Clinical evaluation and antibody responses in sheep after primary and secondary experimental challenges with the mange mite Sarcoptes scabiei var. ovis
P. 109-116In this work the clinical evolution and the specific serum IgG and IgE antibody responses in
sheep after primary (n = 10) and secondary (n = 4) experimental challenges with the
mange mite Sarcoptes scabiei var. ovis were studied. The primary infection was
characterized by the development of mange lesions in all sheep, a detection of live S.
scabiei mites in 70% skin scrapings taken in week 10 post-challenge (PC), strongly raised
and sustained specific IgG levels and a more moderate but continuous rise in specific IgE
levels. Seroconversion was detected for IgG and IgE by ELISA in 90% and 60% of the sheep in
week 8 PC, respectively. By Western-blotting (WB), ten IgG-reactive bands (36–120 kDa)
and four IgE-reactive bands (90–180 kDa) were observed in week 8 PC. Following the
secondary challenge the ewes developed a smaller area of mange lesion than that seen
following primary challenge and live S. scabiei mites were not detected in skin scrapings
collected in week 8 PC, suggesting that sheep had developed immunity to re-infection.
Compared to primary infection, the specific IgG secondary antibody levels were transient,
but in contrast there was an anamnestic IgE response, resulting in an elicitation of specific
serum IgE levels in week 2 PC significantly higher than those demonstrated after primary
infection. WB analysis revealed one additional IgG-reactive band (180 kDa) and no
additional IgE-reactive bands. Determining the immunodiagnostic or vaccination value of
the IgG-reactive antigens and IgE-reactive allergens detected requires further studies.S
Teladorsagia circumcincta beta tubulin: the presence of the E198L polymorphism on its own is associated with benzimidazole resistance.

resistance was measured by the faecal egg count reduction test
(FECRT) and the egg hatch test (EHT) using a discriminating dose
(DD) in 39 sheep flocks. Around 1000 larvae collected before and
after treatment were used for DNA extraction. The resistant
species identified in all flocks was T. circumcincta. The
resistance alleles frequencies were measured for F200Y and
E198A. A 371-bp fragment of the isotype-1 \xCE\xB2-tubulin gene
was analysed, including the three codons of interest, and a new

resistant flocks was 35% by FECRT or 26% by EHT; however, F200Y
and E198A SNPs were absent in T. circumcincta. The amplification
of a 371-bp fragment confirmed the absence of F167Y and F200Y in
6 resistant flocks. Regarding codon 198, all samples after
treatment carried a leucine (CTA). A pyrosequencing assay
analysed the allele frequencies for the first two bases at codon
198 independently, G/C and A/T. The correlation between C and T
frequencies was almost 1 (r\xE2\x80\x89=\xE2\x80\x890.929,
P\xE2\x80\x89<\xE2\x80\x890.0001) and the mean value of both
was calculated to measure the leucine frequency; this value
ranged between 10.4-80.7% before treatment, and 82.3-92.8% after
treatment. High and similar correlations were reported between
the genotypic variables (C frequency, T frequency or mean of
both frequencies) and phenotypic resistance
(r\xE2\x80\x89>\xE2\x80\x890.720,
P\xE2\x80\x89<\xE2\x80\x890.0001), although negatively
associated with the FECRT and positively with the EHT. According
to multivariate linear regression analysis, the T frequency was
the most significant variable influencing the phenotypic
resistance (FECRT or EHT; P\xE2\x80\x89<\xE2\x80\x890.0001).
In the EHT, 67.1% of the phenotypic variability is associated
with the T frequency but in the FECRT only 33.4%; therefore, the
EHT using a DD seems to detect the genotypic resistance more

Modelos de crecimiento y producción en España: historia, ejemplos contemporáneos y perspectivas
En el presente trabajo se presenta una revisión sobre los modelos forestales desarrollados en España durante los últimos años, tanto para la producción maderable como no maderable y, para la dinámica de los bosques (regeneración, mortalidad). Se presentan modelos tanto de rodal completo como de clases diamétricas y de árbol individual. Los modelos desarrollados hasta la fecha se han desarrollado a partir de datos procedentes de parcelas permanentes, ensayos y el Inventario Forestal Nacional. En el trabajo se muestran los diferentes submodelos desarrollados hasta la fecha, así como las plataformas informáticas que permiten utilizar dichos modelos. Se incluyen las principales perspectivas
de desarrollo de la modelización forestal en España.In this paper we present a review of forest models developed in Spain in recent years for both timber and non timber production and forest dynamics (regeneration, mortality). Models developed are whole stand, size (diameter) class and individual-tree. The models developed to date have been developed using data from permanent plots, experimental
sites and the National Forest Inventory. In this paper we show the different sub-models developed so far and the friendly use software. Main perspectives of forest modeling in Spain are presented.The models described in this paper were funded by
different regional, national and European projects, and
some of them were elaborated by the authors. This

