Proteomic analysis of Parietaria judaica pollen and allergen profiling by an immunoproteomic approach
Parietaria judaica pollen is a common
cause of airway allergic disease in the Mediterranean
area. Proteome analysis of mature Parietaria judaica
pollen by two-dimensional gel electrophoresis (2-DE)
and mass spectrometry has established the first
reference proteome map of this weed. Proteins
involved in a variety of cellular functions as well as
the occurrence of allergens were detected. By using
2-DE and immunoblotting with sera from Parietaria
judaica allergic patients we obtained a more detailed
characterization of Parietaria judaica allergen profil
Gold catalysts supported on ceria doped by rare earth metals for water gas shift reaction: Influence of the preparation method
Cataloged from PDF version of article.Gold catalysts based on ceria, doped by various RE metals (La, Sm, Gd, Yb, Y) were studied. The influence of the preparation methods on structure, properties and catalytic activity in the WGS reaction was investigated. The catalysts' supports were prepared using two different methods: co-precipitation (CP) and mechanochemical activation (MA). The catalysts were tested in a wide temperature interval without and after reactivation. All samples were characterized using a combination of X-ray diffraction (XRD), high resolution transmission electron microscopy (HRTEM), Raman spectroscopy (RS) and X-ray photoelectron spectroscopy (XPS) and TPR. It was found that the catalytic activity of MA catalysts is higher than CP ones. The gold catalysts based on ceria doped by Yb and Sm exhibited the highest activity. After reactivation in air the MA samples almost kept the WGS activity same, while the CP catalysts increased it. The catalysts of a single- and double-phase structure are formed as a result of CP and MA preparation, respectively. There are no big differences in the gold particles size (2-3 nm) depending on dopants and on the preparation techniques. The RS spectra analysis indicates that most probably the oxygen vacancies are adjacent to Me(3+) dopant and the ceria structure seems to be better ordered than in the case of alumina as a dopant. There is no distinct correlation between reducibility and WGS activity. The XPS analysis disclose positively charged gold particles in addition to metallic gold within a surface region of fresh samples and only metallic gold on the samples after catalytic processing. Virulence factors in vancomycin-resistant and vancomycin- susceptible Enterococcus faecalis from Brazil
Enterococci are members of commensal flora of animals and insects, but are also important opportunistic pathogens. Our objective was to observe if there was any difference of virulence in several groups of E. faecalis, mainly between vancomycin-resistant E. faecalis (VREFS) of colonization and infection. VREFS and vancomycin-sensitive E. faecalis from Brazil were screened for the presence of virulence factor genes. Phenotypic assays were used to assess in vitro expression, to understand the pathogenic potential of these isolates and to determine whether a correlation exists between virulence and antibiotic resistance. Different virulence profiles were found suggesting that the disseminating clone may have generated several variations. However, our study showed that one constellation of traits appeared most commonly: gelatinase, aggregation substance and esp (GEA). These factors are important because they have been implicated in cell aggregation and biofilm formation. Biofilm formation may promote the conjugation of plasmids harboring resistance and virulence genes, enhancing the probability of entry of new resistance genes into species. Curiously, the profile GEA was not exclusive to VREFS, it was the second most observed in VSEFS isolates from colonization and infection in hospitalized patients and also from rectal swabs of healthy volunteers. Such strains appear to represent the entry gateway to new resistance genes into E. faecalis and may contribute to the spreading of E. faecalis mainly in hospitals

