Desenvolvimento de comportamentos para robô humanoide
Mestrado em Engenharia de Computadores e TelemáticaHumanoid robotics is an area of active research. Robots with human body
are better suited to execute tasks in environments designed for humans.
Moreover, people feel more comfortable interacting with robots that have
a human appearance. RoboCup encourages robotic research by promoting
robotic competitions. One of these competitions is the Standard Platform
League (SPL) in which humanoid robots play soccer. The robot used is
the Nao robot, created by Aldebaran Robotics. The di erence between
the teams that compete in this league is the software that controls the robots.
Another league promoted by RoboCup is the 3D Soccer Simulation
League (3DSSL). In this league the soccer game is played in a computer
simulation. The robot model used is also the one of the Nao robot. However,
there are a few di erences in the dimensions and it has one more
Degree of Freedom (DoF) than the real robot. Moreover, the simulator
cannot reproduce reality with precision. Both these leagues are relevant
for this thesis, since they use the same robot model. The objective of this
thesis is to develop behaviors for these leagues, taking advantage of the
previous work developed for the 3DSSL. These behaviors include the basic
movements needed to play soccer, namely: walking, kicking the ball, and
getting up after a fall. This thesis presents the architecture of the agent
developed for the SPL, which is similar to the architecture of the FC Portugal
team agent from the 3DSSL, hence allowing to port code between both
leagues easily. It was also developed an interface that allows to control a
leg in a more intuitive way. It calculates the joint angles of the leg, using
the following parameters: three angles between the torso and the line connecting
hip and ankle; two angles between the foot and the perpendicular
of the torso; and the distance between the hip and the ankle. It was also
developed an algorithm to calculate the three joint angles of the hip that
produce the desired vertical rotation, since the Nao robot does not have a
vertical joint in the hip. This thesis presents also the behaviors developed
for the SPL, some of them based on the existing behaviors from the 3DSSL.
It is presented a behavior that allows to create robot movements by de ning
a sequence of poses, an open-loop omnidirectional walking algorithm, and
a walk optimized in the simulator adapted to the real robot. Feedback was
added to this last walk to make it more robust against external disturbances.
Using the behaviors presented in this thesis, the robot achieved a forward
velocity of 16 cm/s, a lateral velocity of 6 cm/s, and rotated at 40 deg/s.
The work developed in this thesis allows to have an agent to control the
Nao robot and execute the basic low level behaviors for competing in the
SPL. Moreover, the similarities between the architecture of the agent for
the SPL with that of the agent from the 3DSSL allow to use the same high
level behaviors in both leagues.
Outra liga presente no RoboCup é a 3D Soccer Simulation League (3DSSL).
In vitro and in vivo studies of proatherogenic effects induced by end-products of cholesteryl esters oxidation
ABSTRACT:Atherosclerosis is a chronic inflammatory disease and the major worldwide cause of human
death. It is initiated by the constitutive uptake of modified low density lipoproteins trapped
in the arterial intima by monocyte-derived macrophages, leading to the formation of foam
cells. The oxidation levels and the quantity of lipid loading in macrophages influence their
inflammatory phenotype and function. With time, atherosclerotic macrophages become
dysfunctional and a clear trigger for this process relies on lysosomal malfunction. Eventually
this process leads to apoptotic cells and toxic lipid accumulation in arterial intima, culminating
in the formation of an irreversible advanced atheroma. Thus, the establishment of
a correlation between the composition of lipid species and the irreversibility/instability of
atherosclerotic lesions can contribute to the development of powerful tools for the diagnosis
and treatment of the cardiovascular disease burden. The main objective of this work was
the study of cholesteryl hemiesters (ChE), an oxidized product of cholesteryl esters, as an
atherogenic compound. Here, using the shotgun lipidomics technique, we showed that these
lipids are increased in the plasma of cardiovascular disease patients. We subsequently evaluated
the in vitro and in vivo atherogenic relevance of these oxidized lipids. Monocytes and
macrophages exposed to ChE presented an unusual inflammatory profile, presenting both
pro- and anti-inflammatory markers. ChE-loaded macrophages revealed endocytic trafficking
delays and dysfunctional enlarged lysosomes. The sub cellular location of the observed enlarged
stressed lysosomes was more peripheral than lysosomes from control macrophages.
Furthermore, these lysosomes from ChE-loaded macrophages displayed an increase in their
exocytic capacity. In addition, macrophages stimulated with ChE revealed a metabolic shift,
which, as with the inflammatory response, was dependent on toll-like receptor 4 signaling.
These foamy macrophages also presented an increased proliferative capacity as compared to
the control macrophages. Finally, using zebrafish larvae, several of the in vitro atherogenic
properties of ChE were confirmed in vivo, namely lipid accumulation, inflammation and
macrophage lysosomal dysfunction. Altogether, the work presented here contributes to the
identification of new etiologic compounds in human atherosclerotic lesions and advances
our mechanistic understanding of the alterations observed in atherosclerotic macrophages.RESUMO: A aterosclerose é uma doença inflamatória crónica e a principal causa de morte no mundo
inteiro.
avaliamos a relevância aterogénica destes lípidos oxidados, através de estudos in vitro e in
vivo.
