    StarHorse: A Bayesian tool for determining stellar masses, ages, distances, and extinctions for field stars

    Understanding the formation and evolution of our Galaxy requires accurate distances, ages and chemistry for large populations of field stars. Here we present several updates to our spectro-photometric distance code, that can now also be used to estimate ages, masses, and extinctions for individual stars. Given a set of measured spectro-photometric parameters, we calculate the posterior probability distribution over a given grid of stellar evolutionary models, using flexible Galactic stellar-population priors. The code (called {\tt StarHorse}) can acommodate different observational datasets, prior options, partially missing data, and the inclusion of parallax information into the estimated probabilities. We validate the code using a variety of simulated stars as well as real stars with parameters determined from asteroseismology, eclipsing binaries, and isochrone fits to star clusters. Our main goal in this validation process is to test the applicability of the code to field stars with known {\it Gaia}-like parallaxes. The typical internal precision (obtained from realistic simulations of an APOGEE+Gaia-like sample) are 8%\simeq 8\% in distance, 20%\simeq 20\% in age,6 \simeq 6\ % in mass, and 0.04\simeq 0.04 mag in AVA_V. The median external precision (derived from comparisons with earlier work for real stars) varies with the sample used, but lies in the range of [0,2]%\simeq [0,2]\% for distances, [12,31]%\simeq [12,31]\% for ages, [4,12]%\simeq [4,12]\% for masses, and 0.07\simeq 0.07 mag for AVA_V.     On the relevance of the polar β-phase of poly(vinylidene fluoride) for high performance lithium-Ion battery separators

    Separator membranes based on poly(vinylidene fluoride), PVDF, poly(vinylidene fluoride-co-trifluoroethylene), PVDF-TrFE, poly(vinylidene fluoride-co-hexafluropropylene), PVDF-HFP and poly(vinylidene fluoride-co-chlorotrifluoroethylene), PVDF-CTFE were prepared by solvent casting method using N,N-dimethylformamide (DMF) as solvent. In all cases, the same polymer/solvent ratio and solvent evaporation temperature were used. For all membranes, porous microstructure is achieved with a degree of porosity larger than 50%. The β-phase content as well as degree of crystallinity were different for each membrane, which were lower for the co-polymer membranes when compared with PVDF. On the other hand, the observed ionic conductivity values, electrolyte uptake, tortuosity and MacMullin number were similar for all membranes. The electrochemical performance of the separator membranes was evaluated in Li/C–LiFePO4 half-cell configuration showing good cyclability and rate capability for all membranes. Among the all separator membranes, PVDF-TrFE demonstrate the best electrochemical performance, with a discharge capacity value of 87 mAh.g-1 after 50 cycles with a capacity retention of 78 % at 2C.Finally, the correlation between the β-phase content in the membranes and the cycling performance was demonstrated (which was significant at high-C rates): larger β-phase contents, leading higher polarity, facilitates faster lithium ion migration within the separator for similar microstructures.This work was supported by the Portuguese Foundation for Science and Technology (FCT) in the framework of the Strategic Funding UID/FIS/04650/2013.     Generating real-world evidence on the quality use, benefits and safety of medicines in australia: History, challenges and a roadmap for the future

    Australia spends more than $20 billion annually on medicines, delivering significant health benefits for the population. However, inappropriate prescribing and medicine use also result in harm to individuals and populations, and waste of precious health resources. Medication data linked with other routine collections enable evidence generation in pharmacoepidemiology; the science of quantifying the use, effectiveness and safety of medicines in real-world clinical practice. This review details the history of medicines policy and data access in Australia, the strengths of existing data sources, and the infrastructure and governance enabling and impeding evidence generation in the field. Currently, substantial gaps persist with respect to cohesive, contemporary linked data sources supporting quality use of medicines, effectiveness and safety research; exemplified by Aus-tralia’s limited capacity to contribute to the global effort in real-world studies of vaccine and dis-ease-modifying treatments for COVID-19. We propose a roadmap to bolster the discipline, and population health more broadly, underpinned by a distinct capability governing and streamlining access to linked data assets for accredited researchers. Robust real-world evidence generation requires current data roadblocks to be remedied as a matter of urgency to deliver efficient and equitable health care and improve the health and well-being of all Australians

    Natural Resistance of Leishmania infantum to Miltefosine Contributes to the Low Efficacy in the Treatment of Visceral Leishmaniasis in Brazil

