    Altered antioxidant status in peripheral skeletal muscle of patients with COPD

    AbstractDespite the growing field of interest in the role of pulmonary oxidative stress in chronic obstructive pulmonary disease (COPD), barely any data are available with respect to antioxidant capacity in the peripheral musculature of these patients. The main objective of this study was to assess in detail the antioxidant status in skeletal muscle of patients with COPD. Biopsies from the vastus lateralis of 21 patients with COPD and 12 healthy age-matched controls were analysed. Total antioxidant capacity, vitamin E, glutathione, and uric acid levels were determined and the enzyme activities of superoxide dismutase, glutathione reductase, glutathione peroxidase, and glutathione-S-transferase were measured. Malondialdehyde was measured as an index of lipid peroxidation. The total antioxidant capacity and the uric acid levels were markedly higher in COPD patients than in healthy controls (25%, P=0.006 and 24%, P=0.029, respectively). Glutathione-S-transferase activity was also increased (35%; P=0.044) in patients compared to healthy subjects. Vitamin E level was lower in patients than in controls (P<0.05). The malondialdehyde level was not different between the two groups. It can be concluded that the muscle total antioxidant capacity is increased in patients with COPD. Together with the reduced vitamin E levels, the increased glutathione-S-transferase activity and normal levels of lipid peroxidation products, these findings suggest that the antioxidant system may be exposed to and subsequently triggered by elevated levels of reactive oxygen species

    Evaluation of patients treated with natalizumab for progressive multifocal leukoencephalopathy

    Background: Progressive multifocal leukoencephalopathy (PML) was reported to have developed in three patients treated with natalizumab. We conducted an evaluation to determine whether PML had developed in any other treated patients. Methods: We invited patients who had participated in clinical trials in which they received recent or long-term treatment with natalizumab for multiple sclerosis, Crohn's disease, or rheumatoid arthritis to participate. The clinical history, physical examination, brain magnetic resonance imaging (MRI), and testing of cerebrospinal fluid for JC virus DNA were used by an expert panel to evaluate patients for PML. We estimated the risk of PML in patients who completed at least a clinical examination for PML or had an MRI. Results: Of 3417 patients who had recently received natalizumab while participating in clinical trials, 3116 (91 percent) who were exposed to a mean of 17.9 monthly doses underwent evaluation for PML. Of these, 44 patients were referred to the expert panel because of clinical findings of possible PML, abnormalities on MRI, or a high plasma viral load of JC virus. No patient had detectable JC virus DNA in the cerebrospinal fluid. PML was ruled out in 43 of the 44 patients, but it could not be ruled out in one patient who had multiple sclerosis and progression of neurologic disease because data on cerebrospinal fluid testing and follow-up MRI were not available. Only the three previously reported cases of PML were confirmed (1.0 per 1000 treated patients; 95 percent confidence interval, 0.2 to 2.8 per 1000). Conclusions: A detailed review of possible cases of PML in patients exposed to natalizumab found no new cases and suggested a risk of PML of roughly 1 in 1000 patients treated with natalizumab for a mean of 17.9 months.     New Vesicle-polymer Hybrids: the parachute architecture

    The influence of organized media on polymerization reactions results in many cases in interesting morphologies of the polymeric material. In the present study, vesicle bilayers were used as ordered medium for the free radical polymerization of styrene. Cryo-electron microscopy gives evidence that the polymerization induces phase-separation phenomena leading to parachute-like morphologies. On the basis of general knowledge about vesicles and polymerizations in heterogeneous media, explanations for the observed phenomena are given. Bearing in mind that vesicles are outstanding models for membrane mimetic chemistry, it becomes evident that these findings can be relevant to the investigation of, for example, membrane−protein interactions

    An outflow in the Seyfert ESO 362-G18 revealed by Gemini-GMOS/IFU observations

    Indexación: Scopus.We present two-dimensional stellar and gaseous kinematics of the inner 0.7 × 1.2 kpc2 of the Seyfert 1.5 galaxy ESO 362-G18, derived from optical (4092-7338 Å) spectra obtained with the GMOS integral field spectrograph on the Gemini South telescope at a spatial resolution of ≈ 170 pc and spectral resolution of 36 km s-1. ESO 362-G18 is a strongly perturbed galaxy of morphological type Sa or S0/a, with a minor merger approaching along the NE direction. Previous studies have shown that the [O III] emission shows a fan-shaped extension of ≈ 10′′ to the SE. We detect the [O III] doublet, [N II] and Hα emission lines throughout our field of view. The stellar kinematics is dominated by circular motions in the galaxy plane, with a kinematic position angle of ≈ 137° and is centred approximately on the continuum peak. The gas kinematics is also dominated by rotation, with kinematic position angles ranging from 122° to 139°, projected velocity amplitudes of the order of 100 km s-1, and a mean velocity dispersion of 100 km s-1. A double-Gaussian fit to the [O III]λ5007 and Hα lines, which have the highest signal to noise ratios of the emission lines, reveal two kinematic components: (1) a component at lower radial velocities which we interpret as gas rotating in the galactic disk; and (2) a component with line of sight velocities 100-250 km s-1 higher than the systemic velocity, interpreted as originating in the outflowing gas within the AGN ionization cone. We estimate a mass outflow rate of 7.4 × 10-2 M⊙ yr-1 in the SE ionization cone (this rate doubles if we assume a biconical configuration), and a mass accretion rate on the supermassive black hole (SMBH) of 2.2 × 10-2 M⊙ yr-1. The total ionized gas mass within ∼84 pc of the nucleus is 3.3 × 105 M⊙; infall velocities of ∼34 km s-1 in this gas would be required to feed both the outflow and SMBH accretion. © ESO 2018.https://www.aanda.org/articles/aa/abs/2018/06/aa31671-17/aa31671-17.htm

    The effect of substrate clamping on the paraelectric to antiferroelectric phase transition in Nd-doped BiFeO₃ thin films

    Thin films were deposited on Pt/Ti/SiO₂/Si substrates using pulsed laser deposition from a target with a composition (Bi₀.₈₂₅Nd₀.₁₇₅Fe₀.₉₇Ti₀.₀₃O₃) with 5 mol% excess Bi₂O₃ within the antiferroelectric (AFE) region of the NdFeO₃-BiFeO₃ phase diagram. However, Raman spectroscopy and transmission electron microscopy (TEM) revealed that films consisted of a mosaic microstructure in which (AFE), ferroelectric (FE) and paraelectric (PE) phases coexisted. Variation in the spatial distribution of Nd is typically greater in bulk ceramics than in thin films and therefore, the absence of single phase AFE cannot be attributed to local changes in composition. Instead, it is proposed that clamping due to mismatch in thermal expansion coefficient with the substrate suppresses the large volume change associated with the PE-FE and PE-AFE transition in bulk and its absence in the thin film prevents an avalanche-like transition throughout grains, which in bulk sustains single phase AFE, irrespective of local deviations in the Nd concentration

    Gravity wave analogs of black holes

    It is demonstrated that gravity waves of a flowing fluid in a shallow basin can be used to simulate phenomena around black holes in the laboratory. Since the speed of the gravity waves as well as their high-wavenumber dispersion (subluminal vs. superluminal) can be adjusted easily by varying the height of the fluid (and its surface tension) this scenario has certain advantages over the sonic and dielectric black hole analogs, for example, although its use in testing quantum effects is dubious. It can be used to investigate the various classical instabilities associated with black (and white) holes experimentally, including positive and negative norm mode mixing at horizons.