ChE observadas in vitro foram confirmadas in vivo, nomeadamente, a acumulação de lípidos,
With technologies such as SDN and NFV pushing the the development of the next generation networks, new paradigms, such as Fog Computing, appeared in the network scene. However, these technologies have been associated with the network infrastructure, such as the datacenter. In order for these technologies to be used, for instance, in a Fog Computing scenario it is necessary to, therefore, study and
develop these technologies to form new control and operation mechanisms. So, a Fog Computing scenario composed by resource-constrained devices, typical in these types of situations, was developed, and, a solution proposal is presented. The solution consists in customizing an existent VIM, OpenVIM, to this kind of devices, after the implementation of the solution, where a Raspberry Pi is used to exemplify this type of devices. Tests are done to measure and compare this devices to more powerful ones. The tests are comprised by benchmarks runs, focusing on instantiation times, and power consumption. The results show some drawbacks inherent to this kind of devices when compared to more powerful ones. However,
it is possible to see the potential that this kind of devices might have in the near future.
LULC Map Comparison: Comparison and validation of land use land cover maps derived from satellite imagery
Dissertation submitted in partial fulfilment of the requirements for the Degree of Master of Science in Geospatial TechnologiesThe technological evolution of remote sensing techniques has allowed for the ever-growing creation of land use land cover maps. Nowadays mapping entities from all around the globe are capable of producing their own products, be it for their region or country, or for a whole continent or even the whole world. However, this raises an issue regarding the comparison of these maps, in one sense direct comparison is difficult due to the lack of harmonization of the data, be it the nomenclature’s classification, or technical specifications of the imagery. In another sense, the creation of global cover maps, or continent-wide maps, hinges the ability to accurately classify LULC appropriately due to the complexity of land cover, often leaving specific regions of the map with a less accurate classification, even though the overall one is good. Throughout this study, five maps from five different mapping entities will be compared and evaluated, these maps are COSsim, ELC10, ESA Worldcover, ESRILC and S2GLC. The study area is Continental Portugal, and the main objective is to understand how the international mapping entities’ maps compare with the Portuguese map of COSsim, by observing nomenclature differences and accuracy scores. As well as understand what the impact in accuracy is, in European cover or World cover maps, when only analyzing them for the study area of Continental Portugal. The results obtained showed that most international maps proved to have a much smaller accuracy score for Continental Portugal, most of these even having a 20% to 30% drop in their overall accuracy. This research helps understand the necessity for the harmonization of nomenclatures, and at the same time investment necessary for national mapping entities to create their own more accurate maps
Immunotherapy in melanoma : the role of nk cells
Durante a progressão do
produzir interleucina 2 (IL-2).
contexto do cancro.
60%.
marcadores de prognóstico e estratégias terapêuticas.ABSTRACT: Melanoma is the fifth most common cancer in men (seventh in women) and its incidence is
increasing more rapidly than that of any other malignancy. It is the most aggressive type of
skin cancer; although it accounts for 2% of all skin cancers it leads to more than 70% of their
deaths. In fact, melanoma responds poorly to systemic therapy and the overall prognosis of
patients with distant metastasized melanoma remains poor; the 2-year overall survival rate
ranges from 18 to 40%.
In the context of cancer two opposite immune responses exist: an acute T helper (Th) 1 antitumoral
response that requires a substantial autoimmune attack; and a pro-tumorigenic
inflammatory response, characterized by an immune tolerance via active immunosuppressive
mechanisms. During tumor progression there is an imbalance with a stronger pro-tumorigenic
inflammatory response, while the anti-tumoral Th1 cells display a dysfunctional phenotype.
The new concept of “T cell exhaustion”, that has been developped in the past few years, can
explain T cell dysfunction. This “T cell exhaustion”, either caused by environmental influence
or other factors, was first described in chronic infectious diseases and more recently in
different types of cancer, particularly in melanoma. This phenotype is characterized by early
loss of proliferative capacity, cytotoxic potential, and the ability to produce interleukin (IL)-2.
The concept of T cell exhaustion in the context of metastatic cancer has been reinforced by
the recent success of immunotherapies targeting the exhaustion markers cytotoxic Tlymphocyte-
associated protein 4 (CTLA-4) and programmed (cell) death 1 (PD-1) in
advanced melanoma. However, the phenotype and function of natural killer (NK) cells in
different phases of tumor progression and their relationship with prognostic factors is still
unknown.
