    This study examined the beliefs about English teaching and learning of six Chinese secondary school EFL teachers, whether any changes occurred in their beliefs during the first three years in the profession, and why beginning teachers changed or did not change their beliefs. Data were collected over a period of 10 months using semi-structured interviews, concept mapping, and journal entries. The findings showed how teachers’ learning in the workplace influenced change or no change in beliefs, how their schools supported learning, how teachers elected to engage in activities and how these affected the nature of their beliefs

    Identity-based edge computing anonymous authentication protocol

    With the development of sensor technology and wireless communication technology, edge computing has a wider range of applications. The privacy protection of edge computing is of great significance. In the edge computing system, in order to ensure the credibility of the source of terminal data, mobile edge computing (MEC) needs to verify the signature of the terminal node on the data. During the signature process, the computing power of edge devices such as wireless terminals can easily become the bottleneck of system performance. Therefore, it is very necessary to improve efficiency through computational offloading. Therefore, this paper proposes an identity-based edge computing anonymous authentication protocol. The protocol realizes mutual authentication and obtains a shared key by encrypting the mutual information. The encryption algorithm is implemented through a thresholded identity-based proxy ring signature. When a large number of terminals offload computing, MEC can set the priority of offloading tasks according to the user’s identity and permissions, thereby improving offloading efficiency. Security analysis shows that the scheme can guarantee the anonymity and unforgeability of signatures. The probability of a malicious node forging a signature is equivalent to cracking the discrete logarithm puzzle. According to the efficiency analysis, in the case of MEC offloading, the computational complexity is significantly reduced, the computing power of edge devices is liberated, and the signature efficiency is improved

    Integration of scRNA-Seq and bulk RNA-Seq uncover perturbed immune cell types and pathways of Kawasaki disease

    IntroductionKawasaki disease (KD) is an acute febrile illness primarily affecting children and characterized by systemic inflammation and vasculitis that can lead to coronary artery complications. The aim of this study was to gain a comprehensive understanding of immune dysregulation in KD.MethodsTo this end, we employed integration of single-cell RNA sequencing (scRNA-Seq) and bulk RNA sequencing (bulk RNA-Seq) data. Furthermore, we conducted flow cytometry analysis for a cohort of 82 KD patients.ResultsOur analysis revealed significant heterogeneity within immune cell populations in KD patients, with distinct clusters of T cells, B cells, and natural killer (NK) cells. Importantly, CD4+ naĂŻve T cells in KD patients were found to predominantly differentiate into Treg cells and Th2 cells, potentially playing a role in the excessive inflammation and vascular damage characteristic of the disease. Dysregulated signaling pathways were also identified, including the mTOR signaling pathway, cardiomyopathy pathway, COVID-19 signaling pathway, and pathways involved in bacterial or viral infection.DiscussionThese findings provide insights into the immunopathogenesis of KD, emphasizing the importance of immune cell dysregulation and dysregulated signaling pathways. Integration of scRNA-Seq and bulk RNA-Seq data offers a comprehensive view of the molecular and cellular alterations in KD and highlights potential therapeutic targets for further investigation. Validation and functional studies are warranted to elucidate the roles of the identified immune cell types and pathways in KD pathogenesis and to develop targeted interventions to improve patient outcomes

    Testing the Hypothesis of Multiple Origins of Holoparasitism in Orobanchaceae: Phylogenetic Evidence from the Last Two Unplaced Holoparasitic Genera, Gleadovia and Phacellanthus

    Orobanchaceae is the largest family among the parasitic angiosperms. It comprises non-parasites, hemi- and holoparasites, making this family an ideal test case for studying the evolution of parasitism. Previous phylogenetic analyses showed that holoparasitism had arisen at least three times from the hemiparasitic taxa in Orobanchaceae. Until now, however, not all known genera of Orobanchaceae were investigated in detail. Among them, the unknown phylogenetic positions of the holoparasites Gleadovia and Phacellanthus are the key to testing how many times holoparasitism evolved. Here, we provide clear evidence for the first time that they are members of the tribe Orobancheae, using sequence data from multiple loci (nuclear genes ITS, PHYA, PHYB, and plastid genes rps2, matK). Gleadovia is an independent lineage whereas Phacellanthus should be merged into genus Orobanche section Orobanche. Our results unambiguously support the hypothesis that there are only three origins of holoparasitism in Orobanchaceae. Divergence dating reveals for the first time that the three origins of holoparasitism were not synchronous. Our findings suggest that holoparasitism can persist in specific clades for a long time and holoparasitism may evolve independently as an adaptation to certain hosts

