    Telomere dynamics in the Pacific crown-of-thorns seastar (Acanthaster cf. solaris): effect of age, diet, and tissue type

    Population irruptions of crown-of-thorns seastar (CoTS, Acanthaster spp.) represent a perennial threat to Indo-Pacific coral reefs. Age determination of CoTS is challenging, thereby hindering understanding and management of this nuisance species. Telomeres, which are protective DNA structure found at the ends of eukaryotic chromosomes that shorten at each cell division, have been used to estimate age in wild animals. To investigate the use of telomeres in CoTS, we optimized a quantitative PCR protocol to measure relative telomere length (rTL) in CoTS for the first time. Comparing rTL among four age groups (4, 7, 16, > 24 months post-settlement), we found that adult CoTS generally exhibit shorter rTL than juveniles, which is the first evidence of age-related telomere attrition in CoTS. However, there was large within-age class variation, and no significant relationships were found between adult CoTS rTL and potential age-indicating external features. Furthermore, we found accelerated telomere attrition under sub-optimal diet, where individuals that were fed crustose coralline algae for 16 months exhibited shorter rTL than their counterparts fed on coral. A positive correlation was found between rTL of tube feet and pyloric caeca, suggesting synchronization of telomere dynamics across somatic tissues in CoTS. Overall, our results suggest that rTL could be used to classify CoTS into broad age groups, though individual variation constrains the ability to resolve specific cohorts. The present study contributes to the understanding of telomere dynamics in marine invertebrates, while laying the groundwork for future research into rTL as biomarker for age and potentially stress for CoTS

    Hydrogen production by chemical-looping reforming in a circulating fluidized bed reactor using Ni-based oxygen carriers

    7 pages, 11 figures,.- Available online November 18, 2008.This work presents the experimental results obtained during auto-thermal chemical-looping reforming (CLR) in a 900 Wth circulating fluidized bed reactor under continuous operation using methane as fuel. Two oxygen carriers based on NiO and supported on γ-Al2O3 and α-Al2O3 were used during more than 50 h of operation with each oxygen carrier. During operation the effect of different operating variables, like fuel reactor temperature, H2O/CH4 molar ratio and solid circulation rate, on CH4 conversion and gas product distribution was analyzed. It was found that in all operating conditions CH4 conversion was very high (>98%) and the most important variable affecting to the gas product distribution was the solid circulation rate, that is, NiO/CH4 molar ratio. Similar gas product distribution was obtained working with both oxygen carriers although at different NiO/CH4 molar ratios. The oxygen carrier of NiO on α-Al2O3 needed lower NiO/CH4 molar ratio to reach the same gas product composition than the oxygen carrier of NiO on γ-Al2O3. Working at optimal operating conditions, 2.5 moles of H2 per mol of CH4 could be obtained in this process. During operation the oxygen carrier particles maintained their physical and chemical properties. These results suggest that these oxygen carriers could have a high durability, being suitable oxygen carriers for a CLR system. © 2008 Elsevier B.V.     Fabrication of chitosan-flax composites with differing molecular weights and its effect on mechanical properties

    An aqueous fabrication method is investigated for a composite reinforced with chitosan and flax fibers. The composite is characterized structurally, mechanically and chemically. A strong influence of molecular weight (MW) is identified on the composite properties. A strong fiber-matrix interface, which is associated with porosity and effective fiber impregnation, is achieved by applying low molecular weight (LMW) solution followed by casting using LMW or medium molecular weight (MMW) solution. Porosity is analyzed using μ-CT analysis. Increasing porosity with increasing molecular weight results in a decline of the tensile and flexural properties of the composites. The chitosan-flax composites have a low density compared to synthetic and natural fiber composites, which is a competitive advantage as a replacement material for particle board or plyboard in suspended ceilings, furniture compartments, sports or leisure equipment. A multiscale simulation is carried out to compute the directional effective elastic properties and predicts a potential 21% improvement of the tensile modulus if the process is optimized. This work shows the potential of chitosan-flax composites as a sustainable green material with an aqueous fabrication procedure and useful mechanical properties

    Divergent electrophysiologic action of dapagliflozin and empagliflozin on ventricular and atrial tachyarrhythmias in isolated rabbit hearts

    BackgroundThe use of SGLT-2 inhibitors has revolutionized heart failure therapy. Evidence suggests a reduced incidence of ventricular and atrial arrhythmias in patients with dapagliflozin or empagliflozin treatment. It is unclear to what extent the reduced arrhythmia burden is due to direct effects of the SGLT2 inhibitors or is solely a marker of improved cardiac function.MethodsOne hundred five rabbit hearts were allocated to eight groups and retrogradely perfused, employing a Langendorff setup. Action potential duration at 90% of repolarization (APD90), QT intervals, effective refractory periods, conduction velocity, and dispersion of repolarization were obtained with monophasic action potential catheters. A model for tachyarrhythmias was established with the IKr blocker erythromycin for QT prolongation associated proarrhythmia as well as the potassium channel opener pinacidil for a short-QT model. An atrial fibrillation (AF) model was created with isoproterenol and acetylcholine. With increasing concentrations of both SGLT2 inhibitors, reductions in QT intervals and APD90 were observed, accompanied by a slight increase in ventricular arrhythmia episodes. During drug-induced proarrhythmia, empagliflozin succeeded in decreasing QT intervals, APD90, and VT burden whereas dapagliflozin demonstrated no significant effects. In the presence of pinacidil induced arrhythmogenicity, neither SGLT2 inhibitor had a significant impact on cardiac electrophysiology. In the AF setting, perfusion with dapagliflozin showed significant suppression of AF in the course of restitution of electrophysiological parameters whereas empagliflozin showed no significant effect on atrial fibrillation incidence.ConclusionIn this model, empagliflozin and dapagliflozin demonstrated opposite antiarrhythmic properties. Empagliflozin reduced ventricular tachyarrhythmias whereas dapagliflozin showed effective suppression of atrial arrhythmias