    In India visceral leishmaniasis (VL) caused by Leishmania donovani has been successfully treated with miltefosine with a cure rate of > 90%. To assess the efficacy and safety of oral miltefosine in L. infantum-causing Brazilian VL patients, a phase II, open-label, dose-escalation study of oral miltefosine was conducted in children (ages 2-12) and adolescent-adults (ages 13-60). Definitive cure was assessed at a 6 month follow-up visit. The cure rate was only 42% (6 out of 14 patients) with the recommended 28 days of therapy and 68% (19 out of 28 patients) with an extended treatment of 42 days. The in vitro miltefosine susceptibility profile of intracellular amastigote stages of the pre-treatment isolates, from cured and relapsed patients, showed a positive correlation with clinical outcome. The IC50 mean (SEM) of eventual cures was 5.1 (0.4) µM whereas that of eventual failures was 12.8 (1.9) µM (P = 0.0002). An IC50 below or above 8.0 µM predicts cure or failure, respectively with 82% sensitivity and 100% specificity. The finding of L. infantum amastigotes resistant to miltefosine in isolates from patients who eventually failed treatment, strongly suggests natural resistance to this drug, as miltefosine had never been used in Brazil before this trial was carried out

    Efeito da equoterapia no equilíbrio postural de idosos

    OBJETIVO: Verificar se a equoterapia é capaz de produzir alterações no equilíbrio de idosos. MÉTODOS: Desenvolveu-se um estudo experimental controlado. A amostra foi composta de 17 idosos, divididos em grupo experimental (GE), sete sujeitos e grupo controle (GC), dez sujeitos. A aquisição dos dados da estabilometria foi realizada por meio da plataforma de força da marca AMTI (Force Measurement Systems). Para análise clínica do equilíbrio sentado, transferências de sentado para a posição em pé, estabilidade na deambulação e mudanças do curso da marcha, utilizou-se o teste Timed Up and Go (TUG). Foram realizadas 16 sessões de equoterapia. RESULTADOS: Na comparação das médias entre os grupos por meio do teste de Mann-Whitney, não houve diferença significativa nos parâmetros estabilométricos analisados. Já na comparação das médias intragrupo por meio do teste de Wilcoxon, verificou-se aumento significante sobre as variáveis COPy e área (p=0,02). Nas médias entre o GE e o GC, por meio do teste de Mann-Whitney para análise do teste TUG, verificou-se efeito significante (p=0,04) da equoterapia. Na comparação das médias intragrupo pelo teste de Wilcoxon, verificou-se efeito significante (p=0,04) sobre a variável TUG. CONCLUSÕES: A senescência tende a normalizar as medidas estabilométricas, sendo insuficiente, com esse número de sessões de equoterapia, apontar diferenças ligadas a essa intervenção. No entanto, essa frequência de tratamento foi suficiente como preditor de menor risco de quedas em idosos, uma vez que o teste de TUG mostrou diminuição significativa do tempo necessário para executá-lo. METHODS: The sample included 17 older adults who were divided into experimental (7 subjects) and control (10 subjects) groups. Stabilometry data were acquired with a force platform. The Timed Up and Go test (TUG) was used for clinical analysis of seated balance, transfer from a seated to a standing position, walking stability and changes in gait. Sixteen equine-assisted therapy sessions were carried out. RESULTS: Mann-Witney was used to compare the means between groups and no significant differences were found in the analyzed stabilometric parameters. In intragroup comparison with the Wilcoxon test, a significant increase in the variables COPy and Area (p=0.02) was observed. Equine-assisted therapy significantly affected (p=0.04) TUG test means between the experimental and control groups (Mann-Witney). Intragroup TUG test means were also significantly affected (p=0.04) according to the Wilcoxon test. CONCLUSIONS: Because senescence tends to normalize stabilometric measures, the number of equine-assisted therapy sessions was insufficient to determine any differences. Nevertheless, the significant improvement in TUG test scores demonstrates that this treatment frequency was a predictor of reduced fall risk in the elderly.     Integrative multi-kinase approach for the identification of potent antiplasmodial hits

    Malaria is a tropical infectious disease that affects over 219 million people worldwide. Due to the constant emergence of parasitic resistance to the current antimalarial drugs, the discovery of new antimalarial drugs is a global health priority. Multi-target drug discovery is a promising and innovative strategy for drug discovery and it is currently regarded as one of the best strategies to face drug resistance. Aiming to identify new multi-target antimalarial drug candidates, we developed an integrative computational approach to select multi-kinase inhibitors for Plasmodium falciparum calcium-dependent protein kinases 1 and 4 (CDPK1 and CDPK4) and protein kinase 6 (PK6). For this purpose, we developed and validated shape-based and machine learning models to prioritize compounds for experimental evaluation. Then, we applied the best models for virtual screening of a large commercial database of drug-like molecules. Ten computational hits were experimentally evaluated against asexual blood stages of both sensitive and multi-drug resistant P. falciparum strains. Among them, LabMol-171, LabMol-172, and LabMol-181 showed potent antiplasmodial activity at nanomolar concentrations (EC50 15 folds. In addition, LabMol-171 and LabMol-181 showed good in vitro inhibition of P. berghei ookinete formation and therefore represent promising transmission-blocking scaffolds.     Does severe maternal morbidity affect female sexual activity and function? Evidence from a brazilian cohort study

