Phosphatidylinositol-4,5-Biphosphate-Dependent Rearrangement of TRPV4 Cytosolic Tails Enables Channel Activation by Physiological Stimuli
Most transient receptor potential (TRP) channels are regulated by phosphatidylinositol-4,5-biphosphate (PIP), although the structural rearrangements occurring on PIP binding are currently far from clear. Here we report that activation of the TRP vanilloid 4 (TRPV4) channel by hypotonic and heat stimuli requires PIP binding to and rearrangement of the cytosolic tails. Neutralization of the positive charges within the sequence KRWRK, which resembles a phosphoinositide-binding site, rendered the channel unresponsive to hypotonicity and heat but responsive to 4α-phorbol 12,13-didecanoate, an agonist that binds directly to transmembrane domains. Similar channel response was obtained by depletion of PIP from the plasma membrane with translocatable phosphatases in heterologous expression systems or by activation of phospholipase C in native ciliated epithelial cells. PIP facilitated TRPV4 activation by the osmotransducing cytosolic messenger 5′-6’-epoxyeicosatrienoic acid and allowed channel activation by heat in inside-out patches. Protease protection assays demonstrated a PIP-binding site within the N-tail. The proximity of TRPV4 tails, analyzed by fluorescence resonance energy transfer, increased by depleting PIP mutations in the phosphoinositide site or by coexpression with protein kinase C and casein kinase substrate in neurons 3 (PACSIN3), a regulatory molecule that binds TRPV4 N-tails and abrogates activation by cell swelling and heat. PACSIN3 lacking the Bin-Amphiphysin-Rvs (F-BAR) domain interacted with TRPV4 without affecting channel activation or tail rearrangement. Thus, mutations weakening the TRPV4–PIP interacting site and conditions that deplete PIP or restrict access of TRPV4 to PIP—in the case of PACSIN3—change tail conformation and negatively affect channel activation by hypotonicity and heat.Molecular and Cellular Biolog
Methylglyoxal Produced by Amyloid- Peptide-Induced Nitrotyrosination of Triosephosphate Isomerase Triggers Neuronal Death in Alzheimer’s Disease
Amyloid-β peptide (Aβ) aggregates induce nitro-oxidative stress, contributing to the characteristic neurodegeneration found in Alzheimer's disease (AD). One of the most strongly nitrotyrosinated proteins in AD is the triosephosphate isomerase (TPI) enzyme which regulates glycolytic flow, and its efficiency decreased when it is nitrotyrosinated. The main aims of this study were to analyze the impact of TPI nitrotyrosination on cell viability and to identify the mechanism behind this effect. In human neuroblastoma cells (SH-SY5Y), we evaluated the effects of Aβ42 oligomers on TPI nitrotyrosination. We found an increased production of methylglyoxal (MG), a toxic byproduct of the inefficient nitro-TPI function. The proapoptotic effects of Aβ42 oligomers, such as decreasing the protective Bcl2 and increasing the proapoptotic caspase-3 and Bax, were prevented with a MG chelator. Moreover, we used a double mutant TPI (Y165F and Y209F) to mimic nitrosative modifications due to Aβ action. Neuroblastoma cells transfected with the double mutant TPI consistently triggered MG production and a decrease in cell viability due to apoptotic mechanisms. Our data show for the first time that MG is playing a key role in the neuronal death induced by Aβ oligomers. This occurs because of TPI nitrotyrosination, which affects both tyrosines associated with the catalytic center
La resistencia a tracción indirecta del hormigón por doble punzonamiento. Influencia del tamaño máximo del árido y de la excentricidad
After reminding the interest, or the need, to know the concrete tensile strength, and having analyzed critically normal test methods, it /s briefly described the test method of indirect tension by "double punch test", proposed by W. F. Chen.
The experimental results obtained on three different concretes, two among them of a common density and a third with expanded clay as coarse aggregate, are shown. It has been proved that for normal density concretes, the tensile strength by "double punch test" has little sensibility to the variation of the aggregate maximum size, opposite to which it takes place in the splitting test, and the error of the centred of the "punch" has a minimum influence on the results, till very high eccentricities are reached. In conclusion, indirect tension test by double punch test is simple, practical and reproducible, responding to the needs to the needs of a commercial routine.Tras recordar el interés o la necesidad de conocer en ocasiones ¡a resistencia a tracción del hormigón y realizar un análisis crítico de los métodos de ensayo al uso, se describe brevemente el método de ensayo de tracción indirecta por "doble punzonamiento" propuesto por W. F. Chen.