    Coexistence of adenomyosis uteri and endometrial cancer is associated with an improved prognosis compared with endometrial cancer only

    The present study aimed to identify differences in protein expression in cases of endometrioid endometrial cancer (EEC) with and without coexisting adenomyosis uteri (AM), and to evaluate the histopathological and prognostic distinctions. The total cohort included 22 patients in Group A (patients with concomitant AM and EEC) and 35 patients in Group B (patients affected only by EEC). Evaluation of the following factors was performed: Tumour grade, International Federation of Gynaecology and Obstetrics (FIGO) stage, survival, and expression of estrogen receptor beta (ER beta), glycodelin and inhibin beta B. Group A (AM and EEC) was associated with a lower tumour grade (G1, 90.9 vs. 45.7%;P=0.001) and a lower FIGO stage (FIGO stage I, 100 vs. 80%;P=0.002) compared with Group B (EEC only). In the survival analysis, Group A was associated with a significantly higher 5-year survival rate (95 vs. 82%;P=0.024) than Group B. In addition, the expression of ER beta in Group A was significantly higher (P<0.001), whereas the expression of glycodelin is significantly lower (P=0.028), compared with Group B. The results of the present study indicate that the presence of AM in cases of EEC may be a positive prognostic factor

    Cardiac Myxoma and Cerebrovascular Events: A Retrospective Cohort Study

    Background: Cardiac myxoma (CM) is the most frequent, cardiac benign tumor and is associated with enhanced risk for cerebrovascular events (CVE). Although surgical CM excision is the only curative treatment to prevent CVE recurrence, in recent reports conservative treatment with antiplatelet or anticoagulant agents in high-risk patients with CM-related CVE has been discussed.Methods: Case records at the University Hospital of Tübingen between 2005 and 2017 were screened to identify patients with CM-related CVE. Clinical features, brain and cardiac imaging findings, histological reports, applied treatments and long-term neurological outcomes were assessed.Results: 52 patients with CM were identified and among them, 13 patients with transient ischemic attack, ischemic stroke or retinal ischemia were included to the (to our knowledge) largest reported retrospective study of CM-related CVE. In all identified patients, CVE was the first manifestation of CM; 61% suffered ischemic stroke, 23% transient ischemic attack and 15% retinal ischemia. In 46% of the patients, CVE occurred under antiplatelet or anticoagulation treatment, while 23% of the patients developed recurrent CVE under bridging-antithrombotic-therapy prior to CM surgical excision. Prolonged time interval between CVE and CM-surgery was significantly associated with CVE recurrence (p = 0.021). One patient underwent i.v. thrombolysis, followed by thrombectomy, with good post-interventional outcome and no signs of hemorrhagic transformation.Discussion: Our results suggest that antiplatelet or anticoagulation treatment is no alternative to cardiac surgery in patients presenting with CM-related CVE. We found significantly prolonged time-intervals between CVE and CM surgery in patients with recurrent CVE. Therefore, we suggest that the waiting- or bridging-interval with antithrombotic therapy until curative CM excision should be kept as short as possible. Based on our data and review of the literature, we suggest that in patients with CM-related CVE, i.v. thrombolysis and/or endovascular interventions may present safe and efficacious acute treatments

    Measurement of the top quark forward-backward production asymmetry and the anomalous chromoelectric and chromomagnetic moments in pp collisions at √s = 13 TeV

    Abstract The parton-level top quark (t) forward-backward asymmetry and the anomalous chromoelectric (d̂ t) and chromomagnetic (μ̂ t) moments have been measured using LHC pp collisions at a center-of-mass energy of 13 TeV, collected in the CMS detector in a data sample corresponding to an integrated luminosity of 35.9 fb−1. The linearized variable AFB(1) is used to approximate the asymmetry. Candidate t t ¯ events decaying to a muon or electron and jets in final states with low and high Lorentz boosts are selected and reconstructed using a fit of the kinematic distributions of the decay products to those expected for t t ¯ final states. The values found for the parameters are AFB(1)=0.048−0.087+0.095(stat)−0.029+0.020(syst),μ̂t=−0.024−0.009+0.013(stat)−0.011+0.016(syst), and a limit is placed on the magnitude of | d̂ t| &lt; 0.03 at 95% confidence level.
