    Impact of rotavirus vaccination on rotavirus genotype distribution and diversity in England, September 2006 to August 2016.

    Introduction Rotavirus vaccination with the live-attenuated monovalent (a G1P[8] human rotavirus strain) two-dose Rotarix vaccine was introduced in England in July 2013. Since then, there have been significant reductions in rotavirus gastroenteritis incidence. Aim We assessed the vaccine's impact on rotavirus genotype distribution and diversity 3 years post-vaccine introduction. Methods Epidemiological and microbiological data on genotyped rotavirus-positive samples between September 2006 and August 2016 were supplied by EuroRotaNet and Public Health England. Multinomial multivariable logistic regression adjusting for year, season and age was used to quantify changes in genotype prevalence in the vaccine period. Genotype diversity was measured using the Shannon's index (H') and Simpson's index of diversity (D). Results We analysed genotypes from 8,044 faecal samples. In the pre-vaccine era, G1P[8] was most prevalent, ranging from 39% (411/1,057) to 74% (527/709) per year. In the vaccine era, G1P[8] prevalence declined each season (35%, 231/654; 12%, 154/1,257; 5%, 34/726) and genotype diversity increased significantly in 6-59 months old children (H' p < 0.001: D p < 0.001). In multinomial analysis, G2P[4] (adjusted multinomial odds ratio (aMOR): 9.51; 95% confidence interval (CI): 7.02-12.90), G3P[8] (aMOR: 2.83; 95% CI: 2.17-3.81), G12P[8] (aMOR: 2.46; 95% CI: 1.62-3.73) and G4P[8] (aMOR: 1.42; 95% CI: 1.02-1.96) significantly increased relative to G1P[8]. Conclusions In the context of reduced rotavirus disease incidence, genotype diversity has increased, with a relative change in the dominant genotype from G1P[8] to G2P[4] after vaccine introduction. These changes will need continued surveillance as the number and age of vaccinated birth cohorts increase in the future

    Con el objetivo de generar un conjunto de datos que fijen las l&iacute;neas prioritarias para la puesta en valor de los sitios arqueol&oacute;gicos de la Provincia de Catamarca; el presente trabajo se elaboro en base a la aplicaci&oacute;n de una metodolog&iacute;a capaz de generar una serie de datos duros sobre el estado actual de 12 sitios y/o yacimientos, tratando de fijar en el proceso, los principios b&aacute;sicos para su gesti&oacute;n de manera sostenible, respetando las condiciones del entorno, as&iacute; como a las comunidades del lugar. El debate respecto de las t&eacute;cnicas de control, limitando el impacto de uso, ofreciendo al mismo tiempo el m&aacute;ximo disfrute a todos los visitantes posibles, sin contravenir las limitaciones impuestas por las condiciones ambientales y sociales, es una de las ambiciones a lograr; as&iacute; como el diagn&oacute;stico sobre diferentes enfoques o zonificaciones para la planificaci&oacute;n e implantaci&oacute;n de infraestructura m&iacute;nima de servicios tur&iacute;sticos en el &aacute;rea protegida de cada sitio. La orientaci&oacute;n sobre definici&oacute;n, evaluaci&oacute;n, gesti&oacute;n y uso de los datos de impacto tur&iacute;stico obtenido en cada uno de los sitios, ha respondido en todos los casos en la tendencia a mejorar la calidad de la experiencia tur&iacute;stica, de la conservaci&oacute;n de la diversidad biol&oacute;gica y cultural, y de contribuir al desarrollo de las comunidades locales. Estrat&eacute;gicamente, y a los efectos de considerar la puesta en valor de cada sitio arqueol&oacute;gico, se propone el marco de acci&oacute;n del Limite de Cambio Aceptable (LAC en su sigla inglesa), como una manera de trabajo concreta para la gesti&oacute;n de visitantes, con relaci&oacute;n a establecer metas y cualquier acci&oacute;n posterior a su establecimiento para el turismo de &aacute;reas protegidas, ofreciendo una manera de acci&oacute;n que no se centra tanto en las relaciones entre los niveles de uso y de impacto, sino en determinar las condiciones ambientales y sociales deseables para la actividad de los visitantes y las actividades gestoras requeridas para alcanzar esas condiciones, utilizando un proceso sistem&aacute;tico, expl&iacute;cito, defendible y racional.Tópico 7: Patrimonio Arqueológico.     Genetic diversity of porcine group A rotavirus strains in the UK

