    Separase prevents genomic instability by controlling replication fork speed

    Proper chromosome segregation is crucial for preserving genomic integrity, and errors in this process cause chromosome mis-segregation, which may contribute to cancer development. Sister chromatid separation is triggered by Separase, an evolutionary conserved protease that cleaves the cohesin complex, allowing the dissolution of sister chromatid cohesion. Here we provide evidence that Separase participates in genomic stability maintenance by controlling replication fork speed. We found that Separase interacted with the replication licensing factors MCM2-7, and genome-wide data showed that Separase co-localized with MCM complex and cohesin. Unexpectedly, the depletion of Separase increased the fork velocity about 1.5-fold and caused a strong acetylation of cohesin's SMC3 subunit and altered checkpoint response. Notably, Separase silencing triggered genomic instability in both HeLa and human primary fibroblast cells. Our results show a novel mechanism for fork progression mediated by Separase and thus the basis for genomic instability associated with tumorigenesis

    ABSTRACT Autoimmune and inflammatory disorders of the central nervous system can result in significant morbidity and mortality. Through the recognition of syndromes using diagnostic biomarkers, the clinician is now able to use immune suppressive therapies to improve outcomes. However, the therapeutic decision-making process is complex. The clinician has to balance the risk of disease, with the risk of treatment side effects. To achieve this balance, it is important to understand the natural history of disease, the risk of residual disability, the risk of relapse, and risk of a fatal outcome. It is also important to have some understanding of the pathological processes, as some of the entities have more reversible processes, whereas others have destructive processes. This review will assess the dynamic nature of this decision-making process, and compare some of the more severe diseases such as neuromyelitis optica, anti-N-methyl-D-aspartate receptor encephalitis and opsoclonus myoclonus ataxia syndrome, with disorders with more favourable outcomes such as Sydenham chorea and post-infectious cerebellar ataxia

    Defining a roadmap for harmonizing quality indicators in Laboratory Medicine: A consensus statement on behalf of the IFCC Working Group "laboratory Error and Patient Safety" and EFLM Task and Finish Group "performance specifications for the extra-analytical phases"

    The improving quality of laboratory testing requires a deep understanding of the many vulnerable steps involved in the total examination process (TEP), along with the identification of a hierarchy of risks and challenges that need to be addressed. From this perspective, the Working Group “Laboratory Errors and Patient Safety” (WG-LEPS) of International Federation of Clinical Chemistry and Laboratory Medicine (IFCC) is focusing its activity on implementation of an efficient tool for obtaining meaningful information on the risk of errors developing throughout the TEP, and for establishing reliable information about error frequencies and their distribution. More recently, the European Federation of Clinical Chemistry and Laboratory Medicine (EFLM) has created the Task and Finish Group “Performance specifications for the extraanalytical phases” (TFG-PSEP) for defining performance specifications for extra-analytical phases. Both the IFCC and EFLM groups are working to provide laboratories with a system to evaluate their performances and recognize the critical aspects where improvement actions are needed. A Consensus Conference was organized in Padova, Italy, in 2016 in order to bring together all the experts and interested parties to achieve a consensus for effective harmonization of quality indicators (QIs). A general agreement was achieved and the main outcomes have been the release of a new version of model of quality indicators (MQI), the approval of a criterion for establishing performance specifications and the definition of the type of information that should be provided within the report to the clinical laboratories participating to the QIs project

