    A data mining approach for classifying DNA repair genes into ageing-related or non-ageing-related

    <p>Abstract</p> <p>Background</p> <p>The ageing of the worldwide population means there is a growing need for research on the biology of ageing. DNA damage is likely a key contributor to the ageing process and elucidating the role of different DNA repair systems in ageing is of great interest. In this paper we propose a data mining approach, based on classification methods (decision trees and Naive Bayes), for analysing data about human DNA repair genes. The goal is to build classification models that allow us to discriminate between ageing-related and non-ageing-related DNA repair genes, in order to better understand their different properties.</p> <p>Results</p> <p>The main patterns discovered by the classification methods are as follows: (a) the number of protein-protein interactions was a predictor of DNA repair proteins being ageing-related; (b) the use of predictor attributes based on protein-protein interactions considerably increased predictive accuracy of attributes based on Gene Ontology (GO) annotations; (c) GO terms related to "response to stimulus" seem reasonably good predictors of ageing-relatedness for DNA repair genes; (d) interaction with the XRCC5 (Ku80) protein is a strong predictor of ageing-relatedness for DNA repair genes; and (e) DNA repair genes with a high expression in T lymphocytes are more likely to be ageing-related.</p> <p>Conclusions</p> <p>The above patterns are broadly integrated in an analysis discussing relations between Ku, the non-homologous end joining DNA repair pathway, ageing and lymphocyte development. These patterns and their analysis support non-homologous end joining double strand break repair as central to the ageing-relatedness of DNA repair genes. Our work also showcases the use of protein interaction partners to improve accuracy in data mining methods and our approach could be applied to other ageing-related pathways.</p

    Visual ageing of human faces in three dimensions using morphable models and projection to latent structures

    We present an approach to synthesising the effects of ageing on human face images using three-dimensional modelling. We extract a set of three dimensional face models from a set of two-dimensional face images by fitting a Morphable Model. We propose a method to age these face models using Partial Least Squares to extract from the data-set those factors most related to ageing. These ageing related factors are used to train an individually weighted linear model. We show that this is an effective means of producing an aged face image and compare this method to two other linear ageing methods for ageing face models.     Raman Phonons and Ageing-Related Surface Disorder in NaxCoO2

    The polarized Raman spectra from ab and ac surfaces of single crystal NaxCoO2 (x~0.7), parent compound of recently discovered superconductor NaxCoO2.yH2O, are reported and discussed. The crystals were hexagon platelets of typical size 3x3x0.1 mm. Three of the five (A1g+E1g+3E2g) Raman active phonons were unambiguously identified at 458 (E1g), 494(E2g) and 574 (A1g) cm-1. The spectra from ab and ac surfaces differ significantly and provide evidence that within hours after preparation the ac surface, unlike the ab one, is strongly disordered.     Music for the ageing brain : Cognitive, emotional, social, and neural benefits of musical leisure activities in stroke and dementia

    Music engages an extensive network of auditory, cognitive, motor, and emotional processing regions in the brain. Coupled with the fact that the emotional and cognitive impact of music is often well preserved in ageing and dementia, music is a powerful tool in the care and rehabilitation of many ageing-related neurological diseases. In addition to formal music therapy, there has been a growing interest in self- or caregiver-implemented musical leisure activities or hobbies as a widely applicable means to support psychological wellbeing in ageing and in neurological rehabilitation. This article reviews the currently existing evidence on the cognitive, emotional, and neural benefits of musical leisure activities in normal ageing as well as in the rehabilitation and care of two of the most common and ageing-related neurological diseases: stroke and dementia.Peer reviewe

    Cognitive Dispersion Predicts Grip Strength Trajectories in Men but not Women in a Sample of the Oldest Old Without Dementia

    Background and Objectives: Grip strength is a reliable marker of biological vitality and it typically demonstrates an expected decline in older adults. According to the common-cause hypothesis there is also a significant association between cognitive and physical function in older adults. Some specific cognitive functions have been shown to be associated with grip strength trajectories with most research solely focused on cut-off points or mean cognitive performance. In the present study we examine whether a measure of cognitive dispersion might be more informative. We therefore used an index that quantifies dispersion in cognitive scores across multiple cognitive tests, shown to be associated with detrimental outcomes in older adults. Research Design and Methods: Using repeated grip strength measures from men and women aged 80 and older, free of dementia in the OCTO-Twin study, we estimated ageing-related grip strength trajectories. We examined the association of cognitive dispersion and mean cognitive function with grip strength level and ageing-related rate of change, accounting for known risk factors. Results: Cognitive dispersion was associated with grip strength trajectories in men and the association varied by mean cognitive performance, whereas we found no association in women. Discussion and Implications: Our results provide evidence of a sex-specific vitality association between cognitive dispersion and ageing-related trajectories of grip strength. Our results support the call for integration of sex and gender in health promotion and intervention research

    Study of the Exposome Ageing-related Factors in the Spanish Population

    The authors thank Dr Almudena Fuster-Manzano and Dr Blanca Piedrafita for providing scientific support.To characterize the exposome of the Spanish population and its association with skin ageing a cross-sectional epidemiological study was conducted in men and women (18–60 years old). A total of 1,474 participants were included. Mean age (± standard deviation) was 40.84 ± 10.26 years. Most participants had Fitzpatrick skin phototype II (44.1%) and skin ageing in accordance with their current age (69.0%). In the logistic model, age, smoking habit, use of sunscreen and use of cosmetics were all significant independent predictors of skin ageing. Thus, tobacco consumption increased the score of the model towards presenting skin ageing above that expected for age, while the opposite occurred with the use of sunscreen and a complete skin care routine. The exposome therefore has an impact on skin ageing, with age, smoking habit, use of sunscreen and the use of cosmetics identified as predictors of skin ageing. Skin care routines and sunscreen use may help to prevent this process.Vich