Se presentan los resultados experimentales obtenidos sobre tres hormigones distintos, dos de densidad ordinaria y un tercero con arcilla expandida como árido grueso. Se ha comprobado que, para hormigones de densidad normal, la resistencia a tracción por "doble punzonamiento" es muy poco sensible a la variación del tamaño máximo del árido, contrariamente a lo que sucede con el ensayo brasileño, y que el error de centrado de los "punch" tiene una influencia mínima sobre los resultados hasta que se alcanzan excentricidades muy altas. Se concluye que el ensayo de tracción indirecta por "doble punzonamiento" es simple, práctico y reproducible, respondiendo a las necesidades de una rutina comercial
A precision medicine test predicts clinical response after idarubicin and cytarabine induction therapy in AML patients
Complete remission (CR) after induction therapy is the first treatment goal in acute myeloid leukemia (AML) patients and has prognostic impact. Our purpose is to determine the correlation between the observed CR/CRi rate after idarubicin (IDA) and cytarabine (CYT) 3 + 7 induction and the leukemic chemosensitivity measured by an ex vivo test of drug activity. Bone marrow samples from adult patients with newly diagnosed AML were included in this study. Whole bone marrow samples were incubated for 48 h in well plates containing IDA, CYT, or their combination. Pharmacological response parameters were estimated using population pharmacodynamic models. Patients attaining a CR/CRi with up to two induction cycles of 3 + 7 were classified as responders and the remaining as resistant. A total of 123 patients fulfilled the inclusion criteria and were evaluable for correlation analyses. The strongest clinical predictors were the area under the curve of the concentration response curves of CYT and IDA. The overall accuracy achieved using MaxSpSe criteria to define positivity was 81%, predicting better responder (93%) than non-responder patients (60%). The ex vivo test provides better yet similar information than cytogenetics, but can be provided before treatment representing a valuable in-time addition. After validation in an external cohort, this novel ex vivo test could be useful to select AML patients for 3 + 7 regimen vs. alternative schedules
Bone Marrow-Derived Cells from Male Donors Do Not Contribute to the Endometrial Side Population of the Recipient
Accumulated evidence demonstrates the existence of bone marrow-derived cells origin in the endometria of women undergoing bone marrow transplantation (BMT). In these reports, cells of a bone marrow (BM) origin are able to differentiate into endometrial cells, although their contribution to endometrial regeneration is not yet clear. We have previously demonstrated the functional relevance of side population (SP) cells as the endogenous source of somatic stem cells (SSC) in the human endometrium. The present work aims to understand the presence and contribution of bone marrow-derived cells to the endometrium and the endometrial SP population of women who received BMT from male donors. Five female recipients with spontaneous or induced menstruations were selected and their endometrium was examined for the contribution of XY donor-derived cells using fluorescent in situ hybridization (FISH), telomapping and SP method investigation. We confirm the presence of XY donor-derived cells in the recipient endometrium ranging from 1.7% to 2.62%. We also identify 0.45–0.85% of the donor-derived cells in the epithelial compartment displaying CD9 marker, and 1.0–1.83% of the Vimentin-positive XY donor-derived cells in the stromal compartment. Although the percentage of endometrial SP cells decreased, possibly being due to chemotherapy applied to these patients, they were not formed by XY donor-derived cells, donor BM cells were not associated with the stem cell (SC) niches assessed by telomapping technique, and engraftment percentages were very low with no correlation between time from transplant and engraftment efficiency, suggesting random terminal differentiation. In conclusion, XY donor-derived cells of a BM origin may be considered a limited exogenous source of transdifferentiated endometrial cells rather than a cyclic source of BM donor-derived stem cells
El cultivo de garbanzo (Cicer arietinum L.) en Argentina
El Programa Transferencia de Resultados de Investigación y
Comunicación Pública de la Ciencia (PROTRI), de la Secretaría de
Ciencia y Tecnología del Gobierno de la Provincia de Córdoba,
financió la realización del libro El cultivo de garbanzo en Argentina
con el objeto de promover la transferencia de resultados,
experiencias o saberes entre las áreas del sector social y productivo
para una mejor calidad de vida.
Para la ejecución de esta obra han sido convocados técnicos e
investigadores de las Ciencias Agropecuarias, Biológicas y
Económicas, quienes en una forma clara y sencilla, aunque no menos
consistente, ponen a disposición del lector sus experiencias
adquiridas a lo largo de varios años de trabajo.
El desarrollo de los diferentes temas se realiza a través de
dieciséis Capítulos que abarcan desde la domesticación de Cicer y su
llegada a la Argentina (Capítulo 1), pasando por el estudio
morfológico de las diferentes partes de la planta en relación a los
cultivares locales (Capítulo 2), además del manejo del suelo y sus
nutrientes, en cuanto a requerimientos edáficos en los sistemas
productivos (Capítulo 3). También se contempla el análisis de la
influencia de los diversos factores ambientales para la determinación
de zonas productivas (Capítulo 4). En el Capítulo 5 se hace referencia
a la ecofisiología del cultivo, sus requerimientos en las diferentes
etapas fenológicas, y cuándo y cómo se expresan en los cultivares.