    U radu su prikazane aktivnosti menadžmenta poduzeća koje doprinose reorganizaciji istoga. Motiv reorganizacije u ovom slučaju jest dvojak. S jedne strane, dugogodišnja nestručna politika rukovođenja devastirala je poduzeće, u financijskom, kadrovskom, razvojnom, stručnom i percepcijskom smislu. S druge strane, nova zakonska regulativa nametnuta prvenstveno pridruživanjem republike Hrvatske europskoj uniji, ali i potrebom za održivim razvojem i smanjenjem nepotrebnih troškova zajednice, dovodi do imperativa promjene. Pokazuje se da je sustavan pristup rješavanju zadataka i problema taj koji donosi uspjeh. Metode organizacije procesa unutar poduzeća – u razvojnom, kadrovskom, financijskom, i svakom drugom smislu – zdravorazumske su; predstavljaju primjere uhodane pozitivne poslovne prakse. Prepoznavanje problema, definiranje ciljeva, određivanje ekonomski najoptimalnijih puteva za dosizanje istih, kvalitetna kadrovska politika, pozitivan pristup razvoju, a sve uz prepoznavanje uloge poduzeća u smislu društvene odgovornosti i brige za kako vlasništvo tako i dobrobit lokalne zajednice, aktivnosti su s kojima se poduzeće uspješno uhvatilo u koštac. Nakon manje od 3 godine rada pod novom upravom, Peovica d.o.o. ponos je lokalne javnosti; oslonac gradskim službama i građanima grada Omiša, po svim brojnim djelatnostima koje obavlja. Osnovna djelatnost doživjela je korijenite promjene, pritužbe korisnika svedene su na praktično nulu. Poslovanje je dovedeno u korak za zakonskim odredbama. Financijska situacija je stabilna, osigurani su novi poslovi. Zaposleno je 15-ak novih djelatnika, mahom srednje i mlađe životne dobi, čime je mnogima osiguran početak dugoročnog planiranja vlastite i obiteljske egzistencije. Započet je investicijski ciklus koji će idućih godina potpuno modernizirati poduzeće. Peovica postaje lider i u stručnoj javnosti: poduzeće koje služi kao primjer dobre prakse, inovativno i ekonomično. Broj projekata koje je Peovica d.o.o. prijavila za bespovratno sufinanciranje u Fond za zaštitu okoliša otprilike je isti kao i broj projekata kojih su prijavila sva ostala komunalna poduzeća u županiji.The paper presents organizational and process modifications required of a communal company whose core business is waste collection and disposal. Modifications are requirements in accordance with the Sustainable Waste Management Law passed by Croatian Assembly in summer of 2013, which, in turn, are derived from EU legislative and positive practices. The paper illustrates the results achieved by a systematic approach of menagement to problem solving. All the methods demonstrated (goal defining, determination of optimum path, efficiency and effectiveness, human resources policy, social responsibility, …) are well known in modern business and represent positive practices

    Berdasarkan data tahun 2007 keatas di Amerika diperkirakan akan terjadi ledakan gerontologi lansiayang berusia 65 tahun keatas dari berbagai etnik dan ras. Data dari Departemen Pendidikan danKesejahteraan Amerika melaporkan bahwa pada tahun 2006 terdapat sekitar 35 juta pasien rematik.Sedangkan pada tahun 2006 Mendapatkan data berdasarkan penelitian bahwa prevalensi nyerirematik di Indonesia mencapai 23,6-31,3%. Sedangkan jumlah lansia di PSTW Budi Luhur Jambisebanyak 72 lansia, yang menderita rematik sebanyak 40 lansia dan pengambilan sampelberdasarkan kriteria inklusi dan ekslusi sebanyak 20 lansia.Penelitian ini merupakan penelitian pre experimental dengan pendekatan atau desain “one group pretest dan post test”, pengumpulan data dilakukan dengan wawancara menggunakan kuesioner yangdilakukan pada 03 – 21 Agustus 2015. Populasi dalam penelitian ini adalah seluruh lansia yangmenderita rematik di PSTW Budi Luhur Jambi yaitu sebanyak 38 lansia dan Sampel dalam penelitianini adalah lansia yang mengalami nyeri rematik yang sesuai dengan kriteria inklusi yaitu sebanyak 20responden.Hasil penelitian ini didapatkan sebelum dilakukan latihan rutinitas senam rematik (pre test) yaitu nyeriringan 4 lansia (20%), nyeri sedang 13 lansia (65%), dan nyeri berat terkontrol 3 lansia (15%).Sedangkan sesudah dilakukan latihan rutinitas senam rematik (post test) yaitu tidak ada nyeri 1 lansia(5%), nyeri ringan 10 lansia (50%) dan nyeri sedang 9 lansia (45%).Berdasarkan hasil penelitian di atas didapatkan nilai rata-rata sebelum dilakukan senam 5 dansetelah dilakukan senam 3,5 dengan p-value 0,00 artinya ada pengaruh yang signifikan antarasebelum dan sesudah latihan. Dan diharapkan latihan rutinitas senam rematik dapat diterapkansecara teratur sebagai terapi penurunan tingkat nyeri rematik pada lansia

