    Salmonella spp. in pet reptiles in Portugal : prevalence and chlorhexidine gluconate antimicrobial efficacy

Existing data on Salmonella spp. transmission by reptiles in Portugal is extremely scarce. The aim of the present work was to evaluate the prevalence of Salmonella spp. in pet reptiles (snakes, turtles, and lizards), as well as evaluate the isolates’ antimicrobial resistance and virulence profiles, including their ability to form biofilm in the air-liquid interface. Additionally, the antimicrobial effect of chlorhexidine gluconate on the isolates was tested. Salmonella was isolated in 41% of the animals sampled and isolates revealed low levels of antimicrobial resistance. Hemolytic and lypolytic phenotypes were detected in all isolates. The majority (90.63%) of the Salmonella isolates were positive for the formation of pellicle in the air-liquid interface. Results indicate chlorhexidine gluconate is an effective antimicrobial agent, against the isolates in both their planktonic and biofilm forms, demonstrating a bactericidal effect in 84.37% of the Salmonella isolates. This study highlights the possible role of pet reptiles in the transmission of non-typhoidal Salmonella to humans, a serious and increasingly relevant route of exposure in the Salmonella public health framework.info:eu-repo/semantics/publishedVersio

    In the postero-lateral hypothalamus are located two neuronal systems that utilize the neuropeptides melanin-concentrating hormone (MCH) and hypocretins (also called orexins) as neuromodulators. These systems have reciprocal connections between them, and project throughout the central nervous system. MCH has been involved in the generation of sleep, mainly REM sleep, while hypocretins have a critical role in the generation of wakefulness. MCHergic activity is also involved in the pathophysiology of major depressive disorder (MD). In this regards, intracerebral administration of MCH promotes pro-depressive behaviors (i.e., immobility in the forced swimming test) and REM sleep hypersomnia, which is an important trait of depression. Furthermore, the antagonism of the MCHR-1 receptor has a reliable antidepressant effect, suggesting that MCH is a pro-depressive factor. Hypocretins have been also involved in mood regulation; however, their role in depression is still on debate. Taking these data into account, we explored whether systemic subchronical treatment with Fluoxetine (FLX), a serotonergic antidepressant, modifies the concentration of MCH in the cerebrospinal fluid (CSF), as well as the preproMCH mRNA expression. We also evaluated the hypocretinergic system by quantifying the hypocretin-levels in the CSF and the preprohypocretin mRNA expression. Compared to control, FLX increased the levels of preprohypocretin mRNA without affecting the hypocretin-1 CSF levels. On the contrary, FLX significantly decreased the MCH CSF concentration without affecting the preproMCH gene expression. This result is in agreement with the fact that MCH serum level diminishes during the antidepressant treatment in MD, and supports the hypothesis that an increase in the MCHergic activity could have pro-depressive consequences.     Effect of chemical treatments on respiratory process and physical parameters of melon minimally processed

    Neste trabalho avaliou-se o efeito da aplicação de tratamentos químicos na atividade respiratória e no impacto sobre os parâmetros físicos de melões Amarelo minimamente processados. Frutos selecionados, lavados e sanificados foram minimamente processados em forma de cubos, divididos em quatro lotes que constaram de testemunha, cubos tratados com soluções de cloreto de cálcio (1%), ácido ascórbico (1%) e revestidos com alginato de sódio (1%); os quais foram acondicionados em bandejas com tampa, de tereftalato de polietileno (PET) e armazenados a 5±1ºC e 73±5% UR, por 12 dias. Os produtos foram avaliados quanto à taxa respiratória, produção de etileno, perda de massa fresca, coloração, textura e teor de umidade. Verificou-se que, durante o período de armazenamento, independente do tratamento, houve redução da respiração e não se detectou produção de etileno pelos frutos. A perda de massa, a textura e o teor de umidade não foram influenciados pelos tratamentos químicos, porém o uso de película à base de alginato de sódio resultou em melões de coloração mais escura da polpa, em decorrência da cor da solução filmogênica.The effect of the application of chemical treatments was evaluated on the respiratory activity and the impact on the physical parameters of fresh cut melons. Selected fruits were washed and sanitized and minimally processed in cubes, divided in four lots which consisted of: control, treated with calcium chloride (1%), with ascorbic acid (1%) and coated with sodium alginate (1%). The cubes were packed in trays with cover of polyethylene terephtalate (PET) and stored at 5±1ºC and 73±5% HR, during 12 days. The cubes were evaluated on their respiratory rate, ethylene production, loss of fresh mass, coloration, texture and humidity content. During the period of storage, independent of the treatment, a reduction of the respiratory rate occurred; ethylene production was not detected in fruits. Chemical treatments had no influence on loss of fresh mass, texture and humidity content; however the film of sodium alginate resulted in darker coloration of melon pulp due to the film solution color.Cnp

    Synergistic activity of a novel class of azoimidazole dyes with polyvinylpyrrolidone-silver nanoparticles for the development of antibacterial textiles

