    Prototipo de construcción modular con ecomateriales

    Para dar respuesta a la necesidad real de crear edificios sostenibles, se hace necesario adoptar una nueva manera de construir que igualmente dé respuesta a las nuevas exigencias técnicas y funcionales. Los nuevos sistemas constructivos basados en la prefabricación responden muy favorablemente a la implantación de los conceptos mencionados, y más concretamente en el caso de la construcción mediante módulos prefabricados, al tratarse de un campo en el que, de una manera más directa y sencilla, se pueden implementar nuevas técnicas y materiales destinados a esa necesaria mejora de la eficiencia en la construcción, en el más amplio sentido de la palabra. El objetivo de este trabajo es mostrar las posibilidades que ofrece la construcción modular en el campo de la sostenibilidad, para lo cual se plantea un desarrollo basado en prototipos modulares prefabricados, que permita aprovechar la capacidad de los ecomateriales y de la tecnología ambiental, al tiempo que posibilita de manera asequible la tan deseable flexibilidad y diversidad funcional. Para ello se parte de un profundo proceso de análisis, tanto de la construcción modular, como del empleo de nuevos materiales ecológicos, lo que ha permitido seleccionar los elementos constructivos más adecuados para aunar políticas de ahorro energético y bienestar, hasta alcanzar una propuesta de Edificación Prefabricada Sostenible y Eficiente energéticamente.     Induced Systemic Resistance (ISR) and Fe Deficiency Responses in Dicot Plants

    Plants develop responses to abiotic stresses, like Fe deficiency. Similarly, plants also develop responses to cope with biotic stresses provoked by biological agents, like pathogens and insects. Some of these responses are limited to the infested damaged organ, but other responses systemically spread far from the infested organ and affect the whole plant. These latter responses include the Systemic Acquired Resistance (SAR) and the Induced Systemic Resistance (ISR). SAR is induced by pathogens and insects while ISR is mediated by beneficial microbes living in the rhizosphere, like bacteria and fungi. These root-associated mutualistic microbes, besides impacting on plant nutrition and growth, can further boost plant defenses, rendering the entire plant more resistant to pathogens and pests. In the last years, it has been found that ISReliciting microbes can induce both physiological and morphological responses to Fe deficiency in dicot plants. These results suggest that the regulation of both ISR and Fe deficiency responses overlap, at least partially. Indeed, several hormones and signaling molecules, like ethylene (ET), auxin, and nitric oxide (NO), and the transcription factor MYB72, emerged as key regulators of both processes. This convergence between ISR and Fe deficiency responses opens the way to the use of ISR-eliciting microbes as Fe biofertilizers as well as biopesticides. This review summarizes the progress in the understanding of the molecular overlap in the regulation of ISR and Fe deficiency responses in dicot plants.     Article Interferon-Alpha Decreases Cancer Stem Cell Properties and Modulates Exosomes in Malignant Melanoma

    Malignant melanoma (MM) can spread to other organs and is resistant in part due to the presence of cancer stem cell subpopulations (CSCs). While a controversial high dose of interferon-alpha (IFN-α) has been used to treat non-metastatic high-risk melanoma, it comes with undesirable side effects. In this study, we evaluated the effect of low and high doses of IFN-α on CSCs by analyzing ALDH activity, side population and specific surface markers in established and patient-derived primary cell lines. We also assessed the clonogenicity, migration and tumor initiation capacities of IFN-α treated CSCs. Additionally, we investigated genomic modulations related to stemness properties using microRNA sequencing and microarrays. The effect of IFN-α on CSCs-derived exosomes was also analyzed using NanoSight and liquid chromatography (LC-HRMS)-based metabolomic analysis, among others. Our results showed that even low doses of IFN-α reduced CSC formation and stemness properties, and led to a significant decrease in the ability to form tumors in mice xenotransplants. IFN-α also modulated the expression of genes and microRNAs involved in several cancer processes and metabolomics of released exosomes.     Efficient, resilient, and sustainable electricity delivery is a key cornerstone in increasingly large and complex urban environments, where citizens expect to keep or rise their living standards. In this context, cost-effective and ubiquitous digital technologies are driving the transformation of existing electrical infrastructures into truly smart systems capable of better providing the services a low-carbon society is demanding. The goal of this paper is twofold: 1) to review the dramatically evolving landscape of power systems, from the old framework based on centralized generation and control, aimed at serving inelastic customers through alternating current (ac) transmission networks and one-way distribution feeders, to a new paradigm centered mainly around two main axes: renewable generation, both centralized and distributed, and active customers (prosumers), interacting with each other through hybrid ac/dc smart grids; 2) to illustrate, through featured success stories, how several smart grid concepts and technologies have been put into practice in Spain over the last few years to optimize the performance of urban electrical assets

    On the Combination of Remote Sensing and Geophysical Methods for the Digitalization of the San Lázaro Middle Paleolithic Rock Shelter (Segovia, Central Iberia, Spain)

