    DIGITAL SKILLS AND PROFILE OF EACH GENERATION: A REVIEW

    The technological development is changing, involving and conditioning everyday life, making it faster, hyper-connected and immediate. New technologies can have a double outcome: positive, given that they take on the role of facilitator, or negative, when people tend to make excessive and unconscious use of it or technology repeatedly replaces human beings. Today, therefore, new skills are needed for the entire population, the digital skills, both in everyday life and in the labour market. These are transversal skills that allow human beings to choose the right technologies: to research, analyze data and choose reliable information from those that require further study, to check and verify the health of people, to interact with different and distant subjects in the world, to improve school learning, ... The plurality of people that this technological development is involving allows us to analyze the population in terms of generations, from Silent Generation to Alpha Generation. In this article we want to present a literature review within the Google Scholar database in the last 10 years using the terms digital skills and different generation. This preliminary research allows us to analyze the different theories that make for the Digital Skills a definition, dimensions and evaluation model, and the generational profiles in order to have a shared and common theory among the different professionals. Having an accurate background of digital skills and generational profiles favours the inclusion of all people, and the improvement of the quality of life in the creation of psycho-pedagogical projects and in the design of technological products

    Conformational Assignement, Absolute Configuration and Chiral Separation of all the Stereoisomers Created by the Combined Presence of Stereogenic Centers and Stereogenic Conformational Axes in a Highly Hindered 1,5-Naphthyl Sulfoxide.

    The presence of two stereogenic centers and of two stereogenic conformational axes in 2,6-dimethyl-1,5-bis(2-methyl-2-propylsulfinyl)naphthalene (1) entails the existence of 10 stereoisomers. In particular, both the meso form (1a) and the racemic form (1b) are constituted by three atropisomers; in the case of the latter (1b) each of them entails a pair of enantiomers (total of six species), whereas owing to the symmetry only one of the three atropisomers of the meso form (1a) yields a pair of enantiomers (a total of four species). Despite the low conformational interconversion barrier (18.5 kcal/mol) all of them have been separated by low temperature (-45°C) chiral HPLC. Their configurational and conformational assignment has been achieved by a combined use of NMR (both in solution and solid state) and on-line CD-detected chiral HPLC. The single crystal X-ray diffraction yielded the absolute configuration of one of the stereoisomers ((ZR,ER)-1b) from which all the others could be obtained by CD relationship

    Cystic Fibrosis Defective Response to Infection Involves Autophagy and Lipid Metabolism

    Cystic fibrosis (CF) is a hereditary disease, with 70% of patients developing a proteinopathy related to the deletion of phenylalanine 508. CF is associated with multiple organ dysfunction, chronic inflammation, and recurrent lung infections. CF is characterized by defective autophagy, lipid metabolism, and immune response. Intracellular lipid accumulation favors microbial infection, and autophagy deficiency impairs internalized pathogen clearance. Myriocin, an inhibitor of sphingolipid synthesis, significantly reduces inflammation, promotes microbial clearance in the lungs, and induces autophagy and lipid oxidation. RNA-seq was performed in Aspergillusfumigatus-infected and myriocin-treated CF patients' derived monocytes and in a CF bronchial epithelial cell line. Fungal clearance was also evaluated in CF monocytes. Myriocin enhanced CF patients' monocytes killing of A. fumigatus. CF patients' monocytes and cell line responded to infection with a profound transcriptional change; myriocin regulates genes that are involved in inflammation, autophagy, lipid storage, and metabolism, including histones and heat shock proteins whose activity is related to the response to infection. We conclude that the regulation of sphingolipid synthesis induces a metabolism drift by promoting autophagy and lipid consumption. This process is driven by a transcriptional program that corrects part of the differences between CF and control samples, therefore ameliorating the infection response and pathogen clearance in the CF cell line and in CF peripheral blood monocytes

    Increased incidence of breast cancer in postmenopausal women with high body mass index at the modena screening program