    Circulating tumor DNA clearance predicts prognosis across treatment regimen in a large real-world longitudinally monitored advanced non-small cell lung cancer cohort

    Background: Although growth advantage of certain clones would ultimately translate into a clinically visible disease progression, radiological imaging does not reflect clonal evolution at molecular level. Circulating tumor DNA (ctDNA), validated as a tool for mutation detection in lung cancer, could reflect dynamic molecular changes. We evaluated the utility of ctDNA as a predictive and a prognostic marker in disease monitoring of advanced non-small cell lung cancer (NSCLC) patients.Methods: This is a multicenter prospective cohort study. We performed capture-based ultra-deep sequencing on longitudinal plasma samples utilizing a panel consisting of 168 NSCLC-related genes on 949 advanced NSCLC patients with driver mutations to monitor treatment responses and disease progression. The correlations between ctDNA and progression-free survival (PFS)/overall survival (OS) were performed on 248 patients undergoing various treatments with the minimum of 2 ctDNA tests.Results: The results of this study revealed that higher ctDNA abundance (P=0.012) and mutation count (P=8.5x10(-4)) at baseline are associated with shorter OS. We also found that patients with ctDNA clearance, not just driver mutation clearance, at any point during the course of treatment were associated with longer PFS (P=2.2x10(-1)6, HR 0.28) and OS (P=4.5x10(-6), HR 0.19) regardless of type of treatment and evaluation schedule.Conclusions: This prospective real-world study shows that ctDNA clearance during treatment may serve as predictive and prognostic marker across a wide spectrum of treatment regimens

    GRIK3 rs490647 is a Common Genetic Variant between Personality and Subjective Well-being in Chinese Han Population

    Personality and subjective well-being (SWB) have been suggested to be strongly related in previous studies. This study was intended to confirm the relationship between personality and SWB and tried to seek out the genetic variants which underlie both personality and SWB. The subjects were 890 participants from Chinese Han population. We evaluated their personality using the Big Five Inventory (BFI) and used the Satisfaction With Life Scale (SWLS) to reflect their SWB. Five single nucleotide polymorphisms (SNPs) were selected from the literature (rs1426371, rs2164273, rs322931, rs3756290, rs490647) and genotyped for genetic association study. We found negative correlations between neuroticism and SWB. On the contrary, extraversion and agreeableness were positively associated with SWB. Three SNPs (rs2164273, rs3756290, rs490647) out of the five were found to connect with personality (extraversion, neuroticism, conscientiousness and openness to experience) and rs490647 variants of GRIK3 was also associated with SWB. Individuals carrying G allele at this site were predisposed to have lower risk to be neuroticism and greater chance to be extraverted, open and satisfied with their life. In summary, our study revealed that rs490647 might be a good candidate genetic variant for personality and SWB in Chinese Han population

    Understanding the causality between energy consumption, carbon emissions and economic growth is helpful for policymakers to formulate energy, environmental and economic policies. For the first time, based on nonlinear dynamics, this paper employs multispatial convergent cross mapping (CCM) to revisit the energy-carbon-economy causation for China, India and the G7 countries using both aggregate data and per capita data. The findings indicate that there are significant differences between developing countries and developed countries. A bidirectional nexus between energy consumption, carbon emissions and economic growth is found in China and India, but various causal relationships are identified in the G7 countries, including bidirectional, unidirectional and neutral nexus. The results confirm that the decoupling phenomenon is common in most G7 countries. By leveraging a variety of samples and a new approach, this study provides new evidence for policy authorities to formulate country-specific policies to obtain better environmental quality while achieving sustainable economic growth
