    Analysis Expression of ZIP1 and Caspase-3 Protein in Adenocarsinoma of the Prostate

    Background: Carcinogenesis of adenocarcinoma of the prostate occurs due to dysregulation of zinc level within the cells. Intracellular zinc molecules influx is regulated by a transporter protein ZIP1, whose non-presence is predicted to inhibit apoptosis, thus leads to the development of prostate adenocarcinoma. Methods: This study was aimed to analyse the correlation of ZIP1 and Caspase-3 expression in prostate adenocarcinoma on its grading as represented by Gleason Score. This was a cross-sectional, retrospective analytical study on 31 Formalin-fixed, paraffin-embedded tissue that meets inclusion criteria. The specimen was stained using the immune-histochemistry technique for ZIP1 and Caspase-3. Protein expression of each case was counted using ImageJ analysis. Gleason score was acquired as secondary data from the cases' reports. The correlation of their expression with respect to Gleason score was analysed with Pearson's correlation using SPSS 11.5. Results: Mean expression level of ZIP1 and Caspase-3 in prostate adenocarcinoma were 35% and 33%, respectively. There was a significantly positive correlation between ZIP1 and Caspase-3 expression (r=0.379; p=0.018). However, their correlation was stronger in intermediate-grade group (r=0.73; p=0.01) and the correlation was much weaker in high-grade group (r=0.04; p=0.48). Conclusions: There was a positive correlation between ZIP1 and Caspase-3 expression in prostate adenocarcinoma.&nbsp

    Association of B-cell Lymphoma Protein-2 and Caspase-3 Expression in Ovarian Cancer

    Ovarian cancer remains a major problem of women's health in the world, including Indonesia, and is associated with high rates of incidence and mortality. There are many efforts in early diagnosis on ovarian cancer, but until now there have not been found any satisfactory method. On the other hand, knowledge and research in the field of molecular biology become more advance, one of them is a mechanism to control the growth of cells in ovarian cancer through a process of programmed cell death or apoptosis. B-cell lymphoma protein 2 (Bcl-2) and caspase-3 are proteins that play a role on the mechanism of apoptosis. The purpose of this study was to determine the expression of Bcl-2 and caspase-3 and their association with ovarian cancer. Materials and method: The design of this study was a cross-sectional study. Expression of Bcl-2 and caspase-3 examined by immunohistochemistry under light microscope with 400x light power field and expression as a negative when the protein expressed in 10% or less of cells and as a positive when the protein expressed in more than 10% of cells. A number of 45 subjects were recruited in this study. Thirthy one of 45 subjects showed the expression of Bcl-2 positive (68.9%), while the positive expression of caspase-3 present in 20 subjects (44.4%). There was a significant association between the expression of Bcl-2 with the expression of caspase-3 in ovarian cancer patients (p=0.002; lambda=0.4). There was also a significant association between stage of disease with expression of Bcl-2 (p=0.002; lambda=0.3) dan expression of caspase-3 (p=0.001; lambda=0.3). Conclusion: It concluded that there is a significant association between the expression of Bcl-2 and the expression of caspase-3 in ovarian cancer

    The Influence of Moderate Exercise on Caspase-3 Expression in Inhibiting Transformation of Oral Squamous Epithelial Cells

    The prevalence of cancer is increasing nowadays. In Indonesia, the prevalency of cancer is 1,4% out of 1000 people. Cancer is mainly caused by two factors; genetic and environmental, one of them being is a carcinogenic agent benzopyrene. Physical exercise can decrease blood glucose and fatty acid levels within the blood but its effect on cancer prevention, especially on caspase-3 expression, is still unknown. This study examines the expression of caspase-3 produced by moderate exercise in preventing the formation of transform cell epithelial of the oral squamous cell. 18 mice were divided into three study groups; control group 1 (K1) not given any physical exercise and benzopyrene, control group 2 (K2) not given any physical exercise but induced with 0,08 mg benzopyrene, treatment group (P) were given physical exercise in moderate intensity and induced with 0,08 mg benzopyrene. Buccal mucosa tissue samples were taken and stained for immunohistochemistry and examined under a light microscope at 400x at 10 different angles. The mean of caspase-3 expression was found highest in group P (13.33), meanwhile in group K1 and K2 were 3.00 and 8.33, respectively. There was significant difference of caspase-3 expression between the groups (p = 0.000) Conclusion: Moderate exercise could increase caspase-3 expression in preventing the formation of the transform cell epithelial of the oral squamous cell

    The Administration of Topical Aloe vera Extract Reduce the Number of Sunburn Cells and Expression of Caspase-3 on Post UVB-light-exposure Epidermis

