    PRODUCTION PERFORMANCE OF SOME WHEAT VARIETIES IN THE PEDOCLIMATE CONDITIONS FROM SCDA CARACAL

    Knowing the particularities of the reaction of the new varieties and hybrids of the crop plants, under different conditions of experimentation is one of the basic activities of the experimental agricultural research and its aim is their judicious choice for cultivation in certain areas. The paper presents the production results obtained from two Romanian varieties of autumn wheat grown under pedoclimatic conditions at SCDA Caracal, under non-irrigation regime, during three years of experimentation. Taking into consideration the variations of the climatic conditions over the years of experimentation, at the same level of fertilization, the productions obtained for the two varieties of wheat recommend their cultivation in the area of the Caracal Plain

    Soft skills and psychological well-being: A study on Italian urban and rural NEETs

    Soft skills retain a certain importance in fully understanding the NEET phenomenon: however, only few researchers have focused on them specifically. The aim of this work is two-fold: a) to detect the differences in terms of soft skills and psychological well-being between urban and rural NEETs; and b) to evaluate which of the soft skills analysed may be predictors of psychological well-being. A sample of young 6998 18−34 years old representative of the Italian population was used. Although gender and educational attainment play a key role in determining NEET status, the degree of urbanisation must be considered because it appears to influence the well-being and perceived soft skills of a group of NEETs. The present study shows that females with low educational attainment residing in rural areas have lower levels of well-being than females with low educational attainment residing in urban areas. A similar influence exists in relation to one particular soft skill: positive vision. Furthermore, soft skills predict psychological well-being wherein degree of urbanisation and gender seems to play a determining role.     STUDIES ON SOME QUALITY COMPONENTS IN A LANDRACE COLLECTION OF CLIMBING BEANS (PHASEOLUS VULGARIS VAR. COMMUNIS)

    The study aimed at evaluating a landraces collection of climbing beans regarding some characters that contribute to quality of grains or green pods. The biological material was formed of 56 landraces and 2 varieties of climbing beans from which the grains or green pods can be used in food. The landraces were collected from western and southwestern Romania. The experimentation was performed over three years in a collection-type experience, arranged in three repetitions. As elements of quality we determined: the percentage of husks from beans, boiling coefficient, the percentage of bean total protein, 1000 grain weight, the percentage of sugar in green pods. The experimental data were processed by analysis of variance and applying the t-test for determination of differences from the variety ‘Aurie de Bacau’ used as a control. The collection includes precious populations for all studied characters, but their number is reduced. For the percentage of grain husks and the protein content of grain, the most populations are below the control variety. For sugar percentage in green pod and the coefficient of boiling, the landraces are similar to control variety. In the collection, there are populations that can be processed by selection or can be used as parents in hybridization programs

    Interface Analysis of the Complex between ERK2 and PTP-SL

    The activity of ERK2, an essential component of MAP-kinase pathway, is under the strict control of various effector proteins. Despite numerous efforts, no crystal structure of ERK2 complexed with such partners has been obtained so far. PTP-SL is a major regulator of ERK2 activity. To investigate the ERK2–PTP-SL complex we used a combined method based on cross-linking, MALDI-TOF analysis, isothermal titration calorimetry, molecular modeling and docking. Hence, new insights into the stoichiometry, thermodynamics and interacting regions of the complex are obtained and a structural model of ERK2-PTP-SL complex in a state consistent with PTP-SL phosphatase activity is developed incorporating all the experimental constraints available at hand to date. According to this model, part of the N-terminal region of PTP-SL has propensity for intrinsic disorder and becomes structured within the complex with ERK2. The proposed model accounts for the structural basis of several experimental findings such as the complex-dissociating effect of ATP, or PTP-SL blocking effect on the ERK2 export to the nucleus. A general observation emerging from this model is that regions involved in substrate binding in PTP-SL and ERK2, respectively are interacting within the interface of the complex

    C-Terminus Glycans with Critical Functional Role in the Maturation of Secretory Glycoproteins