    Joint segmentation of color and depth data based on splitting and merging driven by surface fitting

    This paper proposes a segmentation scheme based on the joint usage of color and depth data together with a 3D surface estimation scheme. Firstly a set of multi-dimensional vectors is built from color, geometry and surface orientation information. Normalized cuts spectral clustering is then applied in order to recursively segment the scene in two parts thus obtaining an over-segmentation. This procedure is followed by a recursive merging stage where close segments belonging to the same object are joined together. At each step of both procedures a NURBS model is fitted on the computed segments and the accuracy of the fitting is used as a measure of the plausibility that a segment represents a single surface or object. By comparing the accuracy to the one at the previous step, it is possible to determine if each splitting or merging operation leads to a better scene representation and consequently whether to perform it or not. Experimental results show how the proposed method provides an accurate and reliable segmentation

    318-324Ayurveda has given prime importance to Mutravaha srotas (urinary system) and Srotogata vikaras (urinary disorders). Acharyas of Ayurveda has described many aspects of Mutravaha srotasa (urinary system) and described diseases pertaining to it. Mulaka (Raphanus sativus Linn.) belongs to family Brasicassea, traditionally being used in kidney disorders. Mulaka was firstly introduced in Charak Samhita and used for urinary system related problems. Hence, present study was planned with an aim to prove the traditional claims on Ayurvedic formulations of Mulaka i.e., powder of Mulaka panchanga and Mulaka kshara. Mulaka panchanga (450 and 900 mg/kg) and Mulaka kshara (90 and 180 mg/kg) were evaluated for nephroprotective activity by using cisplatin (5 mg/kg, ip) –induced nephrotoxicity in albino rats. Cisplatin increases the levels of blood urea, uric acid and serum creatinine which suggest the kidney damage in rats. Cisplatin–induced the hepatotoxicity was confirmed by increase in the level of serum transaminases, alkaline phosphatase and bilirubin with increase in the level of serum cholesterol and triglyceride. Both formulations reversed the serum biochemical parameters and urine creatinine levels with an increase in the urine volume. Further, powder of Mulaka panchanga and Mulaka kshara, attenuated the level of free radicals with concomitantly increase in antioxidant parameters in kidney tissue homogenate and thus, protect the kidney cytoarchitecture in histopathological study. The result of present study indicates that Mulaka panchanga and Mulaka kshara both having nephroprotective and antioxidant properties in dose dependent manner in cisplatin–induced nephrotoxicity in rats

    Predmet proučavanja ovog rada su troškovi koji se javljaju u poslovanju komunalnih poduzeća . Troškovi su središnja ekonomska kategorija i pojavljuju se u svakom poslovnom procesu. Poznavanje i praćenje troškova poslovanja pomaže poduzeću da sagleda ukupne i pojedinačne troškove i bitna je pretpostavka svakog poslovnog odlučivanja. Komunalne su službe važan element sustava upravljanja gradom i pružanja javnih usluga građanima o čemu ovisi njihovo šire zadovoljstvo javnom upravom i javnim sektorom u cjelini. Cilj rada je, u teorijskom dijelu, definirati pojam troška, prikazati njegove osnovne podjele i značenje troškova za planiranje i nadzor poslovanja te prikazati specifičnosti trgovačkog društva u komunalnoj djelatnosti. U empirijskom dijelu cilj je prikazati posebnosti u način evidentiranja i planiranja troškova poslovanja na primjeru komunalnog poduzeća Podi Dugopolje d.o.o.The subject of this study are costs that appear in the operations of municipal companies. The costs are central economic category and appear in every business process. Knowing and tracking of operating costs is helping company to look at the total and individual costs and is an important assumption of any business decision. Municipal services are important element of the system of city management and provision of public services to citizens about what depends their wider satisfaction in public administration and public sector as a whole. The aim of the paper, in the theoretical part, is to define cost, demonstrate its main classifications and meaning costs for planning and control of operations and demonstrate the specificity of the company in the municipal activities. In the empirical part the goal is to show the specificities in the method of recording and planning costs in the case of municipal company Podi Dugopolje d.o.o.