    Introduction: Ultraviolet B (UVB) radiation triggers the formation of free radicals that cause apoptosis and sunburn cells (SBC) formation. Aloe vera contains anti-inflammatory and antioxidant compounds that can potentially inhibit this process. Objective: to assess the effect of topical Aloe vera extract administration on the decrease of SBC number and Caspase-3 expression on the epidermis after UVB light exposure.Methods: In a post-test only group design study, a single dose of 3 doses of Erythema (DEM) was performed on two groups of 6 week old BALB/c female rats. Group A (control) did not receive any topical treatment, and group B (treatment) were smeared with 75% Aloe vera extract before irradiation. Each group was divided into 4 sub-groups based on post-exposure time of 6 hours (A1 and B1), 12 hours (A2 and B2), 24 hours (A3 and B3), and 48 hours (A4 and B4). The expression of caspase-3 was assessed by immunohistochemical staining while the SBC number was measured using a microscope. The expression of caspase-3 was analyzed using the Kruskal-Wallis and Mann-Whitney statistical tests, while the number of SBC was analyzed using the one-way ANOVA statistical test and the post-hoc LSD test.Results: The results of the Kruskal-Wallis analysis showed a significant difference in Caspase-3 expression between groups, p <0.05. The results of Mann-Whitney analysis showed a significant difference in Caspase-3 expression between the A1-A3 and B1-B3 subgroups (p <0.05). One-way ANOVA analysis showed a significant difference in the number of SBC between groups (p <0.05). Post-hoc LSD analysis showed significant differences in SBC counts between groups A and B across all subgroups, p <0.05.Conclusion: Topical application of Aloe Vera extract decreased Caspase-3 expression and the number of SBC in UVB light-exposed skin

    Macrophage and Schwann cells are target cell on leprosy disease, where apoptotic is assumed as one of elimination mechanism of macrophage previously infected by M.leprae. Several study showed various result in apoptotic on leprosy disease. Apoptotic level can be evaluated by observing caspase-3 activity, an executioner caspase on cell death. This study is aimed to observe the relationship of caspase-3 expression with paucibacillary and multibacillary types of leprosy. We used observational analytic and cross sectional study with consecutive sampling method. The subject was leprosy patient which had been diagnosed on dermatovenereology division outpatient clinic Saiful Anwar Hospital, and fullfilled the inclusion-exclusion criteria. Total subject was 19 persons (11 multibacillary and 8 paucibacillary). Sampling was taken with punch biopsy on skin lession, followed by immunohistochemical staining with caspase-3 antibody, and caspase-3 expression was measured by ImmunoRatio method. Comparation test revealed significant difference (p&lt;0.05%) between caspase-3 expression mean on paucibacillary patient (84.46%) compared to multibacillary patient (65.39%). Pearson correlation test revealed caspase-3 expression tend to be higher in paucibacillary patient than multibacillary (coefficient correlation -0.759). In conclusion, there is a significance relationship between caspase-3 expression with leprosy type

    Caspase-3 and caspase-8 expression in breast cancer: caspase-3 is associated with survival

    Impaired apoptosis is one of the hallmarks of cancer. Caspase-3 and -8 are key regulators of the apoptotic response and have been shown to interact with the calpain family, a group of cysteine proteases, during tumorigenesis. The current study sought to investigate the prognostic potential of caspase-3 and -8 in breast cancer, as well as the prognostic value of combinatorial caspase and calpain expression. A large cohort (n = 1902) of early stage invasive breast cancer patients was used to explore the expression of caspase-3 and -8. Protein expression was examined using standard immunohistochemistry on tissue microarrays. High caspase-3 expression, but not caspase-8, is significantly associated with adverse breast cancer-specific survival (P = 0.008 and P = 0.056, respectively). Multivariate analysis showed that caspase-3 remained an independent factor when confounding factors were included (hazard ratio (HR) 1.347, 95% confidence interval (CI) 1.086–1.670; P = 0.007). The analyses in individual subgroups demonstrated the significance of caspase-3 expression in clinical outcomes in receptor positive (ER, PR or HER2) subgroups (P = 0.001) and in non-basal like subgroup (P = 0.029). Calpain expression had been previously assessed. Significant association was also found between high caspase-3/high calpain-1 and breast cancer-specific survival in the total patient cohort (P = 0.005) and basal-like subgroup (P = 0.034), as indicated by Kaplan–Meier analysis. Caspase-3 expression is associated with adverse breast cancer-specific survival in breast cancer patients, and provides additional prognostic values in distinct phenotypes. Combinatorial caspase and calpain expression can predict worse prognosis, especially in basal-like phenotypes. The findings warrant further validation studies in independent multi-centre patient cohorts

