    Isolation and Characterization of an Electricity-producing Strain Shewanella sp. S2 from Marine

    以厦门白城海域的潮间带表面沉积物为菌种来源筛选得到一株具有电催化活性的菌株S2,该菌株的16S rrnA和gyrb基因发育树与SHEWAnEllA OnEIdEnSIS Mr-1同支,相似性分别为98.5%和87%,葡萄糖、木糖、半乳糖等碳源利用及最佳生长的nACl浓度与S.OnEIdEnSIS Mr-1有显著差别,因此初步鉴定为SHEWAnEllA属菌株,命名为SHEWAnEllA SP.S2。初步研究了菌株S2产电活性,在以乳酸作为碳源产电时,电压最高为150MV,相应的电流密度为66.1MA/M2。A new electrochemically active bacterium (exoelectrogen),strain S2,was isolated from coastal marine sediments of Xiamen.The phylogenetic trees based on 16S rRNA and gyrB gene sequences showed that strain S2 formed a lineage within the genus Shewanella and had high similarity (98.5% and 87.0%,respectively) with strain S.oneidensis MR-1.The phenotypic characteristics indicated strain S2 could be distinguished easily from S.oneidensis MR-1 by its culture conditions such as pH,NaCl tolerance and carbon source utilization.Based on these results,it is identified as Shewanella sp.S2.The preliminary current generation experiments showed that S2 could use lactate and xylose for electricity production.When lactate was used as a fuel,the MFC had highest voltage (150 mV) and current density (66.1 mA/m2).中国大洋协会项目(No.DYXM-115-02-2-15

    通过对人发样品中22种元素含量的数据进行变量扩维及压缩筛选处理,选出了影响性别判断较显著的变量,用PlS法处理这些变量组成的数据,得到男性与女性分类清晰的二维判别图及预报模型,并根据所建立的预报模型及人发微量元素的含量判别人的性别,准确率为81%.The data of 22 trace elements concentrations in human hair samples were obtained by ICP AES and GFAAS.The variables which have significant influence on discriminating the sex are selected through the treatment of the concentration data by the variable dimension expansion and the variable selection methods.The discrimination plane figure with the good classification is obtained through the treatment of the data with selected variables by PLS method.The prediction models are built and used to distinguish the human sex according to the element concentrations data in human hair.The accuracy of the prediction is 81%.国家教育委员会留学回国人员科研启动费;福建省自然科学基

    Simultaneous Determination of Five Catechins and Caffeine in Tea by HPLC and Differentiation of Teas by Cluster Analysis

    建立高效液相色谱/二极管阵列检测器(HPLC/DAD)同时测定茶叶中(-)-没食子儿茶素(GC),(-)-表没食子儿茶素(EGC),(-)-表没食子儿茶素没食子酸酯(EGCG),(-)-表儿茶素(EC),(-)-表儿茶素没食子酸酯(ECG),咖啡碱(caffeine)6种组分的分析方法,并采用聚类分析探讨以这6种活性成分为指标对茶叶进行分类的方法。采用C18柱,甲醇和0.05%三氟乙酸水溶液为流动相,梯度洗脱,DAD双波长(210、278 nm)同时检测,采用标准物质保留时间和电喷雾飞行时间质谱(ESI TOF-MS)双重定性。结果表明,各组分的色谱峰均达到基线分离,在210 nm对(-)-没食子儿茶素(GC)定量,278 nm对其它组分定量准确。该法重复性好,灵敏度高,回收率高,已用于不同种类的33种实际茶叶样品的测定。以这6种活性成分的含量为指标,采用聚类分析法可对33个红茶、黑茶、绿茶、乌龙茶样本进行合理分类,并能反映茶叶品质的差异。A method was developed for simultaneous determination of five catechins((-)-gallocatechin,(-)-epigallocatechin,(-)-epigallocatechin-3-gallate,(-)-epicatechin,(-)-epicatechingallate) and caffeine by HPLC with diode array detection(DAD).The tea extract was separated on a C18 column which was then eluted with methanol and 0.05% trifuoroacetic acid solution in gradient elution mode.The chromatograms were simultaneously detected by DAD at λ210 nm for(-)-gallocatechin(GC) and at λ278 nm for the other components.Time of flight mass spectroscopy(TOF-MS) was used for qualitative analysis of real tea sample,the results showed that five catechins and caffeine were baseline separated,indicating that the six components could be determined accurately.The method has been used to determine the catechins and caffeine in 33 tea samples with satisfactory results.Cluster analysis has been used to classify 33 tea samples reasonably and to differentiate the qualities of the tea by using the concentrations of the six components as indices.青岛“2004将才计划”(04-3-JJ-11);; 崂山区崂山茶标准化技术研究(LS-05-KJ1-40