Anthelmintic activity of aminoalcohol and diamine derivatives against the gastrointestinal nematode Teladorsagia circumcincta
11 páginas, 5 figuras, 4 tablas.Gastrointestinal nematodes (GIN) infections are a serious problem in livestock production due to the great economic losses they cause. Their control is increasingly difficult because of the rapid development of drug resistance and the limited number of available drugs. Therefore, this study evaluated 18 aminoalcohol and 16 diamine derivatives against eggs, first and third stage larvae from a susceptible and a resistant isolate of Tela-dorsagia circumcincta collected from sheep. The effectiveness of the in vitro anthelmintic activity of the com-pounds was evaluated using three different procedures: Egg Hatch Test (EHT), Larval Mortality Test (LMT) and Larval Migration Inhibition Test (LMIT). Those compounds with activities higher than 90 % in the initial screening at 50 μM were selected to determine their half maximal effective concentration (EC50). In parallel, cytotoxicity assays were conducted on Caco2 and HepG2 cell lines to calculate Selectivity Indexes (SI) for each compound. The diamine 30 presented the best results in preventing egg hatching, displaying the lowest EC50 value (1.01 ±0.04 μM) of all compounds tested and the highest SI (21.21 vs. Caco-2 cells). For the LMIT, the diamine 34 showed the highest efficacy, with EC50 values of 2.67 ±0.08 and 3.02 ±0.09 μM on the susceptible and resistant isolate of the parasite, respectively.We are grateful to Dave Bartley, from Moredun Research Institute for providing the triple-resistant isolate of T. circumcincta. Prevalence of disability in a composite ≥75 year-old population in Spain: A screening survey based on the International Classification of Functioning
<p>Abstract</p> <p>Background</p> <p>The prevalence and predictors of functional status and disability of elderly people have been studied in several European countries including Spain. However, there has been no population-based study incorporating the International Classification of Functioning, Disability and Health (ICF) framework as the basis for assessing disability. The present study reports prevalence rates for mild, moderate, and severe/extreme disability by the domains of activities and participation of the ICF.</p> <p>Methods</p> <p>Nine populations surveyed in previous prevalence studies contributed probabilistic and geographically defined samples in June 2005. The study sample was composed of 503 subjects aged ≥75 years. We implemented a two-phase screening design using the MMSE and the World Health Organization-Disability Assessment Schedule 2<sup>nd </sup>edition (WHO-DAS II, 12 items) as cognitive and disability screening tools, respectively. Participants scoring within the positive range of the disability screening were administered the full WHO-DAS II (36 items; score range: 0-100) assessing the following areas: Understanding and communication, Getting along with people, Life activities, Getting around, Participation in society, and Self-care. Each disability area assessed by WHO-DAS II (36 items) was reported according to the ICF severity ranges (No problem, 0-4; Mild disability, 5-24; Moderate disability, 25-49; Severe/Extreme disability, 50-100).</p> <p>Results</p> <p>The age-adjusted disability prevalence figures were: 39.17 ± 2.18%, 15.31 ± 1.61%, and 10.14 ± 1.35% for mild, moderate, and severe/extreme disability, respectively. Severe and extreme disability prevalence in mobility and life activities was three times higher than the average, and highest among women. Sex variations were minimal, although life activities for women of 85 years and over had more severe/extreme disability as compared to men (OR = 5.15 95% CI 3.19-8.32).</p> <p>Conclusions</p> <p>Disability is highly prevalent among the Spanish elderly. Sex- and age-specific variations of disability are associated with particular disability domains.</p
Measurement of the cosmic ray spectrum above eV using inclined events detected with the Pierre Auger Observatory
A measurement of the cosmic-ray spectrum for energies exceeding
eV is presented, which is based on the analysis of showers
with zenith angles greater than detected with the Pierre Auger
Observatory between 1 January 2004 and 31 December 2013. The measured spectrum
confirms a flux suppression at the highest energies. Above
eV, the "ankle", the flux can be described by a power law with
index followed by
a smooth suppression region. For the energy () at which the
spectral flux has fallen to one-half of its extrapolated value in the absence
of suppression, we find
