    The TriForC database : a comprehensive up-to-date resource of plant triterpene biosynthesis

    Triterpenes constitute a large and important class of plant natural products with diverse structures and functions. Their biological roles range from membrane structural components over plant hormones to specialized plant defence compounds. Furthermore, triterpenes have great potential for a variety of commercial applications such as vaccine adjuvants, anti-cancer drugs, food supplements and agronomic agents. Their biosynthesis is carried out through complicated, branched pathways bymultiple enzyme types that include oxidosqualene cyclases, cytochrome P450s, and UDP-glycosyltransferases. Given that the number of characterized triterpene biosynthesis enzymes has been growing fast recently, the need for a database specifically focusing on triterpene enzymology became eminent. Here, we present the TriForC database (http://bioinformatics. psb. ugent. be/triforc/), encompassing a comprehensive catalogue of triterpene biosynthesis enzymes. This highly interlinked database serves as a user-friendly access point to versatile data sets of enzyme and compound features, enabling the scanning of a complete catalogue of experimentally validated triterpene enzymes, their substrates and products, as well as the pathways they constitute in various plant species. The database can be accessed by direct browsing or through convenient search tools including keyword, BLAST, plant species and substructure options. This database will facilitate gene mining and creating genetic toolboxes for triterpene synthetic biology

    Construing Worst Experiences of the COVID-19 Pandemic in the USA: A Thematic Analysis

This is an Open Access article distributed under the terms of the Creative Commons Attribution-Non Commercial-No Derivatives License (http://creativecommons.org/licenses/by-nc-nd/4.0/.The COVID-19 pandemic has not only resulted in millions of deaths but, together with the strategies imposed to contain the spread of the disease, it has had significant psychological and social effects. This paper considers these effects in residents of the USA, the country that has reported the highest number of deaths from COVID-19. Between April and May, 2020, responses were obtained to an on-line survey, which included asking participants, recruited by snowball sampling, to describe their worst experience of the pandemic. The responses of 741 participants, primarily female and Caucasian, were subjected to a thematic content analysis which used a primarily deductive approach in which these responses were viewed in terms of transitions in construing. The transition themes identified were anxiety; threat; loss of role; sadness; contempt; and stress. Various subthemes were also identified. The study provided further evidence of the utility of a personal construct framework in conceptualizing experiences associated with illness and the risk of this. Implications of its findings are considered at both an individual and a societal level.Peer reviewedFinal Published versio

    Brouage, « Maison Champlain » : un îlot urbain moderne, DFS

    Suite à l’expertise 2005 de la maison Champlain, une fouille programmée a été effectuée à la demande du Syndicat Mixte pour l’Aménagement et l’Animation de Brouage, en tenant compte de sa volonté d’intégrer les vestiges à l’aménagement des futurs jardins. La partie ouest des jardins a été ouverte. La fouille de cette zone a révélé une forte densité de vestiges archéologiques. Tous les éléments constitutifs d’une unité d’habitation moderne s’y retrouvent : • Intérieurs dotés de cheminées, ouvertures avec seuils, sols de chaux, cloisons de séparation• Extérieurs de jardins, cours closes avec sols de galets et caniveau, rampes d’accès, murets de terrasse.Cette année, les niveaux les plus anciens n’ont pas été atteints. Les premières maisons semblent assez grandes lusieurs pièces sur rue, cours intérieures. Durant la première moitié du XVIIe siècle, cette première habitation connaît des modifications intérieures séparations de pièces en deux (salle 2), déplacement des portes… La vision de cette unité d’habitation est altérée puisqu’elle est à cheval sous la parcelle des jardins de la maison Champlain et celle plus à l’ouest.Au milieu du XVIIe siècle, cette première maison se divise en deux, forcement plus petites en surface. La prochaine fouille devrait permettre de compléter leurs plans. En deux siècles, les nivaux de circulation gagnent 1 m. à 1,5. de hauteur pour permettre de rester hors d’eau tout au long de l’année. Le caractère irrégulier de ce remblaiement génère des difficultés de circulation contournées par l’aménagement de rampes (salles 5).Des aménagements à vocation hydraulique et peut-être artisanale sont installés en fin d’occupation, courant XVIIIe siècle, alors que la ville portuaire s’envase et se vide de ses habitants au profit du nouvel arsenal de Rochefort

    Integration of differentiation signals during indirect flight muscle formation by a novel enhancer of Drosophila vestigial gene

    AbstractThe gene vestigial (vg) plays a key role in indirect flight muscle (IFM) development. We show here that vg is controlled by the Notch anti-myogenic signaling pathway in myoblasts and is regulated by a novel 822 bp enhancer during IFM differentiation. Interestingly, this muscle enhancer is activated in developing fibers and in a small number of myoblasts before the fusion of myoblasts with the developing muscle fibers. Moreover, we show that this enhancer is activated by Drosophila Myocyte enhancing factor 2 (MEF2), Scalloped (SD) and VG but repressed by Twist, demonstrating a sensitivity to differentiation in vivo. In vitro experiments reveal that SD can directly bind this enhancer and MEF2 can physically interact with both SD and TWI. Cumulatively, our data reveal the interplay between different myogenic factors responsible for the expression of an enhancer activated during muscle differentiation

