    AbstractWe obtain a local characterization of the convex point of continuity property for every closed, bounded and convex subset not containing sequences equivalent to the standard unit basis of ℓ1. As a consequence, we prove, in the setting of Banach spaces without ℓ1-copies, that the convex point of continuity property is determined on subspaces with a Schauder basis, which is a partial answer to a well-known problem

    Monitoring groundwater nitrate attenuation in a regional system coupling hydrogeology with multi-isotopic methods: the case of Plana de Vic (Osona, Spain)

    This paper describes an integrated application of classical hydrogeological methods and multi-isotopic methods (d15N, d18ONO3, d34S, d18OSO4, d13C) to assess the fate of groundwater nitrate in the Osona area, declared vulnerable to nitrate pollution by the Catalan Government in 1998, where nitrate is derived from intensive pig farming activities. Previous studies, involving a small area, indicated the occurrence of denitrification processes and their relationship with pyrite oxidation [Vitòria, L., Soler, A., Canals, A., Otero, N., 2008. Environmental isotopes (N, S, C, O, D) to determine natural attenuation processes in nitrate contaminated waters: example of Osona (NE Spain). Appl. Geochem. 23, 3597-3611]. For the present study, groundwater samples were collected at 60 production wells at three different periods between April 2005 and May 2006 to confirm that denitrification takes place in a larger area than that studied by Vitòria et al. [Vitòria, L., Soler, A., Canals, A., Otero, N., 2008. Environmental isotopes (N, S, C, O, D) to determine natural attenuation processes in nitrate contaminated waters: example of Osona (NE Spain). Appl. Geochem. 23, 3597-3611]. The aim of the study was to characterize the denitrification processes that control natural attenuation and to study their spatial and temporal variations. Nitrate concentration ranged from 10 to 529 mg/l, with 82% of the wells above the drinking water threshold of 50 mg NO3/l. Nitrate isotopic composition ranged from +5.3% to +35.3% for d15N and from +0.4% to +17.6% for d18ONO3 , and the samples showed a positive correlation between d15N and d18ONO3 , with a eN/eO ratio of 1.8, consistent with denitrification processes. The link between denitrification and pyrite oxidation is demonstrated by coupling chemical data with nitrate and sulfate isotopes. Furthermore, a spatial distribution of samples with significant denitrification was observed, allowing us to determine two main hydrogeological zones where natural attenuation was most effective. In several of the studied points, denitrification processes related to pyrite oxidation predominated and an estimation of the isotopic enrichment factors was performed using the temporal variations of nitrate concentration and the isotopic composition of dissolved nitrate (d15NNO3 and d18ONO3). Finally, using estimated isotopic enrichment factors, an approximation of the degree of natural attenuation of nitrate was performed on those samples showing clear denitrification, and a median value of 30% of contaminant diminution was obtained

    Estudio del apoyo metatarsal en bipedestación mediante podoscopio electrónico de alta resolución.

    Con el jin de estudiar el apoyo metatarsal en bipedestación se han realizado 780 registros en la plataforma PDS 93, correspondientes a 390 individuos, no seleccionados, sin patología de la marcha, 234 varones y 156 mujeres, con edades comprendidas entre 24 y 41 años (edad media de 29,9 años). En cada registro se ha recogido la presión ejercida por cada metatarsiano, relacionando la carga obtenida con el peso, conjiguración mecánica del pie y huella plantar: Los resultados obtenidos se han procesado estadísticamente por medio del programa SPSS. El porcentaje de las cargas medias obtenidas para cada metatarsiano han sido: 16,85, 21,25, 23,88, 20,24 y 17,78%, existiendo dqerencias sign$cativas para el apoyo de los metatarsianos centrales (p < 0,001). Todos los metatarsianos soportan carga en bipedestación, de acuerdo con las teorías actuales. Sin embargo, aunque la distribución del peso para el primer metatarsiano se aproxima al doble de los otros, la carga máxima en g/cm2 se realiza en los metatarsianos centrales

    In vivo potential of recombinant granulysin against human melanoma

    9-kDa granulysin is a protein expressed into the granules of human cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. It has been shown to exert cytolysis on microbes and tumors. We showed previously that 9-kDa granulysin exerted cell death by apoptosis in vitro on hematological tumor cell lines and also on cells from B-cell chronic lymphocytic leukemia (B-CLL) patients. In addition, we have shown the anti-tumor efficiency of granulysin as a single agent in two in vivo models of human tumor development in athymic mice, the MDA-MB-231 mammary adenocarcinoma and the NCI-H929 multiple myeloma, without signs of overt secondary effects by itself. In this work, we have tested recombinant 9-kDa granulysin in an in vivo and especially aggressive model of melanoma development, xenografted UACC62 cells in athymic mice. Recombinant granulysin was administered once UACC62-derived tumors were detectable and it substantially retarded the in vivo development of this aggressive tumor. We could also detect apoptosis induction and increased NK cell infiltration inside granulysin-treated tumor tissues. These observations are especially interesting given the possibility of treating melanoma by intra-tumor injection

Suele presentarse en animales jóvenes de pocos meses de edad. El diagnóstico se basa en la sintomatología y hallazgos radiológicos. El tratamiento es sintomático. Consiste en la administración de analgésicos y antiinflamatorios.

    Conjugation of the 9-kDa isoform of Granulysin with liposomes potentiates its cytotoxicity

    Nine kDa granulysin (GRNLY) is a human cytolytic protein secreted by cytotoxic T lymphocytes (CTL) and NK cells of the immune system whose demonstrated physiological function is the elimination of bacteria and parasites. In previous studies by our group, the anti-tumor capacity of recombinant granulysin was demonstrated, both in vitro and in vivo. In the present work, we developed lipid nanoparticles whose surfaces can bind recombinant granulysin through the formation of a complex of coordination between the histidine tail of the protein and Ni2+ provided by a chelating lipid in the liposome composition and termed them LUV-GRNLY, for granulysin-bound large unilamellar vesicles. The objective of this formulation is to increase the granulysin concentration at the site of contact with the target cell and to increase the cytotoxicity of the administered dose. The results obtained in this work indicate that recombinant granulysin binds to the surface of the liposome with high efficiency and that its cytotoxicity is significantly increased when it is in association with liposomes. In addition, it has been demonstrated that the main mechanism of death induced by both granulysin and LUV-GRNLY is apoptosis. Jurkat-shBak cells are resistant to GRNLY and also to LUV-GRNLY, showing that LUV-GRNLY uses the mitochondrial apoptotic pathway to induce cell death. On the other hand, we show that LUV-GRNLY induces the expression of the pro-apoptotic members of the Bcl-2 family Bim and especially PUMA, although it also induced the expression of anti-apoptotic Bcl-xL. In conclusion, we demonstrate that binding of GRNLY to the surfaces of liposomes clearly augments its cytotoxic potential, with cell death executed mainly by the mitochondrial apoptotic pathway
