    Ryhmäilmiöt ja opettaja ryhmän toiminnan kehittäjänä liikunnassa

    Tiivistelmä. Tämä kandidaatintutkielma on systemaattinen kirjallisuuskatsaus, jossa tarkasteltiin ryhmäilmiöihin liittyviä teemoja sekä opettajan persoonan, vuorovaikutustaitojen ja pedagogisten ratkaisujen vaikutusta ryhmän toimintaan ja sen yksilöihin. Tutkimuksen tavoitteena oli selvittää, mitä asioita liikunnanopettajan tulee ottaa huomioon organisoidessaan liikuntatunteja, jotta ryhmän toiminta olisi tavoitteellista ja mielekästä. Tässä kirjallisuuskatsauksessa on käytetty sekä kotimaisia että ulkomaisia lähteitä. Kerätyn aineiston pohjalta tarkasteltiin ryhmän toiminnan kannalta keskeisimpiä teemoja ja kuinka opettaja voi kehittää ryhmän toimintaa sekä huomioida sen jäsenten tarpeet, siten että kaikki saisivat positiivisia kokemuksia liikunnasta. Jotta liikuntatunneilla saataisiin oppilaille onnistumisen kokemuksia sekä pätevyyden tuntemuksia, on opettajan syytä huomioida ryhmän toimintaan, sen välisiin suhteisiin ja vuorovaikutukseen liittyviä tekijöitä, ja että nämä tekijät ovat jatkuvassa vuorovaikutuksessa keskenään. Opettajan on myös tärkeää tiedostaa, että yksittäisellä ryhmän jäsenellä on merkittävä vaikutus ryhmään ja sen motivaatioilmastoon. Yksilöiden vaikutusten myötä ryhmä on dynaaminen kaiken aikaa muuttuva ilmiö, jonka vuoksi opettajalla tulee olla valmiuksia muokata opetustaan ja omaa toimintaansa tilanteisiin sopivilla tavoilla. Liikuntatuntien suunnittelussa ja toteuttamisessa tulee ottaa huomioon monia ryhmän ja sen yksilöiden toimintaan liittyviä asioita. Ryhmäilmiöt huomioiden ja erilaisia pedagogisia ratkaisuja käyttäen opettaja voi vaikuttaa liikuntatuntien motivaatioilmastoon, tavoitteiden mukaiseen toimintaan ja liikunnallisen elämäntavan omaksumiseen. Myös opettajan oma persoona sekä hänen vuorovaikutus- ja tunnetaidot ovat merkittäviä tekijöitä, jotka vaikuttavat ryhmän ja sen yksilöiden toimintaan liikuntatunnilla

    CHARACTERISATION AND IN VITRO AND IN VIVO EVALUATION OF SUPERCRITICAL-CO2-FOAMED B-TCP/PLCL COMPOSITES FOR BONE APPLICATIONS

    Most synthetic bone grafts are either hard and brittle ceramics or paste-like materials that differ in applicability from the gold standard autologous bone graft, which restricts their widespread use. Therefore, the aim of the study was to develop an elastic, highly porous and biodegradable beta-tricalciumphosphate/poly(L-lactide-co-epsilon-caprolactone) (beta-TCP/PLCL) composite for bone applications using supercritical CO2 foaming. Ability to support osteogenic differentiation was tested in human adipose stem cell (hASC) culture for 21 d. Biocompatibility was evaluated for 24 weeks in a rabbit femur-defect model. Foamed composites had a high ceramic content (50 wt%) and porosity (65-67 %). After 50 % compression, in an aqueous environment at 37 degrees C, tested samples returned to 95 % of their original height. Hydrolytic degradation of beta-TCP/PLCL composite, during the 24-week follow-up, was very similar to that of porous PLCL scaffold both in vitro and in vivo. Osteogenic differentiation of hASCs was demonstrated by alkaline phosphatase activity analysis, alizarin red staining, soluble collagen analysis, immunocytochemical staining and qRT-PCR. In vitro, hASCs formed a pronounced mineralised collagen matrix. A rabbit femur defect model confirmed biocompatibility of the composite. According to histological Masson-Goldner's trichrome staining and micro-computed tomography, beta-TCP/PLCL composite did not elicit infection, formation of fibrous capsule or cysts. Finally, native bone tissue at 4 weeks was already able to grow on and in the beta-TCP/PLCL composite. The elastic and highly porous beta-TCP/PLCL composite is a promising bone substitute because it is osteoconductive and easy-to-use and mould intraoperatively.Peer reviewe

