    Stochastic signalling rewires the interaction map of a multiple feedback network during yeast evolution

    During evolution, genetic networks are rewired through strengthening or weakening their interactions to develop new regulatory schemes. In the galactose network, the GAL1/GAL3 paralogues and the GAL2 gene enhance their own expression mediated by the Gal4p transcriptional activator. The wiring strength in these feedback loops is set by the number of Gal4p binding sites. Here we show using synthetic circuits that multiplying the binding sites increases the expression of a gene under the direct control of an activator, but this enhancement is not fed back in the circuit. The feedback loops are rather activated by genes that have frequent stochastic bursts and fast RNA decay rates. In this way, rapid adaptation to galactose can be triggered even by weakly expressed genes. Our results indicate that nonlinear stochastic transcriptional responses enable feedback loops to function autonomously, or contrary to what is dictated by the strength of interactions enclosing the circuit

    Bistability, Probability Transition Rate and First-Passage Time in an Autoactivating Positive-Feedback Loop

    A hallmark of positive-feedback regulation is bistability, which gives rise to distinct cellular states with high and low expression levels, and that stochasticity in gene expression can cause random transitions between two states, yielding bimodal population distribution (Kaern et al., 2005, Nat Rev Genet 6: 451-464). In this paper, the probability transition rate and first-passage time in an autoactivating positive-feedback loop with bistability are investigated, where the gene expression is assumed to be disturbed by both additive and multiplicative external noises, the bimodality in the stochastic gene expression is due to the bistability, and the bistability determines that the potential of the Fokker-Planck equation has two potential wells. Our main goal is to illustrate how the probability transition rate and first-passage time are affected by the maximum transcriptional rate, the intensities of additive and multiplicative noises, and the correlation of additive and multiplicative noises. Our main results show that (i) the increase of the maximum transcription rate will be useful for maintaining a high gene expression level; (ii) the probability transition rate from one potential well to the other one will increase with the increase of the intensity of additive noise; (iii) the increase of multiplicative noise strength will increase the amount of probability in the left potential well; and (iv) positive (or negative) cross-correlation between additive and multiplicative noises will increase the amount of probability in the left (or right) potential well

    Becoming original: effects of strategy instruction

    Visual arts education focuses on creating original visual art products. A means to improve originality is enhancement of divergent thinking, indicated by fluency, flexibility and originality of ideas. In regular arts lessons, divergent thinking is mostly promoted through brainstorming. In a previous study, we found positive effects of an explicit instruction of metacognition on fluency and flexibility in terms of the generation of ideas, but not on the originality of ideas. Therefore, we redesigned the instruction with a focus on building up knowledge about creative generation strategies by adding more complex types of association, and adding generation through combination and abstraction. In the present study, we examined the effects of this intervention by comparing it with regular brainstorming instruction. In a pretest-posttest control group design, secondary school students in the comparison condition received the brainstorm lesson and students in the experimental condition received the newly developed instruction lesson. To validate the effects, we replicated this study with a second cohort. The results showed that in both cohorts the strategy instruction of 50 min had positive effects on students' fluency, flexibility and originality. This study implies that instructional support in building up knowledge about creative generation strategies may improve students' creative processes in visual arts education

    Manufacturing, characterisation and mechanical analysis of polyacrylonitrile membranes

    To investigate the effect of polyvinylpyrrolidone (PVP) addition and consequently porosity, two different sets of membranes are manufactured, since PVP is a widely used poring agent which has an impact on the mechanical properties of the membrane material. The first set (PAN 1) includes polyacrylonitrile (PAN) and the necessary solvent while the second set (PAN 2) is made of PAN and PVP. These membranes are put through several characterisation processes including tensile testing. The obtained data are used to model the static behaviour of the membranes with different geometries but similar loading and boundary conditions that represent their operating conditions. This modelling process is undertaken by using the finite element method. The main idea is to investigate how geometry affects the load-carrying capacity of the membranes. Alongside membrane modelling, their materials are modelled with representative elements with hexagonal and rectangular pore arrays (RE) to understand the impact of porosity on the mechanical properties. Exploring the results, the best geometry is found as the elliptic membrane with the aspect ratio 4 and the better RE as the hexagonal array which can predict the elastic properties with an approximate error of 12%
