Оценка коллатеральной активности CRISPR/Cas13a-рибонуклеазы на биоанализаторе Agilent 2100
The paper describes an original technique for detecting the collateral activity of CRISPR/Cas 13a ribonuclease based on the assessment of ribosomal RNA degradation. The Agilent 2100 bioanalyzer is used as an analyzing device. This approach is an alternative to existing detection methods and has a number of advantages over them in the case when a quantitative assessment of activity is not required. On the example of the test sample, the optimal concentrations and ratios of the components of the reaction mixture, which are necessary to obtain the most indicative result, were determined. The proposed technique can be used for qualitative assessment of the activity of recombinant ribonuclease Cas13a preparations obtained under different conditions of heterologous protein expression and purification, as well as for testing guide RNAs.В работе описывается оригинальная методика детекции коллатеральной активности CRISPR/Cas13a-рибонуклеазы, основанная на оценке деградации рибосомальной РНК. В качестве анализирующего прибора используется биоанализатор Agilent 2100. Данный подход является альтернативой существующим методам детекции и имеет ряд преимуществ по сравнению с ними в том случае, когда не требуется количественная оценка активности. На примере тестового образца определены оптимальные концентрации и соотношения компонентов реакционной смеси, необходимые для получения наиболее показательного результата. Предлагаемая методика может быть использована для качественной оценки активности препаратов рекомбинантной рибонуклеазы Cas13а, полученных при разных условиях гетерологической экспрессии белка и его очистки, а также для тестирования направляющих РНК
Mannose-specific intercellular aggregation of rat thymocytes and Escherichia coli cells triggered by temperature
D-mannose was shown to induce disaggregation of rat thymocytes and E. coli AB1157 cells at any stage of the process. Mannose-specific aggregation of thymocytes and E. coli was inhibited by heating as opposed to concanavalin A-induced thymocyte aggregation. The process was completely reversible at the rapid temperature changes, indicating its membrane not intracellular mechanism. Taken together, the results demonstrate that mannose-specific intercellular contacts can be affected differently by temperature.Встановлено, що D-маноза викликає дезагрегацію тимоцітів щурів i бактеріальних клітин Е. coli 1157 на будь-якій стадіі процесу міжклітинної агрегації. Манноспецифічна агрегація тимоцитів i E. coli пригнічувалася із підвищенням температури на противагу прискоренню агрегації тимоцитів, викликаної конканаваліном А. Процес міжклітинної агрегації тимоцитів i бактеріальних клітин був повністю зворотним за різкої зміни температури, внаслідок чого нагрівання суспензії до 50 °С призводило до дезагрегації клітин, а подальше зниження температури до 16 °С – до запуску процесу агрегації. Отримані результати свідчать про те, що манозоспецифічні міжклітинні контакти тимоцитів щурів за участю конканаваліну A i бактеріальних клітин виявляють різну чутливість до дії температури.Установлено, что D-манноза вызывает дезагрегацию тимоцитив щуривi бактериальных клеток Е. coli 1157 на любой стадии процесса межклеточной агрегации . Манноспецифична агрегация тимоцитов и E. coli подавлялась с повышением температуры в противоположность ускорению агрегации тимоцитов, вызванной конканавалин А. Процесс межклеточной агрегации тимоцитов i бактериальных клеток был полностью обратным резкой смены температуры , в результате чего нагрев суспензии до 50 °С приводило к дезагрегации клеток , а снижение температуры до 16 °С – до запуска процесса агрегации . Полученные результаты свидетельствуют о том , что манозоспецифичные межклеточные контакты тимоцитов крыс с участием конканавалин A и бактериальных клеток обнаруживают разные чувствительность к воздействию температуры
Anti-corrosion ceramic coatings on the surface of Nd-Fe-B repelling magnets
The results of vacuum-arc deposition of thin ZrO₂ coatings to protect the surface of Nd-Fe-B permanent magnets used as repelling devices in orthodontics are presented. The structure, phase composition and mechanical properties of zirconium dioxide films have been investigated by means of SEM, XRD, EDX, XRF, and nanoindentation method. It was revealed the formation of polycrystalline ZrO₂ films of monoclinic modification with average grain size 25 nm. The influence of the ZrO₂ coating in terms of its barrier properties for corrosion in quasi-physiological 0.9 NaCl solution has been studied. Electrochemical measurements indicated good barrier properties of the coating on specimens in the physiological solution environment.Представленo результати вакуумно-дугового осадження тонких покриттів ZrO₂ для захисту поверхонь постійних магнітів Nd-Fe-B, що використовуються як стягувальні пристрої в ортодонтії. Структура, фазовий склад та механічні властивості плівки діоксиду цирконію були досліджені засобами SEM, XRD, EDX, XRF та наноіндентування. Було виявлено утворення полікристалічних плівок ZrO₂ моноклінної модифікації з середнім розміром зерна 25 нм. Вивчено вплив покриття ZrO₂ як бар’єру для захисту магнітів від корозії в квазіфізіологічному розчині 0,9 NaCl. Електрохімічні виміри встановили хороші бар'єрні властивості покриття на зразках у середовищі фізіологічного розчину.Представлены результаты вакуумно-дугового осаждения тонких покрытий ZrO₂ для защиты поверхности постоянных магнитов Nd-Fe-B, используемых в качестве стягивающих устройств в ортодонтии. Структура, фазовый состав и механические свойства пленок двуокиси циркония были исследованы методами SEM, XRD, EDX, XRF, наноиндентирования. Было обнаружено образование поликристаллических пленок ZrO₂ моноклинной модификации со средним размером зерна 25 нм. Изучено влияние покрытия ZrO₂ в качестве барьера для защиты магнитов от коррозии в квазифизиологическом растворе 0,9 NaCl. Электрохимические измерения показали хорошие барьерные свойства покрытия на образцах в среде физиологического раствора
Longitudinal double-spin asymmetry and cross section for inclusive neutral pion production at midrapidity in polarized proton collisions at sqrt(s) = 200 GeV
We report a measurement of the longitudinal double-spin asymmetry A_LL and
the differential cross section for inclusive Pi0 production at midrapidity in
polarized proton collisions at sqrt(s) = 200 GeV. The cross section was
measured over a transverse momentum range of 1 < p_T < 17 GeV/c and found to be
in good agreement with a next-to-leading order perturbative QCD calculation.
The longitudinal double-spin asymmetry was measured in the range of 3.7 < p_T <
11 GeV/c and excludes a maximal positive gluon polarization in the proton. The
mean transverse momentum fraction of Pi0's in their parent jets was found to be