    Gonadotropin-releasing hormone increased pregnancy risk in suckled beef cows not detected in estrus and subjected to a split-time artificial insemination program

Journal of Animal Science, 94(9), 3722-3728. doi:10.2527/jas2016-0582We hypothesized that GnRH would increase pregnancy risk (PR) in a split-time AI program for cows in which estrus was not detected. A total of 1,236 suckled beef cows at 12 locations in 3 states (Colorado, Kansas, and North Dakota) were enrolled. Before applying the fixed-time AI program, BCS was assessed. Cows were treated on d -7 with a progesterone insert concurrent with 100 mu g GnRH and on d 0 with 25 mg PGF(2 alpha) plus removal of the insert. Estrus-detection patches were affixed to cows at insert removal. Estrus was defined to have occurred when an estrus-detection patch was >50% colored (activated). Cows in estrus by 65 h (n = 758; 61.3% of all cows) were randomly allocated to 2 treatments: 1) 100 mu g GnRH and early + GnRH (E+G; n = 373) or 2) AI only at 65 h (early -no GnRH [E-G]; n = 385). The remaining cows were randomly allocated to 2 treatments: 1) 5(L+G; n = 252) or 2) AI only at 84 h (late no GnRH [L-G]; n = 226). Pregnancy was determined 35 d after AI via transrectal ultrasound. Pregnancy risk did not differ (P = 0.68) between E+G and E-G cows (61.9 vs. 60.4%, respectively). Conversely, for cows inseminated at 84 h, PR was greater (P = 0.01) in cows that received GnRH (L+G) compared with their herd mates not receiving GnRH (L-G; 41.7 vs. 30.8%, respectively). Of those cows not detected in estrus by 65 h, 42.1% were detected by 84 h, for a total expression of estrus by all cows of 77.6%. Administration of GnRH increased (P < 0.01) PR in cows not detected in estrus by 84 h (+ GnRH = 33.4% [n = 146] vs. no GnRH = 15.0% [n = 128]) but had no effect in cows expressing estrus by 84 h (+ GnRH = 65.3% [n = 103] vs. no GnRH = 61.7% [n = 97]). Neither estrus expression by 65 or 84 h nor PR was influenced by BCS, parity, or days postpartum at AI. Cows had greater PR when they had been detected in estrus before AI, and PR was improved by administration of GnRH at 65 h after insert removal in cows that were not detected in estrus and inseminated at 84 h

    Personal identity (de)formation among lifestyle travellers: A double-edged sword?

    This article explores the personal identity work of lifestyle travellers – individuals for whom extended leisure travel is a preferred lifestyle that they return to repeatedly. Qualitative findings from in-depth semi-structured interviews with lifestyle travellers in northern India and southern Thailand are interpreted in light of theories on identity formation in late modernity that position identity as problematic. It is suggested that extended leisure travel can provide exposure to varied cultural praxes that may contribute to a sense of social saturation. Whilst a minority of the respondents embraced a saturation of personal identity in the subjective formation of a cosmopolitan cultural identity, several of the respondents were paradoxically left with more identity questions than answers as the result of their travels

    Exacerbation of acute kidney injury by bone marrow stromal cells from rats with persistent renin-angiotensin system activation

    Abstract Hypertension and persistent activation of the renin-angiotensin system (RAS) are predisposing factors for the development of acute kidney injury (AKI). Although bone-marrow-derived stromal cells (BMSCs) have shown therapeutic promise in treatment of AKI, the impact of pathological RAS on BMSC functionality has remained unresolved. RAS and its local components in the bone marrow are involved in several key steps of cell maturation processes. This may also render the BMSC population vulnerable to alterations even in the early phases of RAS pathology. We isolated transgenic BMSCs (TG-BMSCs) from young end-organ-disease-free rats with increased RAS activation [human angiotensinogen/renin double transgenic rats (dTGRs)] that eventually develop hypertension and die of end-organ damage and kidney failure at 8 weeks of age. Control cells (SD-BMSCs) were isolated from wild-type Sprague-Dawley rats. Cell phenotype, mitochondrial reactive oxygen species (ROS) production and respiration were assessed, and gene expression profiling was carried out using microarrays. Cells&apos; therapeutic efficacy was evaluated in a rat model of acute ischaemia/reperfusion-induced AKI. Serum urea and creatinine were measured at 24 h and 48 h. Acute tubular damage was scored and immunohistochemistry was used for evaluation for markers of inflammation [monocyte chemoattractant protein (MCP-1), ED-1], and kidney injury [kidney injury molecule-1 (KIM-1), neutrophil gelatinase-associated lipocalin (NGAL)]. TG-BMSCs showed distinct mitochondrial morphology, decreased cell respiration and increased production of ROS. Gene expression profiling revealed a pronounced pro-inflammatory phenotype. In contrast with the therapeutic effect of SD-BMSCs, administration of TG-BMSCs in the AKI model resulted in exacerbation of kidney injury and high mortality. Our results demonstrate that early persistent RAS activation can dramatically compromise therapeutic potential of BMSCs by causing a shift into a pro-inflammatory phenotype with mitochondrial dysfunction

