Regulation of RasâMAPK pathway mitogenic activity by restricting nuclear entry of activated MAPK in endoderm differentiation of embryonic carcinoma and stem cells
In response to retinoic acid, embryonic stem and carcinoma cells undergo differentiation to embryonic primitive endoderm cells, accompanied by a reduction in cell proliferation. Differentiation does not reduce the activation of cellular MAPK/Erk, but does uncouple mitogen-activated protein kinase (MAPK) activation from phosphorylation/activation of Elk-1 and results in inhibition of c-Fos expression, whereas phosphorylation of the cytoplasmic substrate p90RSK remains unaltered. Cell fractionation and confocal immunofluorescence microscopy demonstrated that activated MAPK is restricted to the cytoplasmic compartment after differentiation. An intact actin and microtubule cytoskeleton appears to be required for the restriction of MAPK nuclear entry induced by retinoic acid treatment because the cytoskeletal disrupting agents nocodazole, colchicine, and cytochalasin D are able to revert the suppression of c-Fos expression. Thus, suppression of cell proliferation after retinoic acidâinduced endoderm differentiation of embryonic stem and carcinoma cells is achieved by restricting nuclear entry of activated MAPK, and an intact cytoskeleton is required for the restraint
Landscape elements and river chemistry as affected by river regulation â a 3-D perspective
We tested the hypothesis whether individual land classes within a river catchment contribute equally to river loading with dissolved constituents or whether some land classes act as "hot spots" to river loading and if so, are these land classes especially affected by hydrological alterations. The amount of land covered by forests and wetlands and the average soil depth (throughout this paper soil refers to everything overlying bedrock i.e. regolith) of a river catchment explain 58â93% of the variability in total organic carbon (TOC) and dissolved silicate (DSi) concentrations for 22 river catchments in Northern Sweden. For the heavily regulated LuleĂ€lven, with 7 studied sub-catchments, only 3% of the headwater areas have been inundated by reservoirs, some 10% of the soils and aggregated forest and wetland areas have been lost due to damming and further hydrological alteration such as bypassing entire sub-catchments by headrace tunnels. However, looking at individual forest classes, our estimates indicate that some 37% of the deciduous forests have been inundated by the four major reservoirs built in the LuleĂ€lven headwaters. These deciduous forest and wetlands formerly growing on top of alluvial deposits along the river corridors forming the riparian zone play a vital role in loading river water with dissolved constituents, especially DSi. A digital elevation model draped with land classes and soil depths which highlights that topography of various land classes acting as hot spots is critical in determining water residence time in soils and biogeochemical fluxes. Thus, headwater areas of the LuleĂ€lven appear to be most sensitive to hydrological alterations due to the thin soil cover (on average 2.7â4.5 m) and only patchy appearance of forest and wetlands that were significantly perturbed. Hydrological alterations of these relatively small headwater areas significantly impacts downstream flux of dissolved constituents and their delivery to receiving water bodies
Perception of differentiation cues by GATA factors in primitive endoderm lineage determination of mouse embryonic stem cells
AbstractThe formation of the primitive endoderm covering the inner cell mass of early mouse embryos can be simulated in vitro by the differentiation of mouse embryonic stem (ES) cells in culture following either aggregation of suspended cells or stimulation of cell monolayers with retinoic acid. The developmentally regulated transcription factors GATA-4 and GATA-6 have determining role in mouse extraembryonic endoderm development. We analyzed the in vitro differentiation of mouse embryonic stem cells deficient of GATA factors and conclude that GATA-4 is required for ES cells to perceive a cell positioning (cell aggregation) signal and GATA-6 is required to sense morphogenic (retinoic acid) signal. The collaboration between GATA-6 and GATA-4, or GATA-6 and GATA-5 which can substitute for GATA-4, is involved in the perception of differentiation cues by embryonic stem cells in their determination of endoderm lineage. This study indicates that the lineage differentiation of ES cells can be manipulated by the expression of GATA factors
Initial State Dependence of the Breakup of Weakly Bound Carbon Isotopes
The one-neutron nuclear breakup from the Carbon isotopes C and
C, is calculated as an example of application of the theory of transfer
to the continuum reactions in the formulation which includes spin coupling.
The effect of the energy sharing between the parallel and transverse neutron
momentum distributions is taken into account thus resulting in a theory which
is more general than sudden eikonal approaches. Both effects are necessary to
understand properly the breakup from not too weakly bound orbitals.
Breakup which leaves the core into an excited state below particle threshold is
also considered. The core-target interaction is treated in the smooth cut-off
approximation. By comparing to presently available experimental data we show
how to make some hypothesis on the quantum numbers and occupancy of the neutron
initial state. Possible ambiguities in the interpretation of inclusive cross

Curbside Recycling in the U.S.a.: Convenience and Mandatory Participation
This research examines the relationship between the success of a residential curbside recycling program (RCRP), measured as material recovery rate (MRR), and two program factors: (1) whether or not participation is mandated; and (2) convenience, measured by container provision, collection frequency and collection day relative to municipal solid waste collection day. Residential curbside recycling programs, with correct strategies and program design, can be an important part of solid waste management plans world-wide. While residential curbside recycling programs are growing in popularity, many basic design questions lie unanswered and successful program strategies are not always obvious. Data from 357 residential curbside recycling programs in the United States are used to test the hypotheses. Mandatory participation residential curbside recycling programs are seen to collect more material than voluntary participation residential curbside recycling programs. Container provision appears effective for voluntary, but not mandatory, residential curbside recycling programs. Increasing collection frequency appears to have a small positive effect on residential curbside recycling program success, while collection day has little effect on material recovery rate.Yeshttps://us.sagepub.com/en-us/nam/manuscript-submission-guideline
Elucidating halo structure by ÎČ decay: ÎČÎł from the Li-11 decay

Special emphasis is put on the determination of the Gamow-Teller transition Li-11 --> Be-11 (1/2(-), 320 keV) to the only bound excited state in Be-11. Womenâs beliefs about medicines and adherence to pharmacotherapy in pregnancy: Opportunities for community pharmacists?
Background During pregnancy women might weigh benefits of treatment against potential risks to the unborn child. However, non-adherence to necessary treatment can adversely affect both mother and child. To optimize pregnant womenâs beliefs and medication adherence, community pharmacists are ideally positioned to play an important role in primary care. Objective This narrative review aimed to summarize the evidence on 1) pregnant womenâs beliefs, 2) medication adherence in pregnancy, and 3) community pharmacistsâ counselling during pregnancy. Method Three search strategies were used in Medline and Embase to find original studies evaluating womenâs beliefs, medication adherence and community pharmacistsâ counselling during pregnancy. All original descriptive and analytic epidemiological studies performed in Europe, North America and Australia, written in English and published from 2000 onwards were included. Results We included 14 studies reporting on womenâs beliefs, 11 studies on medication adherence and 9 on community pharmacistsâ counselling during pregnancy. Women are more reluctant to use medicines during pregnancy and tend to overestimate the teratogenic risk of medicines. Risk perception varies with type of medicine, level of health literacy, education level and occupation. Furthermore, low medication adherence during pregnancy is common. Finally, limited evidence showed current community pharmacistsâ counselling is insufficient. Barriers hindering pharmacists are insufficient knowledge and limited access to reliable information. Conclusion Concerns about medication use and non-adherence are widespread among pregnant women. Community pharmacistsâ counselling during pregnancy is insufficient. Further education, training and research are required to support community pharmacists in fulfilling all the opportunities they have when counselling pregnant women
