    Combining high pressure and electric fields towards nannochloropsis oculata eicosapentaenoic acid-rich extracts

    Nannochloropsis oculata is naturally rich in eicosapentaenoic acid (EPA). To turn this microalga into an economically viable source for commercial applications, extraction efficiency must be achieved. Pursuing this goal, emerging technologies such as high hydrostatic pressure (HHP) and moderate electric fields (MEF) were tested, aiming to increase EPA accessibility and subsequent extraction yields. The innovative approach used in this study combined these technologies and associated tailored, less hazardous different solvent mixtures (SM) with distinct polarity indexes. Although the classical Folch SM with chloroform: methanol (PI 4.4) provided the highest yield concerning total lipids (166.4 mglipid/gbiomass), diethyl ether: ethanol (PI 3.6) presented statistically higher values in terms of EPA per biomass, corresponding to 1.3-fold increase. When SM were used in HHP and MEF, neither technology independently improved EPA extraction yields, although the sequential combination of technologies did result in 62% increment in EPA extraction. Overall, the SM and extraction methodologies tested (HHP—200 MPa, 21 °C, 15 min, followed by MEF processing at 40 °C, 15 min) enabled increased EPA extraction yields from wet N. oculata biomass. These findings are of high relevance for the food and pharmaceutical industries, providing viable alternatives to the “classical” extraction methodologies and solvents, with increased yields and lower environmental impact.info:eu-repo/semantics/publishedVersio

    The compound Poisson limit ruling periodic extreme behaviour of non-uniformly hyperbolic dynamics

    We prove that the distributional limit of the normalised number of returns to small neighbourhoods of periodic points of non-uniformly hyperbolic dynamical systems is compound Poisson. The returns to small balls around a fixed point in the phase space correspond to the occurrence of rare events, or exceedances of high thresholds, so that there is a connection between the laws of Return Times Statistics and Extreme Value Laws. The fact that the fixed point in the phase space is a repelling periodic point implies that there is a tendency for the exceedances to appear in clusters whose average sizes is given by the Extremal Index, which depends on the expansion of the system at the periodic point. We recall that for generic points, the exceedances, in the limit, are singular and occur at Poisson times. However, around periodic points, the picture is different: the respective point processes of exceedances converge to a compound Poisson process, so instead of single exceedances, we have entire clusters of exceedances occurring at Poisson times with a geometric distribution ruling its multiplicity. The systems to which our results apply include: general piecewise expanding maps of the interval (Rychlik maps), maps with indifferent fixed points (Manneville-Pomeau maps) and Benedicks-Carleson quadratic maps.Comment: To appear in Communications in Mathematical Physic

    Sweet whey cheese matrices inoculated with the probiotic strain Lactobacillus paracasei LAFTI® L26

    Consumption of dairy products containing viable probiotic strains has increased dramatically in recent years, owing to general health claims associated therewith. This trend has boosted diversification of the portfolio of said products, including whey cheese matrices. However, taking into account the relatively poor organoleptic and textural features of these matrices, improvement is in order via incorporation of selected additives, provided that viability of the strains is duly assayed. Lactobacillus paracasei LAFTI® L26 was accordingly incorporated into whey protein solid matrices, in the presence of several additives aimed at enhancing their organoleptic appeal and textural performance. These matrices were produced from a combination of either ovine or bovine whey (or a mixture thereof) with ovine milk, and were inoculated at 10% (v/v) with the probiotic strain. Sugar, sugar and aloe vera, sugar and chocolate, and sugar and jam were further added, and the resulting products were then stored at 7 ◦C for 21 d. In general, viable cell numbers remained high in all experimental matrices throughout storage. Despite the observed low extents of breakdown, proteolytic activities by the end of storage were higher in matrices containing jam. Furthermore, L. paracasei partially converted lactose into lactic acid in these matrices. Additives enhanced the organoleptic features of whey cheeses, and produced different textural patterns. The higher sensory scores were attained by matrices containing sugar: sugar and aloe vera received the best scores by 3 d of storage, but these scores decreased as storage time elapsed

    Diversity of plant growth-promoting bacteria associated with sugarcane

    The sugarcane (Saccharum spp) presents economic importance, mainly for tropical regions, being an important Brazilian commodity. However, this crop is strongly dependent on fertilizers, mainly nitrogen (N). This study assessed the plant growth-promoting bacteria (PGPB) associated with sugarcane that could be used as a potential inoculant to the crop. We evaluated the genetic diversity of PGPB in the plant tissue of sugarcane varieties (RB 867515, RB 1011, and RB 92579). The primer BOX-A1R was used to differentiate the similar isolated and further sequencing 16S rRNA ribosomal gene. The 16S rRNA gene showed the presence of seven different genera distributed into four groups, the genus Bacillus, followed by Paenibacillus (20%), Burkholderia (14%), Herbaspirillum (6%), Pseudomonas (6%), Methylobacterium (6%), and Brevibacillus (3%). The molecular characterization of endophytic isolates from sugarcane revealed a diversity of bacteria colonizing this plant, with a possible biotechnological potential to be used as inoculant and biofertilizers

    Sea bass (Dicentrarchus labrax) : a model organism for the screening of estrogenic chemicals in marine surface waters?

