Artificial membrane-like environments for in vitro studies of purified G-protein coupled receptors
AbstractFunctional reconstitution of transmembrane proteins remains a significant barrier to their biochemical, biophysical, and structural characterization. Studies of seven-transmembrane G-protein coupled receptors (GPCRs) in vitro are particularly challenging because, ideally, they require access to the receptor on both sides of the membrane as well as within the plane of the membrane. However, understanding the structure and function of these receptors at the molecular level within a native-like environment will have a large impact both on basic knowledge of cell signaling and on pharmacological research. The goal of this article is to review the main classes of membrane mimics that have been, or could be, used for functional reconstitution of GPCRs. These include the use of micelles, bicelles, lipid vesicles, nanodiscs, lipidic cubic phases, and planar lipid membranes. Each of these approaches is evaluated with respect to its fundamental advantages and limitations and its applications in the field of GPCR research. This article is part of a Special Issue entitled: Membrane protein structure and function
Differential gene expression profiles in foetal skin of Rex rabbits with different wool density
[EN] This study investigated the mechanisms controlling hair follicle development in the Rex rabbit. The Agilent rabbit gene expression microarray was used to determine differentially expressed genes in Rex rabbit foetuses with different wool densities. The expression patterns of selected differentially-expressed genes were further investigated by quantitative real-time PCR. Compared to low wool density rabbits, 1342 differentially expressed probes were identified in high wool density rabbits, including 950 upregulated probes and 392 downregulated probes. Gene ontology analysis revealed that the most upregulated differentially expressed probes belonged to receptors and the most downregulated differentially expressed probes belonged to DNA binding molecules. Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that the differentially expressed probes were mainly involved in the sonic hedgehog (Shh) and Eph signalling pathways. The results also suggest that transforming growth factor-beta 1, growth hormone receptor, and the keratin-associated protein 6.1 genes, as well as the Shh and Eph signalling pathways, may be involved in the regulation of hair follicle developmental in Rex rabbits. The authors thank Ms. Libo Huang for her technical assistance. Using SPOT Data and FRAGSTAS to Analyze the Relationship between Plant Diversity and Green Space Landscape Patterns in the Tropical Coastal City of Zhanjiang, China
Urban green spaces provide a host of ecosystem services, the quantity and structure of which play an important role in human well-being. Rapid urbanization may modify urban green spaces, having various effects on plant diversity. Tropical coastal cities have urbanized rapidly in recent decades, but few studies have been conducted with a focus on their green spaces. We studied the responses of cultivated and spontaneous plants, both key components of urban flora, to the landscape structure of urban green spaces and possible social drivers. We analyzed existing relationships between plant diversity indices, urban green space landscape metrics (using Systeme Probatoire d’Observation de la Terre (SPOT) data,), and social factors, including the type, population density, construction age, and GPS coordinates of each Urban Functional Unit, or UFU. We found that UFUs with more green space patches had higher cultivated and spontaneous species richness than those with fewer green space patches. Spontaneous species richness decreased when green space patches became fragmented, and it increased when green space patches were more connected (e.g., via land bridges). Conversely, cultivated species richness increased with green space patch fragmentation. The phylogenetic diversity of both cultivated and spontaneous plants were weakly associated with green space structure, which was strongly driven by land use. Old UFUs and those with larger populations had more green space patches overall, although they tended to be small and fragmented. Green space patch density was found to increase as the UFU age increased. From the viewpoint of knowledge transfer, understanding the effects and drivers of landscape patterns of urban green spaces could inform the development of improved policies and management of urban green space areas.Peer Reviewe
Investigation of ferrofluid cooling in modular permanent magnet machines
