    Global analysis of the sugarcane microtranscriptome reveals a unique composition of small RNAs associated with axillary bud outgrowth

    Axillary bud outgrowth determines shoot architecture and is under the control of endogenous hormones and a fine-tuned gene-expression network, which probably includes small RNAs (sRNAs). Although it is well known that sRNAs act broadly in plant development, our understanding about their roles in vegetative bud outgrowth remains limited. Moreover, the expression profiles of microRNAs (miRNAs) and their targets within axillary buds are largely unknown. Here, we employed sRNA next-generation sequencing as well as computational and gene-expression analysis to identify and quantify sRNAs and their targets in vegetative axillary buds of the biofuel crop sugarcane (Saccharum spp.). Computational analysis allowed the identification of 26 conserved miRNA families and two putative novel miRNAs, as well as a number of trans-acting small interfering RNAs. sRNAs associated with transposable elements and protein-encoding genes were similarly represented in both inactive and developing bud libraries. Conversely, sequencing and quantitative reverse transcription-PCR results revealed that specific miRNAs were differentially expressed in developing buds, and some correlated negatively with the expression of their targets at specific stages of axillary bud development. For instance, the expression patterns of miR159 and its target GAMYB suggested that they may play roles in regulating abscisic acid-signalling pathways during sugarcane bud outgrowth. Our work reveals, for the first time, differences in the composition and expression profiles of diverse sRNAs and targets between inactive and developing vegetative buds that, together with the endogenous balance of specific hormones, may be important in regulating axillary bud outgrowth

    Avaliação comparativa da eficácia de fitoterápicos e produtos químicos carrapaticidas no controle do Boophilus micropulus (Canestrini, 1887) por meio do carrapaticidograma

    Neste estudo objetivou-se avaliar a  eficácia de fitoterápicos e produtos químicos carrapaticidas no controle do Boophilus microplus. Foram avaliados, in vitro, três produtos comerciais para o controle de Boophilus microplus, sendo dois à base de cipermetrina + DDVP, um à base de triclorfon + coumafós + ciflutrin e quatro extratos de produtos vegetais,  Cymbopogon  citratus (capim santo),  Lippia  alba (erva cidreira),  Ipomoea  asarifolia (salsa) e o Azadirachta  indica (óleo de nim a 1%), em três diferentes concentrações. Os resultados revelaram eficácia inferior a 95% para os produtos químicos testados. Os fitoterápicos apresentaram indícios de atividade biológica na mortalidade de fêmeas ingurgitadas e inibição de eclosão, porém, o Azadirachta  indica alcançou índices de eficácia superiores a 95% em todas as concentrações testadas, sendo a melhor opção dentre os princípios ativos testados neste experimento.&nbsp

    PHYTOCHEMICAL PROFILE WITH ANTI-TUMOR ACTIVITY ESTIMATION OF CRUDE EXTRACT, ESSENTIAL OIL AND D-LIMONENE FROM CITRUS AURANTIUM L. AGAINST EHRLICH CARCINOMA

    Objective: Plant based drugs have been a solution in the search for more cost-effective and less harmful drugs for the treatment of neoplasia. Citrus aurantium L. (Rutaceae) is abundant in Brazil and D-limonene, a monoterpene used in the prevention and treatment of neoplasia, was identified as a major compound in the oil of this specie. Objective of current study includes estimation of anti-tumor activity of Citrus aurantium L. (Rutaceae) (crude extract, essential oil and D-limonene) against Ehrlich carcinoma, as well as their phytochemical evaluation (D-limonene and essential oil). Methods: There was a randomized non-clinical trial in which were used adult male mice (Balb-C). Four groups of animals were used having 6 numbers of animal in each group. All groups were inoculated with the Ehrlich tumor and then received the treatment (control, crude extract, essential oil and D-limonene) by oral route daily (28 day treatment). Essential oil was obtained by hydro-distillation and analyzed by the means of GC (Gas Chromatography) that was attached to mass spectrometry. In last of the observations  hemogram was obtained. Results: Animals treated with the essential oil has shown no significant difference compared to the group treated with D-limonene. The group treated with crude extract had a growth inhibition close to the essential oil and D-limonene groups. Conclusion: It´s concluded that the essential oil and the crude extract of Citrus aurantium, L. (Rutaceae) can become therapeutic agents because of their anti-tumor activity with no toxicity to the blood cells and have low cost of production. Further studies are necessary, so they can be used in the treatment of neoplasia in humans.     Determination of Cellular Lipids Bound to Human CD1d Molecules

    CD1 molecules are glycoproteins that present lipid antigens at the cell surface for immunological recognition by specialized populations of T lymphocytes. Prior experimental data suggest a wide variety of lipid species can bind to CD1 molecules, but little is known about the characteristics of cellular ligands that are selected for presentation. Here we have molecularly characterized lipids bound to the human CD1d isoform. Ligands were eluted from secreted CD1d molecules and separated by normal phase HPLC, then characterized by mass spectroscopy. A total of 177 lipid species were molecularly identified, comprising glycerophospholipids and sphingolipids. The glycerophospholipids included common diacylglycerol species, reduced forms known as plasmalogens, lyso-phospholipids (monoacyl species), and cardiolipins (tetraacyl species). The sphingolipids included sphingomyelins and glycosylated forms, such as the ganglioside GM3. These results demonstrate that human CD1d molecules bind a surprising diversity of lipid structures within the secretory pathway, including compounds that have been reported to play roles in cancer, autoimmune diseases, lipid signaling, and cell death