    To assess Female Sexual Function Index (FSFI) scores and delay to resume sexual activity associated with a previous severe maternal morbidity. This was a multidimensional retrospective cohort study. Women who gave birth at a Brazilian tertiary maternity between 2008 and 2012 were included, with data extraction from the hospital information system. Those with potentially life-threatening conditions and maternal near miss episodes (severe maternal morbidity) were considered the exposed group. The control group was a random sample of women who had had uncomplicated pregnancy. Female sexual function was evaluated through FSFI questionnaire, and general and reproductive aspects were addressed through specific questions. Statistical analyses were performed using Mann-Whitney and Pearson's Chi-square for bivariate analyses. Logistic regression was used to identify variables independently associated with lower FSFI scores. 638 women were included (315 at exposed and 323 at not exposed groups). The majority of women were under 30 years-old in the control group and between 30 and 46 years-old in the exposed group (p = 0.003). Women who experienced severe maternal morbidity (SMM) had statistically significant differences regarding cesarean section (82.4% versus 47.1% among deliveries without complications, p< 0.001), and some previous pathological conditions. FSFI mean scores were similar among groups ranging from 24.39 to 24.42. It took longer for exposed women to resume sexual activity after index pregnancy (mean 84 days after SMM and 65 days for control group, p = 0.01). Multiple analyses showed no significant association of FSFI below cut-off value with any predictor. FSFI scores were not different in both groups. However, they were lower than expected. SMM delayed resumption of sexual activity after delivery, beyond postpartum period. However, the proportion of women in both groups having sex at 3 months after delivery was similar. Altered sexual response may be evaluated as one of possible long-term consequences after SMM episodes.     Nasal bot : an emerging parasitic disease in small ruminants in the Brazilian Northeast

    No período de janeiro de 2011 a dezembro de 2014 foram diagnosticados 9 surtos (A, B, C, D, E, F, G, H e I) de Oestrus ovis em pequenos ruminantes no estado da Bahia. No surto A obteve-se 0,5% (1/200); B 2,2% (2/90); C 0,8% (1/120); D 2% (2/100); E 1% (1/100); F 3% (1/33); G 0,6% (1/150); H 2,5% (5/200); I com 11,4% (8/70) em ovinos e 5% (2/40) em caprinos. Os sinais clínicos associados ao parasitismo pelas larvas nos surtos foram respiração ruidosa, espirro seguido de secreção nasal catarral, inquietação, movimentação excessiva da cabeça e andar em círculo. Macroscopicamente havia nos seios e conchas nasais hiperemia, edema da mucosa e presença de larvas. Todas as larvas coletadas dos cornetos e conchas nasais variavam desde o primeiro ao terceiro estágio de desenvolvimento. Algumas larvas L3 coletadas nas necropsias foram incubadas e o imago obtido das pupas mediram aproximadamente 10mm de cor acinzentada e abdômen escurecido. Realizada análise descritiva das condições climáticas, ano e positividade de casos de oestrose, demonstrou que a ocorrência tem tendência de crescimento com os anos (p˂0,001) e que houve casos com menor média de temperatura mínima (p˂0,001), possibilitando o desenvolvimento da mosca de O. ovis, demonstrando que houve a introdução da mosca enTtre o rebanho de ovinos e caprinos do estado da Bahia, e que as condições climáticas são ideais para perpetuação da espécie.From January 2011 to December 2014 were diagnosed 9 outbreaks of Oestrus ovis infection in small ruminants (Outbreaks A-I) in the State of Bahia. The incidence of oestrosis in sheep in outbreak A was 0.5% (1/200), in B 2.2% (2/90), in C 0.8% (1/120), in D 2% (2/100), in E 1% (1/100), in F 3% (1/33), in G 0.6% (1/150), in H 2.5% (5/200), and in I 11.42% (8/70), and 5% (2/40) in goats. Clinical signs associated with parasitism were wheezing, sneezing followed by catarrhal nasal secretion, some restlessness, excessive head movement and walking in circles. The breasts and turbinates were hyperemic, with mucosal edema and presence of O. ovis larvae. All larvae collected from the turbinates ranged from the first to the third stage of development. Some L3 larvae collected at necropsy were incubated and the gray colored Imago with dark abdomen obtained from the pupae measured about 10mm. A descriptive analysis of the climatic conditions was carried out; in the year of investigation the incidence of O. ovis infection has grown (p<0.001), and the lowest mean minimum temperature (p<0.001) caused the development the O. ovis fly, so that there was an introduction of an increased number of these flies into the sheep and goat flocks in state of Bahia with the ideal climatic conditions for their perpetuation
