    Biometric skills in developing countries are inadequate to support the changing priorities of agricultural research with the consequence that the quality of research aimed at increasing food security and alleviating poverty is often deficient because it lacks good statistical design" (CTA, 1997). It was on this basis that the workshop was planned, realising that not only do national scientists and statisticians (biometricians) need training in applied biometrics but they also need to be exposed to the range of problems and complexity of agricultural study design necessary for the developing world. Few courses in applied biometrics exist in universities in East and southern Africa. Training in biometrics provided hitherto by overseas institutions has been beneficial, but lecturers do not often have a full appreciation, or practical hands-on experience, of the special features of agricultural research and development in Africa. The subjects of experimental design, statistical analysis, interpretation and presentation of results are still recognised to be among the top priorities for training by national agricultural research system (NARS) scientists participating in collaborative networks co-ordinated by the International Livestock Research Institute (ILRI) and the International Centre for Research in Agroforestry (ICRAF). There is thus a major need to establish strong training programmes in the region itself with links to existing programmes abroad

    目的研究子宫内膜异位症发生进程中腹腔巨噬细胞吞噬功能的变化规律。方法构造小鼠子宫内膜异位症模型,用流式细胞术检测造模前后腹腔巨噬细胞吞噬荧光微球数,计算吞噬率和吞噬指数代表巨噬细胞的吞噬能力。结果巨噬细胞吞噬率和吞噬指数分别为:未造模组[(10.1±0.82)%,0.17±0.01]、造模后第1d[(32.78±2.43)%,0.60±0.02]、第2d[(33.82±1.23)%,0.61±0.02]、第3d[(35.93±2.81)%,0.72±0.03]、第4d[(27.92±1.24)%,0.51±0.03]、第5d[(24.34±0.91)%,0.40±0.02]、第6d[(17.91±1.03)%,0.28±0.01]、第9d[(17.56±0.80)%,0.26±0.01]、第12d[(19.42±1.02)%,0.36±0.01]、第15d[(26.78±2.05)%,0.54±0.02]、第18d[(27.46±1.61)%,0.50±0.02]、第21d[(25.99±2.31)%,0.54±0.03]。造模后的小鼠腹腔巨噬细胞吞噬能力均显著高于未造模组,且呈现出三个阶段的改变:第1-5d为第1阶段,第6-12d为第2阶段,第15-21d为第3阶段,第1和第3阶段显著高于第2阶段(P<0.01)。结论子宫内膜异位症小鼠腹腔巨噬细胞的吞噬功能增强,且随时间呈现规律性变化特征,可能与子宫内膜的清除和异位存活有关

    日本囊对虾人工诱变子一代遗传变异的RAPD分析

    以性腺成熟的日本囊对虾(Marsupenaeus japonicus)作为亲代,进行不同剂量的60Co-γ射线照射.采用RAPD技术对诱变子一代的无节幼体基因组DNA多态性进行了检测.从20个随机寡核苷酸引物共检测177个位点,其中多态位点152个,占85.9%.单个引物获得的标记为6~12个,平均8.85个,分子量在150~2 500 bp之间.遗传相似系数用Ne i的计算方法进行计算,遗传距离则用Lynch的计算方法进行计算.实验数据表明:诱变子代与正常对照组相比,遗传多样性水平较高,诱变产生了较为显著的变异.研究结果证实:在对虾大规模养殖,种质质量严重退化的情况下,人工诱变能够促进基因组更加广泛的变异,为新品种的选育打下坚实的基础;同时证明RAPD技术在检测遗传差异方面具有较高的灵敏度和分辨率

    Expression of Fusion Protein of Parathyroid Hormone and Transferrin N-terminal Half-molecule in Pichia pastoris

    利用重叠PCR技术将PTH(parathyroidhormone,甲状旁腺激素)基因与TFN(transferrinN_terminalhalf_molecule,转铁蛋白N端半分子)基因在体外融合,融合基因克隆至真核表达载体pPIC9中,转化毕赤酵母GS115。转化子经甲醇诱导后,融合蛋白得到了表达并分泌到发酵上清液中。经SPSepharoseFF阳离子交换层析、PhenylSepharoseFastFlow疏水层析纯化获得了纯度大于95%的PTH_TFN样品。Westernblot分析及腺苷酸环化酶实验证明融合蛋白中的PTH具有与抗PTH抗体结合能力及刺激腺苷酸环化酶的活性,铁饱和实验证明融合蛋白中的TFN和单独的TFN具有相同铁结合能力。因而TFN可望作为PTH的天然运输载体。The fused gene (PTH_TFN) of parathyroid hormone (PTH) gene and transferring N_terminal half_molecule (TFN) gene was amplified by multiple PCR and inserted into pPIC9 vector. The recombinant plasmid pPIC9_PTH_TFN was transformed into Pichia pastoris GS115 by PEG. After methanol induction, the target protein was expressed in fermentation supernatant at high level.The fused protein PTH_TFN with purity being higher than 95% was finally obtained after purification through two_step chromatography : SP Sepharose Fast Flow and Phenyl Sepharose Fast Flow.Western blot analysis and adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis and the ability to bind Fe ~3+ in the Fe ~3+ saturation study as the recombinant TFN did indicating that TFN could be used as the transcellar carrier of PTH.国家高技术研究与发展项目基金资助(No.2004AA215172)。~

    本文为武夷山自然保护区生物多样性GEF项目内容之一 ,调查了保护区内不同海拔高度的代表性森林生态系统的物种多样性 .小叶黄杨 (Buxussinicavar .parvifolia)林是中山矮曲林的代表性类型之一 .应用Shannon Wiener多样性指数、Pielou均匀度、Simpson指数和PIE对武夷山小叶黄杨群落高等植物进行了物种多样性研究 .结果表明前两者是较好的多样性指标 ,该群落的Shannon Wiener指数值为 1.80 6 8,Pielou均匀度为 49.6 7% ,Simpson指数值为 2 .9146 ,PIE为 0 .6 5 6 9.群落乔木层与藤本植物的物种多样性较高 ,前者各项指数值分别为 2 .2 312 ,71.16 % ,5 .0 783,0 .80 31;后者各项指数值分别为 1.3371,83.0 8% ,3.5 32 7,0 .716 9