    An Emergent Bilingual Child\u27s Multimodal Choices in Sociodramatic Play

    In this case study, situated in a preschool classroom within an early childhood Spanish/English dual language programme, we examine how an emergent bilingual child engages with multimodal resources to participate in sociodramatic play discourses. Guided by sociocultural and critical discourse perspectives on multimodality, we analysed ways in which Anthony, a four-year-old emergent bilingual child, engaged in meaning-making during play through verbal, visual and actional modes and in conjunction with additional subcategories in his transmodal repertoire (e.g. translanguaging, sentence types, actual versus signified use of artefacts). Our results revealed differences in the ways Anthony engaged his verbal modes (e.g. monolingual languaging versus translanguaging; varying sentence types) together with actional and visual modes to accomplish adult-centric tasks versus creatively engaging in child-centric play. His translanguaging furthered his communication in tandem with the affordances of his visual and actional resources, depending on his play purposes and collaborators. Anthony’s case illustrates how emergent bilingual children access a variety of modes to participate in literate discourses in complex and varied ways. This article concludes with a discussion on the importance of thoroughly accounting for the contexts and multimodal supports in interactive learning spaces

    Comparison of four skin preparation strategies to prevent catheter-related infection in intensive care unit (CLEAN trial): a study protocol for a randomized controlled trial

    International audienceBackgroundCatheter-related infection is the third cause of infections in intensive care units (ICU), increasing the length of stay in ICU and hospital, mortality, and costs. Skin antisepsis is one of the most prevalent preventive measures. In this respect, it would appear preferable to recommend the use of alcoholic povidone iodine or chlorhexidine rather than aqueous povidone iodine. However, the data comparing chlorhexidine to povidone-iodine, both of them in alcoholic solutions, remain limited. Moreover, the benefits of enhanced cleaning prior to disinfection of skin that is not visibly soiled have yet to be confirmed in a randomized study.MethodsA prospective multicenter, 2Ă—2 factorial, randomized-controlled, assessor-blind trial will be conducted in 11 intensive care units in six French hospitals. All adult patients aged over 18 years requiring the insertion of at least one peripheral arterial catheter and/or a non-tunneled central venous catheter and/or a hemodialysis catheter and/or an arterial pulmonary catheter will be randomly assigned to have all their catheters cared with one of four skin preparation strategies (2% chlorhexidine/70% isopropyl alcohol or 5% povidone iodine/69% ethanol with or without prior skin scrubbing). At catheter removal, catheter tips will be quantitatively cultured. Sets of aerobic and anaerobic blood cultures will be routinely obtained when a patient has fever, hypothermia, or other indications. In case of suspected catheter-related infection the patient's form will be reviewed by an independent adjudication committee. We plan to enroll 2,400 patients (4,800 catheters). The main objective is to demonstrate that use of 2% alcoholic chlorhexidine compared to 5% alcoholic povidone iodine in skin preparation lowers the rate of catheter-related infection. The second endpoint is to demonstrate that enhanced skin cleaning prior to disinfection of skin that is not visibly soiled does not reduce catheter colonization. Other outcomes include comparison of skin colonization at catheter insertion site, comparison of catheter colonization and catheter-related bacteremia taking place during implementation of the four strategies of skin preparation, and cutaneous tolerance, length of hospitalization, mortality, and costs.DiscussionThis study will help to update recommendations on the choice of an antiseptic agent to use in skin preparation prior to insertion of a vascular catheter and, by extension, of an epidural catheter and it will likewise help to update recommendations on the usefulness of skin scrubbing prior to disinfection when the skin is not visibly soiled.Trial registrationClinicaltrials.gov number NCT0162955

    A dual sgRNA approach for functional genomics in Arabidopsis thaliana

    Reverse genetics uses loss-of-function alleles to interrogate gene function. The advent of CRISPR/Cas9-based gene editing now allows the generation of knock-out alleles for any gene and entire gene families. Even in the model plant Arabidopsis thaliana, gene editing is welcomed as T-DNA insertion lines do not always generate null alleles. Here, we show efficient generation of heritable mutations in Arabidopsis using CRISPR/Cas9 with a workload similar to generating overexpression lines. We obtain for several different genes Cas9 null-segregants with bi-allelic mutations in the T2 generation. While somatic mutations were predominantly generated by the canonical non-homologous end joining (cNHEJ) pathway, we observed inherited mutations that were the result of synthesis-dependent microhomology-mediated end joining (SD-MMEJ), a repair pathway linked to polymerase theta (PolQ). We also demonstrate that our workflow is compatible with a dual sgRNA approach in which a gene is targeted by two sgRNAs simultaneously. This paired nuclease method results in more reliable loss-of-function alleles that lack a large essential part of the gene. The ease of the CRISPR/Cas9 workflow should help in the eventual generation of true null alleles of every gene in the Arabidopsis genome, which will advance both basic and applied plant research

    The transcriptional repressor complex FRS7-FRS12 regulates flowering time and growth in Arabidopsis

    Most living organisms developed systems to efficiently time environmental changes. The plant-clock acts in coordination with external signals to generate output responses determining seasonal growth and flowering time. Here, we show that two Arabidopsis thaliana transcription factors, FAR1 RELATED SEQUENCE 7 (FRS7) and FRS12, act as negative regulators of these processes. These proteins accumulate particularly in short-day conditions and interact to form a complex. Loss-of-function of FRS7 and FRS12 results in early flowering plants with overly elongated hypocotyls mainly in short days. We demonstrate by molecular analysis that FRS7 and FRS12 affect these developmental processes in part by binding to the promoters and repressing the expression of GIGANTEA and PHYTOCHROME INTERACTING FACTOR 4 as well as several of their downstream signalling targets. Our data reveal a molecular machinery that controls the photoperiodic regulation of flowering and growth and offer insight into how plants adapt to seasonal changes