Un tema de indudable importancia para las leguminosas en general, y
para el garbanzo en particular, es la simbiosis con las bacterias
fijadoras de nitrógeno y su influencia en la productividad, el cual se
desarrolla en el Capítulo 6.
La mejora genética en el país es abordada en el Capítulo 7,
donde se exponen sus inicios, desarrollo, disponibilidad de recursos,
bondades y potencial del germoplasma disponible.
A lo largo del ciclo biológico de la planta, el cultivo es visitado
por insectos e infectado por hongos. Identificarlos y conocer sus
ciclos biológicos y comportamientos es un aspecto importante para
seleccionar las medidas de manejo y control más adecuadas
(Capítulos 8 y 9).
Quizás pocos sepan que el volumen de semilla genética de un
cultivar, alcanza sólo unos pocos gramos (alrededor de 20). Para
llevar este pequeño volumen a toneladas, se requiere del trabajo y
tesón de investigadores, fitomejoradores y productores. Resultado de
ese esfuerzo es la difusión de los seis cultivares obtenidos en el país,
y se sigue trabajando para lograr nuevos materiales que se adapten a
las diversas áreas y sistemas de producción (Capítulo 10).
El desarrollo de los diferentes cultivares, que dan lugar a
diversas arquitecturas de plantas, sumado a la adopción de diversos
sistemas de producción, hace que el tema de la mecanización ocupe
un lugar importante. Maquinas pequeñas y grandes intervienen en las
etapas de siembra y de cosecha tratando de lograr la mayor
eficiencia posible y un producto de calidad (Capítulo 11).
Un cuello de botella para la expansión del cultivo es su
comercialización, tanto para consumo interno como externo. En el
Capítulo 12 se analizan los diferentes mercados y la necesidad de
lograr un producto rentable de alta calidad, para mercados muy
diversos.
En el Capítulo 13 se hace un recorrido por las diferentes
Provincias que actualmente producen garbanzo. Sus autores
comentan como se incorporó el cultivo a los sistemas productivos de
la región, sobre posibilidades y limitaciones, manejos y potencial de
rendimiento, entre otros aportes.
Los Capítulos 14 y 15 presentan dos temas de relevancia actual:
la composición química del grano y las posibilidades de brindar valor
agregado a éste, aspectos reforzados en la última década por el auge
de las tendencias que promueven un nuevo estilo de vida y una
alimentación sana, con alimentos naturales, bajos en grasa y con un
buen balance nutricional.
En el último Capítulo (16) se presentan experiencias de
investigación en las que se utilizó al garbanzo como materia prima o
como sustrato para diversas experimentaciones. La interacción
docente-investigador-alumno permitió que vieran la luz diversos
trabajos que, además de la formación de recursos humanos, brindan
una información útil y novedosa al incursionar en temas tales como
manejo de fechas de siembra, riego, alimento para pollos, cerdos y
abejas.
Estimado lector, tiene en sus manos un libro que es una
invitación a un viaje con dieciséis estaciones. En cada una de ellas
encontrará información sobre el cultivo del garbanzo en la Argentina.
