    Curcumin Modulates Nitrosative Stress, Inflammation, and DNA Damage and Protects against Ochratoxin A-Induced Hepatotoxicity and Nephrotoxicity in Rats.

    Ochratoxin A (OTA) is a fungal toxin of critical concern for food safety both for human health and several animal species, also representing a cancer threat to humans. Curcumin (CURC) is a natural polyphenol that has anti-apoptotic, anti-inflammatory, and antioxidant effects. The aim of this study was to investigate the cytoprotective effect of CURC against OTA-induced nephrotoxicity and hepatotoxicity through the study of the nitrosative stress, pro-inflammatory cytokines, and deoxyribonucleic acid (DNA) damage. Sprague Dawley rats were daily treated with CURC (100 mg/kg b.w.), OTA (0.5 mg/kg b.w), or CURC with OTA by oral gavage for 14 days. Our results demonstrated that OTA exposure was associated with significant increase of pro-inflammatory and DNA oxidative-damage biomarkers. Moreover, OTA induced the inducible nitric oxide synthase, (iNOS) resulting in increased nitric oxide (NO) levels both in kidney and liver. The co-treatment OTA + CURC counteracted the harmful effects of chronic OTA treatment by regulating inflammation, reducing NO levels and oxidative DNA damage in kidney and liver tissues. Histology revealed that OTA + CURC treatment determinates mainly an Iba1+ macrophagic infiltration with fewer CD3+ T-lymphocytes in the tissues. In conclusion, we evidenced that CURC exerted cytoprotective and antioxidant activities against OTA-induced toxicity in rats

    The Na I D resonance lines in main sequence late-type stars

    We study the sodium D lines (D1: 5895.92 \AA; D2: 5889.95 \AA) in late-type dwarf stars. The stars have spectral types between F6 and M5.5 (B-V between 0.457 and 1.807) and metallicity between [Fe/H] = -0.82 and 0.6. We obtained medium resolution echelle spectra using the 2.15-m telescope at the argentinian observatory CASLEO. The observations have been performed periodically since 1999. The spectra were calibrated in wavelength and in flux. A definition of the pseudo-continuum level is found for all our observations. We also define a continuum level for calibration purposes. The equivalent width of the D lines is computed in detail for all our spectra and related to the colour index (B-V) of the stars. When possible, we perform a careful comparison with previous studies. Finally, we construct a spectral index (R_D') as the ratio between the flux in the D lines, and the bolometric flux. We find that, once corrected for the photospheric contribution, this index can be used as a chromospheric activity indicator in stars with a high level of activity. Additionally, we find that combining some of our results, we obtain a method to calibrate in flux stars of unknown colour.Comment: 12 pages, including 14 figures and 4 tables.     Coordination within the remote sensing payload on the Solar Orbiter mission

    Context. To meet the scientific objectives of the mission, the Solar Orbiter spacecraft carries a suite of in-situ (IS) and remote sensing (RS) instruments designed for joint operations with inter-instrument communication capabilities. Indeed, previous missions have shown that the Sun (imaged by the RS instruments) and the heliosphere (mainly sampled by the IS instruments) should be considered as an integrated system rather than separate entities. Many of the advances expected from Solar Orbiter rely on this synergistic approach between IS and RS measurements. / Aims. Many aspects of hardware development, integration, testing, and operations are common to two or more RS instruments. In this paper, we describe the coordination effort initiated from the early mission phases by the Remote Sensing Working Group. We review the scientific goals and challenges, and give an overview of the technical solutions devised to successfully operate these instruments together. / Methods. A major constraint for the RS instruments is the limited telemetry (TM) bandwidth of the Solar Orbiter deep-space mission compared to missions in Earth orbit. Hence, many of the strategies developed to maximise the scientific return from these instruments revolve around the optimisation of TM usage, relying for example on onboard autonomy for data processing, compression, and selection for downlink. The planning process itself has been optimised to alleviate the dynamic nature of the targets, and an inter-instrument communication scheme has been implemented which can be used to autonomously alter the observing modes. We also outline the plans for in-flight cross-calibration, which will be essential to the joint data reduction and analysis. / Results. The RS instrument package on Solar Orbiter will carry out comprehensive measurements from the solar interior to the inner heliosphere. Thanks to the close coordination between the instrument teams and the European Space Agency, several challenges specific to the RS suite were identified and addressed in a timely manner