    The high demand for novel antimicrobial textiles by the medical, health care, hygiene, sportswear, personal protective equipment, and filtration sectors promoted the growth of functional textiles. However, the efficacy of antimicrobial agents against different pathogens is a considerable challenge due to the distinctive mechanisms of action and resistance. The development of novel synergistic antimicrobial agents may offer numerous opportunities to enhance antimicrobial effectiveness, namely boost the activity of individual agents, reduce dosages, minimize toxicity, and amplify the activity spectrum. On the one hand, azo dyes containing a heterocycle present good tinctorial strength and brightness of shades. In particular, the imidazole ring also has interesting antimicrobial, analgesic, and anti-inflammatory properties. On the other hand, silver nanoparticles (AgNPs) are renowned antimicrobial agents against a wide range of microorganisms, but their application is limited by the toxicity observed for effective concentrations. In this work, a novel class of azoimidazoles (AzoIz) and corresponding precursors (AmIz) were conjugated with polyvinylpyrrolidone-coated AgNPs, and their synergistic effect was assessed against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. The results showed interesting antimicrobial properties of the novel AzoIz molecules when combined with a very small concentration of AgNPs. Thus, the application of these conjugates in textiles may lead to highly colored materials with remarkable antimicrobial properties, which worth to be further explored

    Cynara cardunculus: Use in Cheesemaking and Pharmaceutical Applications

    Cynara cardunculus L. is the most widespread species of Cynara genus (f. Asteraceae). This herbaceous perennial plant is native to the Mediterranean region and invasive in other parts of the world, growing naturally in harsh habitat conditions. There are three subspecies: globe artichoke; cultivated cardoon and the progenitor of the two, the wild cardoon. The culture of Cynara cardunculus L. follows an annual growth cycle, emerging in autumn and harvesting in summer. Cynara cardunculus has been considered as a multi-purpose crop due to its relevant biochemical profiles. Inflorescences have been used as food, whereas leaves are a rich source of bioactive compounds. Consequently, larger plants without spines have been selected for technological purposes. Due to its high cellulose and hemicellulose content, the lignocellulosic fraction has been used as solid biofuel, biogas and bioethanol. Both pulp fibers production and seeds oil are suitable for biodiesel production. Over the centuries, the inflorescence pistils of Cynara cardunculus L. have been widely used for cheesemaking. The present chapter gives an overview of the Cynara cardunculus L. emphasizing recent knowledge regarding the use, conservation, preparation and application of Cynara cardunculus in ovine milk cheesemaking, as well as other biotechnological applications

    In Portugal there are 8000 children and young people in Residential Care (RC). The concept of RC quality emerges as the adequacy of the characteristics of the centres to the needs of children. Being a complex and multidimensional construct requires different perspectives and an evaluation by various sources, assuming the voice of children as the main reference in measuring the quality. The quality of the RC is closely related to psychological adjustment, well-being and satisfaction with life of children and adolescents. Until now it has not been performed in Portugal an assessment of the quality of the RC system. At this time there is an ongoing study that aims to assess the quality of Portuguese RC system through needs assessment and psychological adjustment of children in RC, the evaluation of the quality of services provided by centres and understanding of the relationship between these two variables. This symposium aims to present some emerging results of the pilot study of national character research, still in progress, which aims to assess the quality of the RC Portuguese system. In this sense, a brief contextualization of the RC will be in Portugal, followed by the presentation of the main methodology that is used for this assessment – A Comprehensive Evaluation System for Residential Care in Portugal (ARQUA-P). Subsequently, it will be presented the data related to the psychological adjustment of adolescents that are in RC, their self-esteem, well-being and happiness subjective, relating these variables with the quality of the RC.info:eu-repo/semantics/publishedVersio

    XRD Identification of Ore Minerals during Cruises: Refinement of Extraction Procedure with Sodium Acetate Buffer

    The on-board identification of ore minerals during a cruise is often postponed until long after the cruise is over. During the M127 cruise, 21 cores with deep-seafloor sediments were recovered in the Trans-Atlantic Geotraverse (TAG) field along the Mid Atlantic Ridge (MAR). Sediments were analyzed on-board for physicochemical properties such as lightness (L*), pH and Eh. Selected samples were studied for mineral composition by X-ray powder diffraction (XRD). Based on XRD data, sediment samples were separated into high-, low- and non-carbonated. Removal of carbonates is a common technique in mineralogical studies in which HCl is used as the extraction agent. In the present study, sequential extraction was performed with sodium acetate buffer (pH 5.0) to remove carbonates. The ratio between the highest calcite XRD reflection in the original samples (Iorig) vs its XRD-reflection in samples after their treatment with the buffer (Itreat) was used as a quantitative parameter of calcite removal, as well as to identify minor minerals in carbonated samples (when Iorig/Itreat > 24). It was found that the lightness parameter (L*) showed a positive correlation with calcite XRD reflection in selected TAG samples, and this could be applied to the preliminary on-board determination of extraction steps with acetate buffer (pH 5.0) in carbonated sediment samples. The most abundant minerals detected in carbonated samples were quartz and Al- and Fe-rich clays. Other silicates were also detected (e.g., calcic plagioclase, montmorillonite, nontronite). In non-carbonated samples, Fe oxides and hydroxides (goethite and hematite, respectively) were detected. Pyrite was the dominant hydrothermal mineral and Cu sulfides (chalcopyrite, covellite) and hydrothermal Mn oxides (birnessite and todorokite) were mineral phases identified in few samples, whereas paratacamite was detected in the top 20 cm of the core. The present study demonstrates that portable XRD analysis makes it possible to characterize mineralogy at cored sites, in particular in both low- and high-carbonated samples, before the end of most cruises, thus enabling the quick modification of exploration strategies in light of new information as it becomes available in near-real time