    Rotavirus is endemic in pig farms where it causes a loss in production. This study is the first to characterise porcine rotavirus circulating in UK pigs. Samples from diarrheic pigs with rotavirus enteritis obtained between 2010 and 2012 were genotyped in order to determine the diversity of group A rotavirus (GARV) in UK pigs. A wide range of rotavirus genotypes were identified in UK pigs: six G types (VP7); G2, G3, G4, G5, G9 and G11 and six P types (VP4); P[6], P[7], P[8], P[13], P[23], and P[32]. With the exception of a single P[8] isolate, there was less than 95% nucleotide identity between sequences from this study and any available rotavirus sequences. The G9 and P[6] genotypes are capable of infecting both humans and pigs, but showed no species cross-over within the UK as they were shown to be genetically distinct, which suggested zoonotic transmission is rare within the UK. We identified the P[8] genotype in one isolate, this genotype is almost exclusively found in humans. The P[8] was linked to a human Irish rotavirus isolate in the same year. The discovery of human genotype P[8] rotavirus in a UK pig confirms this common human genotype can infect pigs and also highlights the necessity of surveillance of porcine rotavirus genotypes to safeguard human as well as porcine health

    Assignment of the group A rotavirus NSP4 gene into genotypes using a hemi-nested multiplex PCR assay: a rapid and reproducible assay for strain surveillance studies

    The rotavirus non-structural protein NSP4 has been implicated in a number of biological functions during the rotavirus cellular cycle and pathogenesis, and has been addressed as a target for vaccine development. The NSP4 gene has been classified into six genotypes (A-F). A semi-nested triplex PCR was developed for genotyping the major human NSP4 genotypes (A-C), which are common in human rotavirus strains but are also shared among most mammalian rotavirus strains. A total of 192 previously characterized human strains representing numerous G and P type specificities (such as G1P[8], G1P[4], G2P[4], G3P[3], G3P[8], G3P[9], G4P[6], G4P[8], G6P[4], G6P[9], G6P[14], G8P[10], G8P[14], G9P[8], G9P[11], G10P[11], G12P[6] and G12P[8]) were tested for NSP4 specificity by the collaborating laboratories. An additional 35 animal strains, including the reference laboratory strains SA11 (simian, G3P[2]), NCDV (bovine, G6P[1]), K9 and CU-1 (canine, G3P[3]), together with 31 field isolates (canine, G3P[3]; feline, G3P[9]; porcine, G2P[23], G3P[6], G4P[6], G5P[6], G5P[7], G5P[26], G5P[27], G9P[6] and G9P[7]) were also successfully NSP4-typed. Four human G3P[9] strains and one feline G3P[9] strain were found to possess an NSP4 A genotype, instead of NSP4 C, suggesting a reassortment event between heterologous strains. Routine NSP4 genotyping may help to determine the genomic constellation of rotaviruses of man and livestock, and identify interspecies transmission of heterologous strain

    Emergence of unusual human rotavirus strains in Salento, Italy, during 2006–2007

    <p>Abstract</p> <p>Background</p> <p>In recent years, rotavirus genotyping by RT-PCR has provided valuable information about the diversity of rotaviruses (RV) circulating throughout the world.</p> <p>The purpose of the present study was to monitor the prevalence of the different G and P genotypes of rotaviruses circulating in Salento and detect any uncommon or novel types.</p> <p>Methods</p> <p>During the period from January 2006 to December 2007, a total of 243 rotavirus positive stool samples were collected from children with diarrhoea admitted to four Hospitals in the province of Lecce (Copertino, Galatina, Gallipoli and Tricase).</p> <p>All the specimens were tested for RV by real time PCR and genotyped for VP7 (G-type) and VP4 (P-type) gene by reverse transcription (RT) and multiplex PCR using different type specific primers.</p> <p>Results</p> <p>In course of this study we identified 4 common G&P combinations viz. G2P[8], G1P[8], G2P[4] and G9P[8] amongst 59.8% of the typeable rotavirus positives.</p> <p>Rotavirus G2P[8] was recognized as the most widespread genotype during the sentinel-based survey in Salento.</p> <p>The detection of other novel and unusual strains, such as G2P[10], G4P[10], G8P[4], G9P[11] and G10P[8] is noteworthy.</p> <p>Furthermore, a significant number of mixed infections were observed during the survey period but G3P[8] rotaviruses were not detected.</p> <p>Conclusion</p> <p>This study highlights the genetic diversity among rotaviruses isolated from children in Salento and the emergence of some novel strains. Therefore, it is highly essential to continuously monitor for these strains so as to assess the impact of vaccines on RV strains circulating in Salento and understand the effect of strain variation on efficacy of presently available vaccines.</p

    Burden of disease and circulating serotypes of rotavirus infection in sub-Saharan Africa: systematic review and meta-analysis.

