10 Mh = 13.40+0.02-0.02, and a linear bias of blin = 1.93+0.06-0.04. For LRGs in 0.6 < z < 0.8, we find fsat = 14+-1%, log10 Mh = 13.24+0.02-0.02, and blin = 2.08+0.03-0.03. For QSOs, we infer fsat = 3+8-2%, log10 Mh = 12.65+0.09-0.04, and blin = 2.63+0.37-0.26 in redshift range 0.8 < z < 2.1. Using these fits, we generate a large suite of high-fidelity galaxy mocks. We also study the redshift-evolution of the DESI LRG sample from z = 0.4 up to z = 1.1, revealing significant and interesting trends in mean halo mass, linear bias, and satellite fraction.Comment: Submitted to MNRAS, comments welcom

Rotavirus Rearranged Genomic RNA Segments Are Preferentially Packaged into Viruses Despite Not Conferring Selective Growth Advantage to Viruses

The rotavirus (RV) genome consists of 11 double-stranded RNA segments. Sometimes, partial sequence duplication of an RNA segment leads to a rearranged RNA segment. To specify the impact of rearrangement, the replication efficiencies of human RV with rearranged segments 7, 11 or both were compared to these of the homologous human wild-type RV (wt-RV) and of the bovine wt-RV strain RF. As judged by viral growth curves, rotaviruses with a rearranged genome (r-RV) had no selective growth advantage over the homologous wt-RV. In contrast, r-RV were selected over wt-RV during competitive experiments (i.e mixed infections between r-RV and wt-RV followed by serial passages in cell culture). Moreover, when competitive experiments were performed between a human r-RV and the bovine wt-RV strain RF, which had a clear growth advantage, rearranged segments 7, 11 or both always segregated in viral progenies even when performing mixed infections at an MOI ratio of 1 r-RV to 100 wt-RV. Lastly, bovine reassortant viruses that had inherited a rearranged segment 7 from human r-RV were generated. Although substitution of wt by rearranged segment 7 did not result in any growth advantage, the rearranged segment was selected in the viral progenies resulting from mixed infections by bovine reassortant r-RV and wt-RV, even for an MOI ratio of 1 r-RV to 107 wt-RV. Lack of selective growth advantage of r-RV over wt-RV in cell culture suggests a mechanism of preferential packaging of the rearranged segments over their standard counterparts in the viral progeny

Study of J/ψ azimuthal anisotropy at forward rapidity in Pb-Pb collisions at √sNN=5.02 TeV

The second (v2) and third (v3) flow harmonic coefficients of J/ψ mesons are measured at forward rapidity (2.5 < y < 4.0) in Pb-Pb collisions at √ sNN = 5.02 TeV with the ALICE detector at the LHC. Results are obtained with the scalar product method and reported as a function of transverse momentum, pT, for various collision centralities. A positive value of J/ψ v3 is observed with 3.7σ significance. The measurements, compared to those of prompt D0 mesons and charged particles at mid-rapidity, indicate an ordering with vn(J/ψ) < vn(D0 ) < vn(h±) (n = 2, 3) at low and intermediate pT up to 6 GeV/c and a convergence with v2(J/ψ) ≈ v2(D0 ) ≈ v2(h±) at high pT above 6–8 GeV/c. In semicentral collisions (5–40% and 10–50% centrality intervals) at intermediate pT between 2 and 6 GeV/c, the ratio v3/v2 of J/ψ mesons is found to be significantly lower (4.6σ) with respect to that of charged particles. In addition, the comparison to the prompt D0 -meson ratio in the same pT interval suggests an ordering similar to that of the v2 and v3 coefficients. The J/ψ v2 coefficient is further studied using the Event Shape Engineering technique. The obtained results are found to be compatible with the expected variations of the eccentricity of the initial-state geometry.publishedVersio

Measurement of isolated photon-hadron correlations in √sNN = 5.02 TeV pp and p-Pb collisions

    Get PDF
This paper presents isolated photon-hadron correlations using pp and p-Pb data collected by the ALICE detector at the LHC. For photons with |Ρ|<0.67 and 12<pT<40 GeV/c, the associated yield of charged particles in the range |Ρ|<0.80 and 0.5<pT<10 GeV/c is presented. These momenta are much lower than previous measurements at the LHC. No significant difference between pp and p-Pb is observed, with Pythia 8.2 describing both data sets within uncertainties. This measurement constrains nuclear effects on the parton fragmentation in p-Pb collisions, and provides a benchmark for future studies of Pb-Pb collisions.publishedVersio
