Distinct Microglial Responses in Two Transgenic Murine Models of TAU Pathology

Microglial cells are crucial players in the pathological process of neurodegenerative diseases, such as Alzheimer’s disease (AD). Microglial response in AD has been principally studied in relation to amyloid-beta pathology but, comparatively, little is known about inflammatory processes associated to tau pathology. In the hippocampus of AD patients, where tau pathology is more prominent than amyloid-beta pathology, a microglial degenerative process has been reported. In this work, we have directly compared the microglial response in two different transgenic tau mouse models: ThyTau22 and P301S. Surprisingly, these two models showed important differences in the microglial profile and tau pathology. Where ThyTau22 hippocampus manifested mild microglial activation, P301S mice exhibited a strong microglial response in parallel with high phospho-tau accumulation. This differential phospho-tau expression could account for the different microglial response in these two tau strains. However, soluble (S1) fractions from ThyTau22 hippocampus presented relatively high content of soluble phospho-tau (AT8-positive) and were highly toxic for microglial cells in vitro, whereas the correspondent S1 fractions from P301S mice displayed low soluble phosphotau levels and were not toxic for microglial cells. Therefore, not only the expression levels but the aggregation of phospho-tau should differ between both models. In fact, most of tau forms in the P301S mice were aggregated and, in consequence, forming insoluble tau species. We conclude that different factors as tau mutations, accumulation, phosphorylation, and/or aggregation could account for the distinct microglial responses observed in these two tau models. For this reason, deciphering the molecular nature of toxic tau species for microglial cells might be a promising therapeutic approach in order to restore the deficient immunological protection observed in AD hippocampus.CIBERNEDJunta de Andalucía. The aim of this study was to analyze the relationships between physical activity, levels of mental health, and vulnerability to mental disorders. We performed a cross-sectional study with a sample of 1422 people from the Region of Madrid, aged between 15 and 74, assessing levels of physical activity and mental health through questionnaires GPAQv2 (Global Physical Activity Questionnaire Version 2) and GHQ-12 (General Health Questionnaire), respectively. The results reveal that individuals with high or moderate total and recreational physical activity had higher levels of mental health than those who had a low level of physical activity. Furthermore, the level of physical activity during leisure time showed an inverse relationship with vulnerability to mental disorders.O objectivo deste trabalho foi analisar as relações entre a actividade física, os níveis de saúde mental e a vulnerabilidade aos transtornos mentais.

    Get PDF

Influence of tool geometry and numerical parameters when modeling orthogonal cutting of LFRP composites

    Get PDF
The first objective of this paper is to analyze the influence of mesh size and shape in finite element modeling of composite cutting. Also the influence of the level of energy needed to reach complete breakage of the element is considered. The statement of this level of energy is crucial to simulate the material behavior. On the other hand geometrical characteristics of the tool have significant influence on machining processes. The second objective of the present work is to advance in the knowledge concerning tool geometry and its effect in composite cutting. A two-dimensional finite element model of orthogonal cutting has been developed and validated for Glass LFRP composite, comparing with experimental results presented in scientific literature. It was demonstrated that both numerical parameters and tool geometry influence the predicted chip morphology and machining induced damage. The authors are indebted for the financial support of this work, to the Ministry of Science and Innovation of Spain (under Project DPI2008 06746)Publicad

AEMIX: semantic verification of weather forecasts on the web

    Get PDF
Ponencia presentada en: 12th International Conference on Web Information Systems and Technologies celebrada en Roma del 23 al 25 de abril de 2016The main objectives of a meteorological service are the development, implementation and delivery of weather forecasts. Weather predictions are broadcasted to society through different channels, i.e. newspaper, television, radio, etc. Today, the use of theWeb through personal computers and mobile devices stands out. The forecasts, which can be presented in numerical format, in charts, or in written natural language, have a certain margin of error. Providing automatic tools able to assess the precision of predictions allows to improve these forecasts, quantify the degree of success depending on certain variables (geographic areas, weather conditions, time of year, etc.), and focus future work on areas for improvement that increase such accuracy. Despite technological advances, the task of verifying forecasts written in natural language is still performed manually by people in many cases, which is expensive, time-consuming, and subjected to human errors. On the other hand, weather forecasts usually follow several conventions in both structure and use of language, which, while not completely formal, can be exploited to increase the quality of the verification. In this paper, we describe a methodology to quantify the accuracy of weather forecasts posted on the Web and based on natural language. This work obtains relevant information from weather forecasts by using ontologies to capture and take advantage of the structure and language conventions. This approach is implemented in a framework that allows to address different types of predictions with minimal effort. Experimental results with real data are promising, and most importantly, they allow direct use in a real meteorological service. This research work has been supported by the CICYT project TIN2013-46238-C4-4-R, and DGAFS

Global localization based on a rejection differential evolution filter

    Get PDF
Autonomous systems are able to move from one point to another in a given environment because they can solve two basic problems: the localization problem and the navigation problem. The localization purpose is to determine the current pose of the autonomous robot or system and the navigation purpose is to find out a feasible path from the current pose to the goal point that avoids any obstacle present in the environment. Obviously, without a reliable localization system it is not possible to solve the navigation problem. Both problems are among the oldest problems in human travels and have motivated a considerable amount of technological advances in human history. They are also present in robot motion around the environment and have also motivated a considerable research effort to solve them in an efficient way

Multi-objective optimization analysis of cutting parameters when drilling composite materials with special geometry drills

    Get PDF
The cutting tool geometry is strongly influencing damage induced during drilling of composite materials. The searching of new and optimum geometries reducing the damage on the laminates is still a challenge for the scientific community and industry. This study focuses on drilling woven CFRP laminates with four different tool geometries, analyzing the influence of the cutting parameters on the cutting forces and delamination damage. The work is divided in three phases; the first phase carries out a full factorial design of experiments conducted to quantify the significance of each process parameter as well as its interaction, on the generation of delamination at the hole entry and exit as well as the thrust force and torque. The second phase uses the response surface methodology (RSM) to establish the relationships between each output variable and the input variables based on ANOVA results. Finally, a multi-objective optimization strategy has been presented using the fitting equations to select optimum ranges of design parameters that can minimize collectively the aforementioned output variables. Ultimate objective is the process improvement toward negligible defects during drilling of woven CFRP composites. The researchers are indebted to the Ministerio de Economía y Empresa de España (Projects DPI2017-89197-C2-1-R and DPI2017-89197-C2-2-R) for the financial support which permitted to conduct part of this work as well as the Ministerio the Ciencia, Innovación y Universidades (Juan de la Cierva program with the reference FJCI-2017-34910