    Two new rotavirus vaccines have recently been licensed in many countries. However, their efficacy has only been shown against certain serotypes commonly circulating in Europe, North America, and Latin America, but thought to be globally important. To assess the potential impact of these vaccines in sub-Saharan Africa, where rotavirus mortality is high, knowledge of prevalent types is essential because an effective rotavirus vaccine is needed to protect against prevailing serotypes in the community. We did two systematic reviews and two meta-analyses of the most recent published data on the burden of rotavirus disease in children aged under 5 years and rotavirus serotypes circulating in countries in sub-Saharan Africa. Eligible studies were selected from PubMed/Medline, Cochrane Library, EmBase, LILACS, Academic Search Premier, Biological Abstracts, ISI Web of Science, and the African Index Medicus. Depending on the heterogeneity, DerSimonian-Laird random-effects or fixed-effects models were used for meta-analyses. Geographical variability in rotavirus burden within countries in sub-Saharan Africa is substantial, and most countries lack information on rotavirus epidemiology. We estimated that annual mortality for this region was 243.3 (95% CI 187.6-301.7) deaths per 100,000 under 5 years (ie, a total of 300,000 children die of rotavirus infection in this region each year). The most common G type detected was G1 (34.9%), followed by G2 (9.1%), and G3 (8.6%). The most common P types detected were P[8] (35.5%) and P[6] (27.5%). Accurate information should be collected from surveillance based on standardised methods in these countries to obtain comparable data on the burden of disease and the circulating strains to assess the potential impact of vaccine introduction

    Burden of community-acquired and nosocomial rotavirus gastroenteritis in the pediatric population of Western Europe: a scoping review

    <p>Abstract</p> <p>Background</p> <p>Rotavirus affects 95% of children worldwide by age 5 years and is the leading cause of severe dehydrating diarrhea. The objective of this review was to estimate the burden of rotavirus gastroenteritis (RVGE) in the Western European pediatric population.</p> <p>Methods</p> <p>A comprehensive literature search (1999-2010) was conducted in PubMed and other sources (CDC; WHO, others). Data on the epidemiology and burden of RVGE among children < 5 years-old in Western Europe --including hospital-acquired disease--were extracted.</p> <p>Results</p> <p>76 studies from 16 countries were identified. The mean percentage of acute gastroenteritis (AGE) cases caused by rotavirus ranged from 25.3%-63.5% in children < 5 years of age, peaking during winter. Incidence rates of RVGE ranged from 1.33-4.96 cases/100 person- years. Hospitalization rates for RVGE ranged from 7% to 81% among infected children, depending on the country. Nosocomial RVGE accounted for 47%-69% of all hospital-acquired AGE and prolonged hospital stays by 4-12 days. Each year, RVGE incurred 0.540.54- 53.6 million in direct medical costs and 1.71.7-22.4 million in indirect costs in the 16 countries studied. Full serotyping data was available for 8 countries. G1P[8], G2P[4], G9P[8], and G3P[8] were the most prevalent serotypes (cumulative frequency: 57.2%- 98.7%). Serotype distribution in nosocomial RVGE was similar.</p> <p>Conclusions</p> <p>This review confirms that RVGE is a common disease associated with significant morbidity and costs across Western Europe. A vaccine protecting against multiple serotypes may decrease the epidemiological and cost burden of RVGE in Western Europe.</p

    ODS ferritic steels obtained from gas atomized powders through the STARS processing route: Reactive synthesis as an alternative to mechanical alloying

    Oxide Dispersion Strengthened Ferritic Stainless Steels (ODS FS) are candidate materials for structural components in fusion reactors. Their ultrafine microstructure and the presence of a very stable dispersion of Y-Ti-O nanoclusters provide reasonable fracture toughness, high mechanical and creep strength, and resistance to radiation damage at the operation temperature, up to about 750 °C. An innovative route to produce ODS FS with composition Fe-14Cr-2W-0.3Ti-0.3Y2O3 (wt.%), named STARS (Surface Treatment of gas Atomized powder followed by Reactive Synthesis), is presented. This route avoids the mechanical alloying (MA) of the elemental or prealloyed powders with yttria to dissolve the yttrium in the ferritic matrix. In this study, starting powders containing Ti and Y are obtained by gas atomization at laboratory and industrial scale. Then, a metastable Cr- and Fe- rich oxide layer is formed on the surface of the powder particles. During consolidation by HIP the metastable oxide layer at Prior Particle Boundaries (PPBs) dissociates, the oxygen diffuses towards saturated solutions or metallic Ti- and Y-rich particles, and Y-Ti-O nano-oxides (mainly Y2TiO5) precipitate in the ferritic matrix.
