    Supported Evacuation for Disaster Relief through Lexicographic Goal Programming

    Disasters have been striking human-beings from the beginning of history and their management is a global concern of the international community. Minimizing the impact and consequences of these disasters, both natural and human-made, involves many decision and logistic processes that should be optimized. A crucial logistic problem is the evacuation of the affected population, and the focus of this paper is the planning of supported evacuation of vulnerable people to safe places when necessary. A lexicographic goal programming model for supported evacuation is proposed, whose main novelties are the classification of potential evacuees according to their health condition, so that they can be treated accordingly; the introduction of dynamism regarding the arrival of potential evacuees to the pickup points, according to their own susceptibility about the disaster and the joint consideration of objectives such us number of evacuated people, operation time and cost, among which no trade-off is possible. The performance of the proposed model is evaluated through a realistic case study regarding the earthquake and tsunami that hit Palu (Indonesia) in September 2018

    This article describes, albeit briefly, an experience carried out by a colaborative research group in relation to the design, implementation and evaluation of croos-curricular themas within the main subject of Language, for which the action-research methodoly has been used. Afer a brief explanation of the topic and methodology of the research, the stages and procedures for the recolection and analysis of data are described. Finally, some of the conclusions of this experience about the conceptualization and design of cross-curricular themes are put forward, as well as the opportunities which action-research methodology offers to train teachers for their best profssional development.; En este artículo se describe brevemente la experiencia realizada por un equipo de investigación cooperativa, en torno al proceso de diseño globalizado, desarrollo y evaluación de las líneas transversales en el área de Lengua, siguiendo la metodología de la investigación acción. Después de una rápida justificación de la selección del tema y metodología de investigación utilizados, se contemplan las fases y procedimientos de recogida y análisis de la información empleados. Finalmente se avanzan algunas de las conclusiones extraidas de esta experiencia, en relación a la conceptualización y diseño de las líneas transversales, así como a las posibilidades de la investigación acción como modelo de formación del profesorado para su mejor desarrollo profesional

    Equality, Equity, and Diversity: Educational Solutions in the Basque Country

    Public education is one of the greatest achievements of European countries during the twentieth century. While schooling systems neither exclusively form citizens, nor are they sufficient to alleviate all inequalities, education plays an increasingly important strategic role in relieving social problems and promoting the civic and ethical upbringing of our children. Researchers and professors at the UPV/EHU have had the privilege to design and implement important educational projects in conjunction with government of Autonomous Community of the Basque Country, which has the authority over education in its territory. This book presents the timely (in most cases since 2000) observations, research, and programs that have resulted from this cooperation. Our stress—in both our theoretical and analytical dimensions—has been on the importance of diversity, the promotion of social and human values, and respect for basic human rights.     Interaction of silver atomic quantum clusters with living organisms: bactericidal effect of Ag3 clusters mediated by disruption of topoisomerase–DNA complexes

    Essential processes for living cells such as transcription and replication depend on the formation of specific protein–DNA recognition complexes. Proper formation of such complexes requires suitable fitting between the protein surface and the DNA surface. By adopting doxorubicin (DOX) as a model probe, we report here that Ag3 atomic quantum clusters (Ag-AQCs) inhibit the intercalation of DOX into DNA and have considerable influence on the interaction of DNA-binding proteins such as topoisomerase IV, Escherichia coli DNA gyrase and the restriction enzyme HindIII. Ag-AQCs at nanomolar concentrations inhibit enzyme activity. The inhibitory effect of Ag-AQCs is dose-dependent and occurs by intercalation into DNA. All these effects, not observed in the presence of Ag+ ions, can explain the powerful bactericidal activity of Ag-AQCs, extending the knowledge of silver bactericidal properties.     Targets, Mechanisms and Cytotoxicity of Half-Sandwich Ir(III) Complexes Are Modulated by Structural Modifications on the Benzazole Ancillary Ligand

