    Synthesis, Regulation and Degradation of Carotenoids Under Low Level UV-B Radiation in the Filamentous Cyanobacterium Chlorogloeopsis fritschii PCC 6912

    Carotenoids in cyanobacteria play an important role in protecting against and in repairing damage against low level UV-B radiation. Here we use transcriptomics and metabolomic HPLC pigment analysis to compare carotenoid pathway regulation in the filamentous cyanobacterium Chlorogloeopsis fritschii PCC 6912 exposed to white light and to white light supplemented with low level UV-B. Under UV-B changes in carotenoid transcription regulation were found associated with carotenogenesis (carotenoid synthesis), photoprotection and carotenoid cleavage. Transcriptional regulation was reflected in corresponding pigment signatures. All carotenogenesis pathway genes from geranylgeranyl-diphosphate to lycopene were upregulated. There were significant increases in expression of gene homologs (crtW, crtR, cruF, and cruG) associated with routes to ketolation to produce significant increases in echinenone and canthaxanthin concentrations. There were gene homologs for four β-carotene-ketolases (crtO and crtW) present but only one crtW was upregulated. Putative genes encoding enzymes (CruF, CrtR, and CruG) for the conversion of γ-carotene to myxol 2′-methylpentoside were upregulated. The hydroxylation pathway to nostaxanthin via zeaxanthin and caloxanthin (gene homologs for CrtR and CrtG) were not upregulated, reflected in the unchanged corresponding pigment concentrations in zeaxanthin, caloxanthin and nostaxanthin, Transcripts for the non-photochemical quenching related Orange-Carotenoid-Protein (OCP) and associated Fluoresence-Recovery-Protein (FRP) associated with photoprotection were upregulated, and one carotenoid binding Helical-Carotenoid-Protein (HCP) gene homolog was downregulated. Multiple copies of genes encoding putative apocarotenoid related carotenoid oxygenases responsible for carotenoid cleavage were identified, including an upregulated apo-β-carotenal-oxygenase gene homologous to a retinal producing enzyme. Our study provides holistic insight into the photoregulatory processes that modulate the synthesis, photoprotection and cleavage of carotenoids in cyanobacterial cells exposed to low level UV-B. This is important to understanding how regulation of metabolism responds to a changing environment and how metabolism can be modulated for biotechnological purposes

    Spitzer Space Telescope Observations of the Nucleus of Comet 103P/Hartley 2

    We have used the Spitzer Space Telescope InfraRed Spectrograph (IRS) 22-μm peakup array to observe thermal emission from the nucleus and trail of comet 103P/Hartley 2, the target of NASA’s Deep Impact Extended Investigation (DIXI). The comet was observed on UT 2008 August 12 and 13, while 5.5 AU from the Sun. We obtained two 200 frame sets of photometric imaging over a 2.7 hr period. To within the errors of the measurement, we find no detection of any temporal variation between the two images. The comet showed extended emission beyond a point source in the form of a faint trail directed along the comet’s antivelocity vector. After modeling and removing the trail emission, a NEATM model for the nuclear emission with beaming parameter of 0.95 ± 0.20 indicates a small effective radius for the nucleus of 0.57 ± 0.08 km and low geometric albedo 0.028 ± 0.009 (1σ). With this nucleus size and a water production rate of 3 × 10^(28) molecules s^(-1) at perihelion, we estimate that ~100% of the surface area is actively emitting volatile material at perihelion. Reports of emission activity out to ~5 AU support our finding of a highly active nuclear surface. Compared to Deep Impact’s first target, comet 9P/Tempel 1, Hartley 2’s nucleus is one-fifth as wide (and about one-hundredth the mass) while producing a similar amount of outgassing at perihelion with about 13 times the active surface fraction. Unlike Tempel 1, comet Hartley 2 should be highly susceptible to jet driven spin-up torques, and so could be rotating at a much higher frequency. Since the amplitude of nongravitational forces are surprisingly similar for both comets, close to the ensemble average for ecliptic comets, we conclude that comet Hartley 2 must have a much more isotropic pattern of time-averaged outgassing from its nuclear surface. Barring a catastrophic breakup or major fragmentation event, the comet should be able to survive up to another 100 apparitions (~700 yr) at its current rate of mass loss

    Estimating the Reliability of a Soyuz Spacecraft Mission

    Once the US Space Shuttle retires in 2010, the Russian Soyuz Launcher and Soyuz Spacecraft will comprise the only means for crew transportation to and from the International Space Station (ISS). The U.S. Government and NASA have contracted for crew transportation services to the ISS with Russia. The resulting implications for the US space program including issues such as astronaut safety must be carefully considered. Are the astronauts and cosmonauts safer on the Soyuz than the Space Shuttle system? Is the Soyuz launch system more robust than the Space Shuttle? Is it safer to continue to fly the 30 year old Shuttle fleet for crew transportation and cargo resupply than the Soyuz? Should we extend the life of the Shuttle Program? How does the development of the Orion/Ares crew transportation system affect these decisions? The Soyuz launcher has been in operation for over 40 years. There have been only two loss of life incidents and two loss of mission incidents. Given that the most recent incident took place in 1983, how do we determine current reliability of the system? Do failures of unmanned Soyuz rockets impact the reliability of the currently operational man-rated launcher? Does the Soyuz exhibit characteristics that demonstrate reliability growth and how would that be reflected in future estimates of success? NASA s next manned rocket and spacecraft development project is currently underway. Though the projects ultimate goal is to return to the Moon and then to Mars, the launch vehicle and spacecraft s first mission will be for crew transportation to and from the ISS. The reliability targets are currently several times higher than the Shuttle and possibly even the Soyuz. Can these targets be compared to the reliability of the Soyuz to determine whether they are realistic and achievable? To help answer these questions this paper will explore how to estimate the reliability of the Soyuz Launcher/Spacecraft system, compare it to the Space Shuttle, and its potential impacts for the future of manned spaceflight. Specifically it will look at estimating the Loss of Mission (LOM) probability using historical data, reliability growth, and Probabilistic Risk Assessment technique