Estos datos fueron obtenidos por docentes, investigadores,
productores, estudiantes que trabajaron y siguen trabajando para
aportar al conocimiento del cultivo en nuestro país, bajo la realidad
local y el contexto regional, ya que la mayoría de los trabajos y
publicaciones son de origen extranjero y la aplicación de muchas de
las tecnologías de manejo requieren una correcta adaptación y
validación. Esperamos que este libro, además de serle útil, pueda ser
disfrutado, sintiendo la pasión y el entusiasmo de cada uno de los
autores por brindar y compartir sus conocimientos y logros
Análisis comparativo de dos bases de bracket. Un estudio in vitro
El objetivo de este estudio ha sido evaluar las diferencias en la fuerza de adhesión entre dos brackets con bases diferentes: una base presenta la malla fina sin microarenar (bracket de American Orthodontics®) y el otro una malla profundamente ranurada y microarenada (bracket de Olmos®). Todos los brackets fueron soldados a barras de acero, se cementó en la base un composite fotopolimerizable (Eagle®) y se probaron para detectar el punto de fractura entre la base y el composite, mediante un esfuerzo de cizalla, en una máquina universal Instron. Se hallaron diferencias estadísticamente significativas entre ambos tipos de brackets. La malla rugosa (Olmos®) resultó tener una fuerza de adhesión bastante superior en comparación a la fina (American®)
Uncertainty quantification for hybrid random logistic models with harvesting via density functions
[EN] The so-called logistic model with harvesting, p '(t)=rp(t)(1-p(t)/K)-c(t)p(t), p(t(0))=p(0), is a classical ecological model that has been extensively studied and applied in the deterministic setting. It has also been studied, to some extent, in the stochastic framework using the Ito Calculus by formulating a Stochastic Differential Equation whose uncertainty is driven by the Gaussian white noise. In this paper, we present a new approach, based on the so-called theory of Random Differential Equations, that permits treating all model parameters as a random vector with an arbitrary join probability distribution (so, not just Gaussian). We take extensive advantage of the Random Variable Transformation method to probabilistically solve the full randomized version of the above logistic model with harvesting. It is done by exactly computing the first probability density function of the solution assuming that all model parameters are continuous random variables with an arbitrary join probability density function. The probabilistic solution is obtained in three relevant scenarios where the harvesting or influence function is mathematically described by discontinuous parametric stochastic processes having a biological meaning. The probabilistic analysis also includes the computation of the probability density function of the nontrivial equilibrium state, as well as the probability that stability is reached. All these results are new and extend their deterministic counterpart under very general assumptions. The theoretical findings are illustrated via two numerical examples. Finally, we show a detailed example where results are applied to describe the dynamics of stock of fishes over time using real data. (c) 2021 Elsevier Ltd. Flora fúngica transportada por semilla de amaranto (Amaranthus hypochondriacus L.) procedente de Córdoba, Argentina y su efecto sobre la calidad de la semilla
El conocimiento de las características sanitarias de la semilla es fundamental para implementar estrategias efectivas de manejo para reducir las pérdidas por patógenos. El objetivo del trabajo fue evaluar la flora fúngica transportada por semilla de amaranto de Córdoba y su efecto sobre la calidad de la semilla. Los tratamientos fueron T1: semillas usadas para la siembra en el Campo Escuela (FCA-UNC), T2: semillas cosechadas en parcelas sembradas el 1/11/2018 y T3: semillas cosechadas en parcelas sembradas el 19/12/2018. Los hongos se evaluaron mediante "blotter test" y la calidad de la semilla mediante test de poder germinativo (PG) según normas ISTA. Los hongos se identificaron a nivel de género con técnicas de micología y claves taxonómicas y se evaluaron los porcentajes de semillas con hongos. En el test de PG se evaluaron: energía germinativa (EG) y peso seco (PS) de esas plántulas, porcentaje total de plántulas normales (PG), porcentaje de plántulas anormales, semillas frescas y muertas. Ambos ensayos se diseñaron completamente aleatorizados con cuatro repeticiones de 100 semillas. Se analizaron con ANAVA y test comparación de medias LSD Fisher (α=0,05). Los hongos predominantes fueron Alternaria sp. y Fusarium spp. Con menor frecuencia se encontraron Phoma sp., Cladosporium sp., Aspergillus spp. y Penicillium spp. y de manera esporádica Epicoccum sp. y Ulocladium sp. La carga fúngica total aumentó (p=0,0001) en la semilla cosechada (T2:97,75 % y T3:93,5 %), respecto a la utilizada para la siembra (T1:56,25 %), en coincidencia con condiciones ambientales de elevada humedad persistente durante el tiempo de cosecha. Las semillas con Alternaria también aumentaron (p0,0500). En cambio, el porcentaje de semillas muertas aumentó (p=0,0139) en las semillas cosechadas T3:27,25 %, respecto a T1:12,25 % y T2: 7,75 %. La carga fúngica aumentó en ambas semillas cosechadas, pero el PG bajó y el porcentaje de semillas muertas aumentó solo en las semillas cosechadas en las parcelas de segunda fecha de siembra.Fil: Conles, Martha Yolanda. Performance of correlation functionals in ab initio chemisorption cluster-model calculations: Alkali metals on Si(111)
The performance of different correlation functionals has been tested for alkali metals, Li to Cs, interacting with cluster models simulating different active sites of the Si(111) surface. In all cases, the ab initio Hartree-Fock density has been obtained and used as a starting point. The electronic correlation energy is then introduced as an a posteriori correction to the Hartree-Fock energy using different correlation functionals. By making use of the ionic nature of the interaction and of different dissociation limits we have been able to prove that all functionals tested introduce the right correlation energy, although to a different extent. Hence, correlation functionals appear as an effective and easy way to introduce electronic correlation in the ab initio Hartree-Fock description of the chemisorption bond in complex systems where conventional configuration interaction techniques cannot be used. However, the calculated energies may differ by some tens of eV. Therefore, these methods can be employed to get a qualitative idea of how important correlation effects are, but they have some limitations if accurate binding energies are to be obtained