    Updating known distribution models for forecasting climate change impact on endangered species

    To plan endangered species conservation and to design adequate management programmes, it is necessary to predict their distributional response to climate change, especially under the current situation of rapid change. However, these predictions are customarily done by relating de novo the distribution of the species with climatic conditions with no regard of previously available knowledge about the factors affecting the species distribution. We propose to take advantage of known species distribution models, but proceeding to update them with the variables yielded by climatic models before projecting them to the future. To exemplify our proposal, the availability of suitable habitat across Spain for the endangered Bonelli’s Eagle (Aquila fasciata) was modelled by updating a pre-existing model based on current climate and topography to a combination of different general circulation models and Special Report on Emissions Scenarios. Our results suggested that the main threat for this endangered species would not be climate change, since all forecasting models show that its distribution will be maintained and increased in mainland Spain for all the XXI century. We remark on the importance of linking conservation biology with distribution modelling by updating existing models, frequently available for endangered species, considering all the known factors conditioning the species’ distribution, instead of building new models that are based on climate change variables only.Ministerio de Ciencia e Innovación and FEDER (project CGL2009-11316/BOS

    High-Resolution Functional Mapping of the Venezuelan Equine Encephalitis Virus Genome by Insertional Mutagenesis and Massively Parallel Sequencing

    We have developed a high-resolution genomic mapping technique that combines transposon-mediated insertional mutagenesis with either capillary electrophoresis or massively parallel sequencing to identify functionally important regions of the Venezuelan equine encephalitis virus (VEEV) genome. We initially used a capillary electrophoresis method to gain insight into the role of the VEEV nonstructural protein 3 (nsP3) in viral replication. We identified several regions in nsP3 that are intolerant to small (15 bp) insertions, and thus are presumably functionally important. We also identified nine separate regions in nsP3 that will tolerate small insertions at low temperatures (30°C), but not at higher temperatures (37°C, and 40°C). Because we found this method to be extremely effective at identifying temperature sensitive (ts) mutations, but limited by capillary electrophoresis capacity, we replaced the capillary electrophoresis with massively parallel sequencing and used the improved method to generate a functional map of the entire VEEV genome. We identified several hundred potential ts mutations throughout the genome and we validated several of the mutations in nsP2, nsP3, E3, E2, E1 and capsid using single-cycle growth curve experiments with virus generated through reverse genetics. We further demonstrated that two of the nsP3 ts mutants were attenuated for virulence in mice but could elicit protective immunity against challenge with wild-type VEEV. The recombinant ts mutants will be valuable tools for further studies of VEEV replication and virulence. Moreover, the method that we developed is applicable for generating such tools for any virus with a robust reverse genetics system

    Psychosocial stress at work and perceived quality of care among clinicians in surgery

    Abstract Background Little is known about the association between job stress and job performance among surgeons, although physicians' well-being could be regarded as an important quality indicator. This paper examines associations between psychosocial job stress and perceived health care quality among German clinicians in surgery. Methods Survey data of 1,311 surgeons from 489 hospitals were analysed. Psychosocial stress at work was measured by the effort-reward imbalance model (ERI) and the demand-control model (job strain). The quality of health care was evaluated by physicians' self-assessed performance, service quality and error frequency. Data were collected in a nationwide standardised mail survey. 53% of the contacted hospitals sent back the questionnaire; the response rate of the clinicians in the participating hospitals was about 65%. To estimate the association between job stress and quality of care multiple logistic regression analyses were conducted. Results Clinicians exposed to job stress have an increased risk of reporting suboptimal quality of care. Magnitude of the association varies depending on the respective job stress model and the indicator of health care quality used. Odds ratios, adjusted for gender, occupational position and job experience vary between 1.04 (CI 0.70-1.57) and 3.21 (CI 2.23-4.61). Conclusion Findings indicate that theoretical models of psychosocial stress at work can enrich the analysis of effects of working conditions on health care quality. Moreover, results suggest interventions for job related health promotion measures to improve the clinicians' working conditions, their quality of care and their patients' health.</p
