    Qualitative analysis of academic group and discussion forum on Facebook

    In the present study, data was triangulated and two methods of data analysis were used. Qualitative analysis was undertaken of free-text data from students’ reflective essaysto extract socially-related themes. Heuristic evaluation was conducted by expert evaluators, who investigated forum contributions and discourse in line with contemporary learning theory and considered the social\ud culture of participation. Findings of the qualitative analysis of students’ perceptions and results of the\ud heuristic evaluation of forum participation confirmed each other, indicating a warm social climate and a conducive, well-facilitated environment that supported individual styles of participation. It fostered interpersonal relationships between distance learners, as well as study-related benefits enhanced by peer teaching and insights acquired in a culture of social negotiation. The environment was effectively moderated, while supporting student-initiative.\u

    Directed and elliptic flow in heavy ion collisions from Ebeam=90E_{\rm beam}=90 MeV/nucleon to Ec.m.=200E_{\rm c.m.}=200 GeV/nucleon

    Recent data from the NA49 experiment on directed and elliptic flow for Pb+Pb reactions at CERN-SPS are compared to calculations with a hadron-string transport model, the Ultra-relativistic Quantum Molecular Dynamics (UrQMD) model. The rapidity and transverse momentum dependence of the directed and elliptic flow, i.e. v1v_1 and v2v_2, are investigated. The flow results are compared to data at three different centrality bins. Generally, a reasonable agreement between the data and the calculations is found. Furthermore, the energy excitation functions of v1v_1 and v2v_2 from Ebeam=90AE_{\rm beam}=90A MeV to Ecm=200AE_{\rm cm}=200A GeV are explored within the UrQMD framework and discussed in the context of the available data. It is found that, in the energy regime below Ebeam≤10AE_{\rm beam}\leq 10A GeV, the inclusion of nuclear potentials is necessary to describe the data. Above 40A40A GeV beam energy, the UrQMD model starts to underestimate the elliptic flow. Around the same energy the slope of the rapidity spectra of the proton directed flow develops negative values. This effect is known as the third flow component ("antiflow") and cannot be reproduced by the transport model. These differences can possibly be explained by assuming a phase transition from hadron gas to quark gluon plasma at about 40A40A GeV.Comment: 19 pages, minor changes and modified title as published in PR

    Visualisation of PCNA Monoubiquitination In Vivo by Single Pass Spectral Imaging FRET Microscopy

    Monoubiquitination of the DNA sliding clamp, PCNA, plays a central role in the control of damage bypass during replication. By combining a widely-spaced FRET donor/acceptor pair (CFP and mRFP) with spectral imaging, we have developed a simple method for the visualisation of PCNA monoubiquitination in both fixed and live cells with a single imaging pass. We validate the method with genetic controls in the avian cell line DT40 and use it to examine the intracellular dynamics of PCNA ubiquitination following subnuclear UV irradiation. This general approach is likely to be of utility for live imaging of post-translational modifications of a wide range of substrates in vivo

    The pathway to diagnosis of type 1 diabetes in children: a questionnaire study.

    OBJECTIVE: To explore the pathway to diagnosis of type 1 diabetes (T1D) in children. DESIGN: Questionnaire completed by parents. PARTICIPANTS: Parents of children aged 1 month to 16 years diagnosed with T1D within the previous 3 months. SETTING: Children and parents from 11 hospitals within the East of England. RESULTS: 88/164 (54%) invited families returned the questionnaire. Children had mean±SD age of 9.41±4.5 years. 35 (39.8%) presented with diabetic ketoacidosis at diagnosis. The most common symptoms were polydipsia (97.7%), polyuria (83.9%), tiredness (75.9%), nocturia (73.6%) and weight loss (64.4%) and all children presented with at least one of those symptoms. The time from symptom onset to diagnosis ranged from 2 to 315 days (median 25 days). Most of this was the appraisal interval from symptom onset until perceiving the need to seek medical advice. Access to healthcare was good but one in five children presenting to primary care were not diagnosed at first encounter, most commonly due to waiting for fasting blood tests or alternative diagnoses. Children diagnosed at first consultation had a shorter duration of symptoms (p=0.022) and children whose parents suspected the diagnosis were 1.3 times more likely (relative risk (RR) 1.3, 95% CI 1.02 to 1.67) to be diagnosed at first consultation. CONCLUSIONS: Children present with the known symptoms of T1D but there is considerable scope to improve the diagnostic pathway. Future interventions targeted at parents need to address the tendency of parents to find alternative explanations for symptoms and the perceived barriers to access, in addition to symptom awareness.The study was funded by the Royal College of General Practitioners Scientific Foundation Board (SFB-2011-15).     Avoidable Waste in Ophthalmic Epidemiology: A Review of Blindness Prevalence Surveys in Low and Middle Income Countries 2000–2014