    Purpose: We conducted a study to evaluate the relationship between body mass index (BMI) and the risk of breast cancer (BC) and outcome in a population of 14,684 women aged 55 to 69 years eligible to participate in the Mammography Screening Program (MSP) in the Province of Modena, Italy. Methods: The study population was drawn from women who underwent mammography screening between 2004 and 2006 in the Province of Modena. Women were subdivided into obese, overweight, and normal-weight categories according to BMI and followed until July 31, 2010, to evaluate the BC incidence. The clinicopathological characteristics of BC were also evaluated in different groups of patients classified according to BMI. After BC diagnosis, patients were followed for a median period of 65 (range, 2\u2013104) months. Second events (recurrences and second tumors) were recorded, and the 5-year event-free survival (EFS) was calculated. Results: After a period of 73 months, 366 cases of BC were diagnosed. Compared with normal-weight women, obese women had a significantly higher incidence of BC (relative risk [RR], 1.32; p= 0.040) (RR=1), larger tumors (27% of tumors were larger than T2 size), and more nodal involvement (38.5% of tumors were node-positive). Furthermore, a significantly higher rate of total events was seen in obese women compared with overweight and normal-weight patients, respectively (17.9% vs. 11.4% vs. 10.8%, p=0.032). The 5-year EFS was 89.0%, 89.0%, and 80.0% for normal-weight, overweight, and obese patients, respectively. Conclusion: We observed a significantly higher risk of BC in obese women among those eligible to participate in the MSP in the Province of Modena. Finally, obese women had more second events and poorer EFS compared to nono bese women

    EpiHRMAssay, in tube and in silico combined approach for the scanning and epityping of heterogeneous DNA methylation

    Reliable and cost-effective assays with adequate sensitivity are required to detect the DNA methylation profile in plants for scientific and industrial purposes. The proposed novel assay, named EpiHRMAssay, allows to quantify the overall methylation status at target loci and to enable high-throughput analyses. It combines in tube High Resolution Melting Analysis on bisulphite-treated templates with the in silico prediction of the melting profile of virtual epialleles using uMELTSM software. The predicted melting temperatures (Tm-s) of a set of epialleles characterized by different numbers of methylated cytosines (#mC) or different mC configurations were obtained and used to build calibration models, enabling the quantification of methylation in unknown samples using only the in tube observed melting temperature (Tm-o). EpiHRMAssay was validated by analysing the promoter region of CMT3, DDM1, and ROS1 genes involved in the regulation of methylation/demethylation processes and chromatin remodelling within a population of peach plants. Results demonstrate that EpiHRMAssay is a sensitive and reliable tool for locus-specific large-scale research and diagnostic contexts of the regulative regions of genes, in a broad range of organisms, including mammals. EpiHRMAssay also provides complementary information for the assessment of heterogeneous methylation and can address an array of biological questions on epigenetic regulation for diversity studies and for large-scale functional genomics

    Further progress/developments, on surface/bulk treated Constantan wires, for anomalous heat generation by H2/D2 interaction