    Cancers are driven by multiple genetic mutations but evolve to evade treatments targeting specific mutations. Nonetheless, cancers cannot evade a treatment that targets mitochondria, which are essential for tumor progression. Iridium complexes have shown anticancer properties, but they lack specificity for their intracellular targets, leading to undesirable side effects. Herein we present a systematic study on structure-activity relationships of eight arylbenzazole-based Iridium(III) complexes of type [IrCl(Cp*)], that have revealed the role of each atom of the ancillary ligand in the physical chemistry properties, cytotoxicity and mechanism of biological action. Neutral complexes, especially those bearing phenylbenzimidazole (HL1 and HL2), restrict the binding to DNA and albumin. One of them, complex 1[C,NH-Cl], is the most selective one, does not bind DNA, targets exclusively the mitochondria, disturbs the mitochondria membrane permeability inducing proton leak and increases ROS levels, triggering the molecular machinery of regulated cell death. In mice with orthotopic lung tumors, the administration of complex 1[C,NH-Cl] reduced the tumor burden. Cancers are more vulnerable than normal tissues to a treatment that harnesses mitochondrial dysfunction.     Cross-sectoral impacts of the 2018–2019 Central European drought and climate resilience in the German part of the Elbe River basin

    The 2018–2019 Central European drought was probably the most extreme in Germany since the early sixteenth century. We assess the multiple consequences of the drought for natural systems, the economy and human health in the German part of the Elbe River basin, an area of 97,175 km2 including the cities of Berlin and Hamburg and contributing about 18% to the German GDP. We employ meteorological, hydrological and socio-economic data to build a comprehensive picture of the drought severity, its multiple effects and cross-sectoral consequences in the basin. Time series of different drought indices illustrate the severity of the 2018–2019 drought and how it progressed from meteorological water deficits via soil water depletion towards low groundwater levels and river runoff, and losses in vegetation productivity. The event resulted in severe production losses in agriculture (minus 20–40% for staple crops) and forestry (especially through forced logging of damaged wood: 25.1 million tons in 2018–2020 compared to only 3.4 million tons in 2015–2017), while other economic sectors remained largely unaffected. However, there is no guarantee that this socio-economic stability will be sustained in future drought events; this is discussed in the light of 2022, another dry year holding the potential for a compound crisis. Given the increased probability for more intense and long-lasting droughts in most parts of Europe, this example of actual cross-sectoral drought impacts will be relevant for drought awareness and preparation planning in other regions

    Ag2 and Ag3 Clusters: Synthesis, Characterization, and Interaction with DNA

    Subnanometric samples, containing exclusively Ag2 and Ag3 clusters, were synthesized for the first time by kinetic control using an electrochemical technique without the use of surfactants or capping agents. By combination of thermodynamic and kinetic measurements and theoretical calculations, we show herein that Ag3 clusters interact with DNA through intercalation, inducing significant structural distortion to the DNA. The lifetime of Ag3 clusters in the intercalated position is two to three orders of magnitude longer than for classical organic intercalators, such as ethidium bromide or proflavine.Fil: Buceta, David.     L-Arginine Ameliorates Defective Autophagy in GM2 Gangliosidoses by mTOR Modulation.

    AIMS: Tay-Sachs and Sandhoff diseases (GM2 gangliosidosis) are autosomal recessive disorders of lysosomal function that cause progressive neurodegeneration in infants and young children. Impaired hydrolysis catalysed by β-hexosaminidase A (HexA) leads to the accumulation of GM2 ganglioside in neuronal lysosomes. Despite the storage phenotype, the role of autophagy and its regulation by mTOR has yet to be explored in the neuropathogenesis. Accordingly, we investigated the effects on autophagy and lysosomal integrity using skin fibroblasts obtained from patients with Tay-Sachs and Sandhoff diseases. RESULTS: Pathological autophagosomes with impaired autophagic flux, an abnormality confirmed by electron microscopy and biochemical studies revealing the accelerated release of mature cathepsins and HexA into the cytosol, indicating increased lysosomal permeability. GM2 fibroblasts showed diminished mTOR signalling with reduced basal mTOR activity. Accordingly, provision of a positive nutrient signal by L-arginine supplementation partially restored mTOR activity and ameliorated the cytopathological abnormalities. INNOVATION: Our data provide a novel molecular mechanism underlying GM2 gangliosidosis. Impaired autophagy caused by insufficient lysosomal function might represent a new therapeutic target for these diseases. CONCLUSIONS: We contend that the expression of autophagy/lysosome/mTOR-associated molecules may prove useful peripheral biomarkers for facile monitoring of treatment of GM2 gangliosidosis and neurodegenerative disorders that affect the lysosomal function and disrupt autophagy