    Geo-environmental terrain assessments and territorial zoning are useful tools for the formulation and implementation of environmental management instruments (including policy-making, planning, and enforcement of statutory regulations). They usually involve a set of procedures and techniques for delimitation, characterisation and classification of terrain units. However, terrain assessments and zoning exercises are often costly and time-consuming, particularly when encompassing large areas, which in many cases prevent local agencies in developing countries from properly benefiting from such assessments. In the present paper, a low-cost technique based on the analysis of texture of satellite imagery was used for delimitation of terrain units. The delimited units were further analysed in two test areas situated in Southeast Brazil to provide estimates of land instability and the vulnerability of groundwater to pollution hazards. The implementation incorporated procedures for inferring the influences and potential implications of tectonic fractures and other discontinuities on ground behaviour and local groundwater flow. Terrain attributes such as degree of fracturing, bedrock lithology and weathered materials were explored as indicators of ground properties.     Transcriptome changes in newborn goats' skeletal muscle as a result of maternal feed restriction at different stages of gestation

    We investigated how feed restriction at 50% of maintenance requirements during different stages of gestation affects the transcriptome of newborn goats' skeletal muscle. Fourteen pregnant dams were randomly assigned into one of the following dietary treatments: animals fed at 50% of maintenance requirement from 8-84 d of gestation and then fed at 100% of maintenance requirement from day 85 of gestation to parturition (RM, n = 6), and animals fed at 100% of maintenance requirement from 8-84 d of gestation and then fed at 50% of maintenance requirement from day 85 of gestation to parturition (MR, n = 8). At birth, samples of offspring's Longissimus muscle were collected for total RNA extraction and sequencing. Our data showed 66 differentially expressed (DE) genes (FDR < 0.05). A total of 6 genes were upregulated and 60 downregulated (FDR < 0.05) in the skeletal muscle of the newborns resulting from treatment RM compared with MR. Our results suggest that the DE genes upregulated in newborn goats' skeletal muscle from the RM group compared to MR, included genes related to satellite cells, and genes that indicates impaired insulin sensitivity and changes in the composition of intramuscular fat. The DE genes upregulated in newborn goats' skeletal muscle from the MR group compared to RM, are also related to impaired insulin sensitivity, as well as a predominantly oxidative metabolism and cellular oxidative stress. However, protective mechanisms against insulin sensitivity and oxidative stress may have been augmented in the skeletal muscle of offspring from MR treatment compared to RM, in order to maintain cellular homeostasis

    Small airway remodeling in acute respiratory distress syndrome: a study in autopsy lung tissue

    Introduction: Airway dysfunction in patients with the Acute Respiratory Distress Syndrome (ARDS) is evidenced by expiratory flow limitation and dynamic hyperinflation. These functional alterations have been attributed to closure/obstruction of small airways. Airway morphological changes have been reported in experimental models of acute lung injury, characterized by epithelial necrosis and denudation in distal airways. To date, however, no study has focused on the morphological airway changes in lungs from human subjects with ARDS. The aim of this study is to evaluate structural and inflammatory changes in distal airways in ARDS patients. Methods: We retrospectively studied autopsy lung tissue from subjects who died with ARDS and from control subjects who died of non pulmonary causes. Using image analysis, we quantified the extension of epithelial changes (normal, abnormal and denudated epithelium expressed as percentages of the total epithelium length), bronchiolar inflammation, airway wall thickness, and extracellular matrix (ECM) protein content in distal airways. The Student`s t test or the Mann-Whitney test was used to compare data between the ARDS and control groups. Bonferroni adjustments were used for multiple tests. The association between morphological and clinical data was analyzed by Pearson rank test. Results: Thirty-one ARDS patients (A: PaO(2)/FiO(2) <= 200, 45 +/- 14 years, 16 males) and 11 controls (C:52 +/- 16 years, 7 males) were included in the study. ARDS airways showed a shorter extension of normal epithelium (A:32.9 +/- 27.2%, C:76.7 +/- 32.7%, P < 0.001), a larger extension of epithelium denudation (A:52.6 +/- 35.2%, C:21.8 +/- 32.1%, P < 0.01), increased airway inflammation (A:1(3), C:0(1), P = 0.03), higher airway wall thickness (A:138.7 +/- 54.3 mu m, C:86.4 +/- 33.3 mu m, P < 0.01), and higher airway content of collagen I, fibronectin, versican and matrix metalloproteinase-9 (MMP-9) compared to controls (P = 0.03). The extension of normal epithelium showed a positive correlation with PaO(2)/FiO(2) (r(2) = 0.34; P = 0.02) and a negative correlation with plateau pressure (r(2) = 0.27; P = 0.04). The extension of denuded epithelium showed a negative correlation with PaO(2)/FiO(2) (r(2) = 0.27; P = 0.04). Conclusions: Structural changes in small airways of patients with ARDS were characterized by epithelial denudation, inflammation and airway wall thickening with ECM remodeling.
