An Analysis of Putative Roles for the CCR4-NOT Deadenylase-Complex Subunit Regena (NOT2) in microRNA-Mediated Gene Silencing in \u3cem\u3eDrosophila Melanogaster\u3c/em\u3e
microRNAs (miRNAs) are one class of small non-coding ribonucleic acid (RNA) molecules essential to development and homeostasis in plants and animals. miRNAs silence gene expression through complementary base pairing with target gene messenger RNAs and association with the miRNA-induced silencing complex (miRISC). The identification and characterization of cellular factors required for miRNA-mediated gene silencing is incomplete. A forward genetic screen was carried out in Drosophila melanogaster to generate flies defective for gene silencing. Silencing was assayed by expression of a Green Fluorescent Protein (GFP) reporter fused to the Brd gene 3’ UTR, which is regulated by miRNAs. Genetic analysis revealed that the CCR4-NOT deadenylase-complex subunit Regena (NOT2) is required for miRNA-mediated silencing of the reporter. In addition, perturbation of Regena function altered Drosophila eye development and resulting adult eye morphology. miRNAs are thought to silence target gene expression through a combination of translational repression and target mRNA degradation, though the detailed mechanism of this process is a matter of controversy. Novel genetic reagents to explore miRNA function in vivo have been generated and characterized. Ongoing efforts aim to explore whether Regena is required to silence other miRNA targets in vivo, and whether Regena is required for miRNA-mediated gene silencing at different stages of the Drosophila life cycle. Elucidation of the lesion in the Regena (NOT2) gene and the molecular nature of GFP reporter silencing will contribute to an understanding of the mechanism of miRNA-mediated gene silencing in vivo

Rain volume estimation over areas using satellite and radar data
The analysis of 18 convective clusters demonstrates that the extension of the Area-Time-Integral (ATI) technique to the use of satellite data is possible. The differences of the internal structures of the radar reflectivity features, and of the satellite features, give rise to differences in estimating rain volumes by delineating area; however, by focusing upon the area integrated over the lifetime of the storm, it is suggested that some of the errors produced by the differences in the cloud geometries as viewed by radar or satellite are minimized. The results are good and future developments should consider data from different climatic regions and should allow for implementation of the technique in a general circulation model
Combining School and Family-Centered Interventions to Prevent Child Behavior Problems
This poster was presented at the American Psychological Association Conference in Boston, MA, in August 2008.Although school-wide PBS has been shown to reduce problem behaviors among students, approximately 15-20% of students are still in need of more selective interventions due to disruptive behavior problems. Given that both school (see Reinke & Herman, 2002) and family factors (Banks, et al., 1993) contribute to behavior problems in children, combining interventions that target both the school environment and family context is expected to further improve outcomes for those students. An ecological model combining PBS and the Family Check-up is proposed to prevent and reduce behavior problems in children
Exploring and Visualizing A-Train Instrument Data
The succession of US and international satellites that follow each other in close succession, known as the A-Train, affords an opportunity to atmospheric researchers that no single platform could provide: Increasing the number of observations at any given geographic location.. . a more complete "virtual science platform". However, vertically and horizontally, co-registering and regridding datasets from independently developed missions, Aqua, Calipso, Cloudsat, Parasol, and Aura, so that they can be inter-compared can be daunting to some, and may be repeated by many. Scientists will individually spend much of their time and resources acquiring A-Train datasets of interest residing at various locations, developing algorithms to match up and graph datasets along the A-Train track, and search through large amounts of data for areas and/or phenomena of interest. The aggregate amount of effort that can be expended on repeating pre-science tasks could climb into the tens of millions of dollars. The goal of the A-Train Data Depot (ATDD) is to enable free movement of remotely located A-Train data so that they are combined to create a consolidated vertical view of the Earth's Atmosphere along the A-Train tracks. The innovative approach of analyzing and visualizing atmospheric profiles along the platforms track (i.e., time) is accomplished by through the ATDDs Giovanni data analysis and visualization tool. Giovanni brings together data from Aqua (MODIS, AIRS, AMSR-E), Cloudsat (cloud profiling radar) and Calipso (CALIOP, IIR), as well as the Aura (OMI, MLS, HIRDLS, TES) to create a consolidated vertical view of the Earth's Atmosphere along the A-Train tracks. This easy to learn and use exploration tool will allow users to create vertical profiles of any desired A-Train dataset, for any given time of choice. This presentation shows the power of Giovanni by describing and illustrating how this tool facilitates and aids A-Train science and research. A web based display system Giovanni provides users with the capability of creating co-located profile images of temperature and humidity data from the MODIS, MLS and AIRS instruments for a user specified time and spatial area. In addition, Cloud and Aerosol profiles may also be displayed for the Cloudsat and Caliop instruments. The ability to modify horizontal and vertical axis range, data range and dynamic color range is also provided. Two dimensional strip plots of MODIS, AIRS, OM1 and POLDER parameters, co-located along the Cloudsat reference track, can also be plotted along with the Cloudsat cloud profiling data. Center swath pixels for the same parameters can also be shown as line plots overlaying the Cloudsat or Calipso profile images. Images and subsetted data produced in each analysis run may be downloaded. Users truly can explore and discover data specific to their needs prior to ever transferring data to their analysis tools