    Modified Wendan Decoction can Attenuate Neurotoxic Action Associated with Alzheimer's Disease

    We observed the effect of modified Wendan decoction (modified Wen-Dan-Tang) on a cellular model of Alzheimer's disease. Amyloid beta (Aβ) 25–35 segment neurotoxin was employed to induce a PC12 cellular model of Alzheimer's disease. After modified Wendan decoction was fed to rats, the serum containing medicine was prepared and changes in cell morphology observed. Cell mortality and survival rate was examined by trypan blue stain assay and MTT method and caspase-3 expression was detected by western blot, while cell apoptosis was examined by flow cytometry. Cell morphology of prepared serum group was better than that of controls, and cell survival rate in prepared serum group was higher than that in control (P < 0.01 or P < 0.05). Cell mortality, caspase-3 expression and apoptosis rate in prepared serum group were lower than that in control (P < 0.01 or P < 0.05). We conclude that Modified Wendan Decoction can attenuate the neurotoxicity of Aβ 25–35 and rescue neurons via suppressing apoptotic process

    Apoptosis in Gingival Overgrowth Tissues

    Variations in the balance between cell proliferation and apoptosis could contribute to the etiology of gingival overgrowth. The aim of this study was to test the hypothesis that, in fibrotic gingival lesions, fibroblast proliferation is stimulated and apoptosis is decreased. Apoptotic index, caspase 3 expression, the proliferative index, FOXO1 expression, and histological inflammation were measured in situ. Analysis of data showed that apoptosis decreased in all forms of gingival overgrowth examined (p \u3c 0.05), and inflammation caused a small but significant increase compared with non-inflamed tissues (p \u3c 0.05). The greatest decrease of apoptosis occurred in the most fibrotic tissues. Cell proliferation was elevated in all forms of gingival overgrowth tested, independent of inflammation (p \u3c 0.05). To identify potential mechanisms of transcriptional regulation of apoptosis, we assessed FOXO1 and caspase 3 expression levels and found them to correlate well with diminished apoptosis. Analysis of data suggests that increased fibroblast proliferation and a simultaneous decrease in apoptosis contribute to gingival overgrowth

    To evaluate the apoptosis involvement in the angiogenesis as a self-limiting process in patients with temporomandibular joint (TMJ) degenerated disc vessels, we assessed, by immunohistochemistry, the detection of TRAIL, its death receptor DR5 and caspase 3. TRAIL, its death receptor DR5 and caspase 3 expression were studied by immunohistochemistry in 15 TMJ discs displaced without reduction and in 4 unaffected discs. These apoptosis molecules were detected in the intima and media layers of newly formed vessels affected discs. In conclusion, vessels apoptosis activation in TMJ disc with ID could be regarded as a self-limiting process that try to leads to vessel regression; in this way an inhibition of angiogenic vessels may prove a key strategy in limiting pathological angiogenesis, by cutting off blood supply to tumors, or by reducing harmful inflammation

    The Effectiveness of Phaleria Macrocarpa and Chemotherapy in Increasing Caspase 3 and Apoptotic Index in Epidermoid Carcinoma

    Chemotherapy management of epidermoid carcinoma of the skin is relatively expensive and has toxic effects on vital organs. Phaleria macrocarpa is a medicinal plant that is often used as an anti-cancer. This study aims to prove the effectiveness of the Phaleria marcocarpa extract, paclitaxel-cisplatin chemotherapy and the combination of both as a neoadjuvant to increase caspase-3 expression and apoptotic index in epidermoid carcinoma of the skin in Swiss male mice. This study used male mice of the Swiss strain, after induction of epidermoid carcinoma cells, 4 group randomization, K (control), P1 (Phaleria macrocarpa 0.0715 mg/ day), P2 (paclitaxel 175 mg/m2 and cisplatin 50 mg/m2), P3 (the combination of Phaleria macrocarpa and chemotherapy). The caspase-3 expression was examined with immunohistochemical stain and the apoptotic index examination was done by staining the Tunel. The research shows that there is an increase in caspase-3 expression; the order of the increase (from the most significant to the less significant) is P3, P2, and P1 with significant result (p<0.05) compraed to the control group. There is a significant correlation between caspase-3 expression and apoptotic index with a very strong positive correlation. It can be concluded that the Phaleria marcocarpa extract, paclitaxel-cisplatin chemotherapy and the combination of both increased the caspase-3 expression and the apoptotic index of epidermoid carcinoma cell; there was no significant differences between P1 and P2 but there was a synergy effect from the administration of P1 and P2 combination, and there was significant correlation between the caspase-3 expression and apoptotic index with a very strong positive correlation