    Purpose:- Sources of avoidable waste in ophthalmic epidemiology include duplication of effort, and survey reports remaining unpublished, gaining publication after a long delay, or being incomplete or of poor quality. The aim of this review was to assess these sources of avoidable waste by examining blindness prevalence surveys undertaken in low and middle income countries (LMICs) between 2000 and 2014. Methods:- On December 1 2016 we searched MEDLINE, EMBASE and Web of Science databases for cross-sectional blindness prevalence surveys undertaken in LMICs between 2000–2014. All surveys listed on the Rapid Assessment of Avoidable Blindness (RAAB) Repository website (“the Repository”) were also considered. For each survey we assessed i) availability of scientific publication, survey report, summary results tables and/or datasets; ii) time to publication from year of survey completion and journal attributes; iii) extent of blindness information reported; and iv) rigour when information was available from two sources (i.e. whether it matched). Results:- Of the 279 included surveys (from 68 countries) 186 (67%) used RAAB methodology; 146 (52%) were published in a scientific journal, 57 (20%) were published in a journal and on the Repository, and 76 (27%) were on the Repository only (8% had tables; 19% had no information available beyond registration). Datasets were available for 50 RAABs (18% of included surveys). Time to publication ranged from <1–11 years (mean, standard deviation 2.8±1.8 years). The extent of blindness information reported within studies varied (e.g. presenting and best-corrected, unilateral and bilateral); those with both a published report and Repository tables were most complete. For surveys published and with RAAB tables available, discrepancies were found in reporting of participant numbers (14% of studies) and blindness prevalence (15%). Conclusion:- Strategies are needed to improve the availability, consistency and quality of information reported from blindness prevalence surveys, and hence reduce avoidable waste

    Heterogenised catalysts for the H-transfer reduction reaction of aldehydes: influence of solvent and solvation effects on reaction performances

    Heterogenisation of homogeneous catalysts onto solid supports represents a potential strategy to make the homogeneous catalytic function recyclable and reuseable. Yet, it is usually the case that immobilised catalysts have much lower catalytic activity than their homogeneous counterpart. In addition, the presence of a solid interface introduces a higher degree of complexity by modulating solid/fluid interactions, which can often influence adsorption properties of solvents and reactive species and, ultimately, catalytic activity. In this work, the influence of support and solvent in the H-transfer reduction of propionaldehyde over Al((OPr)-Pr-i)(3)-SiO2, Al((OPr)-Pr-i)(3)-TiO2 and Al((OPr)-Pr-i)(3)-Al2O3 heterogenised catalysts has been studied. Reaction studies are coupled with both NMR relaxation measurements as well as molecular dynamics (MD) simulations in order to unravel surface and solvation effects during the reaction. The results show that, whilst the choice of the support does not influence significantly catalytic activity, reactions carried out in solvents with high affinity for the catalyst surface, or able to hinder access to active sites due to solvation effects, have a lower activity. MD calculations provide key insights into bulk solvation effects involved in such reactions, which are thought to play an important role in determining the catalytic behaviour. The activity of the heterogenised catalysts was found to be comparable with that of the homogeneous Al((OPr)-Pr-i)(3) catalysts for all supports used, showing that for the type of reaction studied immobilisation of the homogeneous catalyst onto solid supports is a viable, robust and effective strategy

    Tightly regulated Ca(2+) homeostasis is a prerequisite for proper cardiac function. To dissect the regulatory network of cardiac Ca(2+) handling, we performed a chemical suppressor screen on zebrafish tremblor embryos, which suffer from Ca(2+) extrusion defects. Efsevin was identified based on its potent activity to restore coordinated contractions in tremblor. We show that efsevin binds to VDAC2, potentiates mitochondrial Ca(2+) uptake and accelerates the transfer of Ca(2+) from intracellular stores into mitochondria. In cardiomyocytes, efsevin restricts the temporal and spatial boundaries of Ca(2+) sparks and thereby inhibits Ca(2+) overload-induced erratic Ca(2+) waves and irregular contractions. We further show that overexpression of VDAC2 recapitulates the suppressive effect of efsevin on tremblor embryos whereas VDAC2 deficiency attenuates efsevin\u27s rescue effect and that VDAC2 functions synergistically with MCU to suppress cardiac fibrillation in tremblor. Together, these findings demonstrate a critical modulatory role for VDAC2-dependent mitochondrial Ca(2+) uptake in the regulation of cardiac rhythmicity

    c-di-GMP modulates type IV MSHA pilus retraction and surface attachment in Vibrio cholerae.

    Biofilm formation by Vibrio cholerae facilitates environmental persistence, and hyperinfectivity within the host. Biofilm formation is regulated by 3',5'-cyclic diguanylate (c-di-GMP) and requires production of the type IV mannose-sensitive hemagglutinin (MSHA) pilus. Here, we show that the MSHA pilus is a dynamic extendable and retractable system, and its activity is directly controlled by c-di-GMP. The interaction between c-di-GMP and the ATPase MshE promotes pilus extension, whereas low levels of c-di-GMP correlate with enhanced retraction. Loss of retraction facilitated by the ATPase PilT increases near-surface roaming motility, and impairs initial surface attachment. However, prolonged retraction upon surface attachment results in reduced MSHA-mediated surface anchoring and increased levels of detachment. Our results indicate that c-di-GMP directly controls MshE activity, thus regulating MSHA pilus extension and retraction dynamics, and modulating V. cholerae surface attachment and colonization