Estrogenic responses in fish are the net result of complex chains of events involving the uptake, distribution and metabolism of test agents until they interact with their target sites. Typically these aspects cannot be modelled in short-term cell-based assays, only studies with vertebrates offer the opportunity to assess potential interactions of test compounds at higher organisational levels. However, studies with endocrine disrupting chemicals have been performed mainly with freshwater organisms. The sensitivity of a marine fish species to different estrogenic chemicals was investigated under chronic exposure conditions. This work is part of a study focusing on the combination effects of mixtures of estrogenic chemicals in marine and freshwater organisms (ACE, EVK1-CT-2001-100). As test organism the sea bass (Dicentrarchus labrax) was selected, a common species in European marine systems. Juveniles were exposed under a flow-through system for 14 days for a set of reference chemicals (17Ã -estradiol, ethynylestradiol, nonylphenol, octylphenol, bisphenol A). Effects at subcellular level were analysed using vitellogenesis as endpoint. Its relevance is evaluated by further investigations about the individual fitness (condition factor, hepatossomatic index), as well as the liver cytochrome P450 activity.     Sea bass (Dicentrarchus labrax) as a potential bioindicator of estrogenic contamination in marine surface waters

    Resumo apresentado sob poster apresentado ao 5th International Symposium on Fish Endocrinology, September, 2004, Castellon, Spain.Exposure of aquatic wildlife in surface waters to (xeno-)estrogens is known to cause reproductive dysfunction. Estrogenic responses in fish are the net result of complex chains of events that will depend on a number of factors, such as bioavailability, bioconcentration/bioaccumulation, and biotransformation. Most of known estrogenic chemicals are lipophilic and hydrophobic and therefore have a strong potential to accumulate in aquatic biota. Therefore, determining environmental exposures may be very difficult and not be particularly meaningful. As test organism the sea bass (Dicentrarchus labrax) was selected, a common species in European marine systems. This work is part of a study focusing on the combination effects of mixtures of estrogenic chemicals in marine and freshwater organisms. Juvenile sea bass were used in order to analyse the bioconcentration and distribution among different tissues of the chemical residues of a set of reference estrogenic chemicals such as 17ß-estradiol (E2), ethynylestradiol (EE2), nonylphenol (NP), octylphenol (OP), bisphenol-A (BPA). Fish were exposed for a period of two weeks to environmentally relevant levels of these compounds, after which liver, bile, muscle, gill and kidney were collected and analyzed. Actual concentrations of E2, EE2 and BPA seawater in the tanks were determined by either gas chromatography with ion trap detection or HPLC coupled to diode array detection. In bile, levels of BPA were determined according to a method presented earlier by Houtman et al. (13th Annual Meeting SETAC Europe, 2003). Actual NP and OP concentrations in both water and tissues were determined by HPLC-ESI-MS according to recently developed methods by Pojana et al. (J. Anal. Chem., in press). Bioconcentration and distribution of residual compounds in tissues were correlated to the levels of plasma vitellogenin (results are presented also at this conference) and to actual exposure concentrations.     Neuromuscular assessment of a stand-up paddle stroke

    his study analyzed muscle activity during the stand-up paddle stroke, considering the paddling side and the adjacent and opposing muscles relative to the position of the arms during paddling. Methods: Fourteen male paddleboarders performed three trials covering 195 m in which surface electromyography of the upper trapezius, biceps brachii, triceps brachii, tibialis anterior, and gastrocnemius medialis were recorded (four-cycle strokes on each side). The data were processed according to percentage of maximum voluntary contraction (% MVC). The MVC activation values (µV) for each muscle were then calculated and presented as percentage MVC (% MVC). Results: The recovery phase accounted for 60% of the paddle cycle, while the pull phase represented 39%. During right-side paddling, higher % MVC was found in the opposite-side upper trapezius (24.35%, p < 0.01) during the pulling phase and in the adjacent biceps brachii (8.36%, p < 0.03) during the recovery phase. In left-side paddling, greater % MVC was found in the opposite-side upper trapezius (27.60%, p < 0.01) during the pulling phase and in the opposite-side triceps brachii (42.25%, p < 0.04) during the recovery phase. Furthermore, the pulling phase exhibited higher MVC in the opposite-side upper trapezius compared to the recovery phase, both in the right-side (24.35%, p < 0.03) and left-side (27.60%, p < 0.01) paddling. Conclusions: these findings help establish the muscular activity of both sides of the paddling technique and the differences between the upper and lower limbs.info:eu-repo/semantics/publishedVersio

    Perspetivas complementares de valorização do património em pedra em Almeida (Distrito da Guarda)

    Pode considerar-se que a sua situação da vila de Almeida está enquadrada dentro dos princípios que definem a área temática "Cidade do presente" deste congresso, apresentando um processo de perda por efeito de mudanças estratégicas nacionais, sofrendo, desta forma, de problemas semelhantes. Esta situação promove processos de abandono que podem ter implicações negativas para as povoações e para o cuidado do património histórico, potencialmente com efeitos negativos no ambiente natural circundante. Neste trabalho, com base no caso de estudo de Almeida, pretendemos apresentar e discutir algumas perspetivas de valorização do património em pedra complementares dos aspetos mais históricos e artísticos. A comunicação irá incidir sobre questões de Engenharia, materiais geológicos, processos análogos de processos geológicos, assim como o desenvolvimento de organismos no património construído.