    日本囊对虾人工诱变子一代遗传变异的RAPD分析

    以性腺成熟的日本囊对虾(Marsupenaeus japonicus)作为亲代,进行不同剂量的60Co-γ射线照射.采用RAPD技术对诱变子一代的无节幼体基因组DNA多态性进行了检测.从20个随机寡核苷酸引物共检测177个位点,其中多态位点152个,占85.9%.单个引物获得的标记为6~12个,平均8.85个,分子量在150~2 500 bp之间.遗传相似系数用Ne i的计算方法进行计算,遗传距离则用Lynch的计算方法进行计算.实验数据表明:诱变子代与正常对照组相比,遗传多样性水平较高,诱变产生了较为显著的变异.研究结果证实:在对虾大规模养殖,种质质量严重退化的情况下,人工诱变能够促进基因组更加广泛的变异,为新品种的选育打下坚实的基础;同时证明RAPD技术在检测遗传差异方面具有较高的灵敏度和分辨率

    泛素家族包括泛素及类泛素蛋白,约20种成员蛋白.近年来,泛素家族领域取得了迅猛发展,并已与生物学及医学研究的各个领域相互交叉.泛素家族介导的蛋白质降解和细胞自噬机制的发现分别于2004和2016年获得诺贝尔奖.但是,类泛素蛋白并没有统一规范的中文译名. 2018年4月9日在苏州召开的《泛素家族介导的蛋白质降解和细胞自噬》专著的编委会上,部分作者讨论了类泛素蛋白的中文命名问题,并在随后的\"泛素家族、自噬与疾病\"(Ubiquitinfamily,autophagy anddiseases)苏州会议上提出了类泛素蛋白中文翻译草案,此草案在参加该会议的国内学者及海外华人学者间取得了高度共识.冷泉港亚洲\"泛素家族、自噬与疾病\"苏州会议是由美国冷泉港实验室主办、两年一度、面向全球的英文会议.该会议在海内外华人学者中具有广泛影响,因此,参会华人学者的意见具有一定的代表性.本文介绍了10个类别的类泛素蛋白的中文命名,系统总结了它们的结构特点,并比较了参与各种类泛素化修饰的酶和它们的生物学功能.文章由45名从事该领域研究的专家合作撰写,其中包括中国工程院院士1名,相关学者4名,长江学者3名,国家杰出青年科学基金获得者18名和美国知名高校华人教授4名.他们绝大多数是参加编写即将由科学出版社出版的专著《泛素家族介导的蛋白质降解和细胞自噬》的专家

    Determination of Lead Content in Standard Materials of Lake Sediment and Tea by ID-ICP-MS

    采用2种微波消解体系(HNO3-H2O2和HNO3-H2O2-HF)及3种定量模式(同位素稀释法、标准工作曲线法和标准加入法),对沉积物和茶叶标准物质中的铅进行测定,结果表明:同位素稀释质谱法的各项检测指标显著优于其它两种定量模式(茶叶中铅含量测定值为4.30μg/g),标准加入法次之(3.75μg/g),标准工作曲线法较差(3.61μg/g)。二种微波消解体系中,加HF酸优于不加HF酸,不加HF,茶叶中铅的回收率仅有89%,加入HF则上升为98%。Three different quantitative modes,namely external calibration,standard addition and isotope diluˉtion were compared for the determination of lead contents in lake sediment and tea standard materials with two digestion systems.The results showed that the isotope dilution gave the highest accuracy and precision in comˉparison with the other two modes.But the results were affected by the digestion system.For example,the reˉcoveries of lead content for tea were98%and89%for HNO 3 -H 2 O 2 -HF and HNO 3 -H 2 O 2 digesting sysˉtem,respectively.福建省重大科技项目子课题(2003Y005-04);; 厦门大学现代分析科学教育部重点实验室开放课题(B40402