    The N-glycans of membrane glycoproteins are mainly exposed to the extracellular space. Human tyrosinase is a transmembrane glycoprotein with six or seven bulky N-glycans exposed towards the lumen of subcellular organelles. The central active site region of human tyrosinase is modeled here within less than 2.5 Å accuracy starting from Streptomyces castaneoglobisporus tyrosinase. The model accounts for the last five C-terminus glycosylation sites of which four are occupied and indicates that these cluster in two pairs - one in close vicinity to the active site and the other on the opposite side. We have analyzed and compared the roles of all tyrosinase N-glycans during tyrosinase processing with a special focus on the proximal to the active site N-glycans, s6:N337 and s7:N371, versus s3:N161 and s4:N230 which decorate the opposite side of the domain. To this end, we have constructed mutants of human tyrosinase in which its seven N-glycosylation sites were deleted. Ablation of the s6:N337 and s7:N371 sites arrests the post-translational productive folding process resulting in terminally misfolded mutants subjected to degradation through the mannosidase driven ERAD pathway. In contrast, single mutants of the other five N-glycans located either opposite to the active site or into the N-terminus Cys1 extension of tyrosinase are temperature-sensitive mutants and recover enzymatic activity at the permissive temperature of 31°C. Sites s3 and s4 display selective calreticulin binding properties. The C-terminus sites s7 and s6 are critical for the endoplasmic reticulum retention and intracellular disposal. Results herein suggest that individual N-glycan location is critical for the stability, regional folding control and secretion of human tyrosinase and explains some tyrosinase gene missense mutations associated with oculocutaneous albinism type I

    Long-Term Relations Among Prosocial-Media Use, Empathy, and Prosocial Behavior

    Despite recent growth of research on the effects of prosocial media, processes underlying these effects are not well understood. Two studies explored theoretically relevant mediators and moderators of the effects of prosocial media on helping. Study 1 examined associations among prosocial- and violent-media use, empathy, and helping in samples from seven countries. Prosocial-media use was positively associated with helping. This effect was mediated by empathy and was similar across cultures. Study 2 explored longitudinal relations among prosocial-video-game use, violent-video-game use, empathy, and helping in a large sample of Singaporean children and adolescents measured three times across 2 years. Path analyses showed significant longitudinal effects of prosocial- and violent-video-game use on prosocial behavior through empathy. Latent-growth-curve modeling for the 2-year period revealed that change in video-game use significantly affected change in helping, and that this relationship was mediated by change in empathy

    The consolidated European synthesis of CO2 emissions and removals for the European Union and United Kingdom: 1990-2018

    Reliable quantification of the sources and sinks of atmospheric carbon dioxide (CO2), including that of their trends and uncertainties, is essential to monitoring the progress in mitigating anthropogenic emissions under the Kyoto Protocol and the Paris Agreement. This study provides a consolidated synthesis of estimates for all anthropogenic and natural sources and sinks of CO2 for the European Union and UK (EU27 + UK), derived from a combination of state-of-the-art bottom-up (BU) and top-down (TD) data sources and models. Given the wide scope of the work and the variety of datasets involved, this study focuses on identifying essential questions which need to be answered to properly understand the differences between various datasets, in particular with regards to the less-well-characterized fluxes from managed ecosystems. The work integrates recent emission inventory data, process-based ecosystem model results, data-driven sector model results and inverse modeling estimates over the period 1990-2018. BU and TD products are compared with European national greenhouse gas inventories (NGHGIs) reported under the UNFCCC in 2019, aiming to assess and understand the differences between approaches. For the uncertainties in NGHGIs, we used the standard deviation obtained by varying parameters of inventory calculations, reported by the member states following the IPCC Guidelines. Variation in estimates produced with other methods, like atmospheric inversion models (TD) or spatially disaggregated inventory datasets (BU), arises from diverse sources including within-model uncertainty related to parameterization as well as structural differences between models. In comparing NGHGIs with other approaches, a key source of uncertainty is that related to different system boundaries and emission categories (CO2 fossil) and the use of different land use definitions for reporting emissions from land use, land use change and forestry (LULUCF) activities (CO2 land). At the EU27 + UK level, the NGHGI (2019) fossil CO2 emissions (including cement production) account for 2624 Tg CO2 in 2014 while all the other seven bottom-up sources are consistent with the NGHGIs and report a mean of 2588 (± 463 Tg CO2). The inversion reports 2700 Tg CO2 (± 480 Tg CO2), which is well in line with the national inventories. Over 2011-2015, the CO2 land sources and sinks from NGHGI estimates report-90 Tg C yr-1 ± 30 Tg C yr-1 while all other BU approaches report a mean sink of-98 Tg C yr-1 (± 362 Tg of C from dynamic global vegetation models only). For the TD model ensemble results, we observe a much larger spread for regional inversions (i.e., mean of 253 Tg C yr-1 ± 400 Tg C yr-1). This concludes that (a) current independent approaches are consistent with NGHGIs and (b) their uncertainty is too large to allow a verification because of model differences and probably also because of the definition of "CO2 flux"obtained from different approaches.