    This paper is focused on the Middle Paleolithic rock shelter called “Abrigo de San Lázaro”, placed in the Eresma River valley (Segovia, Spain). In this area, a multisource geomatic approach is used. On the one hand, the external envelope of the shelter has been digitalized by the means of an efficient combination between aerial photogrammetry and laser scanning (static and mobile). On the other hand, the ground penetrating radar and the electric tomography were used with the aim of evaluating the inner disposition of the shelter. The combination of both digitalization (external and internal) has allowed for improving the knowledge of the site characteristics that, in turn, will facilitate the future excavation works. The results of these studies allow archaeologists to know new data for a better understanding of the site formation (geology of the site, sedimentary potential, rock shelter dimensions, etc.) and the events that took place in it (knowing its historical evolution, especially the interaction between man and the environment). Additionally, the information obtained from these studies is very useful to plan future excavation works on the site

    Experience with Sunitinib in metastatic renal cell carcinoma (mRCC) patients: pooled analysis from 3 Spanish observational prospective studies

    [Abstract] Background: A pivotal, randomized, phase III trial demonstrated a statistically significant superiority of sunitinib over interferon-α in metastatic renal cell carcinoma (mRCC) patients. Objective: To evaluate the effectiveness and safety of sunitinib in patients with advanced or mRCC in routine clinical practice. Methods: Retrospective pooled analysis of clinical data from three observational and prospective studies carried out between 2007 and 2011 in 33 Spanish hospitals. Tumor response, Progression-free survival (PFS) and overall survival (OS), and main sunitinib-related toxicities were registered. Results: 224 patients were analyzed. Median PFS 10.6 months (95% CI: 9.02–12.25), median OS 21.9 months (95% CI: 17.2–26.6). Objective response rate (ORR) 43.8% (95% CI: 36.8–50.7). Median time to PR was 3.8 months (95% CI: 3.86–5.99) and to CR 8.2 months (95% CI: 4.75–9.77). The most common ≥ grade-3 AEs were asthenia/fatigue (18.7%), hand-foot syndrome (6.2%), hypertension (5.8%) and neutropenia (4.8%). Hand-foot syndrome, diarrhea and mucositis were confirmed as independent predictors for PFS and/or OS in a multivariate analysis (p < 0.05) Conclusions: Outcomes with sunitinib in daily clinical practice resemble those obtained in clinical trials. Long-term benefit with sunitinib is possible in advanced RCC patients but the appropriate management of toxicities is mandatory to enable patients to remain on treatment

    Simultaneous inhibition of pan-phosphatidylinositol-3-kinases and MEK as a potential therapeutic strategy in peripheral T-cell lymphomas

New, rational approaches are necessary to improve the very poor outcome in these patients. Phosphatidylinositol- 3-kinase is one of the most important pathways in cell survival and proliferation. We hypothesized that phosphatidylinositol- 3-kinase inhibitors could be rationally selected drugs for treating peripheral T-cell lymphomas. Several phosphatidylinositol-3-kinase isoforms were inhibited genetically (using small interfering RNA) and pharmacologically (with CAL-101 and GDC-0941 compounds) in a panel of six peripheral and cutaneous T-cell lymphoma cell lines. Cell viability was measured by intracellular ATP content; apoptosis and cell cycle changes were checked by flow cytometry. Pharmacodynamic biomarkers were assessed by western blot. The PIK3CD gene, which encodes the δ isoform of phosphatidylinositol-3-kinase, was overexpressed in cell lines and primary samples, and correlated with survival pathways. However, neither genetic nor specific pharmacological inhibition of phosphatidylinositol-3-kinase δ affected cell survival. In contrast, the pan-phosphatidylinositol-3-kinase inhibitor GDC-0941 arrested all T-cell lymphoma cell lines in the G1 phase and induced apoptosis in a subset of them. We identified phospho-GSK3b and phospho-p70S6K as potential biomarkers of phosphatidylinositol-3-kinase inhibitors. Interestingly, an increase in ERK phosphorylation was observed in some GDC-0941-treated T-cell lymphoma cell lines, suggesting the presence of a combination of phosphatidylinositol-3-kinase and MEK inhibitors. A highly synergistic effect was found between the two inhibitors, with the combination enhancing cell cycle arrest at G0/G1 in all T-cell lymphoma cell lines, and reducing cell viability in primary tumor T cells ex vivo. These results suggest that the combined treatment of pan-phosphatidylinositol-3-kinase + MEK inhibitors could be more effective than single phosphatidylinositol-3-kinase inhibitor treatment, and therefore, that this combination could be of therapeutic value for treating peripheral and cutaneous T-cell lymphomas.This work was supported by grants from the Asociación Española Contra el Cáncer, Fondo de Investigaciones Sanitarias (PI051623, PI052800 and PI080856), RTICC (RD06/0020/0107) and Ministerio de Ciencia e Innovación (SAF2008-0387-1).