    Comparing extrapolations of the coronal magnetic field structure at 2.5 solar radii with multi-viewpoint coronagraphic observations

    The magnetic field shapes the structure of the solar corona but we still know little about the interrelationships between the coronal magnetic field configurations and the resulting quasi-stationary structures observed in coronagraphic images (as streamers, plumes, coronal holes). One way to obtain information on the large-scale structure of the coronal magnetic field is to extrapolate it from photospheric data and compare the results with coronagraphic images. Our aim is to verify if this comparison can be a fast method to check systematically the reliability of the many methods available to reconstruct the coronal magnetic field. Coronal fields are usually extrapolated from photospheric measurements typically in a region close to the central meridian on the solar disk and then compared with coronagraphic images at the limbs, acquired at least 7 days before or after to account for solar rotation, implicitly assuming that no significant changes occurred in the corona during that period. In this work, we combine images from three coronagraphs (SOHO/LASCO-C2 and the two STEREO/SECCHI-COR1) observing the Sun from different viewing angles to build Carrington maps covering the entire corona to reduce the effect of temporal evolution to ~ 5 days. We then compare the position of the observed streamers in these Carrington maps with that of the neutral lines obtained from four different magnetic field extrapolations, to evaluate the performances of the latter in the solar corona. Our results show that the location of coronal streamers can provide important indications to discriminate between different magnetic field extrapolations.Comment: Accepted by A&A the 20th of May, 201

    Antioxidative Effects of Curcumin on the Hepatotoxicity Induced by Ochratoxin A in Rats

    Ochratoxin A (OTA) is a powerful mycotoxin found in various foods and feedstuff, responsible for subchronic and chronic toxicity, such as nephrotoxicity, hepatotoxicity, teratogenicity, and immunotoxicity to both humans and several animal species. The severity of the liver damage caused depends on both dose and duration of exposure. Several studies have suggested that oxidative stress might contribute to increasing the hepatotoxicity of OTA, and several antioxidants, including curcumin (CURC), have been tested to counteract the toxic hepatic action of OTA in various classes of animals. Therefore, the present study was designed to evaluate the protective effect of CURC, a bioactive compound with different therapeutic properties on hepatic injuries caused by OTA in rat animal models. CURC effects were examined in Sprague Dawley rats treated with CURC (100 mg/kg), alone or in combination with OTA (0.5 mg/kg), by gavage daily for 14 days. At the end of the experiment, rats treated with OTA showed alterations in biochemical parameters and oxidative stress in the liver. CURC dosing significantly attenuated oxidative stress and lipid peroxidation versus the OTA group. Furthermore, liver histological tests showed that CURC reduced the multifocal lymphoplasmacellular hepatitis, the periportal fibrosis, and the necrosis observed in the OTA group. This study provides evidence that CURC can preserve OTA-induced oxidative damage in the liver of rat

    Tyrosine kinase inhibitors (TKIs) have revolutionized the treatment of patients with chronic myeloid leukemia (CML). However, continued use of these inhibitors has contributed to the increase in clinical resistance and the persistence of resistant leukemic stem cells (LSCs). So, there is an urgent need to introduce additional targeted and selective therapies to eradicate quiescent LSCs, and to avoid the relapse and disease progression. Here, we focused on emerging BCR-ABL targeted and non-BCR-ABL targeted drugs employed in clinical trials and on alternative CML treatments, including antioxidants, oncolytic virus, engineered exosomes, and natural products obtained from marine organisms that could pave the way for new therapeutic approaches for CML patients