Longitudinal scaling property of the charge balance function in Au + Au collisions at 200 GeV
We present measurements of the charge balance function, from the charged
particles, for diverse pseudorapidity and transverse momentum ranges in Au + Au
collisions at 200 GeV using the STAR detector at RHIC. We observe that the
balance function is boost-invariant within the pseudorapidity coverage [-1.3,
1.3]. The balance function properly scaled by the width of the observed
pseudorapidity window does not depend on the position or size of the
pseudorapidity window. This scaling property also holds for particles in
different transverse momentum ranges. In addition, we find that the width of
the balance function decreases monotonically with increasing transverse
momentum for all centrality classes.Comment: 6 pages, 3 figure
Measurement of the Bottom contribution to non-photonic electron production in collisions at =200 GeV
The contribution of meson decays to non-photonic electrons, which are
mainly produced by the semi-leptonic decays of heavy flavor mesons, in
collisions at 200 GeV has been measured using azimuthal
correlations between non-photonic electrons and hadrons. The extracted
decay contribution is approximately 50% at a transverse momentum of GeV/. These measurements constrain the nuclear modification factor for
electrons from and meson decays. The result indicates that meson

Azimuthal anisotropy and correlations in p+p, d+Au and Au+Au collisions at 200 GeV
We present the first measurement of directed flow () at RHIC. is
found to be consistent with zero at pseudorapidities from -1.2 to 1.2,
then rises to the level of a couple of percent over the range . The latter observation is similar to data from NA49 if the SPS rapidities
are shifted by the difference in beam rapidity between RHIC and SPS.
Back-to-back jets emitted out-of-plane are found to be suppressed more if
compared to those emitted in-plane, which is consistent with {\it jet
quenching}. Using the scalar product method, we systematically compared
azimuthal correlations from p+p, d+Au and Au+Au collisions. Flow and non-flow

Azimuthal anisotropy: the higher harmonics
We report the first observations of the fourth harmonic (v_4) in the
azimuthal distribution of particles at RHIC. The measurement was done taking
advantage of the large elliptic flow generated at RHIC. The integrated v_4 is
about a factor of 10 smaller than v_2. For the sixth (v_6) and eighth (v_8)

Partonic flow and -meson production in Au+Au collisions at = 200 GeV
We present first measurements of the -meson elliptic flow
() and high statistics distributions for different
centralities from = 200 GeV Au+Au collisions at RHIC. In
minimum bias collisions the of the meson is consistent with the
trend observed for mesons. The ratio of the yields of the to those of
the as a function of transverse momentum is consistent with a model
based on the recombination of thermal quarks up to GeV/,
but disagrees at higher momenta. The nuclear modification factor () of
follows the trend observed in the mesons rather than in
baryons, supporting baryon-meson scaling. Since -mesons are
made via coalescence of seemingly thermalized quarks in central Au+Au
collisions, the observations imply hot and dense matter with partonic

Plasma Wakefield Acceleration with a Modulated Proton Bunch
The plasma wakefield amplitudes which could be achieved via the modulation of
a long proton bunch are investigated. We find that in the limit of long bunches
compared to the plasma wavelength, the strength of the accelerating fields is
directly proportional to the number of particles in the drive bunch and
inversely proportional to the square of the transverse bunch size. The scaling
laws were tested and verified in detailed simulations using parameters of
existing proton accelerators, and large electric fields were achieved, reaching
1 GV/m for LHC bunches. Energy gains for test electrons beyond 6 TeV were found