NK cells are innate immune cells, prone to target tumor cells, whose function depends on the
balance between activating and inhibitory receptors, and on the expression of the respective
ligands in their microenvironment. T cell immunoglobulin domain and mucin domain 3 (Tim-
3), another T cell exhaustion marker along with CTLA-4 and PD-1, behaves as an inhibitory
receptor in T cells and is key in the maintenance of immune tolerance. Tim-3 is also
expressed in NK cells; however its role in modulating the function of these innate effector
cells remains unclear, particularly in human disease. The aims of this study were: to characterize the phenotype and function of NK cells from
melanoma patients; to identify potential players in the mechanism of NK cell exhaustion as
prognostic markers; to define the role of Tim-3 expression in NK cells from advanced
melanoma patients; and to study the effect of checkpoint blockade in NK cell phenotype and
function in the context of advanced melanoma patients.
Our data showed that NK cells from patients with advanced melanoma display an exhausted
phenotype that is characterized by upregulation of inhibitory receptors (Killer cell
Immunoglobulin-like receptor 3DL1 and 2DL3 - KIR3DL1 and KIR2DL3, respectively),
downregulation of activating receptors (natural-killer group 2D (NKG2D), natural killer p46
(NKp46) and DNAX accessory molecule-1 (DNAM-1)), unresponsiveness to IL-2
stimulation, downregulation of the transcription factors T-box (T-bet) and eomesodermin
(Eomes), and defective function. The latter includes the main NK cell functions, cytotoxicity,
interferon gamma (IFN-γ) production and proliferation.
NK cells acquire this exhausted phenotype gradually as disease advances from early (stages I
and II) to later stages (stages III and IV). Moreover, the exhaustion of NK cells is associated
with clinicopathological parameters that have known prognostic value, such as thickness and
presence/absence of metastases (regional and distant). In addition, high serum levels of
soluble MHC class I chain-related genes A (sMICA), an NKG2D ligand (NKG2DL) that may
be involved in the mechanism of NK cell exhaustion, identify patients with shorter diseasefree
and overall survival.
We also identified Tim-3 as an inhibitory receptor over-expressed in NK cells from advanced
melanoma patients that plays a key role in the exhausted phenotype of these cells.
Significantly, we demonstrated that Tim-3 blockade partially reversed this exhausted
phenotype. Therefore, in average, it was possible to enhance cytotoxicity by 20%, IFNγ
production by 15% and proliferation by 60%.
Finally, we characterized the effect of ipilimumab (anti-CTLA-4) on exhausted NK cells from
advanced melanoma patients. Remarkably, NK cells from responders to ipilimumab treatment
have higher levels of IL-2 receptor (IL-2R), and consequently, are more responsive to IL-2
stimulation and more cytotoxic. These data open exciting avenues for new NK cell-based therapies, including targeting Tim-3
in the context of melanoma. Moreover, a better understanding of the mechanism behind NK
cell exhaustion will help defining new prognostic markers and therapeutic strategies
Liver cirrhosis is a major global public health problem with high morbidity and mortality rates. Few epidemiological studies on the subject are available in Brazil, so this study was carried out with the objective of evaluating the profile of mortality from liver cirrhosis and other liver diseases in the state of Santa Catarina and Brazil, in the period from 2009 to 2019. To achieve the objectives, data available on the Global Burden Disease platform were analyzed. Data were collected on sociodemographic variables (age range, gender) as well as the cause of cirrhosis (hepatitis B, hepatitis C, alcohol,non-alcoholic fat disease (NAFLD) and other causes). Over the eleven years evaluated, there was no evident decrease in the mortality rate for both Santa Catarina and Brazil. The main cause of death from liver cirrhosis in Brazil and Santa Catarina was alcohol consumption, followed by hepatitis C and hepatitis B. Regarding the distribution between genders, categorized by age groups, a predominance in males was observed, with even higher mortality rates after 40 years of age. For women, the growth in the mortality rate was gradual, with higher rates from 65 years onwards, and these scenarios were observed both for data from Brazil and Santa Catarina. The study also indicated that the average mortality rate related to cirrhosis and other associated diseases in the state of Santa Catarina is lower than the national average, a situation observed in all years of the study. We believe that the data from this study can contribute to the positive interference of public management through the creation of policies that aim from immunization and treatment of underlying diseases, to awareness and programs to reduce alcohol consumption, drugs, indiscriminate use of pharmaceuticals, as well as better diet and lifestyle habits
The objective of this work was to determine the biological and morphological parameters of Dysdercus maurus fed with cotton (Gossypium hirsutum) seeds with a high or low oil content, as well as to identify genotypes to be used in breeding programs as sources of resistance to this stink bug. The development, survival, and reproduction of the cotton stainer bug were determined in a completely randomized experimental design. The treatments consisted of the insect nymphs being feed with cotton seeds of the CNPA 2001-5581 (high oil content) or CNPA 2001-5087 (low content) genetic line. Survival, weight, and morphological parameters of the bug were determined. The survival of second- and third-instar nymphs and of the total nymph stage of D. maurus was lower with cotton seeds with a low oil content. The body length and head width of D. maurus adults were greater, but pronotum length and width were smaller and the females heavier with cotton seeds with a high oil content.