    The rotation and coma profiles of comet C/2004 Q2 (Machholz)

    Aims. Rotation periods of cometary nuclei are scarce, though important when studying the nature and origin of these objects. Our aim is to derive a rotation period for the nucleus of comet C/2004 Q2 (Machholz). Methods. C/2004 Q2 (Machholz) was monitored using the Merope CCD camera on the Mercator telescope at La Palma, Spain, in January 2005, during its closest approach to Earth, implying a high spatial resolution (50km per pixel). One hundred seventy images were recorded in three different photometric broadband filters, two blue ones (Geneva U and B) and one red (Cousins I). Magnitudes for the comet's optocentre were derived with very small apertures to isolate the contribution of the nucleus to the bright coma, including correction for the seeing. Our CCD photometry also permitted us to study the coma profile of the inner coma in the different bands. Results. A rotation period for the nucleus of P = 9.1 +/- 0.2 h was derived. The period is on the short side compared to published periods of other comets, but still shorter periods are known. Nevertheless, comparing our results with images obtained in the narrowband CN filter, the possibility that our method sampled P/2 instead of P cannot be excluded. Coma profiles are also presented, and a terminal ejection velocity of the grains v_gr = 1609 +/- 48 m/s is found from the continuum profile in the I band.Comment: 11 pages, 9 figures, accepted by A&

    What agro-input dealers know, sell and say to smallholder farmers about pesticides: a mystery shopping and KAP analysis in Uganda

    BACKGROUND: Pesticides can have negative effects on human and environmental health, especially when not handled as intended. In many countries, agro-input dealers sell pesticides to smallholder farmers and are supposed to provide recommendations on application and handling. This study investigates the role of agro-input dealers in transmitting safety information from chemical manufacturers to smallholder farmers, assesses the safety of their shops, what products they sell, and how agro-input dealers abide by laws and recommendations on best practices for preventing pesticide risk situations. METHODS: Applying a mixed-methods approach, we studied agro-input dealers in Central and Western Uganda. Structured questionnaires were applied to understand agro-input dealers' knowledge, attitude and practices on pesticides (n = 402). Shop layout (n = 392) and sales interaction (n = 236) were assessed through observations. Actual behavior of agro-input dealers when selling pesticides was revealed through mystery shopping with local farmers buying pesticides (n = 94). RESULTS: While 97.0% of agro-input dealers considered advising customers their responsibility, only 26.6% of mystery shoppers received any advice from agro-input dealers when buying pesticides. 53.2% of products purchased were officially recommended. Sales interactions focused mainly on product choice and price. Agro-input dealers showed limited understanding of labels and active ingredients. Moreover, 25.0% of shops were selling repackaged products, while 10.5% sold unmarked or unlabeled products. 90.1% of shops were lacking safety equipment. Pesticides of World Health Organization toxicity class I and II were sold most frequently. Awareness of health effects seemed to be high, although agro-input dealers showed incomplete hygiene practices and were lacking infrastructure. One reason for these findings might be that only 55.7% of agro-input dealers held a certificate of competency on safe handling of pesticides and even fewer (5.7%) were able to provide a government-approved up-to-date license. CONCLUSION: The combination of interviews, mystery shopping and observations proved to be useful, allowing the comparison of stated and actual behavior. While agro-input dealers want to sell pesticides and provide the corresponding risk advice, their customers might receive neither the appropriate product nor sufficient advice on proper handling. In light of the expected increase in pesticide use, affordable, accessible and repeated pesticide training and shop inspections are indispensable

    A data-driven classification of outcome behaviors in those who cause concern to British public figures

    Fixated individuals pose a significant threat to public figures. Previous research compares individuals labeled ‘approachers’ to those labeled ‘communicators.’ Typically, such studies compare a number of risk factors among the two groups to identify significant differences. This has impactful implications for the threat assessment and management of the pathologically fixated. The present study builds upon this established body of work by considering if more nuance can be disaggregated from a universe of cases referred to the Fixated Threat Assessment Center (FTAC). FTAC is a joint police and mental health unit in the U.K. which applies a public health approach to managing the pathologically fixated. The present study takes a deductive approach to detect profiles of cases from the data. First, we use cluster analysis to detect unmeasured subgroups of concerning behavior in the case files of 2,118 referrals to FTAC. We identify 5 patterns of concerning behavior: incoherent offline communicators, incoherent online communicators, angry/abusive communicators, concerning approachers, and problematic approachers. Second, we examine the rate of diagnosed mental disorder, the nature of concerns evoked, and case management actions taken among each of the five profiles identified. We conclude by highlighting how our results could inform triaging large volumes of cases, the allocation of limited resources, and more generally, how the success of the FTAC model has relevance across the management of grievance-fueled violence in general.     The Claims Culture: A Taxonomy of Industry Attitudes

    This paper presents an analysis of a familiar aspect of construction industry culture that we have dubbed 'the claims culture'. This is a culture of contract administration that lays a strong emphasis on the planning and management of claims. The principal elements of the analysis are two sets of distinctions. The first comprises economic and occupational orders, referring to two kinds of control that are exercised over the construction process; predicated respectively on economic ownership and occupational competence. The second refers to contrasting attitudes towards relationships and problem solving within these orders: respectively 'distributive' and 'integrative'. The concepts of economic and occupational order entail further sub-categories. The various attitudes associated with these categories and sub-categories are described. They are assessed as to their consequences for change initiatives in the industry