    2D non-LTE radiative modelling of He I spectral lines formed in solar prominences

    The diagnosis of new high-resolution spectropolarimetric observations of solar prominences made in the visible and near-infrared mainly, requires a radiative modelling taking into account for both multi-dimensional geometry and complex atomic models. Hereafter we contribute to the improvement of the diagnosis based on the observation of He I multiplets, by considering 2D non-LTE unpolarized radiation transfer, and taking also into account the atomic fine structure of helium. It is an improvement and a direct application of the multi-grid Gauss-Seidel/SOR iterative scheme in 2D cartesian geometry developed by us. It allows us to compute realistic emergent intensity profiles for the He I 10830 A and D3 multiplets, which can be directly compared to the simultaneous and high-resolution observations made at THeMIS.     Curcumin Supplementation Protects Broiler Chickens Against the Renal Oxidative Stress Induced by the Dietary Exposure to Low Levels of Aflatoxin B1

    Aflatoxin B1 (AFB1) causes hepatotoxicity, immunotoxicity, and kidney damage, and it is included in group I of human carcinogens. The European Commission has established maximum limits of AFB1 in feed, ranging from 5 to 20 ÎĽg/kg. Chicken is moderately sensitive to AFB1, which results in reduced growth performance and economic losses. Oxidative stress triggered by AFB1 plays a crucial role in kidney damage and the antioxidant activity of Curcumin (CURC) could help in preventing such adverse effect. Twenty-days-old broilers were treated for 10 days with AFB1 (0.02 mg/kg feed), alone or in combination with CURC (400 mg/kg feed), to explore the effects on the renal tissue. Animals exposed to AFB1 alone displayed alterations of the oxidative stress parameters compared with controls: serum antioxidant capacity, and enzymatic activity of kidney superoxide dismutase, catalase and glutathione peroxidase were decreased, while renal malondialdehyde levels and NADPH oxidase complex expression were increased. The administration of CURC attenuates all the oxidative stress parameters modified by AFB1 in the chicken kidney, opening new perspectives in the management of aflatoxicosis

    A preliminary assessment of the potential health and genetic impacts of releasing confiscated passerines into the wild: A reduced-risk approach

    The illegal capture and trade of wild birds have long been threats to biodiversity. The rehabilitation and release of confiscated animals may be a useful conservation tool in species management. However, differences between populations regarding health (e.g., different pathogens) and adaptation (e.g., local adaptation) must be taken into account, since both can negatively impact the recipient population. In this pilot study, we used two of the most illegally trafficked Brazilian wild passerine species, namely the red-crested cardinal (Paroaria coronata) and green-winged saltator (Saltator similis) as case studies and assessed some of the health threats that the release of confiscated passerines may pose to free-living birds. We also investigated the level of difference in mitochondrial genetic structure among populations living in different ecoregions. Blood, feces, and oropharyngeal swabs from confiscated (n = 115) and free-living (n = 120) passerines from the release sites were tested for the Newcastle disease virus, Salmonella spp., and Mycoplasma gallisepticum. These are considered major avian diseases by the Brazilian National Avian Health Program. We analyzed mtDNA to study the difference in genetic structure between populations using samples from 127 free-living passerines. We found no evidence of the Newcastle disease virus or Salmonella spp. in confiscated or free-living passerines from either species. However, the levels of infection with M. galissepticum detected in our study for red-crested cardinals and green-winged saltators calls for a high degree of caution in captive release programs. The difference in genetic structure between populations occurring in different regions was low, and was not significant between those from the Pampa/Subtropical Grasslands region. These results suggest that it may be possible to establish a cost-effective and sensitive protocol for releasing confiscated songbirds, provided that further genome-wide studies indicate that the functional genetic diversity among (at least some of the) populations is also low.Fil: Cruz, Cláudio E.