Porcine respiratory disease complex in southern Brazil: pathogens and associated histopathology.
Detection of high-velocity material from the wind-wind collision zone of Eta Carinae across the 2009.0 periastron passage
We report near-IR spectroscopic observations of the Eta Carinae massive
binary system during 2008-2009 using VLT/CRIRES. We detect a strong, broad
absorption wing in He I 10833 extending up to -1900 km/s across the 2009.0
spectroscopic event. Archival HST/STIS ultraviolet and optical data shows a
similar high-velocity absorption (up to -2100 km/s) in the UV resonance lines
of Si IV 1394, 1403 across the 2003.5 event. UV lines from low-ionization
species, such as Si II 1527, 1533 and C II 1334, 1335, show absorption up to
-1200 km/s, indicating that the absorption with v from -1200 to -2100 km/s
originates in a region markedly faster and more ionized than the nominal wind
of the primary star. Observations obtained at the OPD/LNA during the last 4
spectroscopic cycles (1989-2009) also display high-velocity absorption in He I
10833 during periastron. Based on the OPD/LNA dataset, we determine that
material with v < -900 km/s is present in the phase range 0.976 < phi < 1.023
of the spectroscopic cycle, but absent in spectra taken at phi < 0.947 and phi
> 1.049. Therefore, we constrain the duration of the high-velocity absorption
to be 95 to 206 days (or 0.047 to 0.102 in phase). We suggest that the
high-velocity absorption originates from shocked gas in the wind-wind collision
zone, at distances of 15 to 45 AU in the line-of-sight to the primary star.
Using 3-D hydrodynamical simulations of the wind-wind collision zone, we find
that the dense high-velocity gas is in the line-of-sight to the primary star
only if the binary system is oriented in the sky so that the companion is
behind the primary star during periastron, corresponding to a longitude of
periastron of omega ~ 240 to 270 degrees. We study a possible tilt of the
orbital plane relative to the Homunculus equatorial plane and conclude that our
data are broadly consistent with orbital inclinations in the range i=40 to 60

First-principles modeling of the polycyclic aromatic hydrocarbons reduction
Density functional theory modelling of the reduction of realistic
nanographene molecules (C42H18, C48H18 and C60H24) by molecular hydrogen
evidences for the presence of limits in the hydrogenation process. These limits
caused the contentions between three-fold symmetry of polycyclic aromatic
hydrocarbon molecules and two-fold symmetry of adsorbed hydrogen pairs.
Increase of the binding energy between nanographenes during reduction is also
discussed as possible cause of the experimentally observed limited

Identification of potential prognostic biomarkers for node-negative breast tumours by proteomic analysis: a multicentric 2004 national PHRC study
We used a 2D-electrophoresis (2-DE) proteomic approach to identify novel biomarkers in node-negative breast cancers. This retrospective study focused on a population of patients with ductal pN0M0 tumours. A subset of patients who developed metastases and in whose tumours were found high levels of uPA and PAI-1 (metastatic relapse, MR: n=20) were compared to another subset in whom no metastatic relapse occurred and whose tumours were found to have low levels of uPA and PAI-1 (no relapse, NR: n=21). We used a 2-DE coupled with MS approach to screen cytosol fractions using two pH-gradient scales, a broad scale (3.0-11.0) and a narrower scale focussing in on a protein rich region (5.0-8.0). This study was conducted on 41 cytosol specimens analyzed in duplicate on two platforms. The differential analysis of more than 2,000 spots in 2-DE gels, obtained on the two platforms, allowed the identification of 13 proteins which were confirmed by western blotting. Two proteins, GPDA and FABP4 were down-regulated in the MR subset whereas all the others were up-regulated. An in silico analysis revealed that GMPS (GUAA), GAPDH (G3P), CFL1 (COF1) and FTL (FRIL), the most informative genes, displayed a proliferation profile (high expression in basal-like, HER2+ and luminal B molecular subtypes). Inversely, similar to FABP4, GPD1 [GPDA] displayed a high expression in luminal A subtype, a profile characteristic of tumour suppressor genes. Despite the small size of our cohort, the 2-DE analysis gave interesting results which were confirmed by the in silico analysis showing that some of the corresponding genes had a strong prognostic impact in breast cancer, mostly because of their link with proliferation: GMPS, GAPDH, FTL and GPD1. A validation phase on a larger cohort is now needed before these biomarkers could be considered for use in clinical practice