    In the framework of those studies aimed to analyze anomalous effects (thermal and/or nuclear) due to the interaction among some specific materials (pure and/or alloys) and H2 (or D2), we focused, since 2011, on a specific alloy called Constantan (Cu55-Ni44-Mn1). We selected such material using our own considerations and intuitions and because, according to a scientific paper [1], it has the largest energy value for dissociation of H2 to 2H, i.e. about 3eV. Among others B. Ahern suggested that Ni-Cu-H can be used for heat generation. We improved the preparation procedure of such wire from simple thermal treatments (up to May 2012 [2])to more sophisticated ones, with more tight control of the multilayered (400-700) surface structures. Some of the results were presented at ICCF17, Aug. 2012 [3]. After [3], several groups asked to make their own experiments using such kind of wires ([phi]=200[mu]m, l=100cm) to cross-check (and possibly improve) our results. Some of such Researchers (group of M. Fleischmann Memorial Project; U. Mastromatteo) made public their (positive) results since Dec. 14, 2012 at Ministry of Aeronautics in Rome, Italy. In short, using an (home-made) apparatus integrated with an acquisition system (type PXi) by National Instruments, we made, since September 2012, not mentioning qualitative reconfirmation of previous results, further and unexpected progress and discoveries: a) We developed a new kind of procedure of measurement (about anomalous excess heat) under dynamic vacuum, to avoid the effect of different thermal conductivity, inside the gas cell, due to type of gas and pressure variation: the wire didn't lose, macroscopically, H even at T=600[degrees]C. b) We developed a new, very simple, type of surface coating (2 layers) that is nano-diamandoidslike; c) We observed, at least 2 times, the phenomenon of water splitting due to catalytic effect of surface treated Constantan. Such phenomenon is larger in comparison with what expected just by thermal splitting (wires temperature of about 300-500[degrees]C); d) We observed a very large variation (about a factor 100) of Resistive Thermal Coefficient (RTC) of the wire used (400 layers) as the amount of H (related to the macroscopic value of resistive ratio R/Ro, normalize to empty wire Ro) increased. As example, with "treated" virgin wire (w/o H2) the RTC was about 5*10-6 and increased to6*10-4 when the R/Ro reduced to 0.68; temperature range 20-300[degrees]C. The RTC is larger with D in respect to H. Experiments are in progress also at 77K. e) Overall results are affected by previous operating conditions

    Deletion of the miR172 target site in a TOE-type gene is a strong candidate variant for dominant double-flower trait in Rosaceae

    Double flowers with supernumerary petals have been selected by humans for their attractive appearance and commercial value in several ornamental plants, including Prunus persica (peach), a recognized model for Rosaceae genetics and genomics. Despite the relevance of this trait, knowledge of the underlying genes is limited. Of two distinct loci controlling the double-flower phenotype in peach, we focused on the dominant Di2 locus. High-resolution linkage mapping in five segregating progenies delimited Di2 to an interval spanning 150858bp and 22 genes, including Prupe.6G242400 encoding an euAP2 transcription factor. Analyzing genomic resequencing data from single- and double-flower accessions, we identified a deletion spanning the binding site for miR172 in Prupe.6G242400 as a candidate variant for the double-flower trait, and we showed transcript expression for both wild-type and deleted alleles. Consistent with the proposed role in controlling petal number, Prupe.6G242400 is expressed in buds at critical times for floral development. The indelDi2 molecular marker designed on this sequence variant co-segregated with the phenotype in 621 progenies, accounting for the dominant inheritance of the Di2 locus. Further corroborating the results in peach, we identified a distinct but similar mutation in the ortholog of Prupe.6G242400 in double-flower roses. Phylogenetic analysis showed that these two genes belong to a TARGET OF EAT (TOE)-type clade not represented in Arabidopsis, indicating a divergence of gene functions between AP2-type and TOE-type factors in Arabidopsis and other species. The identification of orthologous candidate genes for the double-flower phenotype in two important Rosaceae species provides valuable information to understand the genetic control of this trait in other major ornamental plants. Significance Statement We used peach as a model to gain insight into the molecular basis of double flowers, an important trait in many ornamental plants. We propose that a deletion causes a TOE-type transcription factor to escape miR172-mediated repression, in turn resulting in an increased number of petals, as corroborated by results on the orthologous gene in rose

    Streamlined Calibrations of the ATLAS Precision Muon Chambers for Initial LHC Running

    The ATLAS Muon Spectrometer is designed to measure the momentum of muons with a resolution of dp/p = 3% and 10% at 100 GeV and 1 TeV momentum respectively. For this task, the spectrometer employs 355,000 Monitored Drift Tubes (MDTs) arrayed in 1200 Chambers. Calibration (RT) functions convert drift time measurements into tube-centered impact parameters for track segment reconstruction. RT functions depend on MDT environmental parameters and so must be appropriately calibrated for local chamber conditions. We report on the creation and application of a gas monitor system based calibration program for muon track reconstruction in the LHC startup phase.Comment: 25 pages, 21 figure