Statistical Agencies and Responses to Financial Crises: Eurostat, Bad Banks, and the ESM
We demonstrate the important role that the European Statistical Agency - Eurostat - plays in shaping tools for responding to banking crises. From 2009, Eurostat used its position as the interpreter of member state budget statistical rules to implement increasingly stringent rules for how financial crisis responses would affect public budgets. Rather than mere technical details, these rules affected crisis responses. Elected politicians, and especially those under bailout programs, have strong incentives to minimise the direct budgetary effects of aiding failing financial institutions. By establishing and enforcing new rules about which crisis reponses directly hit member state budgets and which did not, Eurostat created incentives to choose certain policies. We explore this process by examining the creation of bad banks and the European Stability Mechanism. Our paper makes an original contribution to both the study of the European banking union and the general role that statistical agencies can play in shaping crisis responses
Predictors and outcomes of crossover to surgery from physical therapy for meniscal tear and osteoarthritis a randomized trial comparing physical therapy and surgery
BACKGROUND: Arthroscopic partial meniscectomy (APM) combined with physical therapy (PT) have yielded pain relief similar to that provided by PT alone in randomized trials of subjects with a degenerative meniscal tear. However, many patients randomized to PT received APM before assessment of the primary outcome. We sought to identify factors associated with crossing over to APM and to compare pain relief between patients who had crossed over to APM and those who had been randomized to APM. METHODS: We used data from the MeTeOR (Meniscal Tear in Osteoarthritis Research) Trial of APM with PT versus PT alone in subjects ≥45 years old who had mild-to-moderate osteoarthritis and a degenerative meniscal tear. We assessed independent predictors of crossover to APM among those randomized to PT. We also compared pain relief at 6 months among those randomized to PT who crossed over to APM, those who did not cross over, and those originally randomized to APM. RESULTS: One hundred and sixty-four subjects were randomized to and received APM and 177 were randomized to PT, of whom 48 (27%) crossed over to receive APM in the first 140 days after randomization. In multivariate analyses, factors associated with a higher likelihood of crossing over to APM among those who had originally been randomized to PT included a baseline Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) Pain Score of ≥40 (risk ratio [RR] = 1.99; 95% confidence interval [CI] = 1.00, 3.93) and symptom duration of <1 year (RR = 1.74; 95% CI = 0.98, 3.08). Eighty-one percent of subjects who crossed over to APM and 82% of those randomized to APM had an improvement of ≥10 points in their pain score at 6 months, as did 73% of those who were randomized to and received only PT. CONCLUSIONS: Subjects who crossed over to APM had presented with a shorter symptom duration and greater baseline pain than those who did not cross over from PT. Subjects who crossed over had rates of surgical success similar to those of the patients who had been randomized to surgery. Our findings also suggest that an initial course of rigorous PT prior to APM may not compromise surgical outcome. LEVEL OF EVIDENCE: Prognostic Level II. The TreaT-Assay: A Novel Urine-Derived Donor Kidney Cell-Based Assay for Prediction of Kidney Transplantation Outcome
Donor-reactive immunity plays a major role in rejection after kidney transplantation, but analysis of donor-reactive T-cells is not applied routinely. However, it has been shown that this could help to identify patients at risk of acute rejection. A major obstacle is the limited quantity or quality of the required allogenic stimulator cells, including a limited availability of donor-splenocytes or an insufficient HLA-matching with HLA-bank cells. To overcome these limitations, we developed a novel assay, termed the TreaT (Transplant reactive T-cells)-assay. We cultivated renal tubular epithelial cells from the urine of kidney transplant patients and used them as stimulators for donor-reactive T-cells, which we analyzed by flow cytometry. We could demonstrate that using the TreaT-assay the quantification and characterization of alloreactive T-cells is superior to other stimulators. In a pilot study, the number of pre-transplant alloreactive T-cells negatively correlated with the post-transplant eGFR. Frequencies of pre-transplant CD161+ alloreactive CD4+ T-cells and granzyme B producing alloreactive CD8+ T-cells were substantially higher in patients with early acute rejection compared to patients without complications. In conclusion, we established a novel assay for the assessment of donor-reactive memory T-cells based on kidney cells with the potential to predict early acute rejection and post-transplant eGFR