Compared with conventional non-modular machines, the flux gaps in alternate stator teeth of the modular machines can increase the winding factor and provide flux focusing effect, and hence can increase the torque/power density. In addition, the flux gaps can also be used as cooling channels to improve the thermal performance. This paper investigates an advanced cooling approach using ferrofluid as coolant to fill in the cavity around end-windings and flux gaps. The multiphysics modelling in this paper has shown that the influence of the flux gaps on machine thermal performances comes from two aspects: (1) the gravitational effect, the flux gaps allow more self-circulating coolant to transfer heat to the housing. This helps to reduce machine temperature by around 5°C for a flux gap width of 2mm; (2) the magnetic body force, i.e., the thermomagnetic effect. This is very effective in non-modular machine cooling. But its efficiency slightly deteriorates in the modular machines. This is because the self-circulation of ferrofluid in the flux gaps due to the gravitational effect has been negatively affected by the thermomagnetic effect
Future Directions in Parity Violation: From Quarks to the Cosmos
I discuss the prospects for future studies of parity-violating (PV)
interactions at low energies and the insights they might provide about open
questions in the Standard Model as well as physics that lies beyond it. I cover
four types of parity-violating observables: PV electron scattering; PV hadronic
interactions; PV correlations in weak decays; and searches for the permanent
electric dipole moments of quantum systems.Comment: Talk given at PAVI 06 workshop on parity-violating interactions,

Harmonic maps from degenerating Riemann surfaces
We study harmonic maps from degenerating Riemann surfaces with uniformly
bounded energy and show the so-called generalized energy identity. We find
conditions that are both necessary and sufficient for the compactness in
and modulo bubbles of sequences of such maps.Comment: 27 page
Public mental health problems during COVID-19 pandemic: a large-scale meta-analysis of the evidence
The coronavirus disease 2019 (COVID-19) pandemic has exposed humans to the highest physical and mental risks. Thus, it is becoming a priority to probe the mental health problems experienced during the pandemic in different populations. We performed a meta-analysis to clarify the prevalence of postpandemic mental health problems. Seventy-one published papers (n = 146,139) from China, the United States, Japan, India, and Turkey were eligible to be included in the data pool. These papers reported results for Chinese, Japanese, Italian, American, Turkish, Indian, Spanish, Greek, and Singaporean populations. The results demonstrated a total prevalence of anxiety symptoms of 32.60% (95% confidence interval (CI): 29.10-36.30) during the COVID-19 pandemic. For depression, a prevalence of 27.60% (95% CI: 24.00-31.60) was found. Further, insomnia was found to have a prevalence of 30.30% (95% CI: 24.60-36.60). Of the total study population, 16.70% (95% CI: 8.90-29.20) experienced post-traumatic stress disorder (PTSD) symptoms during the COVID-19 pandemic. Subgroup analysis revealed the highest prevalence of anxiety (63.90%) and depression (55.40%) in confirmed and suspected patients compared with other cohorts. Notably, the prevalence of each symptom in other countries was higher than that in China. Finally, the prevalence of each mental problem differed depending on the measurement tools used. In conclusion, this study revealed the prevalence of mental problems during the COVID-19 pandemic by using a fairly large-scale sample and further clarified that the heterogeneous results for these mental health problems may be due to the nonstandardized use of psychometric tools.Action Contro
Microwave and terahertz dielectric properties of MgTiO3–CaTiO3 ceramics
The THz dielectric properties of MgTiO3–CaTiO3 ceramics are reported. The ceramics were prepared via a solid-state reaction route and the sintering conditions were optimized to obtain ceramics with high permittivity and low loss in the terahertz frequency domain. The amount of impurities (MgTi2O5) and grain size increased with increasing sintering temperature. The dielectric properties improved with increasing density, and the best terahertz dielectric performance was obtained at 1260 °C, with a permittivity of 17.73 and loss of 3.07×10−3. Ceramics sintered above 1260 °C showed a sharp increase in loss, which is ascribed to an increase in the impurity content
First principles studies of a Xe atom adsorbed on Nb(110) surface
We study adsorption sites of a single Xe adatom on Nb(110) surface using a
density functional theory approach: The on-top site is the most favorable
position for the adsorption. We compare the binding features of the present
study to earlier studies of a Xe adatom on close-packed (111) surface of
face-centered cubic metals. The different features are attributed through a
microscopic picture to the less than half filled d-states in Nb.Comment: 15 pages, 4 figure
